Effect of combined sodium hyaluronate and carboxymethyl cellulose on ocular surface in rat dry eye model

  • This study was conducted to evaluate three different mixed formulations of sodium hyaluronate (SH) and carboxymethyl cellulose (CMC) using a low-humidity air flow-induced rat dry eye model and determine the most suitable mixture. The total thickness of the cornea, corneal epithelial thickness, corneal stroma thickness, damaged corneal epithelium percentage region, thickness of the bulbar conjunctiva epithelium, number of goblet cells, goblet cell occupation percentage region, and damaged bulbar conjunctiva epithelium percentage region were measured by histomorphological evaluation. After 5 h exposure to drying airflow, the thickness of the cornea and conjunctiva was decreased with desquamation of the corneal and conjunctiva epithelium. However, these dry eye symptoms were markedly inhibited by treatment with the reference and test formulations. More favorable effects on decreased thickness were detected in response to the CMC than the SH. However, SH had a greater protective effect against corneal and conjunctiva epithelial damage. The application of a mixture of 0.1% SH and 0.2% CMC showed more favorable effects on the corneal and conjunctival damage and the stabilization of the ocular surface than SH or CMC alone.

Effects of Laser and Electro Acupuncture Treatment with GB30·GB34 on Change in Arthritis Rat (환도·양릉천 레이저 및 전침이 관절염에 미치는 영향)

  • Objectives : This study aimed to investigate the effects of electroacupuncture (EA), the laser acupuncture (LA) and the combination therapy (LA+EA) in a collagenase-induced osteoarthritis rat model. Methods : Osteoarthritis rat model was induced by injection of collagenase into left lower articular cavity (50 μl to knee and 10 μl to ankle). In order to assess the anti-osteoarthritic effects of EA, the 650 nm LA and 650 nm LA+EA, the histopathological findings and plantar withdrawal responses were analyzed. Results : All of the treatment methods used in this study were effective in reducing pain. All treatment groups were effective in decreasing inflammatory cytokines of TNF-a and IL-6; the 650 nm LA and 650 nm LA+EA groups significantly reduced IL-1β. Conclusions : 650 nm LA and EA inhibit the production of collagenase-induced inflammatory mediators of osteoarthritis.

The Effect of Aspalatone, a New Antithrombotic Agent, on the Specific Activity of Antioxidant Enzyme in the Rat Blood

  • The antioxidant efficacy of aspalatone, a new antithrombotic agent, has been recognized in the neurotoxic model and in the cardiotoxic model in proliminary studies. We examined the specific activity of antiosidnat enzyme in the rat blood following administrations of aspirin, maltol, aspirin together with maltol, salicylmaltol (major metabolite of aspalatone) and aspalatone, respectively. Our assessment showed that salicylmaltol, maltol, aspalatone enhanced antiperoxidative activity. In addition, neither aspirin nor combination of aspirin and maltol, showed any significant effect on the activity of antioxidant enzyme. Because $H_{2}$$O_{2}$ accumulation may stimulate the thrombogenesis in blood, the result suggests that the induction of blood antiperoxidative activity produced by aspalatone may have beneficial effects on the thrombogenesis.

Antioxidative, Anti-inflammatory Effects of Jibaekjihwang-tang (zhibaidihuang-tang) on Osteoarthritic Rat Model (지백지황탕(知栢地黃湯)의 골관절염 Rat 모델에 대한 항산화, 항염증 효과)

  • Objectives This study intended to evaluate antioxidative, anti-inflammatory effects of Jibaekjihwang-tang on monosodium iodoacetate (MIA)-induced osteoarthritic rat model and investigate the potential mechanism. Methods Jibaekjihwang-tang (100 or 200 mg/kg body weight) was orally administered once daily for 2 weeks days from day 7 after intra-articular MIA injection. And blood analysis, the histologic examinations were performed. Moreover, protein expressions related to anti-oxidant and cartilage degradation and anti-inflammatory cytokines were measured by western blot analysis in cartilaginous tissue. Results Jibaekjihwang-tang reduced serum inflammatory cytokines such as tumor necosis factors-α and interleukin-6. Furthermore, the increase of anti-oxidant enzymes reversed the oxidative stress caused by MIA. Meanwhile, Jibaekjihwang-tang suppressed MIA-induced inflammation and cartilage degradation in cartilaginous tissue. Conclusions Jibaekjihwang-tang alleviated MIA-induced inflammation. Jibaekjihwang-tang was associated with a protective effect on cartilage and by reducing inflammation and cartilage degradation. These findings provide new approaches for understanding osteoarthritis therapy.

Cerebral Blood Flow Monitoring by Diffuse Speckle Contrast Analysis during MCAO Surgery in the Rat

  • The rodent model has been used frequently to understand stroke pathophysiology, due to its low cost and the large spectrum of genetic strains available. Here, we present a diffuse speckle contrast analysis system (DSCA) with a $1{\times}2$ optical switch that was used to non-invasively assess cerebral blood flow (CBF) changes in the rat during intraluminal suturing for middle cerebral artery occlusion (MCAO) surgery. The blood flow index (BFI) in the left hemisphere was lower than that in the right hemisphere because the left middle cerebral artery was occluded. Furthermore, the performance of the DSCA system was compared with that of commercial laser Doppler flowmetry. The changes in the BFI measured by the two systems were correlated strongly. The DSCA system was less sensitive to motion artifacts and able to measure relatively deep tissue flow in the rat's brain. In conclusion, the DSCA system secured CBF monitoring during surgery in a rodent model without craniotomy.

Effects of Far Infrared on Nitric Oxide level at a rat model of Autoimmune Arthritis Induced (원적외선이 자가면역성 관절염 흰쥐모델의 산화질소정량에 미치는 영향)

  • OBJECTIVE: This study was Far infrared(FIR) irradiating to a rat collagen-induced autoimmune arthritis and observed nitric oxide level of spine. METHODS: Type II collagen(CII) in adjuvant was injected into rats on day 0. The results showed that rats injected with CII plus adjuvant had signs of autoimmune arthritis on day 14. Thereafter, Far infrared(FIR) irradiation was begun on day 15. The rats were irradiated FIR twice per day for 20 min each time. RESULTS: Seven days later(Day 22), the level of NO at FIR irradiated group(FRG; Arth+FIR) was decreased more than FIR-non irradiated group(FNRG; Arth). Twenty-one days later(Day 36), the level of NO at FRG was more closed to the control group's NO level than the FNRG. CONCLUSION: These results indicated that Far infrared irradiating appears to reduce the nitric oxide level at a rat model of autoimmune arthritis induced.

Comparison of Nelumbinis Semen Extract with Hypericum Perforatum and Fluoxetine in Animal Model of Depression (연자육의 항우울 효과 및 프로티옴 분석을 통한 기전 연구)

  • Clinical evidence suggests that Nelumbinis Semen extracts have antidepressive properties and may offer an interesting alternative for the treatment of mood disorders. It was the aim of the present study to compare the effects of Nelumbinis Semen extracts with those of fluoxetine and hypericum perforatum extract in the rat forced swimming test (FST) and chronic mild stress (CMS), a model of depression. In the FST, p.o. administration of Nelumbinis Semen extracts (1 mg) induced a statistically significant reduction of immobility. The active behaviors in that test did not reflect decreased general activity because Nelumbinis Semen extracts failed to alter the locomotor activity of rats, measured in the open field test. Moreover Nelumbinis Semen extracts was superior to fluoxetine and hypericum perforatum extract in the incidence of sexual side-effects. These effects of Nelumbinis Semen extracts on the rat behavior is to be ascribed to increased Cytochrome c oxidase polypeptide Vla-liver, Mitogen-activated protein kinase 1 , Adenylosuccinate synthetase, and Aldehyde dehydrogenase in rat hippocampus.

Hyperkinetic Rat Model Induced by Optogenetic Parafascicular Nucleus Stimulation

  • Objective : The parafascicular nucleus (PF) plays important roles in controlling the basal ganglia. It is not well known whether the PF affects the development of abnormal involuntary movements (AIMs). This study was aimed to find a role of the PF in development of AIMs using optogenetic methods in an animal model. Methods : Fourteen rats were underwent stereotactic operation, in which they were injected with an adeno-associated virus with channelrhodopsin (AAV2-hSyn-ChR2-mCherry) to the lateral one third of the PF. Behavior test was performed with and without optical stimulation 14 days after the injection of the virus. AIM of rat was examined using AIM score. After the behavior test, rat's brain was carefully extracted and the section was examined using a fluorescence microscope to confirm transfection of the PF. Results : Of the 14 rats, seven rats displayed evident involuntary abnormal movements. AIM scores were increased significantly after the stimulation compared to those at baseline. In rats with AIMs, mCherry expression was prominent in the PF, while the rats without AIM lacked with the mCherry expression. Conclusion : AIMs could be reversibly induced by stimulating the PF through an optogenetic method.

Effect of Acupuncture applied to Hand Yang Meridian on the Rat Model of Ankle Sprain Pain (수양경(手陽經) 경혈(經穴) 자침(刺鍼)이 백서(白鼠)의 족과 염좌(捻挫) 통증(痛症)에 미치는 영향(影響))

  • Objectives : In the present study, the effect of electroacupuncture (EA) applied to hand yang meridian on the ankle sprain model was examined. Methods & Results : A common source of persistent pain in humans is the lateral ankle sprain. To model this condition, the rat's right ankle was bent repeatedly, overextending lateral ligaments, for 4 min under halothane anesthesia. The rat subsequently showed swelling of the ankle and a reduced stepping force of the affected limb for the next several days. The reduced stepping force of the limb was presumably due to a painful ankle. EA was applied to the several acupuncture point on the contralateral forelimb for 30 min under gaseous anesthesia. After the termination of EA, behavioral tests measuring stepping force were periodically conducted during the next 4 h. EA applied to SI-6 point produced a significant improvement of stepping force of the sprained foot lasting for at least 2 h. However, neigher LI-4 point nor TE-3 point produced any significant increase of weight bearing force. The improvement of stepping pressure was interpreted as an analgesic effect. The analgesic effect was specific to the acupuncture point since the analgesic effect on the ankle sprain pain model could not be mimicked by EA applied to a nearby point, LI-4 or TE-3. The analgesic effect of EA applied to SI-6 was more powerful when EA was applied by low-frequency and high-intensity stimulation. In addition, this effect need to be stimulated more than 15 min. Conclusions : These data suggest that EA produces a potent analgesic effect on the ankle sprain pain model in the rat. This analgesic effect is produced by applying EA to a Tae-Yang meridian at opposite side from the painful area in a stimulus point-specific way.
