Viability and Luciferase Activity of Freeze-Dried Recombinant Biosensor Cells for Detecting Aromatic Hydrocarbons

  • Aromatic hydrocarbons are of major concern among genotoxic chemicals due to their toxicity and persistence. Some microorganisms can utilize aromatic hydrocarbons as carbon and energy sources by inducing expression of catabolic operon(s). The XylR regulatory protein activates transcription of the catabolic enzymes to degrade BTEX (benzene, toluene, ethylbenzene, and xylene) from its cognate promoters, Pu and Ps upon exposure of the cells to the aromatic hydrocarbons. The activity of XylR on the promoters was previously monitored using luciferase luc reporter system. The xylR, its promoter Pr and the promoter Po for the phenolic compound catabolic operon were introduced upstream of firefly luciferase luc in the pGL3b vector to generate about 7.1 kb of pXRBTEX. Here E. coli harboring the plasmid was freeze-dried under various conditions to fin,d optimal conditions for storage and transport. The cell viability and luciferase activity were maintained better, when the cells were freeze-dried at -7$0^{\circ}C$ in the addition of the 10% skim milk or 12% sucrose. However, coaddition of protectants such as 10% skim milk plus 10% glucose or 12% sucrose plus 10% glucose, resulted in much better viability and bioluminescence activity compared with the effect of single addition of each protectant. In addition, it was shown that the freeze-dried cells maintained almost intact bioluminescent activities and cell viability for at least 1 week after freeze-drying. This work demonstrated that the properly freeze-dried recombinant bacterial cells could be utilized as a whole-cell biosensor for simple and rapid monitoring of BTEX in the environment.

응급구조(학)과 학생들의 윤리적 가치관 관련 요인 (Factors related to ethical values among some paramedic students)

  • Purpose: This study was performed to investigate the level of ethical values and its related factors among some paramedic students. Methods: The subjects of this study were 480 paramedic students from five universities located in G-metropolitan city and J province. Data were collected using a self-administered questionnaire and analyzed using t-test, ANOVA, Pearson's correlations, and multiple regression analysis. Results: The mean scores of level of ethical values, academic self-efficacy and self-esteem were $3.70{\pm}0.45$, $3.15{\pm}0.42$ and $3.60{\pm}0.70$, respectively. In the correlation analysis, ethical value level was significantly positively correlated with self-esteem (r=.195, p< .001) and academic self-efficacy (r=.156, p= .001), of domains in academic self-efficacy, self-regulation efficacy (r=.230, p< .001) and confidence (r=.105, p= .022). In multiple regression analysis, self-esteem (${\beta}=.163$, p= .026), task ease preferences (${\beta}=-.350$, p< .001) and confidence (${\beta}=.403$, p< .001) were significantly related to ethical value levels. In addition, subjects willing to participate in the bioethics education showed higher relevance to ethical values than indifferent subjects (${\beta}=2.751$, p= .003). Conclusion: Concrete and systematic education will be needed in order to foster the establishment of ethical values of the paramedic students. Additionally, to fulfill their roles and obligations as emergency medical technicians, in the field, various educational curricula based on simulated cases in the university should be introduced.

치위생과 학생의 긍정심리자본에 영향을 미치는 요인 (Factors influencing positive psychological capital of dental hygiene students)

  • Objectives: This study aimed to explore ways to improve the level of positive psychological capital of dental hygiene students. After comparing the levels of positive psychological capital, social support, and emotional regulation ability of dental hygiene students and identifying correlations, the sub-factors of social support and emotional regulation ability that affect positive psychological capital were identified. Methods: A survey of 70 questions was conducted targeting 310 dental hygiene students in Gyeonggi-do and Chungcheong-do. The results were subjected to exploratory factor analysis, frequency analysis, t-test, one-way ANOVA, correlation analysis, and multiple regression analysis using the PASW statistics 18.0 program. Results: The level of positive psychological capital was high when students had high grades and perceived positive relationships with professors and peers (p<0.001). Factors highly correlated with positive psychological capital include 'self-emotional regulation ability' among 'emotion regulation abilities' (r=0.665, p<0.001), and 'emotional support related to daily life' recognized 'social support' (r=0.635, p<0.001), followed by 'social support' (r=0.602, p<0.001). Factors affecting positive psychological capital include 'self-emotion recognized ability' (β=0.377, p<0.001), 'quality of professor relationship' (β=0.201, p<0.001), and 'emotion control ability'. Among 'social support', 'emotional support related to daily life' (β=0.201, p=0.003) and 'grades' (β=0.159, p<0.001), and among 'social support', 'social-related support' (β=0.149, p=0.016), followed by 'grade' (β=0.076, p=0.043) were identified. Conclusions: To improve the positive psychological capital of dental hygiene students, efforts should be made to improve self-emotion regulation ability, professor relationships, emotional support related to daily life, and social support.

한국형 중환자간호 분류도구 개발 (Development of Korean Patient Classification System for Critical Care Nurses)

  • Purpose: This study was performed to develop a valid and reliable Korean Patient Classification System for Critical Care Nurses (KPCSC). Methods: Tertiary and general hospitals with various levels of ICU nurse staffing were included. To verify interrater reliability, data collectors and staff nurses of 15 ICUs in 11 hospitals classified 262 patients. To verify construct validity, the staff nurses classified 457 patients according to KPCSC comparing difference by medical department and type of stay in ICU. For conversion index, 195 patients from 10 ICUs in 7 hospitals were classified and nursing time was measured by 174 nurses, 7 head nurses, 18 charge nurses, 37 nurse aids and 1 secretary. Results: The developed KPCSC has 11 categories, 82 nursing activities and 115 criterias. Reliability was found to have high agreement (r=.96). Construct validity was verified by comparing differences in medical department and type of stay in ICU. According to scores, four groups in the KPCSC were identified. One score on the KPCSC indicates 6.12 minutes of nursing time. Conclusion: The findings show that the KPCSC can be used to measure new and complex nursing demands including rehabilitation and the safety of ICU patients.

수면과다증을 동반한 심인성 요실금 환자 호전 1례 (Case Report of a Psychogenic Urinary Incontinence Patient with Hypersomnia Disorder)

  • Objective: This study evaluates the effects of traditional Korean medicine (including Guibi-tang) on a psychogenic urinary incontinence patient with hypersomnia disorder.Method: Herbal medicine, acupuncture, and moxibustion were carried out. The effect of the treatments was then evaluated via the Beck depression inventory (BDI), the Beck anxiety inventory (BAI), the symptom checklist-90-revised (SCL 90-R), the Epworth sleepiness scale (ESS), and the revised urinary incontinence scale (RUIS), as well as the number of urinations, incontinence levels, and sleeping times during the day.Results: After the treatments, both the urinary incontinence and hypersomnia symptoms showed improvement.Conclusion: This study suggests that traditional Korean medicine treatments are effective in caring for patients who suffer from psychogenic urinary incontinence with hypersomnia disorder.

피부 저항점 탐지기를 사용한 근근막 통증 유발점의 진단 (The Use of Skin Resistance Point Finder for the Diagnosis of Myofascial Trigger Points)

  • Background: At present, there is no reliable objective test for the diagnosis of myofascial trigger points (MFTP). We evaluated the usefulness of skin resistance point finder for the diagnosis of MFTP. Methods: 40 subjects with clinical MFTPs at the upper trapezius muscle were included in this study. Using skin resistance point finder (UNIQUE S-2010$^{(R)}$, Seik medical, Korea), we tried to find out the point of low skin resistance. At three different current level, sensitivity and specificity of this method for the diagnosis of clinically identified MFTP was evaluated. Pressure threshold for pain of low skin resistance point was measured using Pressure Threshold Meter$^{(R)}$ (Pain Diagnostics and Thermography, U.S.A.). 3 groups of point detected at different current were compared in mean pressure threshold. Results: Fixed single current of skin resistance point finder showed variable sensitivity and specificity. But, by adjusting current level, skin resistance point finder detected all of 40 clinically identified MFTPs. Although it is partially statistically significant, the mean pressure threshold of points detected at high current was high. Conclusion: Skin resistance finder can be used as objective diagnostic tool of MFTPs. There is possibility that it can be useful in evaluating treatment effect. However, more investigation is necessary.

고기능 자폐스펙트럼장애에서 제한된 관심과 반복적인 행동 유무에 따른 임상 양상의 차이 (Clinical Characteristics According to the Presence of Restricted, Repetitive Behaviors and Interests in Children with High Functioning Autism Spectrum Disorder)

  • Objectives : The objective of this study was to investigate the relationship between restricted, repetitive behaviors and interests (RRBI) and autistic symptoms in Korean high functioning autism spectrum disorder (ASD) children and to examine the structure of RRBI. Methods : Participants included 147 high functioning ASD subjects and 181 unaffected siblings. ASD subjects were divided into two groups based on the presence of RRBI. The domain scores of the Korean version of Autism Diagnostic Interview-Revised (K-ADI-R), Korean version of Asperger Syndrome Diagnostic Scale and total scores of Korean translated version of Social Responsiveness Scale, Korean version of Social Communication Scale were used for comparison of ASD symptoms between the groups. Eleven items from the RRBI domain of the K-ADI-R were used in principal axis factor analysis (PAF). Results : A statistically lower nonverbal IQ score was observed for ASD with RRBI than for ASD without RRBIs, and more social deficit, communication deficit, and behavioral and emotional problems were observed for ASD with RRBI compared to ASD without RRBI. Using PAF, two distinct factors were identified. 'Resistance to trivial changes in environment', 'Difficulty with minor changes in personal routine & environment', and 'Compulsion/ritual' were included as one factor. Conclusion : Analysis of the data suggests that the presence of RRBI in high functioning ASD is associated with a more severe presentation of autistic disorder. In addition, there appears to be heterogeneity within RRBI in autism except insistence on sameness.

수도권 지역 영양사와 비영양사의 영양지식, 영양태도 및 식 행동 비교연구 (A Comparative Study of Nutrition Knowledge, Nutrition Attitudes and Eating Behaviors of Dietitians and Non-Dietitians Working in the Metropolitan Area)

  • This study was carried out to compare the nutrition knowledge, nutrition attitudes and eating behaviors of dietitians and non-dietitians. The subjects of this study were 148 dietitians and 195 non-dietitians who are working in the metropolitan area. The general characteristics, nutrition knowledge, nutrition attitudes and eating behaviors of the subjects were surveyed using a self-administered questionnaire. The results were as follows: The result of anthropometric data and body mass index showed no significant differences between dietitians and non-dietitians groups. The dietitian group ($16.61{\pm}1.82$) showed a significant difference (p < 0.001) compared to the non-dietitian group ($14.60{\pm}2.17$) in nutrition knowledge scores. Nutrition attitudes of the dietitian group ($36.66{\pm}6.91$) was a little higher than the non-dietitian group ($36.21{\pm}7.95$), but not a significant difference. The dietitian group ($61.52{\pm}9.37$) and the non-dietitian group ($62.50{\pm}11.25$) were no significant difference in eating behaviors. The correlation between nutrition knowledge scores and nutrition attitudes score of dietitians and non-dietitians groups were no significantly difference and also the correlation between nutrition knowledge scores and eating behaviors scores were not statistically significance. Whereas, the correlation between nutrition attitudes and eating behaviors showed a significant difference (dietitian r = 0.40829, non-dietitian r = 0.51914). From these results, we could see the necessity of emphasis of nutrition knowledge by mending the education is necessary to enable dietitians to apply that in real life. Also it is important to guide non-dietitians to adjust their nutrition knowledge, nutrition attitudes and eating behaviors.

스켈링 실습실에 내원한 여고생의 구강건강 실태 조사 (A Study of the Actual Conditions of the Dental Health of High School Girls Visiting Scaling Practice Rooms)

  • The researcher worked with a total of 158 high school girls visiting the scaling practice room at the department of dental hygiene in Jinju Health college from November 11, 2002 to December 4, 2002, having their teeth scaled, observing their teeth and interviewing them with the aid of questionnaires. The findings can be summarized as follows. 1. The survey on the understanding of dental health and the attitudes of the subjects shows the following results : "I have never learned how to brush teeth" (65.8%: 104 girls); "I brush my teeth twice a day" (57.0%: 90 girls) : "I brush my teeth for 2 minutes"(44.3%: 70 girls): "I brush my teeth vertically and horizontally" (53.2%: 84 girls): "( brush my teeth, gum and tongue" (462%: 73 girls): "I use one brush for two months" (40.5%: 64 girls); "I don't have any floss" (582%: 92 girls): "I have never had my teeth scaled" (84.2%: 133girls) : "Smoking is bad for the teeth" (77.8%: 123 girls): "Drinking is bad for the teeth" (48.7%: 77 girls): "I have ever visited dental clinics" (92.4%: 146 girls). 2. The study on part of the teeth most exposed to caries shows the following results : Mandibular right first molar(79.8%: 126 girls): Mandibular left first molar(77.5%: 124 girls), Mandibular left second molar(70.2%: 111 girls). Mandibular right second molar(65.7%: 104 girls). Maxillary left first molar(59.5%: 94 girls): Maxillary left second molar(59.5%: 94 girls): Maxillary right first molar(58.2%: 92 girls). Maxillary right second molar(50.7%: 80 girls) 3. The statistics on the subjects' caries show the following results: DMF rate: 943%: DMFT rate: 25.61%: DMFT index: 7.17 teeth : DT rate: 4651%: FT rate: 52.25%: MT rate: 124% 4. The survey on dental caries statistics, dental health, dental health behavior, and the subjects' attitudes shows the following results: The worse the subjects' dental health was(r=-327, p<.001), the more they visited dental clinics(r=.195, p<.05), the higher DMT rate, DMFT index were; The more the subjects visited dental clinics(r =.359, p<.001), the higher DT rate was; The worse the subjects teeth went(r=-.197, p<.05), the higher MT rate was. It is necessary that systematic and substantial dental health education for high school students should be carried out in regard to plaque control, fluoride application, diet control, pit & fissure sealant as a part of developing national health policies designed for the dental health of the teenagers.

가바펜틴 400밀리그람 캡슐의 생물학적동등성시험 (Bioequivalence Test of Gabapentin 400 mg Capsules)

  • Gabapentin, [1-(aminomethyl) cyclohexaneacetic acid], a structural analog of $\gamma$-aminobutyric acid (GABA), is being developed for the treatment of epilepsy. Unlike GABA, gabapentin crosses the blood-brain barrier after systemic administration. Gabapentin is an effective antiepileptic drug in patients with partial and secondarily generalized seizures who are uncontrolled with use of existing anticonvulsant drug therapy. The purpose of the present study was to evaluate the bioequivalence of two gabapentin 400 mg capsules, $Neurontin^{(R)}$ capsule 400 mg (Pfizer Inc.) and Gabatin capsule 400 mg (Korean Drug Co. Ltd), according to the guidelines of the Korea Food and Drug Administration (KFDA). The release of gabapentin from the two gabapentin formulations in vitro was tested using KP VIII Apparatus II method with various dissolution media (pH 1.2, 4.0, 6.8 buffer solution and water). Twenty six healthy male subjects, 23.58$\pm$1.50 years in age and 66.74$\pm$8.31 kg in body weight, were divided into two groups and a randomized 2$\times$2 cross-over study was employed. After one capsule containing 400 mg as gabapentin were orally administered, blood was taken at predetermined time intervals and the concentrations of gabapentin in serum were determined using HPLC with fluorescence detector. The dissolution profiles of two formulations were similar at all dissolution media. In addition, the pharmacokinetic parameters such as $AUC_t$, $C_{max}$ and $T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters using logarithmically transformed $AUC_t$, $C_{max}$ and untransformed $T_{max}$. The results showed that the differences between two formulations based on the reference drug, $Neurontin^{(R)}$ capsule 400 mg, were 2.04, -3.68 and 16.79% for $AUC_t$, $C_{max}$ and $T_{max}$, respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log 0.8 to log 1.25 (e.g., log 0.91$\sim$log 1.16 and log 0.87$\sim$log 1.11 for $AUC_t$ and $C_{max}$, respectively). Thus, the criteria of the KFDA bioequivalence guideline were satisfied, indicating Gabatin capsule 400 mg was bioequivalent to $Neurontin^{(R)}$ capsule 400 mg.
