The purpose of this study was to improve the production of Mozzarella cheese analogues manufactured using mixtures of soy milk and concentrated raw milk by performing ultrafiltration (UF) and to assess the quality of these cheeses during a 30-day storage period at $4^{\circ}C$, relative to that of Mozzarella cheese manufactured with the traditional method. The solid consistency of Mozzarella cheese analogue prepared from milk mixtures was lower than that of cheese manufactured from raw milk or soy milk and increased during storage, which is considered to be the result of decreasing water levels, as well as with increasing soy milk concentrations. In the Mozzarella cheese analogue generated using the milk mixtures, the fat content decreased with increase in the soy milk concentration, while it decreased during the storage period. Lactose levels were lowest in cheese composed of soy milk or raw milk and processed by UF, and decreased during storage in cheese produced using milk mixtures. In milk mixtures containing soy milk, the protein concentration increased with increasing amounts of raw milk and did not change during the storage period. The water-soluble nitrogen compound level was similar between cheeses and increased only slightly during storage. The amount of non-protein nitrogen compounds was higher in the cheese analogue than in the control cheese and tended to increase during storage. Analysis of the physicochemical traits of the Mozzarella cheese analogue yielded the following results: During storage, titratable acidity levels increased while pH tended to decrease. After analysis using electropherograms, it was classified as ${\alpha}$-, ${\beta}$-, or ${\kappa}$-casein. The results of rheometry tests showed that in the Mozzarella cheese analogue prepared from milk mixtures, with raw milk concentrated by UF, increases in concentration rate lead to lowered hardness, elasticity, cohesiveness, and brittleness. When cheese was produced from milk mixtures and concentrated by UF, meltability increased as the concentration rate increased, although to an extent that was less than that observed for the control cheese, and tended to increase during storage. Sensory evaluation showed that the analogue cheese was much better than the control cheese in terms of formation, appearance, and flavor.
A fine-mapping method exploiting linkage disequilibrium was used to detect quantitative trait loci (QTL) on the X chromosome affecting milk production, body conformation and productivity traits. The pedigree comprised 22 paternal half-sib families of Black-and-White Holstein bulls in the Netherlands in a grand-daughter design for a total of 955 sons. Twenty-five microsatellite markers were genotyped to construct a linkage map on the chromosome X spanning 170 Haldane cM with an average inter-marker distance of 7.1 cM. A covariance matrix including elements about identical-by-descent probabilities between haplotypes regarding QTL allele effects was incorporated into the animal model, and a restricted maximum-likelihood method was applied for the presence of QTL using the LDVCM program. Significance thresholds were obtained by permuting haplotypes to phenotypes and by using a false discovery rate procedure. Seven QTL responsible for conformation types (teat length, rump width, rear leg set, angularity and fore udder attachment), behavior (temperament) and a mixture of production and health (durable prestation) were detected at the suggestive level. Some QTL affecting teat length, rump width, durable prestation and rear leg set had small numbers of haplotype clusters, which may indicate good classification of alleles for causal genes or markers that are tightly associated with the causal mutation. However, higher maker density is required to better refine the QTL position and to better characterize functionally distinct haplotypes which will provide information to find causal genes for the traits.
The effects of diacylglycerol O-acyltransferase (DGAT1) candidate gene polymorphism on the economic traits of Hanwoo were studied. Through sequencing analysis, two polymorphism sites at K232A and T11993C were established and were analyzed by PCR-RFLP. The PCR-RFLP analysis for K232A showed that the frequencies of alleles K and A were 0.75 and 0.25, respectively, and the frequencies of genotypes for K/K, K/A and A/A were estimated as 0.509, 0.491 and 0, respectively. In the PCR-RFLP analysis for T11993C, we found allele frequencies of 0.773 and 0.227 for T and A, respectively, and 0.546, 0.454 and 0 for the T/T, T/C and C/C genotype frequencies, respectively. No significant effects on economic traits in Hanwoo were found in the separate analysis of K232A and T11993C polymorphisms, but the interaction between K232A and T11993C showed a significant effect (p<0.005) on marbling score. The DGAT1 candidate gene was found to have a significant effect not only on milk yield and component traits but also on the metabolism of intramuscular fat.
Objective: Water buffalo, an important domestic animal in tropical and subtropical regions, play an important role in agricultural economy. It is an important source for milk, meat, horns, skin, and draft power, especially its rich milk that is the great source of cream, butter, yogurt, and many cheeses. In recent years, long noncoding RNAs (lncRNAs) have been reported to play pivotal roles in many biological processes. Previous studies for the mammary gland development of water buffalo mainly focus on protein coding genes. However, lncRNAs of water buffalo remain poorly understood, and the regulation relationship between mammary gland development/milk production traits and lncRNA expression is also unclear. Methods: Here, we sequenced 22 samples of the milk somatic cells from three lactation stages and integrated the current annotation and identified 7,962 lncRNA genes. Results: By comparing the lncRNA genes of the water buffalo in the early, peak, and late different lactation stages, we found that lncRNA gene lnc-bbug14207 displayed significantly different expression between early and late lactation stages. And lnc-bbug14207 may regulate neighboring milk fat globule-EGF factor 8 (MFG-E8) and hyaluronan and proteoglycan link protein 3 (HAPLN3) protein coding genes, which are vital for mammary gland development. Conclusion: This study provides the first genome-wide identification of water buffalo lncRNAs and unveils the potential lncRNAs that impact mammary gland development.
The breeding data relating to Hariana herd spread over 18 years (1979-96) were analysed to estimate genetic, phenotypic and environmental changes in characters of economic importance which might have taken place during the several years of selective breeding practiced in the herd. The average genetic changes in a given character were estimated by four methods. The phenotypic trends observed for different economic traits were not significant. On changing the method of estimation, magnitude and direction of genetic trends changed. Comparison of estimates of genetic trends by different methods showed that adjustments for biases due to non-random allotment of dams with respect to their age and merit suggested by Powell and Freeman (1974) were useful for increasing the precision of the estimates. Hence, this method was found to be the best method for estimation of genetic trends. The estimate of genetic trends by this method were 4.03${\pm}$6.21 days, 3.24${\pm}$5.33 kg, 0.15${\pm}$0.43 days, 0.09${\pm}$0.59 days, 0.01${\pm}$0.02 kg and 0.01${\pm}$0.01 kg for age at first calving, first lactation milk yield, first lactation length, first calving interval, first lactation milk yield per day lactation length and first lactation milk yield per day of calving interval, respectively.
The experiment was conducted on 69 cows to identify the quantitative variations of some economic traits of five genetic groups as $Local{\times}Friesian $$F_1$, $Local{\times}Sahiwal$$F_1$, Local (indigenous zebu type), $Local{\times}Sindhi $$F_1$, and $Local{\times}Hariana$$F_1$. The traits studied were age at weaning, age at first heat, age at first conception, services per conception, daily milk yield, lactation length, lactation yield and post partum heat period. The records on milking and reproduction performances of cows and heifers were obtained from farm register and by interviewing the farmers. It was observed that the lowest age at weaning, age at first heat and age at first conception were $5.37{\pm}0.24$, $27.17{\pm}1.72$ and $27.83{\pm}1.82$ months respectively in $Local{\times}Hariana$$F_1$. Services per conception were lowest in $Local{\times}Sahiwal$$F_1$$(1.08{\pm}0.18)$ although not significantly (p>0.05) affected by farms, genetic groups and $farm{\times}genetic$ groups interaction. Average daily milk yield was highest in $Local{\times}Friesian$$F_1$$(5.81 {\pm}0.40 kg)$. Lactation length and lactation yield were highest in $Local{\times}Sahiwal$$F_1$ ($299.38{\times}9.74$ days and $1863.00{\pm}141.00kg $ respectively). Average post partum heat period was lowest in $Local{\times}Sindhi$$F_1$ ($3.19{\pm}0.38$ months). Least squares ANOVA showed that farm had significant (p<0.001) effect on age at weaning, age at first heat, age at first conception and post partum heat period. There was insignificant (p>0.05) effect of farm on services per conception, daily milk yield, lactation length and lactation yield, whereas genetic groups had a significant effect for all the traits under review except services per conception. $Farm{\times}genetic$ groups interaction was insignificant for all of the traits under consideration except age at weaning.
Objective: This study was conducted to examine the effects of a mixture of pinecone oil, garlic, and brown seaweed extracts (PGBE) on milk production traits as well as physiological and ethological parameters in Holstein cows during the summer season (24 May to 03 July 2015, Korea). Methods: Among the extract combinations tested, we found that the level of 2,2'-azino-bis (3-ethylberzothiazoline-6-sulphonic acid) cation radical scavenging activity of the 0.16% PBGE complex at ratio of 1:1:1 (vol/vol) was comparable to that of the control (ascorbic acid; 1 mg/mL). Additionally, the PBGE complex reduced lipopolysaccharide-induced COX-2 expression in bovine mammary epithelial cells. Based on these findings, 40 lactating Holstein cows were used to measure the effects of PBGE complex at ratio of 1:1:1 (vol/vol) on milk production, immune response, metabolites, and behavior patterns by dividing the cows into two groups fed diets containing PGBE complex (n = 20; 0.016%/kg feed dry matter basis) or not containing PGBE complex (control, n = 20) for 40 d. Results: Results showed that PGBE complex did not influence milk composition, eating and ear surface temperature patterns, immune response, or metabolic parameters but promoted average milk yield throughout the experimental period. Additionally, a tendency of higher total antioxidant capacity and glutathione in the PGBE group was observed compared to the those in the control. When the temperature-humidity index (THI) exceeded 72 (average THI = 73.8), PGBE complex-fed cows experiencing heat stress showed increased milk yield and a tendency of increased rumination compared to the control. Conclusion: We suggest that incorporation of a combined mixture of 0.016% PGBE (1:1:1 ratio, vol/vol) to diet has the potential to improve milk yield and health status of cows under mild to moderate heat stress, denoting that it might be useful as an alternative anti-stressor in the diet of dairy cows under hot conditions.
Data were collected on productive and reproductive performance of different $F_1$ crossbred cows (Deshi cows joined by A.I. to Sindhi, Sahiwal, Jersey or Holstein-Friesian Bull semen) reared by farmers in and around the Bangladesh Agricultural University campus at Mymensingh. The traits considered were birth weight, age at first heat, age at first calving, gestation length, post-partum heat period, milk yield, lactation length, dry period and calving interval. Data were analyzed statistically using a completely randomized design and Duncan's Multiple Range Test to compare and determine the significance of between individual group means. It was observed that the Holstein $\times$ Deshi had the highest birth weight (21.4 kg) followed by Sahiwal (17.6 kg), Jersey (16.7 kg) and Sindhi (16.1 kg). Shortest age at first heat (854 and 920 days) and age at first calving (1002 and 1201 days) were found for the Jersey $\times$ Deshi and Holstein $\times$ Deshi crosses respectively. The post-partum heat period were significantly different between breeds with Jersey and Holstein sired calves having the shortest intervals. There was no difference in the gestation length of the four crosses. Holstein $\times$ Deshi and Jersey $\times$ Deshi cows gave the highest milk yields due to longer lactation periods and higher daily milk production. Similarly, Holstein $\times$ Deshi and Jersey $\times$ Deshi crosses had significantly shorter dry periods and Jersey $\times$ Deshi had a shorter calving interval. From this study it may be concluded that exotic genotypes such as Holstein-Friesian and Jersey can perform very well under rural conditions in Bangladesh.
The present study analysed the various productive and reproductive characteristics of 1/2 Pabna Milking Cows (PMC)-1/2 Sahiwal (S) and 1/4 PMC-3/4 S collected from the Pabna milkshed area at Baghabarighat, Sirajgonj, Bangladesh. The studied traits were birth weight (BWT), age at puberty (AP), number of services per conception (NSC), post partum heat period (PPHP), gestation period (GP), daily milk yield (DMY), lactational production (LP), lactation length (LL), fat percentage and solids-not-fat percentage (SNFP). Five individual Sahiwal sires were used for the upgrading of Pabna Milking Cows. The used data covered from 1987 to 1994. Least-squares analysis of variance showed that genetic group had a significant effect on BWT (P < 0.05), AP (p < 0.01), DMY (p < 0.01), LP (p < 0.001), LL (p < 0.05), FP (p < 0.05) and SNFP (p < 0.01). Genetic group had non-significant effect on NSC, GP and PPHP. The effect of sire was significant on BWT (p < 0.01), NSC (p < 0.01), LP (p < 0.05) and LL (p < 0.05). The AP, NSC, DMY, LP, LL and SNFP were higher in 1/2 PMC-1/2S cows; BWT and PPHP were higher in 1/4 PMC-3/4S but GP and FP were almost same in both genetic groups. From this study it may be concluded that production and use of 1/2 PMC-1/2S would seem more profitable for commercial milk production in the Bangladesh Milk Producers' Cooperative Union Limited (BMPCUL) area and at the same time emphasis should be given on rigorous sire selection.

The objective of this study was to estimate the environmental factors affecting milk ${\beta}$-hydroxybutyric acid (BHBA) and acetone (Ac) concentrations in Holstein cattle. A total of 264,221 test-day records collected from the Korea Animal Improvement Association (KAIA) during the period of 2012 to 2014 were used in this study. Analysis of variance (ANOVA) was performed to determine the factors significantly affecting ketone body concentrations. Parameters considered in the model were season of test, season of calving, parity, lactation stage, and milk collecting time (AM and PM). According to the ANOVA, the $R^2$ for milk BHBA and Ac were 0.5226 and 0.4961, respectively. 'Season of test' showed a considerable influence on ketone body concentration. Least square (LS) means for milk BHBA concentrations was the lowest ($39.04{\mu}M$) in winter while it increased up to $62.91{\mu}M$ in summer. But Ac concentration did not significantly change along with 'season of test'. The means of milk BHBA and Ac concentrations were high at first lactation stage, low around second lactation stage, and then gradually increased. Cows milked in the morning had lower mean BHBA and Ac concentrations ($48.49{\mu}M$ and $121.69{\mu}M$, respectively) in comparison to those milked in the evening ($53.46{\mu}M$ and $130.42{\mu}M$, respectively). The LS means of BHBA and Ac slightly increased over parities. These results suggest that proper maintenance of milk collection, herd management programs, and evaluation of ketone body levels in milk should be considered for the efficient management of resistance to ketosis.
