The butanol extract of paulownia tomentosa stem showed antibacterial activity against staphyl ococcus aureus (SG511, 285 and 503), Streptococcus pyogenes (A308 and A77) and Streptococcus farcium MD8b etc. The most active compound of the extractg was identified to be campneoside I, which had a minimal inhibitory concentration(MIC) of $150{\;}{\mu}g/ml$ against Strptococcus and Staphylococcus species. From such antibacterial activity, the methoxy group of campneoside I was posulated to be the essential element for the antibacterial activity.
Objectives: The object of this study was to observe the in vitro antibacterial effects of Gagam-seopyoungjeon aqueous extracts (GGSYJ) against Gardnerella vaginalis and the possible synergic combination effects with clindamycin. Methods: Antibacterial activities against Gardnerella vaginalis of GGSYJ were detected using minimal inhibition concentration (MIC), and the effects on the bacterial growth curve were also monitored at MIC and MIC${\times}$2 levels. The combination effects of GGSYJ with clindamycin were observed by checkboard microtiter assay, and the effects of bacterial growth curve treated with GGSYJ MIC+clindamycin MIC, 1/2 MIC and 1/4 MIC, respectively. The effects on the bacterial invasion and intracellular killing of GGSYJ were also observed using human vaginal epithelial (VK2) and murine macrophage (Raw264.7) cells with combination effects with clindamycin after treatment of GGSYJ MIC+clindamycin 1/2 MIC, 1/4 MIC and 1/6 MIC, respectively. Results: The MIC of clindamycin and GGSYJ against Gardnerella vaginalis were detected as $0.012{\pm}0.006$ (0.004~0.016)${\mu}g/ml$ and $1.016{\pm}0.524$ (0.391~1.563) mg/ml, respectively. Clindamycin and GGSYJ were also showed marked dosage-dependent inhibition of bacterial growth, and significant decreases of viable cells were detected in clindamycin MIC+GGSYJ MIC and clindamycin 1/2 MIC+GGSYJ MIC treatment as compared with each of single clindamycin MIC and GGSYJ MIC treatments. And significant decreases of intraepithelial and intra-macrophage viable bacteria numbers were detected in clindamycin 1/2 MIC+GGSYJ 1/2 MIC and clindamycin 1/4 MIC+GGSYJ 1/2 MIC treatment as compared with each of single clindamycin GGSYJ 1/2 MIC treatments, respectively. Conclusions: GGSYJ showed slight antibacterial effects against Gardnerella vaginalis, but they showed dosage-dependent inhibitory effects on the bacterial growth and VK2 epithelial invasions of bacteria with favorable accelerating effects of intracellular killing activities of macrophages. In addition, combination of GGSYJ also increased the inhibitory effects of clindamycin on the epithelial invasions of Gardnerella vaginalis and intracellular killing activities of macrophages against Gardnerella vaginalis as 2-fold higher as compared with clindamycin single treatment, respectively. Therefore, we expected that the clinical dosages of clindamycin can be reduced as 1/2 levels as combination with GGSYJ.
This study was carried out to evaluate cytotoxic effects of Ajuga multiflora Bunge extracts on murine leukemia tumor $(P388D_1)$ cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The comparison of $IC_{50}$ values of Ajuga multiflora Bunge extracts in L1210 and $P388D_1$ cell lines showed that their susceptibility to these extracts decreased in the following order: Adriamycin>methanol extract>chloroform extract>ethyl acetate extract>hexane extract>water extract by the MTT assay. In order to develop an antimicrobial agent, dried Ajuga multiflora Bunge was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extract against microorganisms were also examined. Antimicrobial activities of amocla and ketoconazole as references were compared to those of extracts of $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, gram-positive bacteria and fungi $(MIC>200\;{\mu}g/ml)$. These results suggest that the methanol soluble extract of Ajuga multiflora Bunge may be a valuable choice for the studies on the treatment of murine leukemia tumor cell lines.
This study was focused on the evaluation of MarB alongside with MarA for its regulatory effects upon the efflux function of AcrAB pump, which were induced or not, perhaps as a target. Transductions of marR and/or acrAB mutation which were derived from Mar and/or AcrAB mutants of wild type E. coli K-12, respectively, into the multicopy plasmid in wild type E. coli backgrounds or into the chromosome of isogenic parents were done. Minimal inhibitory concentration (MIC) of transduced mutants was compared with their original mutants. This study reports the indirect evidences that suggests a model in which MarB along with MarA have a putative negative regulatory effect upon the efflux function of AcrAB/TolC pump while MarA alone have a positive regulatory effect to the expression of acrRAB operon at transcription level. The target of MarB with MarA for its putative negative regulator might be the AcrAB efflux pump. Another efflux system (s) might be negatively regulated by MarB with MarA, and be involved in the efflux of antibiotics which were otherwise extruded preferentially by AcrAB efflux pump.

We carried out the enzymatic synthesis of 1, 2-hexanediol galactoside (HD-gal) by transgalactosylation reaction using recombinant Escherichia coli ${\beta}-galactosidase$ (${\beta}-gal$). The amounts of ${\beta}-gal$ and 1, 2-hexanediol (HD), pH, and temperature, respectively, were first optimized (${\beta}-Gal$, 4.8 U/mL; HD, 75 mM; pH, 7.0; temperature, $37^{\circ}C$). Under these optimal conditions, about 96% HD was converted to HD-gal. When we investigated the water holding capacities (WHCs) of HD and HD-gal using pig epidermis in the concentrations of 84.4, 126.6, 168.8, 211.0 mM, WHC of HD-gal was superior to HD. In particular, at 168.8 mM HD and HD-gal, WHC of HD-gal showed about 20% greater than that of HD. However, it was observed that MIC values against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus of HD-gal were about three to ten times greater than those of HD, although MIC value of HD-gal against Enterococcus faecalis was almost the same as that of HD. Finally, it was concluded that the covalent bonding of a galactose molecule to HD (transgalactosylation) resulted in an increase in WHC of HD-gal and a decrease in anti-bacterial activity.

Mastitis is an inflammatory condition of the mammary gland, most often caused by bacterial infections, resulting in significant economic losses to the dairy industry. Antimicrobial resistance has been of great concern because of the extensive clinical use of antibiotics. For this reason, the development of new compounds as an alternative treatment to bovine mastitis is needed. Bee venom has been widely used as an oriental treatment for several inflammatory diseases and bacterial infections. The aim of the present study was to evaluate the antimicrobial activity of bee venom on bacteria isolated from bovine mastitis. A total of 107 isolates from bovine mastitic milk samples collected in 2019 and 2020 in Jeonbuk province. All bacterial isolates were tested for susceptibility to bee venom of the honey bee (Apis mellifera). In order to obtain comprehensive antibacterial activities of the bee venom, we measured the minimal inhibitory concentration (MIC) of the bee venom against bacterial strains. Bee venom showed significant inhibition of bacterial growth of Gram-negative bacteria Citrobacter spp., Escherchia coli, Klebsiella spp., Pseudomonas spp., Serratia spp. and Raoultella with MIC values of 96, 81, 72, 230, and 85 ㎍/mL, respectively, and Gram-positive bacterial Enterococcus spp., Staphylococcus spp. and Streptococcus spp. with MIC values of 29, 21 and 16 ㎍/mL, respectively. The results indicated that the MIC values were different depending on the bacterial strains, and those of Gram-positive bacteria were lower than those of Gram-negative bacteria for bee venom. These findings suggested that bee venom could be an effective antimicrobial treatment for bovine mastitis; however, further research is necessary to evaluate the mechanism underlying the antimicrobial action, its effectiveness/safety in vivo and effective application for therapeutic use.
Two constituents were isolated as the antibacterial principles from the methanolic extract of Crassirhizomae Rhizoma against Streptococcus mutans OMZ176 which is known to be a strong cariogenic bacterium. They were identified as flavaspidic acid PB and flavaspidic acid AB by means of physicochemical methods. In order to investigate antibacterial activities of Zedoariae Rhizoma against Streptococcus mutans ATCC27351, paper disc test, minimal inhibitory concentration (MIC) test and pH check were carried out. The 80% ethanol extract of Zedoariae Rhizoma(ZXE) showed significant activity. The MIC of ZXE was 25.0mg/ml.
In order to study antibiotic activities of basidomycetes (mushroom), 68 species of mushroom were extracted with petroleum ether, 80% ethanol, and distilled water in that order. A total of 204 extracts were obtained. Antibiotic activities against Microsporum gypseum were observed from the petroleum ether extracts of Abortiporus DGU-L6 mushroom, and the water extracts of Clitogbe DGU-7 mushroom. Antibiotic activity against Aspergillus niger were observed from the 80% ethanol extracts of Cortinarius DGU-51 and Marasmminus DGU-L67 mushroom. The petroleum ether extracts of Hetero DGU-L25 mushroom showed various antibiotic activities, particularly strong activities against M. canis. and the minimum inhibitory concentration (MIC) was $40\;{\mu}g/ml$. The extracts also showed antibiotic activities against A. niger (KCTC 2025), A. niger (KCTC 2118), A. versicolor (KCTC 2120), A. flavus (KCTC 2117), M. gypsem, Pyricularia oryzae, and Trichopyto mentagrophytes, and MIC for each fungus was $600\;{\mu}g/ml,\;500\;{\mu}g/ml,\;800\;{\mu}g/ml,\;100\;{\mu}g/ml,\;600\;{\mu}g/ml,\;200\;{\mu}g/ml,\;and\;600\;{\mu}g/ml$, respectively.

Paeonia lactiflora was stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activity of each extract was investigated. Methanol extract of P. lactiflora revealed anti-microbial activity against S. mutans, C. albicans, and S. aureus. Also, hexane fraction revealed anti-bacterial activity against S. mutans and ethyl acetate fraction acted as potent anti-microbial agent on C. albicans and S. aureus. The relative growth ratio(RGR) of hexane fraction of P. lactiflora against S. mutans were determined as 77.8% in concentration of 0.125 mg/ml, 98.46% in 0.25 mg/ml and 100% in 0.5 mg/ml. The ethyl acetate fraction of P. lactiflora revealed RGR against C. albicans as 52.5% in concentration of 0.125 mg/ml, 60.83% in 0.25 mg/ml and 78.33% in 0.5 mg/ml. It indicate that increasing concentration increase RGR. The measured minimal inhibitory concentration(MIC) of hexane fraction on S. mutans KCTC 5316 strain was 0.5 mg/ml and MIC of ethyl acetate fraction on C. albicans KCTC 7270 was 2.0 mg/ml. The experiment of inhibition to growth of KB roll(oral squamous cell carcinoma) result 61.9% in butanol, 76.7% in hexane extract of P. lactiflora. The hexane extract exhibit potent inhibition effect to the growth of KB cell. These results suggest that the hexane extract of Paeonia lactiflora has antimicrobial activity against S. mutans and has preventive effect to dental caries in addition to potent inhibition to KB cell growth.
