The $NAD^+$-specific activity of a dual coenzyme-specific isocitrate dehydrogenase (IDH; EC 1.1.1.41) from the primitive fungus Pythium ultimum was investigated to elucidate the regulatory factors that may influence the intracellular distribution of carbon and the availability of intermediates, e.g. citrate, for fatty acid synthesis. Inhibition of $NAD^+$-IDH activity by diphospho- and triphosphonucleotides (ATP, ADP, and GTP) reflected the sensitivity of this enzyme to cellular energy charge even though monophosphonucleotides (AMP and GMP) had little effect on activity. NADPH, but not NADH, substantially inhibited $NAD^+$-IDH activity, showing noncompetitive inhibition with isocitrate. Oxalacetate and ${\alpha}$-ketoglutarate showed competitive inhibition with isocitrate, while citrate and cis-aconitate showed mixed-noncompetitive inhibition with isocitrate. Inhibition by these substances ranged from 29 to 46% at 10 mM. The inhibitory effect of oxalacetate was increased synergistically by glyoxylate, which alone caused 31% uncompetitive inhibition at 10 mM, and a mixture of the two substances at 1 mM each showed 98% inhibition of $NAD^+$-IDH activity. The regulation of $NAD^+$-IDH in Pythium ultimum seems to be a complex process involving mitochondrial metabolites. The addition of glyoxylate (3 mM) and oxalacetate (3 mM) to the culture medium resulted in the production of 49% more lipid by P. ultimum.

Patients with hemorrhagic fever with renal syndrome (HFRS) often exhibit altered serum lipid and lipoprotein profile during the oliguric phase of the disease. Serum lipid and lipoprotein profiles were assessed during the oliguric and recovery phases in six male patients with HFRS. In the oliguric phase of HFRS, the apolipoprotein (apo) C-III content in high-density lipoproteins (HDL) was elevated, whereas the apoA-I content was lowered. The level of expression and activity of antioxidant enzymes were severely reduced during the oliguric phase, while the cholesteryl ester transfer protein activity and protein level were unchanged between the phases. In the oliguric phase, electromobility of $HDL_2$ and $HDL_3$ was faster than in the recovery phase. Low-density lipoprotein (LDL) particle size was smaller and the distribution was less homogeneous. Patients with HFRS in the oliguric phase had severely modified lipoproteins in composition and metabolism.
Salix koriyanagi is a deciduous shrub and native to Korea. The spatial distribution of multilocus allelic frequencies and geographical distances of the natural population in upper swampy area at the Namgang River in Korea were studied. The species showed a significant positive and negative spatial autocorrelation according to geographical distances as measured by Moran's I. Genetic similarity of individuals was found among subpopulations at up to a scale of a 12 m distance, and this was partly due to a combination of allelic frequencies, and therefore, a significant spatial autocorrelation was composed of a scale of 12 m intervals. Within S. koriyanagi in swampy area at the Namgang River, a strong spatial structure was observed for allozyme markers, indicating a migration within subpopulations.
The structure of cerebrosides from soybean embryo was determined using fast atom bombardment mass spectrometer (FAB-MS), gas chromatography mass spectrometer (GC-MS) and TLC. The components of cerebroside were determined by GC-MS after acid hydrolysis. The molecular weight distribution of cerebroside was measured by positive mode FAB-MS with LiOH saturated 3-nitrobenzylalcohol(3-NBA) matrix. Structures of individual components of complex mixtures can be determined easily by this process. The major constituent of soybean extracted cerebroside was determined as the glucoside of N-2'-hydroxypalmitoyl-sphingadienine.
Gonadogenesis is a complicated process which involves multi-gene interactions. A rainbow trout (Oncorhynchus mykiss) gene spermatogenesis associated 4 (SPATA4) was cloned and characterized from adult rainbow trout testis. The cDNA sequence of rainbow trout SPATA4 contains an open reading frame of 1, 081 nucleatides encoding a putative protein of 259 amino acids. The putative protein from rainbow trout shares a 76.8% homology with zebrafish SPATA4. No trans-membrane regions or signal peptide were detected using bioinformatics methods. Subcellular localization analysis revealed that rainbow trout SPATA4 was a nuclear protein with highest possibility (39.1%). Multi-tissue reverse transcriptase PCR (RT-PCR) was performed to examine the distribution of rainbow trout SPATA4 in eleven organs of adult rainbow trout. The result demonstrated that this gene express specifically in testis and slight amount of expression was detected in ovary. Further analysis of SPATA4 characterization and function in rainbow trout may provide insight into the understanding of gonadogenesis process.
Hizikia fusiformis hydroysates by five carbohydrases (Viscozyme, Celluclast, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) were investigated for their extraction efficacy (yield and total total polyphenolic content) and antioxidative activity (DPPH radical and hydrogen peroxide scavenging activity). Termamyl and Ultraflo of the carbohydrases and Flavourzyme and Alcalase of proteases were selected by their high eficacy of extraction and antioxidative activity. Selected enzymes were used to investigate the optimum enzymatic reaction time and dosage (enzyme/substrate ratio) suitable for hydorolysis. Optimum reaction time for the enzymatic hydrolysis was 3 days and optimum dosage of hydrolysis was observed as 5%. Simultaneously, Ultraflo of the two carbohydrases and Alcalse of the two proteases were selected as the most effective enzymes. Combination of Ultraflo and Alcalase under optimum hydrolysis conditions could intensify the extraction efficacy of antioxidative materials form H. fusiformis. The hydrolysate obtained by combining the enzymes was separated into four different molecular weight fractions (<1kD, 1-10 kD, 10-30 kD and >30 kD) and recorded the polyphenolic content distribution and respective antioxidative ability. The fraction <1kD was identified as less effective and those fractions > 1kD indicated comparatively higher antioxidative activities related to their polyphenolic content.
The behavior of tyrosine(Tyr.) residue of Bombyx mori silk fiber from yellow fluorescent cocoon has been examined for the dependence of pH in aqueous silk solution under the presence of orange II salt. Through the peak separation of angular dependence of spectral pattern of 15N-Tyr. and [1-13C]-Tyr. between the fiber axis and the molecular bond direction, N-H bond in fiber as well as the orientation distribution around the fiber axis were analyzed. Also, and sericin component was obtained from these angular dependence of oriented spectral pattern. The pH dependence of the 13C NMR chemical shift of B. mori silk fibroin was examined in aqueous solution in the presince of orange II are broad at pH$\geq$7.0. However, these become sharper at pH$\geq$8.0 and remain sharp at higher pH. In these higher pH range, a chemical shift change occurs due to the deprotonation of the Tyr. side group of fibroin. At higher pH. such a hydrophobic cluster is destroyed because of the electrostatic interaction according to the deprotonation of the Tyr-OH group.
Phylogenetic analysis of 31 species representing 12 genera in the family Strigidae (Aves: Strigiformes) including 5 species (Bubo bubo, Otus sunia, O. semitorques, Ninox scutulato, Strix aluco) collected from Korea has been undertaken using nucleotide sequences of the mitochondrial cytochrome b gene. Maximum likelihood analysis was performed and pairwise genetic distances were calculated with Kimura's two-parameter and p-distance. Among well-aligned 959 bp used for this study, 459 sites were variable and 398 sites were informative for the phylogenetic analysis. The family Strigidae was divided into three subgroups, Clade I (Aegolius), Clade II (Athene, Micrathene, Glaucidium and Surnia) and Clade III (Bubo, Nycteo, Pulsatrix, Strix, Otus, Ptilopsis, and Ninox). Also, two separated subgroups in the genus Otus were confirmed by the geographical distribution.

Abiotic-stressed tobacco stems as a non-wooden biomass were analyzed for their chemical characteristics. Light-stressed tobacco stems (LST) have a relatively high nitrogen concentration, much more extractive content, and a similar amount of lignin and higher contents of acid sugars than those of Non stressed tobacco (NST). It also has low cellulose crystallinity and a high degree of condensation. Guaiacyl units having a lower molecular weight distribution consist of rich lignin. Potato scab, an important disease that affects developing tubers, causes a major problem in potato cultivation. The major potato cultivation areas in Korea are located in two Northern provinces, Gangwon and Gyeonggi, and two Southern provinces, Jeju island, and South Jeolla. In these areas, potato scab is widely distributed and has caused severe problem in potato cultivation. Therefore, potato-growing areas were surveyed for identification and distribution of potato scab pathogens from 1996 to 1999. Pathogenic Streptomyces strains were isolated from potato scab lesions and six representative Streptomyces species were characterized based on their phenotypic and molecular characteristics including, pathogenicity, physiological and morphological properties, analyses of 16SrRNA genes and 16S-23S ITS region, DNA relatedness, production of thaxtomin A, and the presence of nec1 and ORFtnp gene homologs. Three species were identified as previously described Streptomyces scabies, S. turgidiscabies, and S. acidiscabies, while other three species having distinct phenotypics properties were identified as novel S. luridiscabiei, S. puniciscabiei, and S. niveiscabiei.
