In order to attain a sustained release at targeted organs in a prolonged time which can reduce the side effects and maximize the therapeutic effect, aclarubicin (ACL) was entrap ped into liposomes of different lipid compositions using Microfluidizer, and dry liposomes were prepared by lyophilization. The dry aclarubicin-entrapped liposomes were evaluated in terms of mean particle size and size distribution, entrapment efficiency and in vitro drug release profile. The Entrapment efficiency of liposome, when the concentration of aclarubicin and lipid were 0.5 to 1.0mg/ml and $200{\mu}mol$/ml, respectively, was over 80% using Microfluidizer, in contrast to 70% of entrapment efficiency using hand-shaking method. Mean particle size and size distribution of aclarubicin-entrapped liposomes of various lipid compositions did not change considerably by the freeze drying. The range of particle size was between 80 and 200nm. Among aclarubicin-entrapped liposomes, ACL-liposome of PC/DPPC/CH0L/TA displayed the most significant sustained release. The addition of DPPC appeared to be favorable for the control of release. In general, aclarubicin entrapped in liposomes was less stable than free aclarubicin either in pH 7.4 phosphate buffer or in human plasma. Formulation I($t_{1/2}$, 20.3 hr) devoid of lipid additive was the most unstable in the phosphate-buffer solution while formulation II($t_{1/2}$, 40.7 hr) with cardiolipin was the most stable. Half lives of aclarubicin-entrapped liposomes in human plasma were 43.2, 50.7, 35.9 and 35.3 hr for formulation I. II, III and IV, respectively, in contrast to 57.8 hr for free aclarubicin.

The MLV liposomes have been developed in many drugs and cosmetics fields. The phospholipid base is made from ceramides, cholesterol, cholesteryl ester, lecithin, lanolin ester, and B-sitosterol, and surfactants are made by using (PEG)n-sitosterol(n=5) and K-cetyl phosphate. We made visicles stable by passing samples through Microfludizer and croated multilamellar vesicles to make MLV liposomes similar to the structure of men's skin. In order to make MLV liposomes, we created lipid membrane films which a mixure of phospholipid base and polyol group was reacted above Tc(95$^{\circ}C$) by gelation for 3 hours. As the optimum conditions of Microfluidizer, we figured out 700 bar for the passing pressure of samples, 4$0^{\circ}C$ for its temperature, and 3 times of frequency to pass through samples. Our MLV liposomes is stable on conditions of a low temperature(5$^{\circ}C$) and a high temperature(45$^{\circ}C$), which is not to be split in a large range. We produced our own moisturizing cream which has a good affinity to skin by means of this system.

Nanoemulsions are actively used in several applications for pharmaceutical, cosmetic and chemical industries. In this study, we propose the use of microfluidizer known as high pressure homogenizer to prepare lipid nanoemulsion as a potent cosmetic delivery carrier. The lipid nanoemulsions were prepared by O/W emulsion with hydrogenated lecithin and different type of oils. Effects of oil type on the stability of the lipid nanoemulsion were investigated with Dynamic Light Scattering (DLS) and Zeta-potential. Arbutin was used as model drug for transdermal administration through hairless mouse skin. Transdermal arbutin delivery using the lipid nanoemulsions was studied with HPLC method.

In this context, it should be mentioned that multilamellar vesicles can be prepared with the main compounds of the intercellular lipids, ceramides, cholesterol, cholesteryl ester, squalane, lecithin, wax ester by effect of the wetting. We investigated properties formation of MLV with use of the AHAsomes and Microfluidizer. The multilamellar vesicles are formed merely adding polyol and water phase, followed with the microfluidizer. Formation of a practically pure AHAsomes system, containing the active ingredients directly incorporated without need for preservatives. There were very good encapsulated properties of the active ingredients whether hydrophilic(malic acid, tartaric acid, lactic acid, allantoin, urea) and hydrophobic(vitamin-I acetate, vitamin-A palmitate). Optimum condition (ormatiom of MLV was passed three times in the microfluidizer, particle size of the vesicles should be within range 50-523nm (mean=163.5nm). As application, It was compared that horny layer of the sole of foots removal with the general OM emulsion and the AHAsomes cream. There was used for three months, those got recovery wrinkles about 151.8% and elasticity three times more the AHAsomes than O/W emulsions, It was confirmed with the Image Analyzer and the Cutometer.

The liposomes have been developed in many drugs and cosmetics fields. In this context, it should be mentioned that MLV liposomes can be prepared standing with the main compounds of the intercellular lipids, cholesterol, palmitic acid, cholesteryl ester and lecithin, by swelling reaction. We report properties of the formation of MLV liposomes with use of the lipid base and Microfluidizer. MLV liposomes formed as multilamellar vesicles(MLV). The effect of the gelation of MLV liposomes have been on swelling reaction which have been mixed lipid with polyol and water phase at high temperature(90$\pm$5$^{\circ}C$). MLV liposomes have been prepared in incorporating alpha hydroxy acid ligrediens. Optimum condition of MLV liposomes were passed three times in the microfluidizer, particle size of the vesicles should be within 150-350nm and those confirmed by freeze-fracture electron microscopy.

This study was investigated optimal conditions which were necessary to prepare emulsion of four component systems by the Microfluidizer. Squalane, isopropyl myristate, PPG-15 stearyl benzoate ester were used as an oil and POE(20) sorbitan monolaurate, POE(20) sorbitan monopalmitate, POE(20) sorbitan monostearate, glycerine were selected as a cosurfactant and surfactant. The emulsion was affected by the operating pressure, the recycle frequency and oil, surfactant, cosurfactant phase. The operating pressure and recycle frequency increased, the droplet diameter of emulsion decreased and was constant later. In this study, the optimal pressure and recycle frequency were 1400 bar, 4 cycles. The droplet size was the smallest when the concentration of glycerine was 30 wt.%. As the oil phase is increased, the size is increased.
In a previous study, we investigated the antioxidative and cellular protective effects of Parthenocissus tricuspidata stem extracts. In this study, we prepared nano-emulsion containing P. tricuspidata stem extract to improve skin permeation. The particle size of the nano-emulsion using the microfluidizer was 302 nm. Its loading efficiency was over 86%. The size distribution of the nano-emulsion took a monodispersed form and the nano-emulsion was more stable than typical emulsion without using microfluidizer during a 2 week period. In vitro skin permeation study of nano-emulsion containing P. tricuspidata stem extracts was carried out using Franz diffusion cell. The 1,3-butylene glycol used as a control group had 32.59% skin permeation efficiency. The skin permeation efficiency of the nano-emulsion was 42.47%. Also, we observed the antibacterial activity of the ethyl acetate fraction on skin flora for prospective applications as a natural antimicrobial. The ethyl acetate fraction had antibacterial activities higher than methyl paraben on Staphylococcus aureus, and Bacillus subtilis. These results indicate that nano-emulsion containing P. tricuspidata stem extracts could possess valued applications in cosmetic formulations for improving skin permeation. Also, based on the antibacterial activities on skin flora, antioxidative and cellular protective effects shown in our previous study, we suggest that P. tricuspidata stem extracts could be used as functional cosmetic materials.
In this study, nano-emulsion containing ethyl acetate fraction of Polygonum aviculare (P. aviculare) extract was prepared and skin permeability of the nano-emulsion was evaluated. Nano-emulsion was prepared using homogenizer and microfluidizer by the high-energy method. The droplet size and loading efficiency of nano-emulsion containing ethyl acetate fraction of P. aviculare extract were determined. The mean droplet size was 238 nm and the loading efficiency was more than 98%. The size distribution of nano-emulsion was a monodispersed form and nano-emulsion was more stable than that of using typical emulsion without using the microfluidizer. The in vitro skin permeation study of nano-emulsion containing ethyl acetate fraction of P. aviculare extract was carried out using Franz diffusion cells. Compared to 1,3-butylene glycol, nano-emulsion showed greater values of cumulative permeation of ethyl acetate fraction from P. aviculare extract. These results indicate that the stability and skin permeability of nano-emulsion containing ethyl acetate fraction of P. aviculare exerting anti-oxidative and anti-aging activities were enhanced.

Glycyrrhiza uralensis (licorice root) is very useful medicinal herb because of strong anti-inflammatory and anti-wrinkle effect. Therefore, it is widely used in functional cosmetics. However, it is insoluble and easily decomposed by light, heat, oxygen, etc. In this study, we first prepared NanoSolve-Licorice (30-50nm) using Glycyrrhiza uralensis and propylene glycol! hydrogenated lecithin/caprylic/capric triglyceride/glycerin/water system with microfluidizer. And then, NanoSolve-Licorice and porous PMMA are dispersed in ethanol. Finally, we could get a stabilized system with high-pressure homogenizer (1,000 Bar, 3 passes). According to HPLC measurement for glabridin content, our system is more stable compared with general liposome ones. Capsulated licorice has an enhanced anti-inflammatory effect on account of excellent skin penetration. We also evaluated our final product through image analyzer, particle size analyzer, FF-TEM and chromameter.

The process variables for the manufacture of translucent microemulsion prepared with 2-octyl dodecanol, 12-hydroxy stearic acid cholesteryl , POE(40)HCO and 1,3-butandiol were examined initially (primary emulsion) and following aging for three months. The techniques empolyed in this study were particle size, turbidity, interfacial tension and microfluidizer. Particle size analysis and turbidity measurement to evaluate the emulsion stability were used. It was concluded that the process of the emulsification was an important indicator of the stability of the translucent microemulsion. From the particle size and and turbidity measurement of translucent microemulsion, adding the surfactant to the oil phase before the emulsification was found to be the most important factor for the stability of emulsions. We found that interfacial tension of the adding the surfactant to the oil phase is lower than that of the adding the surfactant to aqueous phase. In spite of hydrophilic surfactant, adding the surfactant to aqueous phase produced inferior emulsion to that to oil phase.
