These experiments were performed to investigate the safety of two herbs-Houttuynia cordata Thunberg and Lycium chinese Miller-irradiated with gamma-rays in respect of genotoxicity. Water extracts from the 10 kGy gamma-irradiated herbs were examined in two short -term in vitro tests ; (1) Salmonella typhimurium reversion assay (Ames test) in strain TA 98 and Ta100 and (2) Micronuclues test on clutured Chinese hamster ovary(CHO) cells. No mutagenicity was detected in the two assays with or without metabolic activation . From these results , the safety of the herbs irradiated with gamma-rays at practical doses could be revealed in further tests of genotoxicity in vivo, chronic and reproductive toxicity.
The ethanol extracts of the mixed vegetables (Bioactive Vegetables, BV) and the mixed fruits (Bioactive Fruits, BF) were evaluated for their in vivo antioxidant activities. Four weeks treatment of oral administration was performed to mice. A $KBrO_3$ as a potent oxidant was used to induce the oxidative stress for in vivo experiment. BV and BF were shown to possess the significant inhibitory effect of lipid peroxidation as measured by the level of malondialdehyde (MDA) formation although the potencies were not higher than those of well-known antioxidants such as vitamin C, trolox and quercetin. Furthermore, BV and BF inhibited DNA damage assessed by single cell gel electrophoresis (comet assay) and reduced the micronucleated reticulocyte (MNRET) formation of peripheral blood. Antioxidants tested also revealed potent inhibitory activities higher than BV and BF. These antigenotoxic activity profiles were similar to that of abovementioned inhibition of lipid peroxidation. Therefore, BV and BF having mild antioxidant activity as functional food candidates may be useful natural antioxidants by the inhibiting of lipid peroxidation and the protecting oxidative DNA and chromosomal damage.

This study was designed to demonstrate the antigenotoxic potential of methyl alcohol extracts from Auricularia mesenterica and Gyrophora esculenta against the frequency of micronucleate polychromatic erythrocyte(MNPCE) produced by benzo($\alpha$) pyrene in vivo. We used the mouse bone marrow test system to measure the effect of single and multiple treatments of each sample. Genotoxicity of benzo ($\alpha$) pyrene(150mg/kg, i.p.) as positive control was the highest at 36 hours. However, each sample per dose was not genotoxic, showing MNPCE values in the range of the control level. Treatments of methyl alcohol extracts both of Auricularia mesenterica and Gyrophora esculenta showed significant decreased frequencies of NMPCE induced by benzo($\alpha$) pyrene within 12 hours by single treatment(100mg/kg, oral). And also, the MNPCE level produced benzo($\alpha$) pyrene was decreased by the treatment of benzo($\alpha$) pyrene(5 to 200mg/kg, oral) of each sample, but significantly different redults were obtained with 100mg/kg. In the multiple treatment, the highest antigentoxic effects were demonstrated with 20mg/kg in the each sample, a range which induced inhibition indices of 54.2 and 56.3%, respectively.
The present work was conducted to examine the effect of tobacco smoke solution, and its components such as nicotine and catechol treated separately on the root tip cells of Allium cepa L The mitotic abnormalities in root tip cells treated with those three components were increased linearly, while the mitotic index was decreased following concentrations of the components. Chromosomal abnormalities showed 29.7%, 15.8% and 13.0% in the treatment of tobacco smoke solution, nicotine, and catechol, respectively. The abonormalities were included polyploid, c-mitosis, stickness at metaphase, laggard, bridge formation at anaphase or telophase, and micronucleus, abnormal nucleus at interphase, after the treatment of the components for 24 hours. hours.
The ethanol extract of the peony root (Paeonia Lactiflora Pall, Paeoniaceae) as well as its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. In addition, the protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were examined. The ethanol extracts of the peony root (PREs) and its active constituents, gallic acid and methyl gallate, exhibited a significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and had an inhibitory effect on lipid peroxidation, as measured by the level of malondialdehyde (MDA) formation. The PREs did not have any pro-oxidant effect. They strongly inhibited the hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, as assessed by single cell gel electrophoresis. Furthermore, the oral administration of 50% PRE (50% ethanol extract of peony root), gallic acid and methyl gallate potently inhibited the formation of micronucleated reticulocytes (MNRET) in the mouse peripheral blood induced by a $KBrO_3$ treatment in vivo. Therefore, PREs containing gallic acid and methyl gallate may be a useful antigenotoxic antioxidant by scavenging free radicals, inhibiting lipid peroxidation and protecting against oxidative DNA damage without exhibiting any pro-oxidant effect.

Three Loxodes ciliates collected from estuarine littoral, wetland and small pond in Korea, were identified as Loxodes kahli Dragesco and Njin$acute{e}$, 1971, L. magnus Stokes, 1887 and L. vorax Stokes, 1885. The descriptions for these species based on living and protargol impregnated specimens were given. Morphometry, illustrations and microphotographs were also provided. Diagnoses of three species are as follows. Loxodes kahli; size in vivo $160-300{\times}40-70\;{\mu}m$; oral area with reddish to brownish pigments; 6-11 macronuclei arranged linearly; 5-9 micronuclei located near macronuclei; 4-12 M$\ddot{u}$ller's vesicles; somatic kineties on right 18-20 and left 2 in number. L. magnus: size in vivo $250-470{\times}87-15\;{\mu}m$; body colored dark brown; 5-13 macronuclei; 8-13 micronuclei; 8-18 M$\ddot{u}$ller's vesicles; somatic kineties on right 23-26 and left 2 in number. L. vorax: size in vivo $70-160{\times}20-35\;{\mu}m$; oral area with brownish pigments; 2 macronuclei; 1 micronucleus located between macronuclei; 2-4 M$\ddot{u}$ller's vesicles; somatic kineties on right 18-20 and left 2 in number.
DA-3002, an authentic recombinant human growth hormone(rhGH), was examined for mutagenicity in the reverse mutation test on bacteria, in the chromosomal aberration test on cultured mammalian cells and in the micronucleous test on mice. The reverse mutation test was performed by a plate incorporation method with or without a metabolic activation system(S9 Mix) using Salmonella typhimurium strain TA100, TA1535, TA98 and TA1537. DA-3002 did not significantly increase revertant colonies in any of the test strains under any conditions at dose levels ranging from 0.0125 to 0.4 IU/plate, compared with the vehicle control. In the chromosomal aberration test using cultured Chinese hamster lung(CHL) cells, DA-3002 did not increase the number of aberrant cells in the presence or absence of S9 mix at concentrations of 0.0125 IU/mι to 0.4 IU/mι, compared with the vehicle control. In the micronucleus test, male ICR mice were given DA-3002 intraperitoneally at a dose level of 20, 40 and 80 lU/kg. The incidence of bone marrow micronucleated polychromatic erythrocytes in the DA-3002 treated mice did not differ from that of the vehicle control. These results indicate that DA-3002 doesn't have mutagenic potential under the present test conditions.

1,2-Dibromoethane(DBE) has been widely used as a soil fumigant, an additive to leaded gasoline and an industrial solvent. In this study, we have carried out in vitro genetic toxicity test of 1,2-dibromoethane and microarray analysis of differentially expressed genes in response to 1,2-dibromoethane. 1,2-Dibromoethane showed mutations in base substitution strain TA1535 both with and without exogenous metabolic activation. 1,2-Dibromoethane showed mutations in frame shift TA98 both with and without exogenous metabolic activation. 1,2-Dibromoethane showed DNA damage based on single cell gel/comet assay in L5178Y cells both with and without exogenous metabolic activation. 1,2-Dibromoethane increased micronuclei in CRO cells both with and without exogenous metabolic activation. Microarray analysis of gene expression profiles in L5178Y cells in response to 1,2-dibromoethane selected differentially expressed 241 genes that would be candidate biomarkers of genetic toxic action of 1,2-dibromoethane.

Tritium is an important nuclide that must be monitored for radiation safety management. In this study, HTO was orally administered to rats at the level of 37 kBq ($1{\mu}Ci$) or 370 kBq ($10{\mu}Ci$) to examine tissue distribution and excretion levels. After sacrifice, wet and dry tissue samples were weighed and analyzed for tissue free-water tritium (TFWT) and organically bound tritium (OBT). The mean tissue concentrations of TFWT (OBT) were 30.9 (17.8) and 4.4 (8.1) Bq/g on days 7 and 13 at the 37 kBq level and 30.8 (64.6) Bq/g on day 17 at the 370 kBq level. To assess the cytogenetic damage due to tritium exposure, a cytokinesis-blocked micronucleus (MN) assay was performed in blood samples from rats exposed to HTO for 14 and 21 days after oral administration. There was no significant difference in the MN frequencies between the control and exposed rats.
Two ciliates of suborder Euplotina collected from the two habitats (estuarine littoral and sewage treatment plant) in Ulsan, Korea were Euplotes charon ($M\ddot{u}ller$, 1773) and Diophrys oligothrix Borror, 1965. These two species are reported for the first time from Korea. The description was based on the observation of living and silver impregnated specimens. Diagnostic characteristics of these species are as follows. E. charon: size in vivo about $90-130\times65-80{\mu}m$, adoral zone of membranelles over 79.5% of cell length with 54-80 adoral membranelles; right margin of the peristome shaped sinusoidal form and passed through adoral zone of membranelles; buccal cavity wide anteriorly; 10 frontoventral, 5 transverse, 4 caudal cirri, 12 dorsal kineties, mid-dorsal kinety with 21 -25 dorsal bristles; silver-line system double-eurystomus type. D. oligothrix: size in vivo about $80-90\times30-70{\mu}m$; body shape ovoid with prominent right concave posterio-lateral end, two irregular elongated macronuclei with one micronucleus, respectively; 7 fronto-ventral, 5 transverse, 2 left marginal and 2 caudal cirri, 4 dorsal kineties with prominent bristles about $9-14{\mu}m$ long in vivo.
