결명자(決明子), 청상자(靑葙子), 밀몽화(密蒙花) 전탕액(前湯液)의 항균성에 대한 실험적 연구 (The Experimental Study on Antibiosis of Decoctions Made by Cassiae Semen, Celosiae Semen and Buddlejae Flos)

  • Objectives : This experimental study was performed to investigate antibiosis of decoctions made by Cassiae Semen(CaS), Celosiae Semen(CeS) and Buddlejae Flos(BF). Methods : Decoctions made by CaS. CeS and BF were prepared. After administering decoctions made by CaS. CeS and BF on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Condida albicans) which cause Keratitis, the size of inhibition zone and MIC(Minimum Inhibition Concentration) were measured Results : The inhibition zone on bacterial species didn't appear, after administering decoctions made by CaS, CeS and BF. Conclusions : This experimental study is showed that decoctions made by CaS, CeS and BF don't have antibiosis on bacterial species which cause Keratitis.

Development of a Blended Corrosion, Scale and Micro-Organism Inhibitor for Open Recirculating Cooling System

  • This paper presents the results of a study that was undertaken to optimize the ratio of the components of a new multi-component inhibitor blend composed of orthophosphate/ phosphonates/ acrylate copolymer/ isothiazolone. The effects of newly developed inhibitor on carbon steel dissolution in synthetic cooling water were studied through weight loss tests, electrochemical tests, scale tests, and micro-organism tests. The obtained results were compared to blank (uninhibited specimen) and showed that developed inhibitor revealed very good corrosion, scale, and micro-organism inhibition simultaneously. All measurements indicated that the efficiency of the blended mixture exceeded 90 %. The inhibitive effects arose from formation of protective films which might contain calcium phosphate, calcium phosphonate, and iron oxide. The nature of protective films formed on the carbon steel was studied by scanning electron microscopy (SEM) and auger electron spe ctroscopy (AES). Inhibitor used in this study appeared to have better performance for scale inhibition due to their superior crystal modification effect and excellent calcium carbonate scale inhibition properties. The effect of inhibitor on microorganisms was evaluated through minimum inhibitory concentration (MIC) test. All kinds of micro-organisms used in this study were inhibited under 78ppm concentration of inhibitor.

Synergistic Inhibition of Membrane ATPase and Cell Growth of Helicobacter pylori by ATPase Inhibitors

  • Helicobacter pylori were found to be resistant to azide but sensitive to vanadate, suggesting that defect in the P-type ATPase activity rather than F-type ATPase would be lethal to cell survival or growth. To elucidate the relationship between this enzyme inhibition and H. pylori death, we determined the effect of omeprazole (OMP) plus vanadate on enzyme activity and cell growth. The minimum inhibitory concentration (MIC; ca. 0.8$\mu$mol/disk) of vanadate for H. pylori growth was lowered over l0-fold with the aid of OMP, whereby its inhibitory potential toward the P-type ATPase activity was diametrically increased. Alternatively, we found that this enzyme activity was essential for active transport in H. pylori. From these observations, we strongly suggest that the immediate cause of the growth inhibition of H. pylori cells with OMP and/or vanadate might be defective in the cell's active transport due to the lack of P-type ATPase activity. From the spectral data with circular dichroism (CD) spectroscopy, we found that activated OMP (OAS) at concentration below MIC did not disrupt helical structures of membrane proteins. Separately, we determined the cytopathic effect of OAS by SDS-PAGE, indicating the change in the production of cytoplasmic protein but not cell membrane.

Antimicrobial Effect of Polyphenon 60 against Streptococcus mutans and Streptococcus sobrinus

  • Polyphenon 60 refers to the mixture of catechins present in green tea. The aim of this study was to investigate the antimicrobial activities of polyphenon 60 against 4 strains of Streptococcus mutans and 2 strains of Streptococcus sorbrinus, which are the major causative bacteria of dental caries. The minimum bactericidal concentration (MBC) values of polyphenon 60 for S. mutans and S. sobrinus were determined and the effect of biofilm formation inhibition of that was evaluated. The MBC value of polyphenon 60 against the bacterial strains was 2.5 mg/ml except for one particular strain, S. mutans KCOM 1128 for which the value was 1.25 mg/ml. The results of biofilm formation inhibition assay revealed that polyphenon 60 inhibited biofilm formation more than 90% at a concentration of 2.5 mg/ml. It was apparent that polyphenon exhibited biofilm formation inhibition activity along with bactericidal effect against S. mutans and S. sobrinus. Therefore, it is proposed that polyphenon 60 as one of the components of bactericidal agents could be useful in developing oral hygiene products, toothpaste or gargling solution.

다양한 山豆根·川烏·木槿皮 혼합물 용액이 足部白癬菌에 미치는 영향 (The Experimental Study on Antifungal effects of mixture of Sophorae Subprostratae Radix, Acoti Radix, and Hibisci Syriaci Cortex by the Three Types of Extraction on three species of the dermatophytes.)

  • This experimental study was performed to investigate the antifungal effects of mixture of Sophorac Subprostratae Radix, Aconti Radix, and Hibisci Syriaci Cortex by the three types of extraction in the dermatophytes such as Trichophyton rubrum, Trichophyton mentagrophytes, and Epidermophyton floccosum. The following results were obtained by using Minimum Inhibition Concentration(MIC) and Inhibition Zone. 1. MIC on Trichophyton mentagrophytes in Sample S-2, Sample S-3, and Sample-Y were 10${\mu}l$, respectively, showing the highest antifungal effect. 2. MIC on Epidermophyton floccosum in Sample S-2, Sample S-3, and Sample-Y were 10㎕ respectively, showing the highest antifungal effect. 3. MIC on Trichophyton rubrum in Sample S-3, Sample-Y were 10${\mu}l$ respectively, showing the highest antifungal effect. 4. The size of inhibition zone on Trichophyton mentagrophytes for Sample S-3 were 22.3mm in 10${\mu}l$, showing the highest antifungal effect. 5. The size of inhibition zone on Epidermophyton floccosum for Sample S-3 was 34.3mm in 10${\mu}l$, showing the highest antifungal effect. 6. The size of inhibition zone on Trichophyton rubrum for Sample-Y was 26.5mm in 10${\mu}l$, showing the highest antifungal effect. Whereas. in extractions, that of Sample S-3 was 12.5mm in 10${\mu}l$, showing the highest antifungal effect. 7. From the above results, Sample S-3 on Trichophyton mentagrophytes, Epidermophyton floccosum and Sample-Y on Trichophyton rubrum showed the highest antifungal effects. This results suggests that vinegar extraction excels other extraction method using water or ethanol to measure the antifungal effect on dermatophytes. In addition, the result shows that the extract of herbal medicines can be used to tinea pedis and if further study is performed, the use of the extract of herbal medicines will be valuable and beneficial in the clinical medicines. Polarization Behavior and Corrosion Inhibition of Copper in Acidic Chloride Solution Containing Benzotriazole

  • Polarization behavior and corrosion inhibition of copper in acidic chloride solutions containing benzotriazole were studied. Pourbaix diagrams constructed for copper in NaCl solutions with different BTAH concentrations were used to understand the polarization behavior. Open circuit potential (OCP) depended not only on chloride concentration, but also on whether a CuBTA layer was formed on the copper surface. Only when the (pH, OCP) was located well in the CuBTA region of the Pourbaix diagram, a stable corrosion inhibiting CuBTA layer was formed, which was confirmed by X-ray Photoelectron Spectroscopy (XPS) and a long-term corrosion test. The OCP for the CuBTA layer decreased logarithmically with increasing [Cl-] activity in the solution. A minimum BTAH concentration required to form a CuBTA layer for a given NaCl concentration and pH were determined from the Pourbaix diagram. It was found that 320 ppm BTAH solution could be used to form a corrosion-inhibiting CuBTA layer inside the corrosion pit in the sprinkler copper tube, successfully reducing water leakage rate of copper tubes. These experimental results could be used to estimate water chemistry inside a corrosion pit.

국화 꽃 휘발성 향기성분의 식중독균에 대한 항균 작용 (Antibacterial Action against Food-Borne Pathogens by the Volatile Flavor of Essential Oil from Chrysanthemum morifolium Flower)

  • 국화 정유의 식중독균에 대한 항균력을 검증하기 위해 국화 정유와 그 주성분인 camphor, chrysanthemyl alcohol, $\alpha$-pinene, $\gamma$-terpinene, ${\alpha}+{\beta}$-thujone를 10종 균주에 대하여 agar-well diffusion 분석법으로 생육 여부를 분서하였다. 실험 결과 국화 정유와 그 지표물질은 Gram 양성균과 음성균 모두에 대해서 탁월한 항균 효과가 나타났다. Agar-well diffusion 분석법 결과 국화 정유와 그 지표물질은 Gram 음성균보다 Gram 양성균에 대해서 강력한 활성을 나타내었다. 국화 정유의 minimum inhibitory concentration은 모든 균주에 대하여 5 ${\mu}g/m{\ell}$에서 균의 생장을 저해하는 효과가 나타났고, minimum bacteriocidal concentration은 S. choleraesuis와 L. monocytogenes에 대해서는 시험농도 내에서 사멸효과가 관찰되지 않았으나, 다른 균주에 대해서는 5~20 ${\mu}g/m{\ell}$에서 살균력이 나타났다. 또한 액체 배양중의 항균효과를 조사한 결과, 국화 정유와 그 지표물질은 24시간 동안 꾸준히 항균력이 지속되었다. 따라서 camphor, chrysanthemyl alcohol, $\alpha$-pinene, $\gamma$-terpinene, ${\alpha}+{\beta}$-thujone를 조성분으로 함유한 국화 정유성분은 10종의 식중독균에 대한 항균활성이 입증되어, 천연 항균제재로서 식품보존료로의 이용이 가능하리라 생각된다.

측백나무 열매 추출물의 항균활성 (Antimicrobial Activities of Organic Extracts from Fruit of Thuja orientalis L.)

  • This study was carried out to investigate the antimicrobial activities of organic extracts obtained from the fruit of Thuja orientalis L. The native fruits in Korea were collected and extracted by 80% ethanol, and the extract was sequentially fractionated with n-hexane, chloroform, ethylacetate, and butanol. The fraction yields of n-hexane, chloroform, ethylacetate, butanol and water of ethanol extract were 10.15%, 10.05%, 1.45%, 45.35% and 27.55%, respectively. n-Hexane-soluble fraction showed the highest antibacterial activity against gram positive bacteria, while the chloroform, ethylacetate, butanol and aqueous fractions did not show any antibacterial activity. Minimum inhibitory concentration (MICs) on Staphyloycoccus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction were $100\;{\mu}g$, $500\;{\mu}g$ and $50\;{\mu}g$/disc, respectively. The antibacterial activity was not destroyed by heating at 80, 100, $120^{\circ}C$ for 30 min and was not affected by pH. In the inhibitory test against the Staphylococcus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction showed potent growth inhibition at the concentration of 0.1 and $0.5\;{\mu}g/mL$ for 12~24 hours and n-hexane-soluble fraction did not show any mutagenic activity.

머스커다인 포도 추출물의 항산화 및 항균 활성 (Antioxidant and antimicrobial activities of muscadine grape extracts)

  • 본 논문은 미국 남부지역 특화작물인 머스커다인 포도의 부분별 추출물의 항산화능 및 항균활성을 구명하기 위한 목적으로, Higgins, Jumbo, Noble 3종의 머스커다인 품종을 대상으로 하여 포도의 과피/과육부분과 종자부분을 추출하여 실험에 사용하였다. 각 추출물에 대하여, 항산화능은 총페놀함량 및 라디칼 소거능(Scavenging activity 및 $EC_{50}$)을, 항균활성은 E.coli K12에 대한 최소생육저해농도(minimum inhibitory concentration, MIC)를 조사하였다. 종자 추출물의 총페놀함량은 231.24~294.81 mg/mL GAE로 과피/과육 추출물에 비하여 높은 함량을 보였다. 또한 Higgins품종의 종자 추출물이 가장 우수한 라디칼소거능($EC_{50}=0.026mg/mL$)을 나타내, 종자추출물이 과피/과육추출물보다 우수한 항산화능을 보유하였다. 반면, E.coli K12에 대한 항균활성은 Higgins 품종의 과피/과육추출물이 40 mg/mL의 MIC를 보임으로써 가장 우수하였다. 따라서, 본 연구는 머스커다인 포도 추출물이 천연 유래의 항산화 및 항균 기능성을 보유한 소재로 활용될 수 있는 잠재적 가치를 제시하였다.

GROWTH INHIBITION OF Acanthamoeba BY HYDROPEROXYNAPHTALIMIDES UPON PHOTOIRRADIATION

  • Acanthamoeba strains were photoirradiated in the presence of light-sensitive organic peroxides (hydroperoxynaphthalimide derivatives) including a Photo-Fenton reagent at 366 nm. The survival rates of Acantharnoeba strains determined after 20 h culture showed a significant decrease only upon photoirradiated conditions. The most effective hydroperoxynaphthalimides among these compounds was the bromohydroperoxynaphthalimide (Br-HPO II). The minimum inhibitory concentration (MIC) of Br-HPO II is 100 times lower than that of hydrogen peroxide.
