Biodegradation is the key process involved in natural lignocellulose biotransformation and utilization. Microbial consortia represent promising candidates for applications in lignocellulose conversion strategies for biofuel production; however, cooperation among the enzymes and the labor division of microbes in the microbial consortia remains unclear. In this study, metagenomic analysis was performed to reveal the community structure and extremozyme systems of a lignocellulolytic microbial consortium, TMC7. The taxonomic affiliation of TMC7 metagenome included members of the genera Ruminiclostridium (42.85%), Thermoanaerobacterium (18.41%), Geobacillus (10.44%), unclassified_f__Bacillaceae (7.48%), Aeribacillus (2.65%), Symbiobacterium (2.47%), Desulfotomaculum (2.33%), Caldibacillus (1.56%), Clostridium (1.26%), and others (10.55%). The carbohydrate-active enzyme annotation revealed that TMC7 encoded a broad array of enzymes responsible for cellulose and hemicellulose degradation. Ten glycoside hydrolases (GHs) endoglucanase, 4 GHs exoglucanase, and 6 GHs β-glucosidase were identified for cellulose degradation; 6 GHs endo-β-1,4-xylanase, 9 GHs β-xylosidase, and 3 GHs β-mannanase were identified for degradation of the hemicellulose main chain; 6 GHs arabinofuranosidase, 2 GHs α-mannosidase, 11 GHs galactosidase, 3 GHs α-rhamnosidase, and 4 GHs α-fucosidase were identified as xylan debranching enzymes. Furthermore, by introducing a factor named as the contribution coefficient, we found that Ruminiclostridium and Thermoanaerobacterium may be the dominant contributors, whereas Symbiobacterium and Desulfotomaculum may serve as "sugar cheaters" in lignocellulose degradation by TMC7. Our findings provide mechanistic profiles of an array of enzymes that degrade complex lignocellulosic biomass in the microbial consortium TMC7 and provide a promising approach for studying the potential contribution of microbes in microbial consortia.

NdgR, a global regulator in soil-dwelling and antibiotic-producing Streptomyces, is known to regulate branched-chain amino acid metabolism by binding to the upstream region of synthetic genes. However, its numerous and complex roles are not yet fully understood. To more fully reveal the function of NdgR, phospholipid fatty acid (PLFA) analysis with gas chromatography-mass spectrometry (GC-MS) was used to assess the effects of an ndgR deletion mutant of Streptomyces coelicolor. The deletion of ndgR was found to decrease the levels of isoleucine- and leucine-related fatty acids but increase those of valine-related fatty acids. Furthermore, the defects in leucine and isoleucine metabolism caused by the deletion impaired the growth of Streptomyces at low temperatures. Supplementation of leucine and isoleucine, however, could complement this defect under cold shock condition. NdgR was thus shown to be involved in the control of branched-chain amino acids and consequently affected the membrane fatty acid composition in Streptomyces. While isoleucine and valine could be synthesized by the same enzymes (IlvB/N, IlvC, IlvD, and IlvE), ndgR deletion did not affect them in the same way. This suggests that NdgR is involved in the upper isoleucine and valine pathways, or that its control over them differs in some respect.
The efficiency of the luminal fermentation process influences overall efficiency of luminal production, animal health and reproduction. Ruminant production systems have a significant impact on the global environment, as well. Animal wastes contribute to pollution of the environment as ammonia volatilized to the air and nitrate leached to ground water. Microbial protein synthesis in the rumen satisfies a large proportion of the protein requirements of animals. Quantifying the microbial synthesis is possible by using markers for lumen bacteria and protozoa such as nucleic acids, purine bases, some specific amino acids, or by isotopic $^{15}N,^{32}P,\;and\;^{35}S$ labelled feeds. All those methods require cannulated animals, they are time-consuming and some methods are very expensive as well. Many attempts have been made to find an alternative method for indirect measurement of microbial synthesis in intact animals. The present investigations aimed to assess possibilities of NIRS for prediction of purine nitrogen excretion and ruminal microbial nitrogen synthesis by NIR spectra of urine. Urine samples were collected from 12 growing sheep,6 of them male, and 6- female. The sheep were included in feeding experiment. The ration consisted of sorghum silage and protein supplements -70:30 on dry matter basis. The protein supplements were chosen to differ in protein degradability. The urine samples were collected daily in a vessel containing $60m{\ell}$ 10% sulphuric acid to reduce pH below 3 and diluted with tap water to 4 liters. Samples were stored in plastic bottles and frozen at $-20^{\circ}C$ until chemical and NIRS analysis. The urine samples were analyzed for purine derivates - allantoin, uric acid, xantine and hypoxantine content. Microbial nitrogen synthesis in the lumen was calculated according to Chen and Gomes, 1995. Transmittance urine spectra with sample thickness 1mm were obtained by NIR System 6500 spectrophotometer in the spectral range 1100-2500nm. The calibration was performed using ISI software and PLS regression, respectively. The following statistical results of NIRS calibration for prediction of purine derivatives and microbial protein synthesis were obtained.(Table Omitted). The result of estimation of purine nitrogen excretion and microbial protein synthesis by NIR spectra of urine showed accuracy, adequate for rapid evaluation of microbial protein synthesis for a large number of animals and different diets. The results indicate that the advantages of the NIRS technology can be extended into animal physiological studies. The fast and low cost NIRS analyses could be used with no significant loss of accuracy when microbial protein synthesis in the lumen and the microbial protein flow in the duodenum are to be assessed by NIRS.
본 연구는 경기도 중소하천에서 하상의 부착미생물군집의 시간에 따른 생장 변화와 부착미생물군집의 성장에 따른 불소의 제거 효율을 파악하기 위해 수행하였다. 방류수가 유입되는 지점인 P1과 그 곳에서 약 2 km 유하한 하류 지점인 P2에서 12주 동안의 현장 모니터링을 실시하고, 조사 지점의 수질과 부착미생물군집의 생체량 분석을 진행하였다. 부착미생물군집의 성장량은 7차 조사 시점까지 증가하다 이후 탈리현상이 발생하는 것으로 관찰되었으며, 부착미생물군집의 성장에 영향을 미치는 요인으로는 유량, 유속, 영양물질(질소, 인) 등으로 조사되었다. 한편, 부착미생물군집 체내의 불소 함유량 또한 7차 조사 시기까지 증가하다 8차 시점부터 감소하는 경향을 보였다. 이는 부착미생물군집의 탈리현상에 의해 불소의 함유량 또한 감소하였음을 사사한다. 이를 통해 하천 관리의 방안으로서 부착미생물군집의 활용법에 대한 평가 및 관리에 기여할 수 있을 것으로 기대된다. 부착미생물군집을 활용한 기법의 적용 시에는 부착미생물군집의 성장에 영향을 미칠 수 있는 요인에 대한 기초 조사 및 탈리 시점에 따른 부착판 교체 방안 등이 포함되어야할 것으로 사료된다.
A commensal thermophile, Symbiobacterium toebii, isolated from hay compost (toebii) in Korea commensally interacted with a thermophilic Geobacillus toebii sp. nov., which was a new species within the genus Geobacillus on the basis of the phenotypic traits and molecular systematic data. S. toebii required the crude extracts and/or culture supernatant of the Geobacillus toebii for axenic growth and could grow on the temperature between 45 and $70^{\circ}C$ (optimum: $60^{\circ}C$; 2.4 h doubling time) and pH 6.0 and 9.0 (optimum: pH 7.5). The G+C content of the genomic DNA was $65 mol\%$, and the major quinones were MK-6 and MK-7. A phylogenetic analysis of its 16S rDNA sequence indicated that Symbiobacterium toebii was closely related with solely reported Symbiobacterium thermophilum. The presence of the commensal thermophile 16S rDNA and accumulation of indole in all the enriched cultures indicate that Symbiobacterium toebii is widely distributed in the various soils. The genome of S. toebii constituted a circular chromosome of 3,280,275 base pairs and there was not an extra-chromosomal element (ECE). It contained about 4,107 predicted coding sequences. Of these protein coding genes, about $45.6\%$ was encoded well-known proteins and annotated the functional assignment of 1,874 open reading frames (ORFs), and the rest predicted to have unknown functions. The genes encoding thermostable tyrosine phenol-lyase and tryptophan indole-lyase were cloned from the genomic DNA of S. toebii and the enzymatic production of L-tyrosine and L-tryptophan was carried out with two thermostable enzymes overexpressed in recombinant E. coli.
갯벌 퇴적토는 미생물 다양성이 매우 높다고 알려져 있다. 하지만 인위적인 교란에 의해 갯벌 퇴적토 내 미생물 다양성이 달라질 수 있다. 지속적인 퇴적토 내 미생물 다양성 모니터링을 위해서는 대표성을 지닌 시료의 채취가 중요하다. 본 연구에서는 강화도 여차리 갯벌을 대상으로 식생이 있는 지역과 식생이 없는 지역의 미생물 다양성과 이화학적 특성치를 지구 통계학적으로 비교분석 하였다. 갯벌 시료에서 측정된 미생물 다양성과 다양한 이화학적 특성치를 지구통계학적으로 분석한 결과, 식생이 존재하는 지역에서의 상관거리가 식생이 존재하지 않는 지역에 비하여 대체로 짧다는 것을 알 수 있었다. 이는 식생이 존재하는 지역의 높은 생태학적 및 이화학적 복잡성과 이질성으로 인해 식생이 존재하는 지역에서는 식생이 존재하지 않는 지역에서 보다 비교적 좁은 간격으로 시료를 채취해야 한다는 것을 의미한다. 데이터 마이닝 기법을 사용하여 미생물 다양성에 영향을 주는 주요 환경영향 인자를 분석한 결과, 식생이 존재하는 지역에서는 유기물 함량과 질산염이온, 식생이 존재하지 않는 지역에서는 pH와 황산염이온이 미생물 다양성에 영향을 끼친다는 것을 알 수 있었다. 이러한 지구통계학 및 데이터 마이닝 분석 결과들을 활용해서 갯벌 퇴적토 내 미생물 다양성 측정을 위한 시료 채취 간격 및 개수 선정 지침을 본 연구에서 제안하였다.
Phytophthora sp.의 균사성장을 특이적으로 저해하는 토양 미생물인 YNB54 균주의 정확한 분류적 위치를 밝히기 위해 Biolog GN2, API 20E와 같은 상업적 생화학 kit, 16S rDNA, DAN-DNA hybridization, GC함량, MIDI 등의 분석을 수행하였다. 다양한 생화학적 kit를 사용한 동정 결과는 이 균주가 다른 어떤 종보다 Enterobacter cloacae와 E. cancerogenus에 보다 더 가까움을 보여주었다. 또한 DAN-DNA hybridization, GC함량, MIDI 분석의 결과들 역시 다른 속 (Citerobacter, Klebsiella, Leclercia)보다 Enterobacter 속에 더 유사함을 암시해 주었다. 그러나 16S rDNA분석에서 이 균주는 Citrobacter freundii(99.4%)와 동일 그룹으로 구분되었지만 Enterobacter, Leclecia, Klebsiella 속 등과도 98%이상의 상동성을 보여주는 polyphyletic 특성을 보였다. 결론적으로 YNB54의 분류 동정을 위한 우리의 조사들은 이 균주가 유전적으로 다양하고 지금까지 아는 것보다 분류학적으로 더 복잡함을 암시해줌에도 불구하고 Enterobacter속임이 가장 유력하다는 것을 보여 주었다.
현재 토양 생태에서 토양미생물은 유기물 분해, 질소 순환, 식물의 질소 이용 등 중요한 역할을 하고 있어, 토양 내 미생물 다양성을 분석하기 위한 연구는 지속적으로 진행되어 오고 있다. 본 연구에서는 논 토양의 미생물 생태 다양성을 조사하기 위한 효과적인 방법으로 denaturing gradient gel electrophoresis (DGGE)를 적용하고자 본 연구를 수행하였다. 논 토양 미생물의 DNA를 분리하기 위하여 lysis buffer method, skim milk bead method, sodium phosphate buffer method, Epicentre SoilMaster DNA extraction kit (Epicentre, USA), Mo Bio PowerSoil kit (Mo Bio, USA)를 이용하여 토양 내 gDNA 최적 추출방법을 확인하였다. 그 결과 Mo Bio PowerSoil kit를 사용하였을 때 Shannon 다양성지수가 세균 3.3870, 진균 3.6254으로 미생물 다양성 분석시에 가장 효과적이었다. DGGE 분석을 위한 조건은 세균의 경우 6% polyacylamide gel, 45-60% denaturing gradient였고, 진균의 경우 6% polyacrylamide gel, 45-80% denaturing gradient에서 최적 분석조건을 보였다. 위의 분석법을 적용하여 논 토양내의 미생물 군집의 변화를 살펴보면 시간의 변화 요인에 의해 미생물 변화가 일어나는 것을 알 수 있었다. 본 연구에서 사용된 DGGE 분석법을 통해 논토양 미생물의 분석 가능성을 제시 할 수 있었다.
미생물 유전체(genome)들 사이의 보존된 유전자 (con-served gene)를 밝히는 것은 생명의 본질을 이해하는데 있어 다양한 의미를 갖는다고 할 수 있을 것이다. 본 연구에서는 보존적 유전자를 찾아내고, distance value를 이용하여 구한 보존성의 정도 C(conservation score)를 이용하여 종간의 유전자 변이의 정도를 단백질 관점에서 분석하였다. 분석에 사용된 자료는 COGs 데이티베이스의 총 43종의 미생물 유전체들이며, 이들은 총 n,009개의 유전자들을 포함하는 3,852 개의 ortholog들로 구성되어있었다. 분석 결과 43종의 미생물 유전체에 대하여 총 $\ulcorner$2개의 유전자들이 보존적인 것으로 나타났으며, 이들 중 72.2%인 52종의 유전자가 단백질 합성에 관련되는 것으로 나타났다. 이들 보존적 유전자들에 대하여 보존성의 정도 C를 계산하여 보존성의 순위를 얻었으며, 가장 잘 보존된 유전자는 CTPase-trans-lation elogation factor (COG0050)로 나타났다. 그리고 72개의 보존적 유전자가 나타내는 CU 모두를 이용한 분석결과 고세균(archaea)과 진정세균(bacteria)이 각각 독자적인 그룹을 형성하는 것을 관찰하였다. 본 연구의 결과에서 도출한 72개의 보존적 유전자는 생명체의 본질적 기능에 중요한 역할을 담당하는 것으로 사료되었고, 생명체의 진화 과정에서 이 유전자들이 보존된 이유와 기능적 연계에 대한 생물학적 연구에 기초 자료를 제공할 것으로 판단되어 진다.

Nivalenol (NIV) and deoxynivalenol (DON) are predominant Fusarium-producing mycotoxins found in grains, which are mainly produced by Fusarium asiaticum and F. graminearum. NIV is found in most of cereals grown in Korea, but the genetic basis for NIV production by F. asiaticum has not been extensively explored. In this study, 12 genes belonging to the trichothecene biosynthetic gene cluster were compared at the transcriptional level between two NIV-producing F. asiaticum and four DON-producing F. graminearum strains. Chemical analysis revealed that time-course toxin production patterns over 14 days did not differ between NIV and DON strains, excluding F. asiaticum R308, which was a low NIV producer. Both quantitative real-time polymerase chain reaction and Northern analysis revealed that the majority of TRI gene transcripts peaked at day 2 in both NIV and DON producers, which is 2 days earlier than trichothecene accumulation in liquid medium. Comparison of the gene expression profiles identified an NIV-specific pattern in two transcription factor-encoding TRI genes (TRI6 and TRI10) and TRI101, which showed two gene expression peaks during both the early and late incubation periods. In addition, the amount of trichothecenes produced by both DON and NIV producers were correlated with the expression levels of TRI genes, regardless of the trichothecene chemotypes. Therefore, the reduced production of NIV by R308 compared to NIV or DON by the other strains may be attributable to the significantly lower expression levels of the TRI genes, which showed early expression patterns.

회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
