A Multilayered microelectrode design is presented for large area travelling wave dielectrophoresis (TwDEP) separators. Most of typical TwDEP chip has been arrayed with 1000 electrodes in $20{\times}20\;mm^2$. However, there is a limitation of the device area that is critical in throughput, because when the area of TwDEP becomes larger, the resistance of microelectrodes for bus bar is also increased. In this paper, we successfully developed a novel TwDEP chip with extremely large area ($31{\times}25\;mm^2$) by a unique multilayered bus bar design. According to the resistance simulation of our microelectrodes, it is possible to realize a TwDEP chip with an infinite longitudinal length. We demonstrated the feasibility of our suggestion with latex microbeads and showed the potential of extremely high throughput separation with TwDEP technique.
The effect of Vanadate on the isometric contraction, membrane potential and intracellular calcium ion activities of rabbit myocardial cells were investigated, using calcium selective microelectrode, filled with neutral calcium ion carrier, ETH-1001. The resting tension, the membrane potential and the intracellular calcium ion activities were recorded in normal Tyrode solution and compared with those in the contracture induced by 10 mM Vanadate. The following results were obtained: 1. The dose-response relationship between the contraction of Vanadate and twitch tension showed near-maximum response in 5mM with no corresponding changes in action potential. 2. The resting tension increased up to the amplitude of a control twitch in 10mM Vanadate with resting membrane potential, hyperpolarized. 3. Increase in intracellular calcium ion activities proceeded the contracture by 10mM Vanadate which were restored to the control level in accordance with a decrease of intracellular calcium ion activities. 4. The amplitude of contractures by 10mM Vanadate were 90-120% of the control twitch tension in which the intracellular calcium ion activities were increased about 70 times from p Ca, 7.1 in the control to p Ca, 5.8 in contractures.
Electrode impedances are measured to quantitatively characterize the electrode-electrolyte or cell-electrode interfaces. In the case of high-density microelectrode arrays(MEAs) that have been developed for brainmachine interface applications, the characterization process becomes a repeating and time-consuming task; a system that can perform the measurement and analysis in an automated fashion with accuracy and speed is required. However, due to the large number of channels, parasitic capacitance and off-capacitance components of the switching system become the major factors that decreased the accuracy for the measurement of high impedance microelectrodes. Here we investigated the implementation of automatic impedance measurement system with analyzing the causes of possible measurement-related problems in multichannel switching configuration. Based on our multi-channel measurement circuit model, we suggest solutions to the problems and introduce a novel impedance measurement scheme using electro-mechanical relays. The implemented measurement system could measure |Z| < 700 $k{\Omega}$ of impedance in - 10% errors, which can be widely applicable to high density neural recording MEAs.
Iridium oxide films (IROFs) have been extensively studied as a material for electrical stimulation of neurons, as iridium oxide has higher charge storage capacity than other metal films. More recently, sputtered iridium oxide film (SIROF) has been studied, because it can be made more conveniently than activated iridium oxide film (AIROF). Typically, the SIROFs are grown at temperatures from 400 to 600 $^{\circ}C$. However, such high temperatures cannot be used when the iridium oxide (IrOx) film is to be deposited on a flexible polymer material, such as polyimide. In this paper, we show that we can still obtain excellent characteristics in SIROFs grown without heating (cold SIROF), by optimizing the growth conditions. We show that the oxygen flow rate is a critical parameter for controlling the surface properties of a cold SIROF. At an oxygen flow rate of 12 seem, the cold SIROF exhibited a charge storage capacity (CSC) of 60 mC/cm$^2$, which is comparable to or better than other published values for iridium oxide films including heated SIROFs. The film produced under these conditions also had the minimum impedance value of all cold SIROFs deposited for this study. A stability test and biocompatibility test also demonstrated the superiority of the optimized cold SIROF.
Membrane potential of cells in the isolated rabbit coronary sinus was measured by conventional glass microelectrode and investigated the effect of $[K^+]_0$ variation in control, 20 mM and Ach-containing Tyrode solution. The results obtained were as follows: 1) The resting membrane potential exposed to normal Tyrode solution containing 3 mM $K^+\;was\;about\;-60{\sim}\;-65mV$. At extracellular $K^+$ concentrations from 1 to 30 mM the resting Potential was reasonably well described by Goldman -Hodgkin -Katz equation on the assumption that $[K^+]_1$ was 150 mM and that the ratio of membrane permeability coefficient for $Na^+\;and\;K^+,\;P_{Na}/P_K\;({\alpha})$ was 0.07. 2) In 20 mM Na-Tyrode solution (replacing by equimolar Tris) the resting membrane potential was hyperpolarized by 15 to 20 mV and showed slightly deviated to depolarized direction compared to the predicted value by Goldman-Hodgkin -Katz equation. 3) In the presence of $10^{-6}M$ Ach, the resting potentials at $[K^+]_0$ levels from 1 to 30 mM were well fitted with the predicted value on the assumption that $P_{Na}/P_K$ was 0.0144. It could be concluded that the low resting membrane potential of coronary sinus cells reflects a relatively high ratio $P_{Na}/P_K$ of about 0.07.
The present study was undertaken in order to investigate effect of ethanol extract of Rehmanniae radix(RREE) on electrophysiology of sinus node and papillary muscle. Rehmanniae radix is a herbal medicine which has been known to have diuretic, antipyretic, hemopoietic and cardiotonic effects. Action potentials were recorded by means of glass capillary microelectrode(technique) in rabbit sinoatrial nodal cells and papillary muscle cells which were superperfused with either tyrode solution or tyrode solutions containing different amount of RREE. The results obtained were as follows ; 1) In both central and peripheral nodal cells maximum diastolic potential (MDP) and amplitude of action potential (APA) were not affected by RREE. 2) Action potential duration as expressed $APD_{60}$(time to 60% repolarization) of central and peripheral pacemaker cells were significantly prolonged following perfusion with tyrode solution containing 0.1% RREE. 3) The rates of spontaneous firing from central pecemaker cell were decreased by RREE at concentration of 0.05% and 0. 1% while spontaneous rhythm of perinodal cell was decreased by 0.1% RREE. 4) The action potential duration of papillary muscle as expressed $APD_{60}$ were prolonged by 0.1% RREE.
In investigating the characteristics of a neural network, the use of planar microelectrode array shows several advantages over normal intracellular recording[1]. A transparent indium tin oxide(ITO) multi-electrode array(MEA) was fabricated and its top surface was insulated with photodefinable polyimide(HD-8001) except the exposed area for interfacing between the ITO electrodes and the neuronal cells. The exposed ITO electrodes were platinized in order to reduce the impedance between the electrodes and electrolyte. The one-minute platinization with $0.99nA/{\mu}m^2$ current density reduced the average impedance of the electrodes from $2.5M\Omega\;to\;90k\Omega$ at 1kHz in normal ringer solution. Cardiac cells were cultured on this MEA as a pilot study before neuron culture. The signals detected by the platinized electrodes had larger amplitudes and improved signal to noise ratio(SNR) compared to non-platinized electrodes. It is clear that microelectrodes need to have lower impedance to make reliable extracellular recordings, and thus platinization is essential part of MEA fabrication. Burst spike of cultured olfactory bulb was also detected with the MEA having platinized electrodes.
Gastrointestinal motility consists of phasic slow-wave contractions and the migrating motor complex (MMC). Eupatilin (Stillen$^{(R)}$) has been widely used to treat gastritis and peptic ulcers, and various cytokines and neuropeptides are thought to be involved, which can affect gastrointestinal motility. We performed a study to identify the effects of eupatilin on lower gastrointestinal motility with electromechanical recordings of smooth muscles in the human ileum and colon. Ileum and colon samples were obtained from patients undergoing bowel resection. The tissues were immediately stored in oxygenated Krebs-Ringer's bicarbonate solution, and conventional microelectrode recordings from muscle cells and tension recordings from muscle strips and ileal or colonic segments were performed. Eupatilin was perfused into the tissue chamber, and changes in membrane potentials and contractions were measured. Hyperpolarization of resting membrane potential (RMP) was observed after administration of eupatilin. The amplitude, AUC, and frequency of tension recordings from circular and longitudinal smooth muscle strips and bowel segments of the ileum and colon were significantly decreased after admission of eupatilin. Eupatilin elicited dose-dependent decreases during segmental tension recordings. In conclusion, eupatilin (Stillen$^{(R)}$) showed inhibitory effects on the human ileum and colon. We propose that this drug may be useful for treating diseases that increase bowel motility, but further studies are necessary.
There is evidence that noradrenaline enhances spontaneous contractions dose-dependently in guinea-pig antral circular muscle. To investigate the mechanism of this excitatory action, slow waves and membrane currents were recorded using conventional microelectrode techniques in muscle strips and the whole cell patch clamp technique in isolated gastric myocytes. On recording slow waves, noradrenaline $(10^{-5}\;M)$ induced the hyperpolarization of the membrane potential, although the shape of the slow waves became tall and steep. Also, spike potentiaIs occurred at the peaks of slow waves. These changes were completely reversed by administration of phentolamine $(10^{-5}\;M),\;an\;{\alpha}-adrenoceptor$ blocker. Noradrenaline-induced hyperpolarization was blocked by apamin $(10^{-7}\;M)$, a blocker of a class of $Ca^{2+}\;-dependent\;K^+$ channels. To investigate the mechanisms for these effects, we performed whole cell patch clamp experiments. Norndrenaline increased voltage-dependent $Ca^{2+}$ currents in the whole range of test potentials. Noradrenaline also increased $Ca^{2+}\;-dependent\;K^+$\;currents, and this effects was abolished by apamin. These results suggest that the increase in amplitude and the generation of spike potentials on slow waves was caused by the activation of voltage-dependent $Ca^{2+}$ channel via adrenoceptors, and hyperpolarization of the membrane potential was mediated by activation of apamin-sensitive $Ca^{2+}\;-dependent\;K^+\;channels$.
