메틸유제놀에 의한 즉시형 과민 반응의 억제 (Suppression of Immediate Hypersensitivity by Methyleugenol)

  • We studied the action of methyleugenol on immediate hypersensitivity. Methyleugenol completely inhibited systemic anaphylaxis induced by compound 48/80 in mice. Methyleugenol al so inhibited local anaphylaxis induced by anti-dinitrophenyl (DNP) IgE. Moreover, methyleugenol dose-dependently inhibited histamine release in peritoneal mast cells activated by compound 48/80 or anti-DNP IgE. These results suggest that the inhibitory effect of methyleugenol on anaphylaxis induced by compound 48/80 or anti-DNP IgE is due to, in part at least, the membrane stabilization of mast cells.

  • $^1H-NMR,\;^{13}C-NMR$, Dept. NMR의 spectrum으로 compound 1은 safrole로, compoud 2은 o-methyleugenol로 추정(推定)되었으며 처리농도별(處理濃度別) 발아억제(發芽抑制) 효과조사(效果調査)에서 발아억제 물질로 추정한 safrole은 25mg/ml의 처리에서도 발아 및 생육억제 효과를 나타내지 않았다. o-methyleugenol의 경우는 5mg/ml의 처리(處理)로 최종발아(最終發芽)는 무처리구에 비해 58% 저해하였으며, 그리고 25mg/ml에서도 92%, 생육(生育)에는 100%의 저해도(沮害度)를 나타내었다. 이상(以上)의 결과(結果)를 보면 o-methyleugenol은 세신의 강(强)한 발아저해활성 물질의 하나로 처음 확인되었으며 천연(天然) 제초제(除草劑)로서의 개발(開發) 가능성(可能性)은 추후 재검증(再檢證)해 보아야 하겠다.

세신의 재배년차에 따른 근수량 및 정유성분 함량변화 (Changes of Root Yield and Essential Oil Content by Cultivated Years in Asaram siebold Mio)

  • 야생 약초인 세신의 재배년수별 생육특성과 수량성을 구명하고 주요 정유 성분인 methyleugenol과 safrole함량 변화를 조사하여 세신의 상품적 가치를 높이고 품질의 규격화를 위한 기초자료를 얻고자 1993년부터∼1997년까지 5년간에 걸쳐 시험을 실시하였던 결과를 요약하면 다음과 같다. 1. 재배년차별 생육은 재배년수가 경과함에 따라 지하부 생육의 신장차이가 크게 나타났으며 특히 근장의 크기에 따라 수량변화에 영향을 미쳤다. 2. 10a당 건근 수량은 3년재배(127kg/10a)에 비하여 4년재배 191%, 5년재배 259% 순으로 증수되었다. 3. 재배기간별 정유함량은 재배 기간에 따라 3년 5%, 4년 4.5%, 5년은 4.3% 순이었다. 4. 주요 정유 조성성분인 methyleugenol은 재배년수가 길어질수록 증가하였으나 safrole함량은 점차 감소하여 methyleugenol과 safrole 함량은 서로 상반된 관계를 나타내었다.

SPME-GC-MS를 이용한 담배와 관련된 향료의 분석 (Analysis of Flavor-related Compounds from Tobacco using SPME-GC-MS)

  • 담배에 포함된 향료 성분을 headspace solid phase microextraction(SPME)를 이용하여 gas chromatography-mass spectrometry(GC-MS)의 selected ion monitoring(SIM)방법으로 분석하였다. 본 연구에서 사용한 담배향과 관련된 성분은 estragole, pulegone, trans-anethole, safrole, piperonal, eugenol, methyleugenol, coumarin, trans-isoeugenol, trans-methylisoeugenol 및 myristicin 등 11종이며 분석결과 모든 담배에서 한가지 또는 그 이상의 담배향 성분을 $0.001-1.3{\mu}g/g$ 검출할 수 있었으며 회수율은 89.1-102.9%로 나타났고 상대표준편차는 2.6-25.2%를 얻었다.

Toxicological Studies on the Essential Oil of Eugenia caryophyllata Buds

  • The essential oil (EC-oil) obtained from the buds of Eugenia caryophyllata (Myrtaceae) was examined for its free radical-scavenging activity, cytotoxicity, and in vivo toxicity. To find the xenobiotic properties of EC-oil, serum thiobarbituric acid reactive substances (TBARS) level and hepatic drug-metabolizing enzyme activities were measured. It was found that EC-oil displayed xenobiotic properties like bromobenzene. The cytotoxicities of eugenol and of the EC-oil were greatly attenuated by the sulfhydryl-containing N-acetyl-L-cysteine (NAC), suggesting that eugenol was susceptible to nucleophilic sulfhydryl. In addition, eugenol also showed potent free radical-scavenging activity in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Moreover, methyleugenol considerably exhibited less cytotoxicity and less potent free radical-scavenging activity than eugenol, and the cell viability of the methyleugenol was more increased with NAC treatment than the eugenol. These results indicate that the phenolic OH in eugenol may play a crucial role in both cytotoxicity and free radical-scavenging activity. The fashion on oxidative stress and hepatic drug-metabolizing enzyme activities of eugenol resembled those of bromobenznene.

  • Eight compounds were isolated from the roots of Asarum sieboldii and their structures were identified as $({\pm})$-car-3-ene-2,5-dione (1), (-)-asarinin (2), (-)-sesamin (3), eucarvone (4), methyleugenol (5), ${\gamma}-asarone$ (6), pellitorine (7) and asarinol A (8) by analysis of spectral data. Among them, (-)-asarinin (2) showed the most potent inhibitory effect on melanogenesis, with inhibition rate of 66%.

증숙 과정을 통한 소청룡탕의 뇌신경세포 보호 활성 변화에 관한 연구 (Neuroprotective Activity of So Cheong Ryong Tang with Steaming Process)

  • So Cheong Ryong Tang (SCRT) has been used as a traditional herbal medicine for the treatment of the bronchial asthma. In this study, the effect of steaming process on changes in 11 major compounds (homogentisic acid, ephedrine HCl, paeoniflorin, cinnamic acid, cinnam aldehyde, glycyrrhizin, 6-gingerol, schizandrin, methyleugenol, gomisin A and gomisin N) content and neuroprotective activity of SCRT were evaluated. Major compound content was slightly different with steaming produce. The contents of paeoniflorin, cinnamic acid, cinnam aldehyde, 6-gingerol and methyleugenol were decreased and homogentisic acid, ephedrine HCl, glycyrrhizin, schizandrin and gomisin A were increased by steaming process. The neuroprotective activity of steaming SCRT was determined in mouse hippocampal HT22 cells by MTT assay. As a result, neuroprotective activity of a steaming SCRT was higher than that of a non steaming SCRT. This study demonstrated that neuroprotective activity of SCRT can be improved through steaming process.

Development of Fingerprints for Quality Control of Acorus species by Gas Chromatography/Mass Spectrometry

  • An effective analytical method of gas chromatography/mass spectrometry (GC/MS) was developed for the rapid determination of essential oils in the crude extract of Acorus species (Acorus gramineus, Acorus tatarinowii, and Acorus calamus). Major phenypropanoids (${\beta}$,${\alpha}$-asarone isomers, euasarone, and methyleugenol) and ${\beta}$-caryophyllene in Acorus species were used as marker compounds and determined for the quality control of herbal medicines. To extract marker compounds, various extraction techniques such as solvent immersion, mechanical shaking, and sonication were compared, and the greatest efficiency was observed with sonication extraction using petroleum ether. The dynamic range of the GC/MS method depended on the specific analyte; acceptable quantification was obtained between 10 and 2000 ${\mu}g/mL$ for ${\beta}$-asarone, 10 and 500 ${\mu}g/mL$ for ${\alpha}$-asarone, 10 and 200 ${\mu}g/mL$ for methyleugenol, and between 5 and 100 ${\mu}g/mL$ for ${\beta}$-caryophyllene. The method was deemed satisfactory by inter- and intra-day validation and exhibited both high accuracy and precision, with a relative standard deviation < 10%. Overall limits of detection were approximately 0.34-0.83 ${\mu}g/mL$, with a standard deviation (${\sigma}$)-to-calibration slope (s) ratio (${\sigma}$/s) of 3. The limit of quantitation in our experiments was approximately 1.13-3.20 ${\mu}g/mL$ at a ${\sigma}$/s of 10. On the basement of method validation, 20 samples of Acorus species collected from markets in Korea were monitored for the quality control. In addition, principal component analysis (PCA) and hierarchical cluster analysis (HCA) were performed on the analytical data of 20 different Acorus species samples in order to classify samples that were collected from different regions.

배초향 지상부에서 얻은 정유의 조성과 항균효과 (Antimicrobial Activity of the Essential Oil of the Herbs of Agastache rugosa and Its Composition)

  • 수증기 증류법으로부터 얻은 배초향(Agastache rugo-sa, Labiatae)의 정유는 부패균을 포함하는 수종의 미생물에 대하여 현저한 항균활성을 나타내었다. 배초향의 유효성분을 확인하기 위하여 정유성분을 GC-MS로 분석한 결과 GC chromatogram에서 얻은 각 피크의 reten-tion time 및 mass spectral data 의표준물질과의 비교에 의해서 isomenthone, methone, dihydrocarvone, anetho-le, vanillin, eugenol, methyleugenol, $\beta$-caryophyllene, $\beta$-caryophyllene oxide 등의 정유성분이 확인되었다. 이 연구에서 밝혀진 여러 성분들의 항균 활성은 잘 알려져 있으므로 이들이 유효성분으로 추측된다. 또한 Korean mint로 세계적으로 알려져 있는 배초향의 정유는 식품 방부제로서 적합할 것으로 생각된다.
