초임계 자소자추출물의 항균효과와 바이오필름, 염증매개인자 생성 억제 효능 (The Effect of Anti-microbial and the Inhibitory Effect of Biofilm Formation and Inflammatory Factors Production of Perillae semen Supercritical Fluid Extracts)

  • 본 연구는 자소자 열수추출물과 압력은 일정하게 유지한 상태로 온도를 조절하여 추출한 3종의 자소자 초임계추출물에 대해 항산화력, 항균활성, biofilm 형성 억제능과 염증매개물질(nitric oxide), 염증성 사이토카인(interleukin-6, interleukin-8)의 생성 억제능을 평가하였다. 자소자 열수추출물과 비교하였을 때 자소자 초임계추출물이 항산화 효과는 떨어졌으나, Staphylococcus aureus(S. aureus)에 대한 minimal inhibitory concentration(MIC)이 월등히 낮은 것으로 확인되었고, biofilm 형성 억제능 또한 더 우수하였다. 염증매개물질과 염증성 사이토카인의 생성도 자소자 초임계추출물이 자소자 열수추출물에 비해 현저히 감소시킬 수 있었다. 따라서 우리는 초임계추출물중 가장 우수한 항균작용과 biofilm 형성 억제, 염증인자 생성 억제 효능을 보인 45℃로 추출한 자소자 초임계추출물이 아토피 피부염 환자의 가려움증과 경표피수분손실을 방지하기 위한 천연 기능성 소재로 적합함을 시사한다.

노인급식에서 제공되는 마늘쫑 새우 볶음의 미생물학적 품질평가 (Assessment of Microbial Quality on the Preparation of Stir-Fried Dried-Shrimp with Garlic stems in the Meal Service Operation for the Elderly)

  • The purpose of this study was to identify HACCP-based CCP and CP from the microbial quality assessment on the process of side dish (stir-fried dried-shrimp with garlic stems) production in the meal service operation for the elderly. Total plate counts (TPC) of fresh garlic stalks were $7.80{\times}10^{3}$ CFU/g and they were above the standard value of microbial growth potential. The TPC, Coliform and E.coli were not detected in the dried shrimps. The TPCs after rinsing and slicing the garlic stems were $2.5{\times}10^{2}$ CFU/g and $5.5{\times}10^{2}$ CFU/g, respectively. The TPC number of cook’s hand and cutting board were also exceeded the standard limit with values of $2.2{\times}10^{2}$ CFU/g and $10.0{\times}10$ CFU/g, respectively. However, the TPC, Coliform and E.coli were not detected in the other cooking instruments. The identified CCP in inspection step was fresh garlic stems and that of prepreparation step was slicing the stems after blanching. Cook’s hand and cutting board were also verified as CCP and the other steps in cooking process and utensils tested were identified as CP’s. These result’s suggest that it is important to control the microbial contamination of raw materials at purchasing step and the sanitary education program should be developed for the employees for continuous supplement of safe and sound meal service for the elderly.

카벤다짐 분해 미생물인 Rhodococcus sp. 3-2의 대량 배양 및 미생물 제제 개발 (Mass Cultivation of Rhodococcus sp. 3-2, a Carbendazim-Degrading Microorganism, and Development of Microbial Agents)

  • Rhodococcus sp. 3-2 strain has been reported to degrade benzimidazole-based pesticides, such as benomyl and carbendazim. Therefore, this study aimed to optimize culture medium composition and culture conditions to achieve cost-effective and efficient large-scale production of the Rhodococcus sp. 3-2 strain. The study identified that the optimal media composition for mass culture comprised 0.5% glucose, 0.5% yeast extract, 0.15% NaCl, 0.5% K2HPO4, 0.5% sodium succinate, and 0.1% MgSO4. Additionally, a microbial agent was developed using a 1.5-ton fermenter, with skim milk (20%), monosodium glutamate (15%), and vitamin C (2%) as key components. The storage stability of the microbial agent has been confirmed, with advantages of low temperature conservation, which helps to sustain efficacy for at least six months. We also assessed the benomyl degradation activity of the microbial agent within field soil. The results revealed an over 90% degradation rate when the concentration of viable cells exceeded 2.65 × 106 CFU/g after a minimum of five weeks had elapsed. Based on these findings, Rhodococcus sp. 3-2 strain can be considered a cost-effective microbial agent with diverse agricultural applications.

Rhizospheric fungi of Panax notoginseng: diversity and antagonism to host phytopathogens

  • Background: Rhizospheric fungi play an essential role in the plantesoil ecosystem, affecting plant growth and health. In this study, we evaluated the fungal diversity in the rhizosphere soil of 2-yr-old healthy Panax notoginseng cultivated in Wenshan, China. Methods: Culture-independent Illumina MiSeq and culture-dependent techniques, combining molecular and morphological characteristics, were used to analyze the rhizospheric fungal diversity. A diffusion test was used to challenge the phytopathogens of P. notoginseng. Results: A total of 16,130 paired-end reads of the nuclear ribosomal internal transcribed spacer 2 were generated and clustered into 860 operational taxonomic units at 97% sequence similarity. All the operational taxonomic units were assigned to five phyla and 79 genera. Zygomycota (46.2%) and Ascomycota (37.8%) were the dominant taxa; Mortierella and unclassified Mortierellales accounted for a large proportion (44.9%) at genus level. The relative abundance of Fusarium and Phoma sequenceswas high, accounting for 12.9% and 5.5%, respectively. In total,113 fungal isolates were isolated from rhizosphere soil. They were assigned to five classes, eight orders (except for an Incertae sedis), 26 genera, and 43 species based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer. Fusarium was the most isolated genus with six species (24 isolates, 21.2%). The abundance of Phoma was also relatively high (8.0%). Thirteen isolates displayed antimicrobial activity against at least one test fungus. Conclusion: Our results suggest that diverse fungi including potential pathogenic ones exist in the rhizosphere soil of 2-yr-old P. notoginseng and that antagonistic isolates may be useful for biological control of pathogens.

상추 및 오이 시설재배 토양의 미생물 다양성 분석 (Microbial Diversity and Community Analysis in Lettuce or Cucumber Cultivated Greenhouse Soil in Korea)

  • 시설재배지의 토양미생물 분포 특성을 밝히고 토양의 건전성을 평가하고자, 전국 주요 시설재배 8개 주산단지에서 각각 5포장을 선정하여 토양화학성, 미생물분포 및 생화학적 특성을 조사하였다. 토양화학성은 가용성인산, 칼륨, 칼슘의 함량이 적정범위보다 크게 상회하여 상당량의 염류 집적을 확인하였다. 토양미생물의 배양적 방법을 통해 조사한 시설재배지의 주요 우점 박테리아는 Bacillus 속, Microbacterium 속, Arthrobacter 속, Lysobacter 속 등이었으며, 형광성 Pseudomonas 속의 밀도는 상추와 오이 재배지에서 각각 $0.018-7.3{\times}10^4\;cfu\;g^{-1}$, $0.0013-9.6{\times}10^4\;cfu\;g^{-1}$으로 시료에 따라 큰 변이를 보였다. 비배양학적 방법을 통한 토양미생물 분포 조사를 위해 수행한 인지질지방산 (PLFA)의 주성분 분석 결과, 작물 및 지역별 군집구조의 큰 차이는 없었다. 따라서 토양화학성 및 미생물군집구조 측면에서 시설재배지 조사지역의 토양은 대체로 건전한 것으로 판단된다.

Dominance of Endospore-forming Bacteria on a Rotating Activated Bacillus Contactor Biofilm for Advanced Wastewater Treatment

  • The bacterial diversity inherent to the biofilm community structure of a modified rotating biological contactor wastewater treatment process, referred to as the Rotating Activated Bacillus Contactor (RABC) process, was characterized in this study, via both culture-dependent and culture-independent methods. On the basis of culture-dependent methods, Bacillus sp. were found to exist in large numbers on the biofilm (6.5% of the heterotrophic bacteria) and the microbial composition of the biofilms was quite simple. Only three phyla were identified-namely, the Proteobacteria, the Actinobacteria (High G+C Gram-positive bacteria), and the Firmicutes (Low G+C Gram-positive bacteria). The culture-independent partial 16S rDNA sequence analysis revealed a considerably more diverse microbial composition within the biofilms. A total of eight phyla were recovered in this case, three of which were major groups: the Firmicutes (43.9%), the Proteobacteria (28.6%), and the Bacteroidetes (17.6%). The remaining five phyla were minor groups: the Planctomycetes (4.4%), the Chlorobi (2.2%), the Actinobacteria (1.1%), the Nitrospirae (1.1%), and the Verrucomicrobia (1.1%). The two most abundant genera detected were the endospore-forming bacteria (31.8%), Clostridium and Bacillus, both of which are members of the Firmicutes phylum. This finding indicates that these endospore-forming bacteria successfully colonized and dominated the RABC process biofilms. Many of the colonies or clones recovered from the biofilms evidenced significantly high homology in the 16S rDNA sequences of bacteria stored in databases associated with advanced wastewater treatment capabilities, including nitrification and denitrification, phosphorus accumulation, the removal of volatile odors, and the removal of chlorohydrocarbons or heavy metals. The microbial community structures observed in the biofilms were found to correlate nicely with the enhanced performance of advanced wastewater treatment protocols.

  • Estrone, which is obtainable from 19-hydroxycholesterol acetate through microbial oxidation, was chemically transformed to orally active estrogens, ethynylestradiol and methylestradiol. For progestin synthesis, $5{\alpha}$-bromo-619-oxidoisoandrosterone, was harvested from the culture broth(ATCC:19170), in which $5{\alpha}$-bromo-619-oxidocholestanol, acetate was added as a carbon source. Methylestrenolone, a potent orally active progestin, was synthesized from the harvested product via several reactions. Norethindrone, a popular progestin, was also prepared from 19-norandrostenedione. 19-Norandrostenedione is also available through microbial oxidation and chemical modification of $5{\alpha}$-chlor-619-oxidocholestanol, acetate.

Culturing the Uncultured in the Ocean

  • Epifluorescence microscopy and direct viable counting methods have shown that only 0.01-0.1% of all the microbial cells from marine environments form colonies on standard agar plates. To culture novel marine microorganisms, high throughput culturing (HTC) techniques were developed to isolate cells in very low nutrient media. This approaches was designed to address microbial metabolic precesses that occur at natural substrate concentrations and cell densities, which are typically about three orders of magnitude less than in common laboratory media. Approximately 5000 cultures of pelagic marine bacteria were examined over the course of 3 years. Up to 14% of cells from coastal seawater were cultured using this method, a number that is 1400 to 140-fold higher than obtained by traditional microbiological culturing techniques. Among the cultured organisms are many unique phylogenetic lineages that have been named as new phyla (7), orders (2, 5, 12), families (3), and genera (1, 4, 6). Over 90% of the cells recovered by this method do not replicate in standard agar plating, the most common method of microbial cell cultivation.

인삼 Saponin이 미생물의 효소활성에 미치는 영향 (The Effect of Gingseng Saponins on Microbial Enzyme Activity)

  • In order to investigate biochemical effects of ginseng saponin on microbial enzyme activity, Aspergillus oryxae-143 and Aspergillus niger-40, which were selected from various sources of samples and were the highest enzyme producing mold strains, were grown in the medium containing various saponin concentration (0mg%, l0mg%, 50mg%, l00mg%, 150mg% and 300mg%). The enzyme activity (amylase, protease) was found most active when the saponin was added in the culture media with the concentration of l0mg%∼l00mg%. But it seemed that the action of microbial enzyme was inhibited by adding more than 300mg% of saponin.

Application of rDNA-PCR Amplification and DGGE Fingerprinting for Detection of Microbial Diversity in a Malaysian Crude Oil

  • Two culture-independent methods, namely ribosomal DNA libraries and denaturing gradient gel electrophoresis (DGGE), were adopted to examine the microbial community of a Malaysian light crude oil. In this study, both 16S and 18S rDNAs were PCR-amplified from bulk DNA of crude oil samples, cloned, and sequenced. Analyses of restriction fragment length polymorphism (RFLP) and phylogenetics clustered the 16S and 18S rDNA sequences into seven and six groups, respectively. The ribosomal DNA sequences obtained showed sequence similarity between 90 to 100% to those available in the GenBank database. The closest relatives documented for the 16S rDNAs include member species of Thermoincola and Rhodopseudomonas, whereas the closest fungal relatives include Acremonium, Ceriporiopsis, Xeromyces, Lecythophora, and Candida. Others were affiliated to uncultured bacteria and uncultured ascomycete. The 16S rDNA library demonstrated predomination by a single uncultured bacterial type by >80% relative abundance. The predomination was confirmed by DGGE analysis.
