Enhanced Production of Human Serum Albumin by Fed-Batch Culture of Hansenula polymorpha with High-Purity Oxygen

  • Fed-batch cultures of Hansenula polymorpha were studied to develop an efficient biosystem to produce recombinant human serum albumin (HSA). To comply with this purpose, we used a high-purity oxygen-supplying strategy to increase the viable cell density in a bioreactor and enhance the production of target protein. A mutant strain, H. polymorpha GOT7, was utilized in this study as a host strain in both 5-l and 30-l scale fermentors. To supply high-purity oxygen into a bioreactor, nearly 100% high-purity oxygen from a commercial bomb or higher than 93% oxygen available in situ from a pressure swing adsorption (PSA) oxygen generator was employed. Under the optimal fermentation of H. polymorpha with highpurity oxygen, the final cell densities and produced HSA concentrations were 24.6 g/l and 5.1 g/l in the 5-l fermentor, and 24.8 g/l and 4.5 g/l in the 30-l fermentor, respectively. These were about 2-10 times higher than those obtained in air-based fed-batch fermentations. The discrepancies between the 5-l and 30-l fermentors with air supply were presumably due to the higher contribution of surface aeration over submerged aeration in the 5-l fermentor. This study, therefore, proved the positive effect of high-purity oxygen in enhancing viable cell density as well as target recombinant protein production in microbial fermentations.

  • 기체크로마토그래프에 직접 연결한 펄스 반응장치를 이용하여 10wt% $Ag/{\alpha}-Al_2O_3$ 촉매상에서 에틸렌 부분산화반응을 수행하였다. 산소가 분자흡착상태로 존재하기 어려운 실험온도에서 산소를 주입한 후 에틸렌을 주입하였을 때 산화에틸렌이 발생하였으며, 이 결과로부터 원자상태의 흡착산소가 산화에틸렌 생성에 관여함을 알 수 있었다. 흡착산소량과 몸체산소량을 감소시키면 산화에틸렌의 선택도는 감소하였다. 생성된 산화에틸렌은 에틸렌과 흡착산소로 분해되거나 아세트알데히드로 이성질화되었으나, 아세트알데히드로의 이성질화반응은 먼저 흡착된 산소에 의해서 크게 억제되었다.

Sustainable Development of Palm Oil: Synthesis and Electrochemical Performance of Corrosion Inhibitors

  • Palm oil production is among the highest worldwide, and it has been mainly used in the food industry and other commodities. Currently, a lot of palm oil production has been destined for the synthesis of biodiesel; however, its use in applications other than the food industry has been questioned. Thereby for a sustainable development, in this paper the use of palm oil of low quality for corrosion inhibitors synthesis is proposed. The performance of the synthesized inhibitors was evaluated by using electrochemical techniques such as open circuit potential measurements, linear polarization resistance and electrochemical impedance spectroscopy. The results indicate that the fatty amides from palm oil are excellent corrosion inhibitors with protection efficiencies greater than 98%. Fatty amides molecules act as cathodic inhibitors decreasing the anodic dissolution of iron. When fatty amides are added, a rapid decrease in the corrosion rate occurs due to the rapid formation of a molecular film onto carbon steel surface. During the adsorption process of the inhibitor a self-organization of the hydrocarbon chains takes place forming a tightly packed hydrophobic film. These results demonstrate that the use of palm oil for the production of green inhibitors promises to be an excellent alternative for a sustainable use of the palm oil production.

  • 수평균 중합도 900 및 교대배열기 함량 63.1%의 교대배열 poly(vinyl alcohol) (s-PVA)을 사용하여 s-PVA/요오드계 편광필름을 제조하였다. 흔성배열 PVA/요오드계 필름과 비교할 때 편광도는 자연신비에서도 99% 이상의 값을 가짐으로써 향상된 결과를 보였으나 투과도는 약간 저하되는 경향을 보였다. 따라서 침지액의 요오드 농도를 보다 낮추고 4배의 연신비를 유지함으로써 우수한 투과도와 편광도를 가지는 필름을 제조할 수 있었다. 한편, s-PVA/요오드 필름의 열수하에서의 탈착은 현저히 억제되었다. 결정화도, (100)면의 결정크기 및 면간거리가 조금씩 감소하거나 더 이상 증가하지 않는 것으로부터 침지시간 초기에는 요오드가 주로 결정영역에서 복합체를 형성하고, 이후에는 주로 결정면 밖에서 복합체를 형성하거나 단순한 물리적인 흡착을 이루고 있음을 확인하였다.

색소 흡착 산화아연 감광체의 전자사진 특성에 관한 연구 (The Application for Electrophotographic Photoreceptors of Zinc Oxide Adsorbed Copper Phthalocyanine and Sunfast Yellow)

  • 가시부 영역에서 산화아연을 광증감시키기 위해 copper phthalocyanine(CuPc)과 sunfast yellow(SY)를 산화아연 분말에 이층 흡착시켰다. 산화아연에 대한 CuPc의 흡착상태를 알기 위하여 ZnO/CuPc의 광음향, IR 및 라만 스펙트럼을 측정한 결과, CuPc는 $\alpha$형 및 $\beta$형의 결정 특성을 유지한 이합체 또는 분자들의 집합체 상태로 산화아연에 흡착된다는 것을 알았다. 산화아연에 CuPc 및 SY를 순차적으로 이층 흡착시킨계(ZnO/CuPc/SY)는 SY를 먼저 흡착시킨 ZnO/SY/CuPc계보다 광기전력이 높게 나타났고, $ZnO/\beta-CuPc/SY$$ZnO/\alpha-CuPc/SY$보다 광기전력이 높게 나타났다. $ZnO/\beta-CuPc/SY$의 전자사진 감도를 측정하였더니 630 nm에서 $$S_{1/2}=2.99{\times}10^{-2}(erg/cm^2)^{-1}$ 이었다.

Characterization of an Extracellular Lipase in Burkholderia sp. HY-10 Isolated from a Longicorn Beetle

  • Burkholderia sp. HY-10 isolated from the digestive tracts of the longicorn beetle, Prionus insularis, produced an extracellular lipase with a molecular weight of 33.5 kDa estimated by SDS-PAGE. The lipase was purified from the culture supernatant to near electrophoretic homogenity by a one-step adsorption-desorption procedure using a polypropylene matrix followed by a concentration step. The purified lipase exhibited highest activities at pH 8.5 and $60^{\circ}C$. A broad range of lipase substrates, from $C_4\;to\;C_{18}$ p-nitrophenyl esters, were hydrolyzed efficiently by the lipase. The most efficient substrate was p-nitrophenyl caproate ($C_6$). A 2485 bp DNA fragment was isolated by PCR amplification and chromosomal walking which encoded two polypeptides of 364 and 346 amino acids, identified as a lipase and a lipase foldase, respectively. The N-terminal amino acid sequence of the purified lipase and nucleotide sequence analysis predicted that the precursor lipase was proteolytically modified through the secretion step and produced a catalytically active 33.5 kDa protein. The deduced amino acid sequence for the lipase shared extensive similarity with those of the lipase family 1.2 of lipases from other bacteria. The deduced amino acid sequence contained two Cystein residues forming a disulfide bond in the molecule and three, well-conserved amino acid residues, $Ser^{131},\;His^{330},\;and\;Asp^{308}$, which composed the catalytic triad of the enzyme.

  • Fish protein hydrolysates (FPHs) with different degrees of hydrolysis by treatment with alcalase, pronase, flavourzyme and trypsin and isolated peptide were prepared from Hwangtae (yellow dried pollack, Theragra chalcogramma). Hwangtae protein hydrolysate was fractionated according to the molecular weight into six major types of APO1 (1.3 kDa), APO2 (1 kDa), APO3 (<1 kDa), APACE (<1 kDa), APG1 (70 kDa) and APG2 (70 kDa) isolated from the hydrolysate using consecutive chromatographic methods. Soluble peptide were produced from Hwangtae and evaluated for their nutritional and functional properties. Some functional properties of FPHs were assessed and compared with those of egg albumin or the soybean protein. APO2 had the highest nitrogen solubility value (94.2%), emulsion capacity and emulsion stability of the Alaska Pollack peptide ranged from 12.4 to 39.5 (mL of oil per 200 mg of protein) and 44.0% to 77.5%, respectively. Highest and lowest fat adsorption values were observed for APG1 (9.9 mL of oil per gram of protein) and APO3 (3.8 mL of oil per gram of protein), respectively.

탈알루미늄화가 SAPO-11 분자체의 구조 및 산성도에 미치는 영향 (The Effect of Dealumination on the Structure and Acidity of SAPO-11 Molecular Sieve)

  • 수열법으로 합성한 SAPO-11 분자체를 $H_4$EDTA로써 골격내의 알루미늄을 부분적으로 탈알루미늄화시킬때 24시간 탈알루미늄화까지는 골격이 안전하였으나 48시간 탈알루미늄화에 의해서는 구조가 붕괴되어 variscite($AIPO_4{\cdot}2H_2O$)와 tridymite($SiO_2$)로 변하였다. SAPO-11의 탈알루미늄화로 인하여 3607$cm^{-1}$과 3453$cm^{-1}$에서 하이드록실기에 의한 IR 흡수띠가 각각 관측되었다. 이들 하이드록실기 피크들의 세기는 탈알루미늄화의 정도에 비례하여 증가하였으며 메틸아민의 흡착에 의하여 피크가 사라지는 것으로 보아 이들은 Bronsted 산 자리임을 알 수 있었다. TPD(Temperature-Programmed Desorption) 및 DSC(Differential Scanning Calorimetry) 실험결과 탈알루미늄화된 SAPO-11에 흡착된 메틸아민과 물 분자는 탈알루미늄화가 되지않은 SAPO-11보다 높은 온도에서 그 피크들이 관측되었으며 메틸아민과 물 분자의 탈착 및 탈수 활성화 에너지는 탈알루미늄화의 정도에 비례하여 증가하였다. 이와 같은 현상은 제올라이트 분자체에 있어서와 유사하게 탈알루미늄으로 인하여 SAPO-11 분자체 내에 생성된 구조 하이드록실(srtuctural hydroxyl)기의 Bronsted 산 자리와 흡착 분자간의 보다 강한 상호작용에 기인되는 것으로 생각되었다.

Surface plasmon resonance 바이오센서를 이용한 재조합 B형 간염 표면항원의 정량분석 (Quantitative Assay of Recombinant Hepatitis B Surface Antigen by Using Surface Plasmon Resonance Biosensor)

  • We performed a basic experiment for rapid, on-line, real-time measurement of HBsAg by using a surface plasmon resonance biosensor to quantify the recognition and interaction of biomolecules. We immobilized the anti-HBsAg polyclonal antibody to the dextran layer on a CM5 chip surface which was pre-activated by N-hydroxysuccinimide for amine coupling. The binding of the HBsAg to the immobilized antibody was measured by the mass increase detected by the change in the SPR signal. The binding characteristics between HBsAg and its antibody followed typical monolayer adsorption isotherm. When the entire immobilized antibody was interacted, there was no additional, non-specific binding observed, which suggested the biointeraction was very specific as expected and independent of the ligand density. No significant steric hindrance was observed at 17.6 nm/$mm^2$ immobilization density. The relationship between the HBsAg concentration in the sample solution and the antigen bound to the chip surface was linear up to ca. $40\mu\textrm{g}$/mL, which is much wider than that of the ELISA method. It appeared the antigen-antibody binding was increased as the immobilized ligand density increased, but verification is warranted. This study showed the potential of this biosensor-based method as a rapid, simple, multi-sample, on-line assay. Once properly validated, it can serve as a more powerful method for HBsAg quantification replacing the current ELISA method.

  • CTAB 혹은 CTAB-나피온 촉매를 속빈 실리콘 튜브형 MCM-41에 담지시켰다. 이들 촉매들을 XRD, SEM, BET 등으로 분석하였다. 이들을 1,7-heptanediol 의 브롬화 반응 촉매로 사용하였고 다른 촉매들과 비교하였다. 이들 촉매들은 CTAB 보다 효과가 좋았으며 CTAB-Nafion/MCM-41 이 성능이 가장 우수하였다.
