Treatment of Acid dye Using Microbial Immobilization (미생물 고정화를 이용한 산성염료의 처리)

  • Strains degrading and decolorizing acid dyes, Nylosan red E-BL 150%. were isolated from natural system, was named as ARK3. The optimal culture conditions of temperature and pH were $35^\circ{C}$, 7.0, respectively. Growth rate of cells in conditions of aerobic shaking more than standing culture conspicuously increased, and optical density of those to strain ARK3 were found as 1.38 and 0.25 after 42 hrs. Decolorization efficiency in batch culture which used as immobilization media to natural zeolite was 15% after 6 hrs, while suspension culture was 5%, also its of immobilization and suspension culture were 90% and 85% after 48 hrs, respectively. Decolorization efficiency of air-lift bioreactor was more than 90% to a dilution rate of $0.038hr^{-1}$, but that was decreased as 70%, when the dilution rate was $0.05hr^{-1}$. Even though at maximum dilution rate of this study, there was not appeared "wash out" phenomienon of biomass. Decolorization efficiency was 97.7% at a dilution rate of $0.025hr^{-1}$, when influent dye concentration was $100mg/\ell$. But if influent dye concentration increased as $150mg/\ell$, even though MLVSS increased, that of treatment water decreased as 93%. Also, when influent dye concentration increased as $200mg/\ell$ and $300mg/\ell$, decolorization efficiencies of treatment water abruptly decreased as 85% and 63%, respectively. Decolorization efficiency was more than 92% to the limit volumetric loading rate of $3.75mg/\ell\cdot{hr}$hr, without regard to variation of influent dye concentration or hydraulic retention time. if volumetric loading rate was more than $3.80mg/\ell\cdot{hr}$, at same condition, decolorization efficiency was lower decrease of retention time than increase of influent dye concentration.entration.

  • Comamonas sp. AEBL-85 was isolated from microbial granules in an activated sludge process of long-term operated for the treatment of reactive azo dye, and characterized its capability to decolorize Reactive Black 5. The effects of adding carbon source and nitrogen source on the extent of decol-orization were analyzed to develop an optimal medium. The optimum initial pH and temperature wire 6.0 and 35$^{\circ}C$, respectively. Reactive Black 5 of 50 mg/l was readily decolorized up to 95% within 40 hr by Comamonas sp. AEBL-85.

A Study on the Effects of Cosubstrates on the Biological Treatment and the Decolorization Mechanisms of Dyeing Wastewater (염색폐수의 생물학적 처리에 미치는 cosubstrates의 영향 및 색도제거 기전 연구)

  • In this research, the decolorization mechanisms of dye wastewater were divided into two pathways, one was physicochemical sorption to biomass flocs and the other was biological removal by microbial metabolisms. Batch tests were conducted to examine the reaction conditions, anaerobic and aerobic conditions, types and dose of cosubstrates, and to confirm the mechanisms of decolorization through the biosorption tests using the activated sludge and the autoclaved deactivated sludge. From the tests, the decolorization efficiencies of dye wastewater were 102 ${\Delta}$unit/g MLSS under the aerobic condition and 123 ${\Delta}$unit/g MLSS under the anaerobic condition, and organic removals were 82 $mg{\Delta}$COD/gMLSS and 75 $mg{\Delta}$COD/gMLSS respectively. Acetate was the more efficient cosubstrate than the domestic wastewater in the decolorization step. In addition the removal of colors and organics was increased with cosubstrates dosage. And $20.3{\sim}37.3$ ${\Delta}$unit/g MLSS was removed by the autoclaved sludge and $102.0{\sim}159.0$ ${\Delta}$unit/g MLSS by the activated sludge. The physicochemical sorption was dominant in the beginning of biosorption tests, and the biological decolorization was increased with a cosubstrate in the course of time.

Molecular identification of dye degrading bacterial isolates and FT-IR analysis of degraded products

  • In the present study, dye decolorizing bacteria were isolated from water and soil samples, collected from textile industries in Jodhpur province, India. Two bacterial species namely, Bacillus pumilis and Paenibacillus thiaminolyticus were screened and identified based on biochemical characterization. The degradation efficiency of these two microorganisms was compared through optimization of pH, incubation time, initial dye concentration and inoculum size. B. pumilis and P. thiominolyticus were able to degrade 61% and 67% Red HE3B, 81% and 75% Orange F2R, 49.7% and 44.2% Yellow ME4GL and 61.6% and 59.5% Blue RC CT dyes of 800mg/l concentration respectively. The optimum pH and time were found to be 8 within 24 hours. The FT-IR analysis confirmed that microorganisms were able to degrade toxic azo dyes into a non-toxic product as proved through structural modifications to analyze chemical functions in materials by detecting the vibrations that characterize chemical bonds. It is based on the absorption of infrared radiation by the microbial product. Therefore, Bacillus pumilis and Paenibacillus thiaminolyticus are a promising tool for decolorization of dyes due to its potential to effectively decolorize higher azo dye concentrations (10-800 mg/L) and can be exploited for bioremediation.

  • As a rare sugar alcohol, L-arabitol can be used in food and can prevent extra fat deposits in the intestinal tract. Commercially, L-arabitol is prepared from pure L-arabinose by hydrogenation, which needs a high temperature and high pressure, leading to a high production cost for Larabitol. Therefore, this study describes a novel L-arabitol production method based on biological purification from the xylitol mother liquor, a cheap and readily available raw material that contains a high concentration of Larabitol. First, a novel Bacillus megaterium strain was screened that can utilize xylitol, sorbitol, and mannitol, yet not L-arabitol. The isolated strain was inoculated into a medium containing the xylitol mother liquor under formulated culture conditions, where a high L-arabitol yield (95%) and high purity (80%) were obtained when the medium was supplemented with 50 g/l of xylitol mother liquor. Upon further purification of the fermentation broth by ion exchange and decolorization, L-arabitol was crystallized with a purity of 98.5%.

Also, because the effect is big to decolorization beside sterilization effect about microorganism, deodorization, disjointing of venomousness hazardous substance, food save, indoor air purge, good hand processing, hydrospace agricultural chemicals processing etc. the use extent is wide. This research ran parallel a sterilization experiment of E.coli colitis germs by ozone that manufacture ozonizer that use doped diamond maund electrode (BDD) by boron and searched special quality electrochemistry enemy of the ozonizer and is created. E.coli. After cultivate E.coli colitis germs during 37C 1 day is LB ship, after do ozonation, was sterilized more than 90% by ozone as result that examine sterilizing power by substance microscope and could confirm excellency of diamond electrode.

Screening of the Antimicrobial Activity against Helicobacter pylori and Antioxidant by Extracts from Mulberry Fruits (Morus albba L.) (품종이 다른 오디추출물의 Helicobacter pylori에 대한 항균활성 탐색과 항산화 효과)

  • The water and $40\%$ ethanol extracts from mulberry fruits were tested for their antimicrobial activities against Helicobacter pylori and antioxidant. Kangwon III (Morus albba L.) was higher phenolic content (2.90 mg/g) than other water extracts. The phenolic contents of $40\%$ ethanol extracts from Kangwon III and Hihak were 3.02 mg/g and 2.46 mg/g, respectively . The ABTS [2,2'-azinobis(3-ethylbenzothiazoline-6-sulfornic acid)] radical decolorization, electron donating ability (EDA), thiobarbituric acid reactive substance (TBARS) and antioxidant protection factor (PF) were determined for water extracts and $40\%$ ethanol extracts from mulberry fruits. EDA was higher in water extracts than ethanol extract. EDA of water extract from Kangwon III was determined as $99.54\%$ while the activity of ethanol extracts was $89.61\%$ in Daechoukmyeun. The water extracts from Cheongilppong showed higher antioxidative activity than another mulberry fruits extract when evaluated by ABTS [2,2'-azinobis(3-ethylbenzothiaznoline-6-sulfornic acid)] radical decolorization, TBARS and antioxidant PF by water extracts. Kangwon III was higher phenolic content (2.90 mg/g) than other that of water extracts. $40\%$ ethanol extracts were determined that phenolic contents of Kangwon III, Hihak were 3.02 mg/g, 2.46 mg/g for each other. Antimicrobial activity showed the high value in water extracts of Cheongilppong, Kuksang 20. The result will be useful nature microbial medicine for mulberry fruits.
