2010년산 맥류의 붉은곰팡이병 발생 및 Fusarium 곰팡이독소 자연발생 (Natural Occurrence of Fusarium Head Blight and Its Mycotoxins in 2010-harvested Barley and Wheat Grains in Korea)

  • 2010년산 쌀보리 43시료, 겉보리 17시료, 맥주보리 11시료, 청보리 14시료, 밀 41시료 등 총 126개 시료를 농가로부터 채집하였다. 이들 시료를 대상으로 Fusarium균 오염정도와 이들 균이 생산하는 주요 독소인 DON, NIV, ZEA의 자연발생을 조사하였다. 2010년산 맥류의 전국 평균 오염율은 23.0%로 2009년 전국 평균 오염율(Yeh 등, 2010)에 비해 10% 이상 높았다. 지역별 오염율은 전남지방의 맥류가 35.8%로 가장 높았고 전북 20.8%, 경기 20.5% 순이었다. 각 독소별 오염빈도와 오염수준을 조사한 결과, DON은 43시료(34%), 0.89 mg/kg 이었고, NIV는 107시료 (84.9%), 1.86 mg/kg, ZEA은 13시료(10.3%), 0.06 mg/kg이 었다. NIV의 오염빈도가 DON과 ZEA에 비해 높았다. 한편 40시료(31.7%)가 DON과 NIV가 중복오염되어 있었다. 맥류 종류별 곰팡이독소의 오염수준을 보면, 밀에서는 DON의 오염수준이 NIV보다 높았으며, 보리에서는 NIV 오염수준이 DON보다 높았다. 보리 종류별 NIV의 오염수준은 청보리, 맥주보리, 쌀보리, 겉보리 순으로 높았다. 독소오염에 대해 지역별로 분석하여보면 경기, 충남북의 중부지방은 DON의 오염수준이 NIV보다 높았으며, 전남북과 경남북의 남부지방은 NIV의 오염수준이 높았다. DON과 NIV 독소의 오염빈도 또한 중부지방에 비해 남부지방이 많았다. 지역별 맥류 곰팡이독소 자연발생 양상의 차이를 처음으로 보고한다.

돈분뇨 슬러리 퇴비화시 부재료 절감형 시설 연구 (Studies on Swine Slurry Composting Facilities with Curtailment of Bulking Agents)

  • This study was carried out to estimate the economic impacts on operation cost and curtail the bulking agent between two kinds of plants in swine farms. Bulking agents and Plants have a variety of roles in the fields of the composting for livestock manure and also represent an economic problem in terms of plant operation costs and compost production. Two farms which have rotary(size of reactor : 10${\times}$35${\times}$1.5m) or bucket(size of reactor : 10${\times}$68${\times}$2m) plants were used for 24 weeks for bucket conveyor system, which are composed of refilling rice hull as a bulking agent every 3 weeks till decreasing volume, for 4 weeks for rotary conveyor system, which has continuous compost production system without refilling rice hull, respectively. Composts was produced in 24 weeks in the bucket conveyor system, in 4 weeks in the rotary conveyor system, respectively. The results are as follows : 1. The tissue change of Rice hull at the composts of 45 days pursuant to composting steps was more crumbling in bucket conveyor system than in rotary conveyor system. 2. Microbial counts of the final composts for safety and quality showed that total bacteria counts was 1.01${\times}$108 cfu/g in bucket conveyor system, 2.82${\times}$108 cfu/g in rotary conveyor system, Salmonella was 0.3${\times}$102 cfu/g in bucket conveyor system, 7.6${\times}$102 cfu/g in rotary conveyor system, colifom bacteria was 0.5${\times}$106 cfu/g in bucket conveyor system, 1.5${\times}$106cfu/g in rotary conveyor system, fungi 1.24${\times}$106 cfu/g in bucket conveyor system, 0.01${\times}$106 cfu/g in rotary conveyor system, respectively. However, Any system used in this trial could not be met the regulation of A grade compost of EPA and USA. 3. C:N ratio according to the composting was more rapidly changed in bucket conveyor system with 64.5 of 5 days compost to 25.4 of final products than in rotary conveyor system with 26.7 of 5 days compost to 25.9 of final products. 4. Based on the mechanical characteristics of plants used in trial and compared with Rotary conveyor system, the Bucket conveyor system in which has 0.72 ㎥/㎥ of bulking agent capacity per slurry could be curtailed 1.78 ㎥of rice hull for disposal of waste, 1㎥. It was proper facilities to produce composts quantitative in Rotary conveyor system, and to treat waste quantitative and obtain good results in compost quality in Bucket conveyor system.

저장조건에 따른 전통고추장의 미생물 및 이화학적 특성 변화 (Effect of Storage Conditions on the Microbiological and Physicochemical Characteristics of Traditional Kochujang)

  • 고추장의 저장성 향상을 목적으로 숙성이 완료된 고추장에 마늘이나 알콜, 키토산, K-sorbate, 겨자를 첨가하거나 저온살균 처리를 하여 포장하고 $30^{\circ}C$에서 24주간 저장하면서 미생물과 이화학적 특성의 변화를 비교하였다. 수분활성도는 저장 12주 이후에 감소하나, 점조성은 저장 중 증가하였고 겨자와 마늘 첨가구에서 높았다. 고추장의 색도는 저장 중에 a 와 b값의 감소가 심하였고, ${\Delta}E$값의 증가는 키토산 첨가구에 비하여 알콜 첨가구에서 높았다. 가스발생은 저장 20일 이내에 대조구와 키토산 첨가구에서 심하였다. Capsanthin의 감소는 알콜이나 마늘, 겨자 첨가 고추장에서 심하였다. 고추장 중의 효모와 호기성 세균수는 저장 12주 전후까지 증가하나 이후 감소하였고, 효모수는 K-sorbate와 알콜 첨가구에서 적은 경향이었다. 고추장의 ${\alpha}-amylase$ 활성은 저장 중 감소하여 미약하였으나 ${\beta}-amylase$ 활성은 알콜이나 마늘, K-sorbate, 저온살균 고추장에서 높았고, protease 활성은 저장 중 낮았고 불규칙한 증감을 보였다.

일반인과 식품취급자의 손 위생관리에 관한 비교 (A Study on Hand Hygiene Practices: A Comparison of Food Handlers with General Population)

  • Objectives: This study was performed to investigate hygienic behavior of food handlers and general population focusing on awareness of hand-washing and the microbial load of their hands. Methods: A questionnaire survey and microbiological analysis were carried out for sixty-four people each. Samples for microbiological analysis were collected through the glove-juice method from the hands, and were analyzed for the presence of aerobic plate counts, total coliform, fecal coliform, Escherichia coli, Staphylococcus aureus, and Salmonella spp. according to the Food Code of Korea. Results: In the survey, significant differences between the food handlers and general population (p < 0.01) were found in hand-washing frequency, duration, use of hand-washing agents, washing parts of hands, hand-drying method, and method of turning off water. In eight different situations among the ten particular situations in their daily life, more food handlers responded to wash their hands than general population (p < 0.05). Bacterial load on hands with general population was consistently higher than with food handlers (p < 0.05), however, percentages of positive hands of S. aureus and Salmonella spp. were not. Conclusions: Poor hand hygiene practices were indicated by the positive results for E. coli, S. aureus, and Salmonella spp. on the hands of some respondents in both groups. This study reveals that there is the need for programs or campaigns to increase hand-washing practices of both groups.

Peptidoglycans Promotes Human Leukemic THP-1 Cell Apoptosis and Differentiation

  • The innate immune system coordinates the inflammatory response to pathogens. To do so, its cells must discriminate self from non-self utilizing receptors that identify molecules synthesized exclusively by microbes. Toll-like receptors have a crucial role in the detection of microbial infection in mammals and insects. In mammals, they have evolved to recognize conserved products unique to microbial metabolism. These include lipopolysaccharide (LPS), lipotechoic acids, and peptidoglycans (PGN). We show here that TLRs, including TLR2, are expressed on the THP-1 human leukemia cell line. Activation of TLR2 signaling in THP-1 by PGN induces the synthesis of various soluble factors and proteins including interleukin-$1{\beta}$, interleukin-8 and TNF-${\alpha}$ and apoptosis of THP-1 with PGN dose and time dependence. Moreover, in this study we show that PGN induces apoptosis of THP-1 cells in a TNF-${\alpha}$-dependent manner. These findings indicate that TLR2 signaling results in a cascade leading to tumor apoptosis and differentiation, which may suggest new clinical prospects using TLR2 agonists as cytotoxic agents in certain cancers.

신갈나무 잎 에탄올 추출물의 식품보존제 효과 (Effect of Ethanol Extract of Quercus mongolica Leaf as Natural Food Preservative)

  • 본 연구는 독성과 안정성의 문제가 없는 천연식품 보존제의 개발을 위하여 신걸나무 잎 추출물을 이용하여 천연보존제로서의 특성 및 간으성을 조사하였다. 신갈나무 잎 에탄올 추출물은 그람양성균에 대해서 $250\;\mu\textrm{g}/mL$, 그람음성균에 대해서는 $500\;\mu\textrm{g}/mL$ 농도에서 40시간 동안 식중독균의 증식을 억제하였다. 신갈나무 잎 추출물의 항균활성 물질은 pH와 열에 매우 안정하였다. 신갈나무 잎 에탄올 추출물과 합성보존제가 솔잎 음료, 당근 쥬스, 된장 및 막걸리의 보존성에 미치는 영향을 비교한 결과, 솔잎 음료 및 당근 쥬스의 경우 0.1% 첨가시 합성보존제와 유사한 미생물 생육억제 효과를 나타내었고, 된장은 추출물을 2~3% 첨가하였을 경우 7주까지는 미생물 중식이 억제되어 에틸알코올과 비슷한 보존효과를 보였다. 막걸리의 경우는 추출물 0.5% 이상 첨가시 합성보존제인 안식향산과 유사한 미생물 생육 억제효과를 나타내었다. 따라서 이와같은 결과로 미루어 보아 신갈나무 잎은 향후 천연보존제의 원료로서 사용할 수 있을 것으로 기대된다.

Helicobacter pylori의 생육에 영향을 미치는 성분과 식품첨가물에 관한 연구 (Studies on Food Components and Food Additives Affecting the Growth Patterns of Helicobacter pylori)

  • The purpose of this study was to choose prinicipal food components contained in diet foods and food additives used for manufacturing processed foods and elucidate their in vivo effects on the growth pattern of Helicobacter pylori. To do this the antibacterial effects of various sources of carbon nitrogen and mineral as an effect agent on Helicobacter pylori were first assessed based upon bacterial growth degree. results show that the source of carbon tested had different effects on bacterial growth of Helicobacter pylori. It was revealed that a promotional effect of monosaccharides resulted in enhanced growth of Helicobacter pylori compared with disaccharides and polysacchrides, in particular glucose was observed to be most effective in growth of Helicobacter pylori among monosaccharides teste whereas mannose to hinder the growth of Helicobacter pylori. Polyols such as sorbitol mannitol maltitol and xylitol was however observed to show no promotion or suppression effect on growth of Helicobacter pylori. Apart from these the sources of amino acid and inorganic nitrogen were chosen and tested to assess the promotion or suppression effect of nitrogen sources on growth of helicobacter pylori. It was found that amino acid such as lysine showed its promotion effect on the growth of Helicobacter pylori while arginine (NH4)2SO4 and NH4Cl showed no effect on its growth. Ammoia and urea were however observed to have a positive effect on the growth of Helicobacter pylori. Among these effect agents lysine and methionine were revealed to show the most positive effect on growth of Helicobacter pylori. Minerals such as MgSO4 KH2PO4 and MgCl2 appered to exert their positive growth effects whereas CaCl2 and CaSo4 had a little effect. In addition FeSO4 FeCl2 and FeCl3 brought suppression on the growth of helicobacter pylori. In studies of the growth of Helicobacter pylori by food additives ascorbic acid showed extreme suppression on its growth,. Sodium nitrate and sodium chloride were also found to be of negative effect on the growth of Helicobacter pylori in rder of degree whereas tocopherol had nothing to do with microbial growth.

Screening the Antibacterial Activities of Streptomyces Extracts against Phytopathogens Xanthomonas oryzae pathovar oryzae, Xanthomonas campestris pathovar vesicatoria, and Pectobacterium carotovorum pathovar carotovorum

  • Xanthomonas oryzae pv. oryzae (Xoo), X. campestris pv. vesicatoria (Xcv), and Pectobacterium carotovorum pv. carotovorum (Pcc) are the causative agents of bacterial blight in rice, bacterial spot in pepper, and bacterial soft rot in carrot and cabbage, respectively. To isolate novel microbial extracts with antimicrobial activities against these bacteria, approximately 5,300 different Streptomyces extracts were prepared and tested. Microbial cultures from various Streptomyces strains isolated from the Jeju Island, Baekam, Mankyoung river, Jiri mountain etc. in Korea were extracted into three different factions -secreted hydrophobic, secreted hydrophilic, and mycelia- using ethyl acetate, water, and methanol. Initially, 34, 29, and 10 extracts were selected as having antibacterial activities against Xoo, Xcv, and Pcc, respectively. Extracts 1169G4, 1172E9, and 1172E10 had the highest growth inhibition activities against both Xoo and Xcv, and extracts 1151H7 and 1152H7 showed the highest growth inhibition activities against Pcc.

NF-κB 조절을 통한 오매추출물의 항염효과 및 작용기작에 관한 연구 (Study on the Anti-inflammatory Effect and Mechanism of Prunus mume Extract Regarding NF-κB)

  • NF-${\kappa}B$ is a transcriptional factor which is involved in many biological processes including immunity, inflammation, and cell survival. Many investigators studied on the mechanism involved in activation of NF-${\kappa}B$ signalling pathway via ubiquitination and degradation of $I{\kappa}B$ regarding skin disease. Some specific molecules including Akt, MEK, p38 MAP Kinase, Stat3, et al. represent convergence points and key regulatory proteins in signaling pathways controlling cellular events such as growth and differentiation, energy homeostasis, and the response to stress and inflammation. Ultraviolet (UV) irradiation has many adverse effects on skin, including inflammation, alteration in the extracellular matrix, cellular senescence, apoptosis and skin cancer. Prunus mume, a naturally derived plant extract, has beneficial biological activities as blood fluidity improvement, anti-fatigue action, antioxidative and free radical scavenging activities, inhibiting the motility of Helicobacter pyolri. Previous reports on various beneficial function prompted us to investigate UVB-induced or other immunostimulated biological marker regarding P. mume extract. P. mume extract suppresses UVB-induced cyclooxygenase-2 (COX-2) expression in mouse skin epidermal JB6 P+ cells. The activation of activator protein-1 and nuclear factor-${\kappa}B$ induced by UVB was dose-dependently inhibited by P. mume extract treatment. This results suggest that P. mume extracts might be used as a potential agents for protection of inflammation or UVB induced skin damage.

토양 방선균 Streptomyces platensis YK-2가 생산하는 Transglutaminase의 정제 및 효소학적 특성 (Purification and Characterization of Transglutaminase from a Newly Isolated Streptomyces platensis YK-2)

  • 본 연구는 transglutaminase 생산능이 우수한 토양유래방선균 strain YK-2를 TGase 최적생산배지에서 $28^{\circ}C$, 5일간 배양하여 얻은 배양여액으로 본 효소의 정제 및 정제된 효소의 효소화학적 특성에 관하여 검토한 것이다. 본 효소의 정제는 50% methanol precipitation, DEAE-Sephadex column chromatography의 정제 절차를 거쳐 56.5%의 수율로 정제되었고, 정제된 효소의 순도는 12.5% SDS-PAGE에서 단일 밴드를 나타내어, 서브유닛트의 분자량이 약 45,000 dalton으로 추정되는 호모형 효소인 것을 알 수 있었다. 정제된 TGase의 생화학적 제 특성을 검토한 결과, 등전점은 pH $6.0{\sim}7.0$ 부근에 있는 것으로 나타났으며, 본 효소의 기질인 CBZ-L-Gln-Gly 농도에 대한 Km치는 18.5 mM으로 추산되었다. 또한 금속이온 및 저해제의 영향으로는 $Hg^{++}$에 의해서 본 효소의 활성이 강하게 저해되었으나, DTT 및 mercaptoethanol에 의해 각각 293% 및 219% 활성이 증가하였다.
