An intensive study of the embryology of Gymnospermium microrrhynchum was conducted to provide information regarding a discussion of the phylogenetic relationships of the genus, which is yet unstudied. Our results indicated that Gymnospermium is similar to other genera of Berberidaceae in terms of its embryological features. Nevertheless, newly reported and unique features are the well-developed endothelium and the undifferentiated seed coat type. Until the study of Gymnospermium, it may have been considered to be closer to Caulophyllum and Leontice in the tribe Leonticeae. These three genera share many morphological features as well as molecular similarities, by which they are kept in the same tribe, Leonticeae. However, very little detailed embryological data regarding these genera have been published thus far. Gymnospermium was characterized according to the basic type of anther wall formation as well as its glandular tapetum, successive cytokinesis in the microspore mother cell, two-celled mature pollen grains, anatropous and crassinucellate ovules with a nucellar cap, well-developed endothelium, its Polygonum type of embryo sac formation, its nuclear type of endosperm formation, and its undifferentiated seed coat type. In comparison with Nandina, there are many differences, such as the dehiscence of the anther, the cytokinesis in the microspore mother cells, the shape of the megaspore dyad, and the seed characteristics. Although we had no available detailed embryological information regarding Caulophyllum and Leontice, which are genera that are more closely related to Gymnospermium, we could deduce from the phylogenetic relationship that Gymnospermium, Caulophyllum, and Leontice are more closely related to each other than other genera of Berberidaceae on the basis of the seed characteristics.
The effect of several factors on efficiency of anther culture of broccoli was studied. The medium supplemented with 14% sucrose, 125mg/L of silver nitrate and 0.1mg/L of each of NAA and 2,4-D was best in embryogenesis in all varieties tested. The effect of auxin was affected by silver nitrate, that is, it lowered embryogenesis when the concentration of NAA and 2,4-D was low, but it enhanced when their concentrations were high. Benzyl aminopurine increased embryogenesis in some varieties, but decreased in others. Although microspore embryogenesis was observed in anthers of which heigest was 1 to 0.6 - 1.0 of petal heigest in the bud, which corresponds to the early uninucleate to early binucleate stage of the microspores, it was much better when the ratio of heigest was 1 : 0.8. More embryos ware inducted from culture in March than in February in general. Because there was a big yearly variation, it was assumed that microspore embryogenesis was influenced greatly by the culture condition and the physiological condition of donor plants.
Kimchi cabbage (Chinese cabbage), radish and Cabbage are major Brassicaceae vegetables in Korea. Especially, we can easily develop whole plant from one microspore in Kimchi cabbage. To develop clubroot resistant doubled-haploid (DH) inbred lines, we pollinated a clubroot resistant turnip of 'IT 033820' with a Kimchi cabbage (Chinese cabbage) inbred of 'BP 079'. More than 85 DH inbred lines were developed from this combination. We screened about 400 materials including these DH inbred lines, commercial cultivars and breeding materials during 3 years using hydroponic system after inoculating single spore isolation race 4(SSI-04) inoculate. One inbred line derived from this combination selected as clubroot resistant and registered as 'Wonkyo20036ho'. We inoculated 26 DH inbred lines derived from 'Zoong-baek 2ho' using SSI-4, the percent of resistant plants varied from 0 to 83%. However the horticultural traits of highly resistant DH inbred line was poor. Thus we selected one DH line showing 77% resistant with yellow inner leaf and maid good head, was registered as 'Wonkyo20034ho'. Another DH inbred line derived from Korean variety of 'Wol-dong' showing 86% resistant was registered as 'Wonkyo20037ho'. Other DH inbred lines were derived from Chinese cultivar of 'Choon-hi-go-hang-wang' and 'Hwang-shim-zo48' showed 80 and 71% resistant, respectively, was also selected for registration. Even though DH inbred lines derived from turnip showed highly resistant to SSI-04 and provincial inoculate, they showed poor characteristics in horticultural traits. However, commercial seed companies showed interesting for adapting these DH inbred lines in commercial breeding.

Ligusticum chuanxiong is receiving much attentions as one of the important medicinal crops with the increasement of the crude drug demands. This study was conducted to obtain the basic informations of breeding of Ligusticum chuanxiong. Embryological characteristics were examined to elucidate the process of male and female gametophytic development and fertilization. Meiosis and nucleus division of megaspore and microspore were proceeded normally. With regard to the formation of female gametophyte, only a half of female gametophyte developed to normal egg apparatus. While another 50% showed abnormal egg apparatus with poly-nuclei or non-nuclei ovule. The pollens developed from the micros pore were formed more than 90 % of normal pollen. It was difficult to observe fertilization because ovule tissue was very compact and cell was extremely tiny, but could be easely observed proembryo and embryo formation. Only 30 percent developed into proembryo and subsequently into embryo, and the others were degenerated.
To optimize the in uitro chromosome doubling procedure in anther cultures of rice, anthers were cultured on callus induction medium with 0.001 to 0.1 mg/L colchicine for 30 days. The addition of colchicine slightly reduced callus formation and plant regeneration in comparison to the colchicine-free medium (control medium). This reduction was greater with higher concentration colchicine. Microspore-derived rice plants in control medium were found to be mainly haploid (50 to 58.8%) and doubled-haploid (31 to 40%) in anther culture of 3 Japonica and 1 Tonsil type cultivars. The application of 0.001 mg/L colchicine was increased to 54.3∼60.0% in the frequency of fertile doubled-haploid plants. These results indicate that the addition of colchicine to the callus induction medium is an efficient means to obtain doubled-haploid plants in anther cultures of rice.
A survey was carried out on the cold-induced sterility of paddy rice in 1988 in the alpine area of Kangweon province when cold spell occurred during late July to early August, During this period minimum temperature as low as 8.5$^{\circ}C$ and 5.7$^{\circ}C$ was recorded at Dunnae and Jinbu, respectively. The rice cultivars, which encountered this cold spe]J at meiotic stage of microspore, were damaged by sterility in most alpine areas of higher than 300m in altitude. To secure spilkelet fertility higher than 80% it was estimated that the minimum and average air temperature accumulated during 13 days of meiotic stage should be higher than 2$25^{\circ}C$ and 285$^{\circ}C$, respectively and/or the duration of lower than 17$^{\circ}C$ in minimum temperature should not exceed fivedays during that period.
The ontogeny of pollen wall in Panax ginseng was studied with transmission and scanning electron microscopy from early tetrad stage until pollen maturity. Initial indication of exine development is undulation of plasma membrane for the preparation of bacular mound. The first recognizable structure of the pollen wall is the cellulosic primexine which is formed outside of the plasma membrane while microspore tetrads are still surrounded by callose wall. As development proceeds, foot-layer and baculum differentiation, callose dissolution and exine formation were progressed. During this process, sporopollenin is deposited into the exine, and then endexine development was followed. The intine, innermost pollen wall layer, is developing form hypertrophic Golgi vesicles. The thickness of exine is very even on all along the pollen wall, but intine thickness of apertural region is thicker than that of nonapertural region. Mature pollen of ginseng is $20{\mu}m$ in size, tricolpate and shows fine reticulate sculpturing.

Unreduced (2n) gametes are meiotic products (pollen or egg) having a sporophytic (somatic) chromosome number, resulting from abnormalities during either microsporogenesis or megasporogenesis. They occur naturally at a low frequency in many plant species. Unreduced (2n) gametes in plants can be identified for four possible ways as follow i) pollen size and/or shape differences between haploid (n) and diploid (2n) pollen, ii) ploidy analysis (chromosome number) of progeny or meiotic analysis (presence of dyads andlor triads at the microspore stage), iii) progeny performance and fertility and iv) dosage of isozyme and DNA markers. Unreduced (2n) gametes can be an effective breeding tool in synthesizing new cultivars, providing a unique method to maximizing heterozygosity, i.e., transferring a large proportion of the non-additive genetic effects (intra- and inter- locus interactions) h m parent to offspring and can also be used to overcome infertility of interploidy crosses. Sexual polyploidization through 2n gametes has been a major route to the formation of naturally occurring polyploids. The three mechanisms of 2n pollen formation in potato have been discovered as follow: i) parallel spindles (ps) or tripolar spindles (ts), ii) premature cytokinesis (pc-I, pc-2) and iii) synaptic mutants (sy-2, sy-3, sy-4). Genetic analysis indicated that the mechanisms of 2n gamete formation were controlled by single recessive gene in potato, alfalfa, red clover, etc., and by two recessive genes in wheat. The use of 2n gametes which can efficiently transfer germplasm fiom wild relatives to cultivated species, especially fiom diploid to tetraploid could make a contribution to the improvement of germplasm base in breeding programs.
Present experiment has been carried out in order to make clear the abnormalities of the female gametophyte formation and its relation to fertility, using the short-style of F. koreana, the results of which are summarized as follows : (1) Anatropous ovule has single integument with thick cell-layer and tiny nucellus consisting of nucella-epidermis and megaspore mother cell. (2) Meiotic division of megaspore mother cell takes place around middle or latter part of March, while that of microspore mother cell occurs from the end of September to the beginning of October. (3) Megaspore mother cell stage is long, and ranges from October to March next year. (4) Formation of mature embryo sac is not completed until the beginning of May, approximately one month after blooming. (5) Normal embryo sac is rare, most of the nucellus being devoid of embryo sac.
xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed.
