Influence of Level of Feed Intake on Concentration of Purine Derivatives in Urinary Spot Samples and Microbial Nitrogen Supply in Crossbred Bulls

  • The potential of the spot urine sampling technique as an alternative to performing a total urine collection to predict the microbial nitrogen supply was evaluated in crossbred bulls. In a completely randomized design, 20 growing crossbred bulls were assigned four levels of feed intake (120, 100, 80 and 60% of voluntary dry matter intake) on diets comprised of wheat straw and concentrate mixture (50:50). After three months of experimental feeding, a metabolism trial was conducted for ten days, during which spot urine collections were performed every 6 h post feeding on days 9 and 10. The daily urinary excretion of allantoin (A) and purine derivatives (PD) decreased with the reduction in feed intake while creatinine (C) excretion remained similar in animals fed at different levels. The microbial nitrogen (MN) supply calculated from the PD excreted in total urine (35.08 to 72.08 g/d) was higher at increased levels of feed intake. PD concentration in spot urine samples had poor correlation with feed intake except at 12 h post feeding. A/C ratio and PD/C ratio in spot urine samples remained similar irrespective of sampling time and significantly (p<0.01) correlated with daily urinary PD excretion, digestible organic matter intake and dry matter (DM) intake. However, no significant differences were evident in these ratios among animals fed at levels 120, 100 and 80% of voluntary dry matter intake (VDMI) at different times post feeding. These results suggests that the spot urine sampling technique to predict the microbial protein supply is not suitable for detecting small differences in MN supply and hence, estimation of PD excreted in total urine (mmol/d) is necessary to assess precisely the MN supply in crossbred bulls.

Survival and Performance of Two Cellulose-Degrading Microbial Systems Inoculated into Wheat Straw-Amended Soil

  • A cellulose-degrading composite microbial system containing a mixture of microbes was previously shown to demonstrate a high straw-degrading capacity. To estimate its potential utilization as an inoculant to accelerate straw biodegradation after returning straw to the field, two cellulose-degrading composite microbial systems named ADS3 and WSD5 were inoculated into wheat straw-amended soil in the laboratory. The microbial survival of the inoculant was confirmed by a denaturing gradient gel electrophoresis (DGGE) analysis, whereas the enhancement of straw degradation in soil was assessed by measuring the mineralization of the soil organic matter and the soil cellulase activity. The results indicated that most of the DGGE bands from ADS3 were detected after inoculation into straw-amended autoclaved soil, yet only certain bands from ADS3 and WSD5 were detected after inoculation into straw-amended non-autoclaved soil during five weeks of incubation; some bands were detected during the first two weeks after inoculation, and then disappeared in later stages. Organic matter mineralization was significantly higher in the soil inoculants ADS3 and WSD5 than in the uninoculated controls during the first week, yet the enhanced degradation did not persist during the subsequent incubation. Similar to the increase in soil organic matter, the cellulase activity also increased during the first week in the ADS3 and WSD5 treatments, yet decreased during the remainder of the incubation period. Thus, it was concluded that, although the survival and performance of the two inoculants did not persist in the soil, a significant enhancement of degradation was present during the early stage of incubation.

Characterization of Cork Biofilter (코르크담체를 이용한 유기휘발성화합물의 제거)

  • The cork have been preferred over the conventional materials, zeolite, ceramics, and lignite as a biofilter medium. During the 6 months of operation, the performance of the cork biofilter was considered good with 150ppm of mixture BTX vapor efficiencies greater than 90% at 60 second of EBCT. It was observed 56 % of removal efficiency under transition conditions at first stage, and then the removal efficiency was increased to above 90 %, and the sustainability of removal efficiency was maintained. At second stage, the performance of cork biofilter was not decreased 90 % efficiency with 150 ppm BTX at 30 sec. EBCT. The production of $CO_2$ due to microbial respiration was increased to the 40 % on the operation of cork biofilter.

  • The microbial and chemical changes, and characterization of the predominant acid-producing bacteria in the swine manure ensilage blended with corn meal at a ratio of 50:50 were studied, The fermentation was dominated by Lactobacilli, which multiplied rapidly for the first 24 hrs. The acid production during the fermentation process caused rapid pH drop to 4.5, and controlled the population of E. coli and other organisms in the feedlot waste.(중략)

  • In order to isolate constituents of Lactarius piperatus$(F_{R.})$ S.F. $G_{RAY}$, the fresh carpophore was homogenized with ethanol in a blendor, stored for three years and subsequently extracted with ethanol. From the concentrated ethanol solution, a mixture of compounds was precipitated, from which a white needle crystal, named 'gultorin', $(mp.160{\sim}162^{\circ})$ was obtained. The physical and chemical characteristics of gultorin were determined.

Effect of Microbial Additives on Metabolic Characteristics in Sheep and Milking Performance of Lactating Dairy Cows (미생물제제의 첨가가 면양의 반추대사 및 젖소의 유생산성에 미치는 영향)

  • Two experiments were conducted to observe the effects of direct fed microbials on metabolic characteristics in sheep and milking performance in dairy cows. A metabolic trial with four ruminally cannulated sheep(60±6kg) was conducted in a 4×4 Latin square design to investigate the supplementation effects of Saccharomyces cerevisiae, Clostridium butyricum or mixed microbes of S. cerevisiae and C. butyricum on ruminal fermentation characteristics and whole tract digestibility. Sheep were fed 1.25 kg of total mixed ration(TMR, DM basis) supplemented with S. cerevisiae (2.5g/day), C. butyricum (1.0g/day) or its mixture(S. cerevisiae 1.25g/day+C. butyricum 1g/day), twice daily in an equal volume. But control sheep were fed only TMR. A feeding trial with 28 lactating Holstein cattle was also conducted for 12 weeks to investigate the effects of the same microbial supplements as for the metabolic trial on milking performance. The cows were fed the TMR(control), and fed S. cerevisiae(50g/day), C. butyricum(15g/day) or its mixture (S. cerevisiae 25g/day + C. butyricum 7.5g/day) with upper layer dressing method. Total VFA concentration and the digestibility of whole digestive tract in the sheep increased by supplementation of S. cerevisiae, C. butyricum or their combined microbials compare to control group. The proportion of propionic acid at 1h(P<0.039) and 3h(P<0.022) decreased by supplementation of S. cerevisiae while tended to increase acetic acid proportion at the same times. Daily dry matter intake(DMI) was not influenced by the microbial treatments, but milk yield(P<0.031) and feed efficiency(milk yield/DMI, P<0.043) were higher for the cow received C. butyricum than those for other treatments. The milk fat content was higher (P<0.085) when cows fed S. cerevisiae(4.11%) than that fed the control (4.08%), the diets with C. butyricum (3.85%) and the microbial mixture. Based on the results obtained from the current experiments, supplementation of C. butyricum or mixture with S. cerevisiae might be increased milk fat content and milk productivity of lactating daily cows.   • Due to internal feeding behavior, the oriental tobacco budworm, Helicoverpa assulta ($Guen\acute{e}e$), infesting hot pepper has been regarded to be effectively controlled by targeting egg and neonate larval stages just before entering the fruits. This study aimed to develop an efficient biological control method focusing on these susceptible stages of H. assulta. An egg parasitoid wasp, Trichogramma evanescens Westwood, was confirmed to parasitize the eggs of H. assulta. A mixture of Gram-positive soil bacterium, Bacillus thuringiensis subsp. kurstaki, and Gram-negative entomopathogenic bacterium, Xenorhabdus nematophila ANU101, could effectively kill neonate larvae of H. assulta. A sex pheromone trap monitored the occurrence of field H. assulta adults. The microbial insecticide mixture was proved to give no detrimental effects on immature development and adult survival of the wasp by both feeding and contact toxicity tests. A combined treatment of egg parasitoid and microbial pesticide was applied to hot pepper fields infested by H. assulta. The mixture treatment of both biological control agents significantly decreased the fruit damage, which was comparable to the chemical insecticide treatment, though either single biological control agent did not show any significant control efficacy. This study also provides morphological and genetic characters of T. evanescens.

Effect of Diet on Enzyme Profile, Biochemical Changes and In sacco Degradability of Feeds in the Rumen of Buffalo

  • Four rumen fistulated Murrah buffaloes were used to study the effect of four diets differing in roughage to concentrate ratio on rumen biochemical changes, microbial enzyme profile and in sacco degradability of feed in a $4{\times}4$ Latin Square design. The animals were fed four diets consisting of 80:20, 70:30, 60:40 and 50:50 ratios of wheat straw and concentrate mixtures, respectively. Wheat straw and concentrate mixture were mixed with water (0.6 l/kg feed) and complete feed mixture was offered to the animals at 8:00 h and 16:00 h in two equal parts. The variation in pH of rumen liquor (difference of maximum and minimum during 0-8 h post feeding) increased with increasing level of concentrate mixture in the diet. There was no effect of diet composition on volatile fatty acids, total nitrogen and trichloro-acetic acid precipitable nitrogen in the rumen liquor, but ammonia nitrogen increased with increasing level of concentrate mixture in the ration. Major portions of all fibre degrading enzymes were present in the particulate material (PM) of the rumen contents, but protease was absent in PM fraction. The activities of micro-crystalline cellulase, acetyl esterase and protease increased with increase in the level of concentrate mixture, but the activities of other enzymes (carboxymethylcellulase, filter paper degrading activity, xylanase, $\beta$-glucosidase and $\beta$-xylosidase) were not affected. The in sacco degradability and effective degradability of feeds increased with increasing level of concentrate mixture in the ration.

Kimchi Preparation with Brined Chinese Cabbage and Seasoning Mixture Stored Separately (분리 저장한 절임배추와 김치속을 이용한 김치의 제조)

  • The brined Chinese cabbages and kimchi seasoning mixture were separately stored and used for preparation of kimchi and the alternative method of conventional kimchi-manufacturing process was investigated. Chinese cabbages brined with 3% and 7% salt contents were stored at $0^{\circ}C$ and $10^{\circ}C$, and kimchi seasoning mixture without salt was stored at $4^{\circ}C$ and $10^{\circ}C$. The changes in pH and total acid of the cabbages were significantly slowly for the brined cabbage of 7% salt and storage at $0^{\circ}C$. Microbial growth including lactic acid bacteria were also increased slowly at $0^{\circ}C$. But, pH and total acid content of the kimchi seasoning mixture were not changed significantly at both temperatures during storage. Sensory evaluation on the kimchi samples showed that the modified method of kimchi preparation with separately stored seasoning mixture and brined cabbages is useful industrially or at home.

The Effects of the Mixture of Herbal Extract on Developing Plaque and Gingivitis (생약복합제재에 의한 구강양치가 치태 및 치은염에 미치는 영향)

  • This double-blind controlled clinical and microbiological study was carried out to determine the effects of mouthwash preparation containing the mixture of herbal extract on developing plaque and gingivitis in the experimental gingivitis model. Following a 2-week normalization period, 34 dental students were distributed randomly into 1 of 3 treatment groups. They rinsed, under supervision, two times daily for 3 weeks with either normal saline(CT), 0.1% chlorhexidine(CH), or the mixture of herbal extract (HT), but refrained from any oral hygiene measures. The Plaque Index(PlI), the Gingival Index(GI), and the amount of Gingival Crevicular Fluid(GCF) were measured at week 0,1,2, and 3 of the experimental period, while the assessment of total wet weight of plaque and the phase contrast microscopic examination of plaque were performed at the end of experimental period(3 weeks). Subjects using mouthrinse preparation containing the mixture of herbal extract demonstrated negligible, if any, changes in the accumulation and microbial composition of plaque compared to those using normal saline, while the reduction of gingival inflammation by this mixture was highly significant and comparable to that of chlorhexidine. The results of this study indicate that the preparation containing the mixture of herbal extract do not provide any antiplaque benefits but is very effective in inhibiting the development of and in reducing existing experimental gingivitis when used as mouthrinse. Further research is needed to determine whether a significant reduction of gingival inflammation without a concomitant decrease in plaque accumulation is of clinical importance.
