Vinylsulfone Activated Agarose 에 Endo- 및 Exoinulinase의 고정화 (Immobilization of Endo- and Exoinulinase on Vinylsulfone Activated Agarose)

  • Inulinase의 효율적인 재사용을 위하여 vinylsulfone activated agarose에 endo- 및 exoinulinase를 고정화시켰다. Gram gel당 exoinulinase는 400U, endoinulinase는 80U까지 고정화시킬 수가 있었고 열안정성은 exoinulinase 에서 증가되었다. 두 고정화 효소의 혼합비율에 따른 synergistic effect는 endo/exo가 0.5-0.1일 대 가장 컸으며, synergistic effect는 혼합되지 않은 상태의 고정화 효소에 비해 그 활성이 약 1.7배 증가하였다. 두 고정화 효소의 최적 pH는 4.4-5.0 범위이었으며 operational stability는 batch reactor에서 20번 반복된 실험결과 어떠한 효소활성의 감소도 보이지 않았다.

Fructosyltransferase와 Glucose oxidase 혼합효소계를 이용한 고순도 Fructo-oligosaccharides 생산에서 반응 메카너즘에 대한 수학적 모델 (Mathematical Model for the Production of High-purity Fructo-oligosaccharides by the Mixed-enzyme System of Fructosyltransferase and Glucose Oxidase)

  • Fructosyltransferase와 glucose oxidase의 혼합효소계를 이용한 고순도 fructo-oligosaccharides 생산반응에서의 수학적 모델을 제안하고 실험적으로 검증한 결과 실험치와 모델 값이 서로 잘 일치하였다. 혼합 효소계에서 두 효소의 kinetic parameters를 구한 결과, fructosyltransferase 단일 효소계에서의 값들에 비해 $K_m$ 값들은 감소하였고, $K_m,\;V_{max}$값들은 증가하였다. 혼합 효소계의 반응메카니즘은 전체적으로 Michaelis-Menten kinetics로 표현할 수 있었고, 제안된 모델을 이용하여 고순도 fructo-oligosaccharides 생산에 이상적인 설탕농도를 예측할 수 있었다.

  • Waste papers(MOW and ONP) were deinked with alkaline cellulolytic enzyme from Coprinus cinereus 2249. The effectiveness of alkaline enzyme on sticky contaminant removal and bleaching of recycling pulp was investigated. The conclusions obtained from the results are as follows. The brightness of deinked pulps MOW and ONP was most high in 0.4IU enzyme treatment. \circled2 The removal effect of sticky contaminant was enhanced with alkaline cellulolytic enzyme treatment. Bleaching with sodium hydrosulfite was most effective in one and two stage bleaching after enzyme treatment of mixed wastepaper of MOW and colored paper. \circled4 The brightness of alkaline enzyme deinked ONP was increased 20% with $H_2O$$_2$ bleaching.

  • The present study investigated the effect of enzyme inclusion on silage quality using meta-analysis tool. A total of 16 research papers reporting the effect of enzyme application on silage quality were employed in the meta-analysis of this study. Mixed model for integrating quantitative results from multiple studies was used first to calculate the predicted error of each study. Individual error from the estimated model was the applied into standard deviation of each study to calculate the mean difference. Finally, summary effect was determined using standard mean difference (SMD) and inversed variance weighting. Mixed model analysis and SMD analysis showed the same effect patterns in all analysis items. Enzyme inclusion in silage significantly (p < 0.05) altered all silage quality characteristics investigated compared to control when enzyme was not included. Our results showed that enzyme treatment increased dry matter content, preserved crude protein effectively, and elevated water soluble carbohydrate content. However, the pH value, acetic acid, propionic acid, neutral detergent fiber, and acid detergent fiber contents in silage with enzyme inclusion were lower than those of the control.

Extraction of β-glucosidase from Bagasse Fermented by Mixed Culture under Solid State Fermentation

  • Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.

  • Development of stabilized enzyme was attempted for cosmetic applications. Papain, a proteolytic enzyme, was stabilized through conjugation with a soluble carbohydrate biopolymer, SC-glucan$^{TM}$ . With a novel structure of the conjugation site, stability of the enzyme was significantly enhanced such that more than 90% of the initial activity retained after a month storage at 45$^{\circ}C$, while no activity were detected in native enzyme or enzyme simply mixed with SC-glucan$^{TM}$ after the storage. Conjugation with SC-glucan$^{TM}$ not only extended the half-life of the enzyme on storage at higher temperature, but was also found to protect enzymes against some components contained in cosmetic products for skin care. Cosmetic lotion containing 1 % papain conjugate was more effective and less irritative in exfoliating stratum corneum of human skin than the lotion containing 5% lactic acid, one of the current popular exfoliating agents.gents.

배추 Myrosinase의 정제 및 효소학적 특성 (Purification and Enzymatic Characteristics of Myrosinase from Korea Cabbage)

  • Myrosinase from Korean cabbage(Bogdoli) was purified and its enzymatic properties were investigated. Myrosinase from the Korean cabbage was purified by DEAE Bio-Gel Sepharose, Concanavalin-A, and Mono-Q column chromatography and exhibited a 55KD molecular weight with a single band on the gel of SDS-PAGE. The enzyme was purified about 21-fold compared to its crude enzyme and a specific activity of purified enzyme was 15, 120units/mg. Optimum pH of the myrosinase was 7.0 in both phosphate and Tris-HCl buffer solutions, the enzyme was stable at pH 6.5~7.0. Optimum temperature of enzyme was 37~38$^{\circ}C$. The enzyme activity was significantly inhibited by Cu2+ and Hg2+, but enhanced by ascorbic acid, resulting in a maximum activity at 1mM ascorbic acid. Among the ascorbic acid analogues, dehydro-ascorbic acid did not affect, whereas others showed a little effect on the enzyme activity, but less than ascorbic acid itself. Reducing agents such as 2-mercaptoethanol and dithiothreitol had no effect on the enzyme activity, but the enzyme activity was enhanced when 2-mercaptoethanol was mixed with ascorbic acid.

  • Alkaline cellulolytic enzymes from cultured medium of Coprinus cinereus 2249 were purified with gel and ion-exchange chromatography and characteristics of those enzyme proteins were investigated. A fiber length distribution and a crystallinity of cellulose and sugar composition of enzyme treated Mixed Office Wastepaper(MOW) and Unbleached Kraft Pulp(UKP) were analysed. The conclusion could summarized as follows; \circled1 Alkaline and acidic, endo- and exo-glucanases were purified from cultured medium of Coprinus cinereus 2249. \circled2 The approximate molecular weight of alkaline endo-glucanase was 42 kDa, and also that of alkaline exo-glucanase was 50 kDa. A fiber length distribution and a crystallization of cellulose and sugar composition of enzyme treated MOW and UKP were not so much changed with original paper and pulp.

전통발효식품과 토양으로부터 분리된 혼합균주의 최적생육조건 및 음식물쓰레기 분해 효과 (Optimal Culture Conditions and Food Waste Decomposition Effects of Mixed Strains Separated from Traditional Fermented Food and Soils)

  • 음식물쓰레기 분해용 균주를 선발하기 위하여 전통발효식품과 토양으로부터 내염성, 내열성을 갖는 균주 분리를 시도하였으며, protease, amylase, cellulase 및 lipase의 효소활성도를 평가하였다. 5% NA 배지에서 분리된 균주는 212종이었으며, 전통발효식품의 경우 4가지 효소활성을 가지고, 토양의 경우 2가지 이상의 효소활성을 가지며, 억제환이 15 mm 이상의 크기를 지닌 79종의 균주를 우선 선발하였다. 선발된 균주를 대상으로 분자유전학적 동정을 진행한 결과, 중복성을 배제하여 11종의 균주를 최종적으로 선발하여 혼합균주로 사용하였다. 혼합균주의 최적 배양조건으로는 배양시간은 24시간, 배양온도는 $30^{\circ}C$ 그리고 pH의 최적 조건은 7.0 부근인 것으로 확인되었다. 최적배양된 혼합균주 처리에 따른 음식물쓰레기 분해능 평가 결과 멸균수 첨가군은 10 mesh ($2000{\mu}m$)에서 103 g, 혼합균주 첨가군은 10 mesh ($2000{\mu}m$)에서 18 g인 것으로 보아 혼합균주로 나타나 음식물의 고형분의 입자가 액상으로 분해되었음을 확인하였다.

Influences of Surfactant Tween 80 on the Gas Production, Cellulose Digestion and Enzyme Activities by Mixed Rumen Microorganisms

  • The surfactant Tween 80 was evaluated for its ability to influence cumulative gas production, cellulose digestion, and enzyme activities by mixed ruminal microorganisms grown on barley grain or Orchardgrass hay. The addition of Tween 80 at a level of 0.10% significantly (p<0.05) decreased the cumulative gas production rate from both barley grain or Orchardgrass hay substrates. However, 0.05% Tween 80 did not affect gas production rates compared to the control treatment. The addition of 0.05% Tween 80 to cultures growing on barley grain resulted in a significant increase in cellulase (90.01%), xylanase (90.73%) and amylase (487.25%) activities after 30 h incubation. Cultures utilizing Orchardgrass hay had a significant increase in cellulase (124.43%), xylanase (108.86%) and amylase (271.22%) activities after 72 h incubation. These increases in activities were also observed with cultures supplemented with 0.10% Tween 80 throughout all the incubation times tested. These results indicated that the addition of 0.05% Tween 80 could greatly stimulate the release of some of key enzymes without decreasing cell growth rate in contrast to trends reported with aerobic microorganism. Our data indicates potential uses of the surfactant Tween 80 as a feed additive for ruminant animals.
