Citric Acid Production from Glucose and Pumpkin by Using Immobilized Bead of Aspergillus niger (고정화 Aspergillus niger Bead를 이용한 포도당과 호박으로부터 구연산 생성)

  • The spore of Aspergillus niger KCTC-6144 was immobilized on alginate gel beads. When pumpkin powder was used with glucose for a medium of citric acid fermentation by Aspergillus niger beads, the beaded Aspergilus niger grew up inside the bead and mycelia penetrated through the pore of the bead membrane. The bead size became largely from 2.0∼2.5mm to 6∼8mm after growing at 30$\^{C}$ for 4 days. Studies of optimum culture conditions on citric acid fermentation using Aspergillus niger beads on pumpkin medium (pumpkin powder 1% +glucose 7%, pH 6.0) were carried out in submerged cultures on 250m1 Erlenmeyer flask. As a result, it was found that to reinforce 12% as carbon source was good for citric acid production and that 1% pumpkin powder was good as nitrogen and mineral source in orbital shaker (150rpm) at 30$\^{C}$ for 5 days. The optimum initial pH on citric acid production was pH 6.0 and it was found that 100 beads of immobilized Aspergillus niger was adequate for citric acid production in a 250ml Erlenmeyer flask containing 50m3 of pumpkin medium solution with orbital shaker at 30$\^{C}$ for 5 days. We also found that maximal production of citric acid was 23.5g/ℓ at optimal condition (at 30$\^{C}$ for 5 days, pH 6.0, and 100 beads and medium containing 1% pumpkin powder plus 12% glucose).

Utilization of Egg-shell for Bread-making (제빵시 난각의 이용에 관한 연구)

  • This study was conducted to investigate whether egg-shell may be used as a mineral sourceor leavening agent in breadmaking. In Korea the waste volume of egg shell has been estimated at about 28,694 tons per year. Carbon dioxide generation maxima were established for barking powder$(153{\pm}3ml/g)$, egg-shell(205in reaction with lactic acid) and yeast$(115{\pm}3ml/sugar\;g)$. Gas release time required for each substance to reach $CO_2$ maximum was, for baking powder 7 minutes, for egg-shell 45 mins and for yeast 240 mins. Particle size of egg-shell in breadmaking was suitable more than 20 mesh (-). When egg-shell only was added to the basic formular without including lactic acid, no leavening effect was observed. However, when lactic acid and egg-shell were used together, the leavening effect was more or less equivalent to that of yeast(control). Addition of egg-shell was found to increase calcium content of bread products without noticeable altering flavor, as compared with control. Joint use of egg-shell was organic acids in breadmaking was shown to have potential in time saving, volume increase and yeast saving.

Chemical Characteristics in Fermented Cordycepin-enriched Cordyceps militaris (발효 동충하초의 이화학적 특성)

  • The comparative effects of fibrinolytic, antioxidative activity and electrophoretical protein patterns with Cordyceps militaris powder (CM) and cordycepin-enriched Cordyceps militaris JLM0636 powder (CCM) and fermented cordycepin-enriched Cordyceps militaris JLM0636 powder by several microscopic organisms were investigated. In addition, nutritional materials such as mineral, protein, and fatty acids were also measured. The protein concentration was higher in CCM than that in CM. The protein concentration in fermented CCM was the highest in CCM treated by Aspergillus kawachii among the various samples. When total protein patterns of CM, CCM, and both fermented CCMs were analyzed by native- and SDS-PAGE, there were slightly varietal differences in electrophoretical protein patterns. Major minerals were K, Ca, Mg, and Zn. Major fatty acids were palmitic acid, stearic acid, oleic acid, linoleic acid, and linolenic acid. Fibrinolytic activity was the highest in the fermented CCM by Bs treatment among the various samples. The ${\alpha},{\alpha}^{\prime}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radical scavenging activity was slightly stronger in the CCM treated with Aspergillus kawachii among the various samples; however, these samples all exhibited relatively low levels of activity compared with the butylated hydroxytoluene (BHT). These results may provide the basic data necessary to understand the biological activities and chemical characteristics of Cordyceps militaris JLM0636 powder fermented by several microscopic organisms to develop functional foods.

Optimal Abrasion Conditions for Separating Aggregate and Cement paste for Using Waste Concrete Fine Powder as Decarbonization Raw Material (폐콘크리트 미분말을 탈탄산 원료로 사용하기 위한 골재와 시멘트페이스트 분리의 최적 마쇄 조건 분석)

  • In this study, we attempted to reduce CO2 generated during manufacturing by replacing limestone (CaCO3), a carbonate mineral used to produce cement clinker, with a decarbonated raw material to which CO2 is not bound. The raw material for decarbonization was cement paste attached to waste concrete, among various industrial by-products. Waste concrete has cement paste adhered to the aggregate, which cannot be separated efficiently by general crushing and grinding methods. Peeling and grinding methods effectively remove only the cement paste without damaging the original aggregate. The abrasion time, steel ball type, and steel ball ratio were selected as effective factors for Abrasion. An optimal abrasion experiment was conducted to produce waste concrete fine powder containing decarbonated CaO as a cement clinker raw material through an experimental design method. The experiment revealed that the optimal conditions for producing waste concrete fine powder were an abrasion time of 7 minutes, a steel ball size for pulverization of 8 mm, and a steel ball ratio for pulverization of 0.6.

Comparison of the Effects of Deer Antler, Old Antler, and Antler Glue on Osteoporosis in Ovariectomized Rats

  • Background: Examination of the effects of deer antler, old antler, and antler glue on postmenopausal osteoporosis in an ovariectomized Sprague-Dawley rat model. Methods: The study involved 7 experimental groups; SHAM (sham-operated rats), OVX (ovariectomized rats), E2 (ovariectomized rats with estradiol $10{\mu}g/kg$ daily, orally), DA (ovariectomized rats with deer antler extract 5.83 mg/kg), OA (ovariectomized rats with old antler extract 3.8 mg/kg), low-AG (ovariectomized rats with low dose of antler glue powder 12.5 mg/kg), high-AG (ovariectomized rats with high dose of antler glue powder 37.5 mg/kg). After 6 weeks of treatment, body weight, blood calcium, phosphorus, estradiol, liver [alkaline phosphatase (ALP), aspartate transaminase (AST), alanine transaminase (ALT)] and kidney [blood urea nitrogen (BUN)/creatinine ratio] function, and femoral bone mineral density (BMD) were measured. Results: The body weights of DA, OA, low-AG, and high-AG groups did not significantly differ from OVX group. Blood estradiol levels were significantly increased in the DA, low-AG, and high-AG groups compared to the OVX group. Blood calcium, phosphorus, ALP, AST, and ALT levels and BUN/creatinine ratio did not show significant changes in the DA, OA, low-AG, and high-AG groups. BMDs of the femur, and femoral head and neck were significantly increased in the low-AG group. In the OA group, the BMD of the femoral head and neck was significantly increased. Conclusion: Treatment with deer antler, or antler glue for 6 weeks was effective for increasing estradiol and femoral BMD in ovariectomized rats, suggesting that this may be of therapeutic benefit for osteoporosis.

  • This study was carried out for analyzing pathological and epidemiological factors of osteoporosis and doing pilot test using trial compounds of tuna bone and oriental herbs based on the factors. Osteoporosis is originated from osteoblast, osteoclast, organic and inorganic factors etc. Therefore the pathology of osteoporosis is not simple because the cytokine, growth factors and hormones of the components are various a lot. Taking a view of epidemiological factors of type I osteoporosis, ageㆍmenarcheㆍcholesterolㆍBMI etc. have definite relation to them. So we can approach to aging or consumptive disease in oriental medicine, specifically differential diagnosis of blood depletion with deficiency of qi, deficiency of kidney, deficiency of yin, bony weakness etc. And it should be considered together with rules for maintaining good health or habit concomitantly. Therefore IL1ㆍ6 or TNF αㆍβ are generally used as molecular biological index for osteoblast and osteoclast because the most important index is bone mineral density and strength, but the factors like collagen and noncollagen protein must be accounted as biomarkers. Trial compounds generally showed favorable effects on accompanying subjective symptoms of osteoporosis in the pilot test for menopausal woman. But if she didn't have specific symptoms of osteoporosis there wasn't any specific change. And osteocalcin was increased in case of being under standard level, but wasn't changed in case of normal level. Therefore these trial compounds can be used as a funcdonal diet for type I osteoporotic patients or preventive measures.

Leaching behavior of Ga and In from MOCVD dust (MOCVD 더스트로부터 Ga과 In의 침출 거동)

  • Leaching of MOCVD dust in the LED industry is an essential stage for hydro-metallurgical recovery of pure Ga and In. To recover Ga and In, the leaching behavior of MOCVD scrap of an LED, which contains significant amounts of Ga, In, Al and Fe in various phases, has been investigated. The leaching process must be performed effectively to maximize recovery of Ga and In metals using the most efficient lixiviant. Crystalline structure and metallic composition of the raw MOCVD dust were analyzed prior to digestion. Subsequently, various mineral acids were tested to comprehensively study and optimize the leaching parameters such as acidity, pulp density, temperature and time. The most effective leaching of Ga and In was observed for a boiling 4 M HCl solution vigorously stirred at 400 rpm. Phase transformation of GaN into gallium oxide by heat treatment also improved the leaching efficiency of Ga. Subsequently high purity Ga and In can be recovered by series of hydro processes.

A study on the effects of fine sludge powder addition on portland cement-limestone sludge system (포틀랜드시멘트-석회석슬러지계에서의 슬러지 미분말첨가반응 효과에 대한 연구)

  • We tested the limestone sludge produced in Pohang Iron & Steel Co., Ltd. as a filler powder for the effective use of portland cement. Hydration process was investigated by measuring the hydration rate, the amounts of non-evaporable water and compressive strength of cement-limestone sludge paste prepared by mixing limes-tone sludge with cement. The results obtained in this study can be summarized as follows: 1. There is no significant difference between the cases of adding up to 10% limestone sludge and those of unmixed cement system. However the reaction rate increases in the 5% limestone sludge system(due to the effects of fine). 2. The compressive strength increases proportionally with increasing the measured amount of non-evaporable water, Adding 5% limestone sludge also increases the strength a little higher, and the compressive strength and calcium silicate hydrates. In the case of the mixed limestone sludge, $2\theta$=$11.7^{\circ}$ peak appears in the samples of 28 days hydration. This peak indicted the presence of calcium carboaluminate hydrate. Although limestone sludge is generally regarded as a inert materials, some kinds of cement can produce a calcium carboaluminate by reacting with aluminate in cement pastes.

Characteristics of Leaves, Roots, and Fruit as Influenced by Energized-Functional Water Supply in Fuji Apple Trees (Energized 기능수 처리에 따른 후지사과의 잎, 뿌리 및 과실특성)

  • Energized-functional water (EFW) and powder (EFP) were manufactured by Kyungwon Institute of Life Science, Seoul, through a series of processes; tap water ultra-purification energy imprinting with catalysts in platinum columns mixing energy-imprinted water + activated zeolites + photosynthetic bacteria fermenting at $25^{\circ}C$ filtering EFW and/or EFP. A single application of EFP to soil under tree canopy before bud burst, combined with three EFW applications to soil during growth of 'Fuji' apples (Malus domestica Borkh.) resulted in a higher Ca concentrations in fruit skins and flesh, and lower Ca and N concentrations in leaves and shoot-bark tissues. EFW also stimulated the net photosynthesis of leaves and root activity. Soluble solid concentrations (SSC) and anthocyanin levels of fruits were also significantly increased at harvest, producing greater firmness and less core browning during storage at $0^{\circ}C$. However, there was no significant difference in titratable acidity of fruit juice between the EFW treatment and the controls.

Nutrients and antioxidant activity of Diospyros lotus L. (고욤나무 열매의 영양성분 및 항산화 활성)

  • This study was conducted to determine whether Diospyros lotus L. fruit could be used as a natural functional food ingredient and whether its use should be increased. D. lotus fruit powder was found to have high acidity in the immature IDL stage and significantly higher sugar content in the mature MDL stage. The levels of crude fat, selenium, and tannins decreased significantly from the IDL stage to the MDL stage in D. lotus fruit powder. However, the levels of magnesium and calcium were significantly greater in the MDL stage than in the IDL stage (p<0.05). The total phenol content was significantly higher in the IDL stage, but the DPPH radical scavenging activity significantly increased in the MDL stage (p<0.05). In conclusion, the fruits of D. lotus. have high nutritional quality in both the immature and mature growth stages. Thus, it is a good candidate for use as a functional food, and its use should be increased because of its high nutritional qualities.
