Chitosan has been known to have an antimicrobial activity. Chitosan samples having different molecular weights were tested for the antimicrobial activity against Escherichia coli in a liquid medium and on the paper sheets coated with different types of chitosan samples. In a liquid medium, Chitosan A $(Mw\;3,000{\sim}5,000)$ and B $(Mw\;200,000{\sim}500,000)$ showed minimal inhibitory concentration (MIC) of 40 and $10\;{\mu}g/ml$, respectively. The MIC of chitosan C $(Mw\;500,000{\sim}1,500,000)$ was similar to that of chitosan B. The paper sheets coated with chitosan A, B, and C also showed growth inhibition against E. coli. As a typical agricultural product, strawberry was tested for the extension of shelf life by using the chitosan-coated packaging papers. Storage experiments showed that chitosan-coated packaging papers extended the shelf life of strawberry at the chilling temperature of $4^{\circ}C$.
The susceptibilities of 45 clinical isolates of bacteroides frogilis to cefaclor, ciproflxacin and imipenem were determined by new method, E-test (AB Bidisk, Solna, Sweden) and were compared with those from conventional agar dilution method by using brain heart infusion, Mueller-Hinton and Wilkins Chalgren agar plates. And the susceptibility of 60 clinical isolates of Bacteroides fragilis group (B. fragilis 45 strains, B. distasonis 6 strains, B. ovatus 5 strains, B. thetaiotaomicron 4 strains) to 5 quinolones (ciprofloxacin, enoxacin, norfloxacin, ofloxacin, pefloxacin) were determined by in vitro agar dilution method. Compared with agar dilution MICs for B. fragilis 45 strains, 90.3% of E-test MICs were within ${\pm}$1 dilution of the agar dilutions, and 98.4% were within 2 dilutions. And there were little effect of different medium bases to determine MICs except Mueller-Hinton agar. On Mueller-Hinton agar, B. fragilis showed have or no growth activity. In vitro susceptibility of B. fragjlis group to quinolones, most of the test strains showed resistant patterns to quinolones except ofloxacin and there was little difference of susceptibility patterns between species of B. fragilis group.
The methanol extracts of 19 commercial herb medicines was analyzed to antibacterial activities against Edwardsiella tarda, causing several fish diseases. Rhus javanica showed most strong antibacterial activity against E. tarda and Escherichia coli. Methanol extract of R. javanica was further extracted using several organic solvents having different polarity. Extract from ethyl acetate fraction showed strong activity against E. tarda as well as E. coli. Minimal inhibitory concentration, MIC of R. javanica extract was measured and resulted showing $64\;{\mu}g/m{\ell}$ for E. tarda and $256\;{\mu}g/m{\ell}$ for E. coli. It is needed that, from these results, further purification and isolation of reposible compound of these activities and further study on the synergy effect using combination with antibiotics against pathogenic bacteria.

Machilus thunbergii has been showed relation to antimicrobial activity on minimal inhibitoty concentration (MIC) and colony forming inhibitory activity (CFIA) test, so that can be used to food preservatives for green vegetable. Nanoparticles has been made of edible materials. 80% of the nanoparticles has been characterized by image analyser and electron microscopy, showing in the range under 300 nm diameter. The sprayed nanoparticles remained on the surface of chicon even after washing by dilution water, then activate biological activities for storage of chicon with storing and releasement system of extracts. Chicon treated nanoparticle has been kept fresh condition about 2 months longer than 3 weeks of the non-treated control. It can be tell that treatment with nanoparticle of M. thunbergii extracts extends storage time of chicon possibly by inhibition of ethylene production through efficiency control on cell breathing.

Mutans streptococci, especially S. mutans and S. sobrinus strongly implicated in pathogenesis of dental caries, the major cause of tooth loss in children. Use of an antibacterial agent controlling dental caries has been rationalized. The present study was performed to observe the antibacterial effect of inorganic polyphosphates (polyP) on S. mutans and S. sobrinus. S. mutans GS5 and S. sobrinus 6715 were grown in brain-heart infusion broth with or without polyP. Minimal inhibitory concentration (MIC) of polyP for S. mutans GS5 was determined to be 0.08% and that for S. sobrius 6715 was 0.17%. PolyP 15 added to the growing culture of S. mutans GS5 and S. sobrinus 6715 at their exponential phase was as effective in inhibiting the growth of S. mutans GS5 and S. sobrinus 6715 as polyP added at the very beginning of the culture. More than 85% of the cells lost their viability determined by viable cell count when polyP 15 was added to the culture of growing S. mutans GS5 at MIC, suggesting that polyP 15 has bacterial effect on the bacterium. And more than 99.9% of the cells lost their viability determined by viable cell count when polyP 15 was added to the culture of growing S. sobrinus 6715 at MIC, suggesting that polyP 15 has bacterial effect on the bacterium. Intracellular nucleotide release from S. mutans CS5 and S. sobrinus 6715 was increased in the presence of polyP 15 for 5h but was not really reversed by the addition of divalent cations like $Ca^{++}\;and\;Mg^{++}$. The majority of the cells appeared to be atypical in their shape, demonstrating accumulation of highly electron-dense granules and ghost cells. The overall results suggest that polyP have a strong bactericidal activity against S. mutans and S. sobrinus in which lysis in relation to chelation may not play the major role but unknown mechanism that possibly affects the viability of the bacterium may be involved. PolyP may be used as an agent for prevention of dental caries.
The expression and secretion of human lactoferrin (hLf) in Sacclnromyces diastaticus were performed. 1. For the secretion of hLf in yeast, recombinant plasmid pYEGLf was constructed using promoter, secretion signal sequence of glucoamylase I gene (STA1) and transcriptional terminator of GAL7 gene. 2. Each correct recombinant plasmid was selected by mini-preparation of plasmid DNA from E coli transformant and restriction enzyme digestion analysis. The selected plasmids, pYEGLf, were transformed into S. diastaticus YIY345 as a expression host, respectively. 3. Western blot analysis using rabbit anti-hLf was carried out to identify expressed hLf. Positive signals were shown in culture supernatant of pYEGLf transformant. 4. About $100{\mu}g-1mg$ of concentrated culture supernatant of positive clone were loaded on paper disc and tested for the antimicrobial activity against E coli. However, no activity was observed. We concluded that this fact results from low concentration of hLf secreted from yeast, compared with the fact that MIC of hLf is as high as $3mg/m{\ell}$. Therefore, the purification of secreted hLf may be require to investigate the antimicrobial activity. From this study, the feasibility of low-cost production of sufficient quantities of human lactofferin for nutritional and therapeutical applications were suggested.
Contamination level and characteristics of Ralstonia solanacearum in soil of greenhouses cultivating tomato plants in Chungbuk province was determined. R. solanacearum was isolated with the semiselective media in 27 greenhouse soil samples out of 133 greenhouses soil investigated, which indicates 20.3 % of tomato cultivating greenhouses in Chungbuk contaminated with the bacterial wilt pathogen. Density of R. solanacearum was estimated to 10$^{2.4}$ cfu/g in the contaminated soil. All 71 isolates of R. solanacearum which containing 12 isolates from the diseased tomato plants were race 1, ann 35 isolates of them were biovar 3 and 36 isolates were biovar 4. More than 88% of 71 isolates were inhibited growth on nutrient agar containing oxolinic acid 0.5 $\mu\textrm{g}$/ml, streptomycin 25 $\mu\textrm{g}$/ml, tetracycline 5 $\mu\textrm{g}$/ml and cupric sulfate 375 $\mu\textrm{g}$/ml (1.5 mM). The 11.3%, 4.2% and 5.6 % of the isolates can grow on nutrient agar containing 10 times more oxolinic acid, streptomycin, tetracycline than minimal inhibitory concentration of the sensitive strains. Five isolates were resistant to 2 bactericides and one isolates was resistant to all 3 bactericides.
Efficacy of different control methods was evaluated for disease management of tomato bacterial wilt caused by $Ralstonia$$solanacearum$. All six chemical pesticides applied to the bacterial suspension showed $in$$vitro$ bactericidal activities against $R.$$solanacearum$. Minimal inhibitory concentrations (MICs) of copper hydroxide (CH), copper hydroxide-oxadixyl mixture (CH+O), and copper oxychloride-dithianon mixture (CO+D) were all 200 ${\mu}g/ml$; MIC of copper oxychloride-kasugamycin (CO+K) mixture was 100 ${\mu}g/ml$; MICs of both streptomycin- validamycin (S+V) and oxine copper-polyoxine B mixture (OC+PB) were 10 ${\mu}g/ml$. Among these chemical pesticides, treatment of the detached tomato leaves with the 5 pesticides (1 mg/ml), except for OC+PB delayed early wilting symptom development caused by the bacterial inoculation ($10^6$ and $10^7$ cfu/ml). Four pesticides, CH, CH+O, CO+K and S+V, showed disease protection in pot analyses. Six plant essential oils, such as cinnamon oil, citral, clove oil, eugenol, geraniol and limonene, differentially showed their antibacterial activities $in$$vitro$ against $R.$$solanacearum$ demonstrated by paper disc assay. Among those, cinnamon oil and clove oil exert the most effective activity for protection from the wilt disease caused by the bacterial infection ($10^6$ cfu/ml). Treatment with cinnamon oil and clove oil also suppressed bacterial disease by a higher inoculum concentration ($10^7$ cfu/ml). Clove oil could be used for prevention of bacterial wilt disease of tomato plants without any phytotoxicity. Thus, we suggest that copper compounds, antibiotics and essential oils have potency as a controlling agent of tomato bacterial wilt.
A total of 108 strains of C jejuni isolated from pigs and chickens were examined for the susceptibility to 10 antimicrobial agents and normal sera of cattle, sheep, guinea pigs and chickens. Minimal inhibitory concentration(MIC) ranges of antimicrobial agents to C jejuni isolates were $${\leq_-}1.56$$ to $${\geq_-}100{\mu}g/ml$$ for erythromycin, rifampin, streptomycin and tetracycline, 50 to $${\geq_-}100{\mu}g$$ for cephalothin, $${\leq_-}1.56$$ to $50{\mu}g$ for ampicillin, $${\leq_-}1.56$$ to $25{\mu}g$ for kanamycin and nalidixic acid, $${\leq_-}1.56$$ to $12.5{\mu}g$ for chloramphenicol and gentamicin. Resistance rates of C jejuni were showed to in order of rifampin(84.7%), tetracycline(56.2 %), erythromycin(17.1%) and ampicillin(3.8%), all of the strains were sensitive to chloramphenicol, gentamicin and kanamycin, and the incidence rates of resistant C jejuni were highly frequent in pig isolates than chicken isolates. The drug resistance patterns of 87 chicken isolates C jejuni to 9 antimicrobial drugs were showed 12 patterns, and Sm Ra Tc(24.1%), Sm Ra(21.8%) and Ra Tc(14.9%) were relatively common, and also 21 pig isolates were showed 6 patterns and Em Sm Ra Tc(57.1%) were most frequent. The majority of the isolates showed multiple drug resistance. Bactericidal activity of 10% normal sera from healthy animals were examined for 60min at $37^{\circ}C$. C jejuni were decreased from 0.4 to 1.0 ${\log}_{10}$(p<0.01), and serum susceptibility were high in order of guinea pig, sheep, chicken and cattle sera. Serum sensitivity of C jejuni Ch-39 strain in increased serum concentation up to 10, 20, 40 and 80% were highly significant. In the normal animal serum, the number of Ch-39 strain were decreased from $1.8{\times}10^4/ml$ to $2.7{\times}10^3/ml$ after 60 min incubation(p<0.01), but the numbers were decreased to $6.6{\times}10^3/ml$ in the heat inactivated normal serum for 30 min at $56^{\circ}C$. Bactericidal activity was restored in the heat inactivated normal serum after the serum of complement source was added.
Vibrio vuln$cus has been recognized as a pathogen of septicemia and wound infection, when the organism attacks high-risk persons with a history of hepatic disease. alcohol abuse. diabetes or any debilitative disease. Forty six strains of K vulnzjicus. isolated from 1025 marine specimens from May to Novemver for three years. from 1985 to 1987. were studied for their biochemical properties. growth requirements, serotype and drug susceptibilities. The isolates were different in their various biochemical reactions. Ninety-five percent of isolated strains were able to ferment lactose, while most strains didn't utilize sucrose in their biochemical test, for example ornithine, gelatin and mannitol were quite dit'ferent composition than those described in other reports. It was found that the biochemical test wasn't useful for identifying strain. The type of somatic 0 antiserum was determined in isolates from marine sources and in patients with Vibrio septicemia. In patient isolates. 1-2 group were 24% and 1-4 group were 42%. However. 02 group(33%) were more abundant in isolates from marine sources. Minimal inhibitory concentrations(M1Cs) of chloramphenicol, tetracycline. erythromycin and ampicillin were determinef for V vuln~ficus by broth dilution method. MIC90 was I , 0.25, :! and 4,ug/ml in patient isolates. 1, 0.25, 2 and 2 ,ug/ml in marine isolates. The divalent chelating agent, IDTA. inhibited the growth of V. vuln!'ficus at 6.25 mMlml of MIC90.
