식품가공소재용 꽃게의 화학적 및 미생물학적 위생 특성 (Chemical and Microbiological Properties on Sanitary of Swimming Crab Portunus trituberculatus as Sources for Seafood Products)

  • Crabs are a popular seafood item. However, they can harbor many microorganisms, heavy metals, radioactivity, and benzo(a)pyrene, which are potential health risks to humans. The objective of this study was to assess the potential of swimming crabs for use in foods such as Ganjang-gejang, by measuring their sanitary biological and chemical properties. Viable microbial cell counts in swimming crab samples were $3.4{\times}10^2-6.7{\times}10^4CFU/g$, but no coliform, Escherichia coli, or pathogenic bacteria, such as Vibrio parahaemolyticus, Enterohemorrhagic E. coli, Listeria monocytogenes, Staphylococcus aureus, or Salmonella spp., were detected. Heavy metal concentrations in swimming crab samples were non-detectable to 0.112 mg/kg for total mercury, non-detectable to 0.435 mg/kg for lead, and 0.115-0.836 mg/kg for cadmium. Benzo(a)pyrene concentrations ranged from $0.025-0.060{\mu}g/kg$, and the volatile basic nitrogen content ranged from 8.7-15.6 mg/100 g. No radioactivity was detected in samples. These results suggest that swimming crabs are viable for use in seafood products.

Efficiency of Hurdle Technology Applied to Raw Cured Meat (Si-Raw)Processing

  • Si-Raw is a raw cured meat (raw, cured meat fermented with steamed rice) produced by the aboriginal people of Taiwan. In order to prevent food poisoning or intoxication from botulism, new methods of monitoring the production base on hurdle technology were investigated. New methods investigated incorporated citric acid, sodium hypophosphite, Monascus anka mash, plum paste or lactic acid bacteria inoculum added separately to meat with steamed rice and salt to lower the Aw (water activity) and pH values of the products to control the microbial growth. Results showed that anaerobic bacterial counts, lactic acid bacterial counts and aerobic bacterial counts for the products of all treatments were less than $10^6$, $10^5$ and $10^2cfu/g$, respectively. Sodium chloride content of all products was above 5.46%, water activity was below 0.939 and pH value was below 4.27. IMP was lower and ATP and hypoxanthine were higher. ATP concentrations were higher in the samples which contained the anka mash. Result of sensory panel test indicated that most people preferred the products with added sodium hypophosphite. Except for the fact that the content of tryptamine in the sample with Monascus anka mash was higher, the amine concentrations for all treatments were lower than those of other fermented meat products. The amino acid nitrogen content was higher in the product made from raw meat treated with citric acid, but lower in the other products. Neither Clostridium botulinum nor Trichinella spiralis were detected in any of the treatments. The result may indicate that hurdle technology is effective for hygiene and safe producing Si-Raw.

  • Oysters Crassostrea gigas are a globally popular shellfish for human consumption. As filter-feeding bivalve mollusks, oysters may harbor many microorganisms and chemicals that could pose potential human health risks. The objective of this study was to investigate the suitability of cultured oysters for raw consumption or use in seafood products by measuring concentrations of harmful microorganisms and chemicals in their flesh. Microbial concentrations in cultured oysters were found to be: $1.0{\times}10^2-6.0{\times}10^4CFU/g$ (viable cell counts), not detected $(ND)-5.4{\times}10^3CFU/g$ (coliform bacteria), $ND-1.3{\times}10^2CFU/g$ (E. coli), and $ND-4.6{\times}10^3CFU/g$ (Vibrio parahaemolyticus). Other pathogenic bacteria, including Enterohemorrhagic E. coli (EHEC), Listeria monocytogenes, Staphylococcus aureus, and Salmonella spp., were not detected in any samples. Heavy metal concentrations of cultured oysters were ND-0.239 mg/kg (total mercury), ND-1.091 mg/kg (lead), ND-0.968 mg/kg (cadmium). The concentrations of benzo(a)pyrene ranged from $0.280-0.880{\mu}g/kg$. Paralytic shellfish poison ranged from ND-0.58 mg/kg, while diarrhetic shellfish poison was not detected. No radioactivity was detected. These results suggest that oysters intended for raw consumption or use in seafood products should be subjected to chemical and biological controls.

오존 및 방사선 조사기술 이용 원부재료의 위생화 후 제조한 김치의 저장기간에 따른 변화 (Quality Changes of Kimchi Manufactured with Sanitized Materials by Ozone and Gamma Irradiation during Storage)

  • 원부재료에 부착된 미생물을 김치제조 전에 오존 및 방사선 조사를 실시하여 미생물을 감균시키고 위생화된 재료로 김치를 제조하였을 때의 미생물학적, 화학적 및 관능적 변화를 측정하였다. 대조군 맛김치의 미생물 변화는 총균수의 경우, 저장기간이 증가할수록 빠른 속도로 증가한 후 저장 10일 이후부터는 서서히 감소하였다. 그러나 오존 및 방사선 처리군의 경우에서는 대조군에 비하여 서서히 증가하는 경향이었다. 젖산균수의 경우에는 대조군은 오존 및 방사선 처리군에 비하여 다소 낮은 변화를 보이는 것으로 나타났다. 효모 및 곰팡이수의 변화는 대조군이 가장 높은 균수를 저장기간 내내 유지하였으나 오존 및 방사선 처리군은 대조군보다 낮은 균수를 보였으며 그 변화는 3 kGy와 5 kGy가 가장 적은 것으로 나타났다. pH 및 산도의 변화에서도 대조군은 빠른 속도로 변화되었으나 오존 및 방사선 처리군의 경우는 서서히 변화되었으며 오존 및 방사선 처리군 중에서는 3 kGy와 5 kGy 처리군이 가장 변화가 적었다. 저장기간에 따른 관능검사 결과는 발효가 진행됨에 따라 대조군은 빠른 품질변화를 보였지만 오존 및 방사선 처리군은 발효의 진행이 서서히 일어나 품질 유지기간을 연장시키는 것으로 나타났다.

  • The demand of packaged baby leaves has been increased for its convenient use as fresh-cut produce. This investigation was aimed to explore the effects of different sanitizers on the quality parameters of 'Tah Tasai' Chinese cabbage (Brassica campestris var. narinosa) baby leaves. Thirteen days old baby leaves were harvested and washed in tap water (TW), 100 ppm chlorine solution (Cl), 2 ppm ozonated water ($O_3$), 15 ppm chlorine dioxide solution ($ClO_2$) and washing with 0.2% citric acid solution followed by 50% ethanol spray (CA+Et). The samples were then packaged in 50 ${\mu}m$ polyethylene bags and stored at $5^{\circ}C$ for 10 days. Off-odor of packaged baby leaves was not detected during storage. There was no significant difference in color parameters among the treatments. Samples treated with $O_3$ showed substantially higher electrolyte leakage throughout the storage. This treatment also rendered a higher accumulation of $CO_2$ in the packages. Samples treated with Cl and CA+Et maintained good overall visual quality with higher scores compared to that of $O_3$ and $ClO_2$. Although Cl treatment showed lower number of total aerobic count at the beginning of storage, citric acid in combination with ethanol treatment was more effective until the end of storage. The combined treatment also showed comparatively lower coliform plate count. This result indicates that citric acid wash followed by ethanol spray could be an alternative to chlorine for environment friendly sanitization of baby leaves.

산란계 농장에서의 생물학적 위해요인과 관리점 분석 (Analysis of biological hazards and control points in layer houses)

  • An egg has been considered as one of the most important food sources because of it's nutritional superiority and reasonable price. With the complexity of egg flow system from production to consumption in Korea, preventive measures for egg safety have been required. Therefore, our study was carried out to analyze hazards for the egg under farm level and develope preventive measures with a purpose of obtaining egg safety. To analyze biological hazards, microbial contamination of egg(normal, dirty and cracked), water, feed, manure and equipments associated with laying were investigated. One isolate of Salmonella enteritidis and S. bardo were detected from the manure and dirty egg shell respectively. The sanitary conditions in the farm were surveyed by questionaries. Confirmation of vaccination for purchasing chicks and establishment of sanitary guidances for water and type of water suppliers ought to be preceded. Feed supplier systems including feed tanks, feed pipes and hoppers were known that their contamination might give it a chance to infect individuals and egg content and shell. The safe and rapid disposal of dead chickens and rodent were reported as more crucial factors to prevent infectious disease and manage good sanitation. Egg selector and collecting belt should be kept properly not to be contaminated from egg fluids and feces. It should be also considered that regular gathering of eggs, removal of dirty or cracked ones, storage under refrigeration and the use of disposable egg tray were continuously fulfilled. Conclusively, Our results suggested that HACCP-based system for providing fresh and safe eggs to consumers should be applied to the farm.

시판 냉동굴(Crassostrea gigas)의 위생 안전성 평가 (Safety Evaluation of Frozen Oyster Crassostrea gigas as a Raw Material for Seafood Products)

  • This study was conducted to investigate the suitability of frozen oysters as a raw material for the preparation of seafood products by measuring the concentrations of harmful microorganisms and chemicals in thawed flesh. The microbial concentrations in thawed oysters were 2.3-5.0 log CFU/g for viable cell counts, not detected (ND)-1.0 log CFU/g for coliform bacteria, and ND for Escherichia coli and pathogenic bacteria such as Staphylococcus aureus, Salmonella spp., Listeria monocytogenes, Vibrio parahaemolyticus, Enterohemorrhagic Escherichia coli (EHEC), and Clostridium perfringens. In frozen oysters, the heavy metal concentration for viable cell counts was ND-0.030 mg/kg, for lead was ND-0.393 mg/kg, and for cadmium was 0.021-0.597 mg/kg. Benzo(a)pyrene, shellfish poison (paralytic shellfish and diarrhetic shellfish poisons), and radioactivity were not detected in the thawed oysters. These results suggest that frozen oysters can be safely used as a raw material for the preparation of seafood products.

냉장 중 품질변화 측정 지표 개발을 위한 냉장고 형태별(개방형 및 도어형) 저장 중 주요 냉장 식품의 품질변화 측정 (Comparative Assessment of Quality Changes in Refrigerated Foods Stored in Open-type and Door-type Refrigerators: Towards Developing Quality Indicators)

  • This study was conducted to investigate the impact of refrigerator type and temperature fluctuations on the quality changes of refrigerated foods. Phycochemical and sensory quality, as well as microbial growth, were measured for various foods stored in open- or door-type refrigerators set at 5℃ during storage. The average temperatures recorded were 11.50±1.14℃ in an open refrigerator, and 6.34±0.97℃ in a closed refrigerator. The average surface temperatures of the food items were 9.60±1.20 and 6.00±0.80℃ for open and closed refrigerators, respectively. Significant changes in color and appearance quality were observed in lettuce, mackerel, ground beef, and cut pineapples when stored in open refrigerators. Ready-to-Eat foods such as gimbap and sandwiches exhibited higher levels of microbiological proliferation when stored in open refrigerators compared to closed refrigerators. Processed foods, such as sterilized milk and packaged tofu, did not show significant differences in quality among various types of refrigerators. The installation of refrigerator doors can effectively minimize temperature fluctuations caused by external factors, thereby reducing variations in food quality. These findings provide essential insights into the quality changes associated with the implementation of refrigerator doors, serving as fundamental data for ensuring optimal food preservation.

포장두부의 가공공정에서 미생물 분석 및 안전성 평가 (Microbial Analysis and Safety Evaluation in the Process of Packaged Tofus)

  • 본 연구는 생산 후 3일 이내인 시판두부의 가공공장별, 두부유형별 위생실태를 파악하여 두부에 대한 미생물 규격의 설정 가능성을 검토하며, 가공단계별 총생균수와 대장균군을 분석하여 오염 원인을 규명코자 실시되었다. 시판중인 두부의 위생성은 동일유형이더라도 가공공장마다 차이가 있었고 동일공장의 시료 간에도 차이를 보였다. 총생균수가 $1.0{\times}10^6CFU/g$ 이상을 보인 일부 포장두부에서는 관능적 부패 변질 현상이 관찰되었다. 총 148개 두부시료 중 총생균수와 대장균군이 각각 $1.0{\times}10^5CFU/g$ 이하와 음성에 적합한 시료는 단순포장두부가 32.0%와 12.0%, 포장가열두부가 86.9%와 83.7%인 것으로 조사되었다. 이러한 가공공장에 따른 위생성 차이는 가열과 열처리사이의 공정인 응고, 압착, 성형, 1차냉각, 내포장공정에서 2차오염 정도의 차이에서 기인하는 것으로 생각되었다. 포장두부의 가공공정 중 미생물이 증가하여 각별한 관리가 요구되는 공정으로는 원료대두의 세척 침지와 마쇄, 응고 후 압착 성형, 1차냉각공정이었다. 따라서 보다 위생적인 두부를 가공하기 위해서는 이들 가공공정에서 사용되는 각종 설비나 작업도구, 성형포 및 작업자 등에 대한 세척 및 소독 강화가 절실하게 요구되었다.

제주도 어류양식장에 대한 미생물학적 오염도 조사 (Survey of the level of Microbial Contamination in Fish Farms on the Jeju-Island)

  • 본 연구는 2006년 3월부터 8월까지 제주도에서 성업 중인 육상 수조식 양식장 21개소를 대상으로 하여 미생물학적 위해요소를 양어 용수, 양어 사료, 양식 넙치에서 조사하였다. 식품 병원성 미생물 중 일반세균과 총 대장균군, 그리고 Salmonella spp., Escherichia coli O157:H7, Staphylococcus aureus, Bacillus cereus, Vibrio parahaemolyticus를 조사하였다. 이번 연구를 진행하는 동안 특이할만한 점은 여름에 들어서면서 집중적이면서도 빈번하게 비가 내렸다. 양어용수를 검사한 결과 S. aureus, B. cereus and Salmonella spp.는 각각 $0-3.3{\times}10^2 CFU/ml,\;0-2.2{\times}10^2CFU/ml\;and\;0-5.1{\times}10^1 CFU/ml$ 검출이 되었다. 또한 사료 내 미생물을 조사하여 본 결과 조사한 MP사료 85%에서 S. aureus, B. cereus and total coliforms이 검출이 되었다 반면 양식 넙치를 조사한 결과 Escherichia coli O157:H7, total coliforms은 검출이 되지 않았다. 추후 이와 관련하여 좀더 체계적인 연구 시스템을 양식현장에 적용한다면 근래 들어 문제가 되고 있는 식품위생상 수산물이 야기하는 문제점들이 해결이 되어 안전한 식품으로서의 가치를 인정받을 수 있을 것 같다.
