Immunosensor for Detection of Escherichia coli O157:H7 Using Imaging Ellipsometry

  • Imaging ellipsometry (IE) for detection of binding of Escherichia coli O157:H7 (E. coli O157:H7) to an immunosensor is reported. A protein G layer, chemically bound to a self-assembled layer of 11-mercaptoundecanoic acid (11-MUA), was adopted for immobilization of monoclonal antibody against E. coli O157:H7 (Mab). The immobilization of antibody was investigated using surface plasmon resonance. To fabricate antibody spots on a gold surface, protein G solution was spotted onto the gold surface modified with an 11-MUA layer, followed by immobilizing Mab on the protein G spot. Ellipsometric images of the protein G spot, the Mab spot, and Mab spots with binding of E. coli O157:H7 in various concentrations were acquired using the IE system. The change of mean optical intensity of the Mab spots in the ellipsometric images indicated that the lowest detection limit was $10^3$CFU/ml for E. coli O157:H7. Thus, IE can be applied to an immunosensor for detection of E. coli O157:H7 as a detection method with the advantages of allowing label-free detection, high sensitivity, and operational simplicity.

  • A new UPLC-DAD method for simultaneous determination of p-coumaric acid and trans-cinnamic acid in propolis was developed using Halo-C18 column and step gradient elution of MeCN and $0.1%H_3PO_4$ in 8 min. The method was validated by specificity, linearity, limit of detection, limit of quantification, precision(intra- and inter-day variability) and recovery tests. The validated method was sufficiently applicable for quantitative analysis of propolis products as well as propolis collected from different regions in Korea. The established method is simple, appropriate and efficient, and can be used for quality assessment of Korean propolis.

  • A novel sampling method of the headspace poly(dimethylsiloxane) (PDMS) mini-disk extraction (HS-PDE) was developed, optimized, validated and applied for the GC/MS analysis of spices flavors. A prototype PDMS mini-disk (8 mm outer diameter, 0.157 mm thickness, 9.4 mg weight) has been designed and fabricated as a sorption device. The technique uses a small PDMS mini-disk and very small volume of organic solvent and less sample size than the solvent extraction. This new HS-PDE method is very simple to use, inexpensive, rapid, requires less labor. Linearities of calibration curves for ${\alpha}$-pinene, ${\beta}$-pinene, limonene and ${\gamma}$-terpinene by HS-PDE combined with GC/MS were excellent having $r^2$ values greater than 0.99 at the dynamic range of 6.06~3500 ng/mL. The limit of detection (LOD) and the limit of quantitation (LOQ) showed very low values. This method exhibited good precision and accuracy. The overall extraction efficiency of this method was evaluated by using partition coefficients ($K_p$) and concentration factors (CF) for several characteristic components from nutmeg and mace. Partition coefficients were in the range from $2.04{\times}10^4$ to $4.42{\times}10^5$, while CF values were 0.88-15.03. HS-PDE was applied successfully for the analysis of flavors compositions from nutmeg, mace and cumin. The HS-PDE method is a very promising sampling technique for the characterization of volatile flavors.

Validation of an analytical method for cyanide determination in blood, urine, lung, and skin tissues of rats using gas chromatography mass spectrometry (GC-MS)

  • This study was conducted to establish the analytical method for the determination of cyanide in blood, urine, lung and skin tissues in rats. In order to detect or quantify the sodium cyanide in above biological matrixes, it was derivatized to Pentafluorobenzyl cyanide (PFB-CN) using pentafluorobenzyl bromide (PFB-Br) and then reaction substance was analyzed using gas chromatography mass spectrometer (GC/MS)-SIM (selected ion monitoring) mode. The analytical method for cyanide determination was validated with respect to parameters such as selectivity, system suitability, linearity, accuracy and precision. No interference peak was observed for the determination of cyanide in blank samples, zero samples and lower limit of quantification (LLOQ) samples. The lowest limit detection (LOD) for cyanide was $10{\mu}M$. The linear dynamic range was from 10 to $200{\mu}M$ for cyanide with correlation coefficients higher than 0.99. For quality control samples at four different concentrations including LLOQ that were analyzed in quintuplicate, on six separate occasions, the accuracy and precision range from -14.1 % to 14.5% and 2.7 % to 18.3 %, respectively. The GC/MS-based method of analysis established in this study could be applied to the toxicokinetic study of cyanide on biological matrix substrates such as blood, urine, lung and skin tissues.

Determination of L-Carnitine in Infant Powdered Milk Samples after Derivatization

  • Herein, a novel analytical method using a high-performance liquid chromatography-fluorescence detector (HPLC/FLD) is developed for rapidly measuring an L-carnitine ester derivative in infant powdered milk. In this study, solid-phase extraction cartridges filled with derivatized methanol and distilled water were used to effectively separate L-carnitine. Protein precipitation pretreatment was carried out to remove the protein and recover the analyte extract with a high recovery (97.16%-106.56%), following which carnitine in the formula was derivatized to its ester form. Precolumn derivation with 1-aminoanthracene (1AA) was carried out in a phosphate buffer using 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC) as the catalyst. Method validation was performed following the AOAC guidelines. The calibration curves were linear in the L-carnitine concentration range of 0.1-2.5 mg/L. The lower limit of quantitation and limit of detection of L-carnitine were 0.076 and 0.024 mg/L, respectively. The intra- and interday precision and recovery results were within the allowable limits. The results showed that our method helped reduce the sample preparation time. It also afforded higher resolution and better reproducibility than those obtained by traditional methods. Our method is suitable for detecting the quantity of L-carnitine in infant powdered milk containing a large amount of protein or starch.

  • Defect inspection system for industrial applications takes the important portion. Non-destructive inspection method has been significantly improved. Infrared thermography, as one of method for non-destructive inspection, can provide relatively precise data and quick inspection time. This study, it was performed to measure defect according to the measurement limit of the non-visible areas such as the back surface of the pipe using reflection plate using reflection plate based on Infrared thermography. The materials of the reflection plate were determined in consideration of the space limitation and the thermal characteristics, and defects were detected by the manufactured reflection plate. Detection of defect in non-visible area using the candidate materials for reflection plate was conducted.

  • The atmospheric concentration of dimethylsulfide (DMS), known as the predominant volatile organic. sulfur compound, is determined at subnanogram level by a combined application of non-cryogenic preconcentration method and gas chromatography with flame photometric detection (GC/FPD). The volatile DMS in air is preconcentrated using a trapping tube containing adsorbent like Molecular Sieve 5A (or gold-coated sands). The tube is then connected to the GC/FPD system via a six-way rotary valve, thermally desorbed at 40$0^{\circ}C$, separated on OV101 column, and detected by a flame photometric detector. The DMS peak elutes at about 2.5 mins and is integrated electronically. The analytical precision, if expressed in terms of relative standard error, is around 5%. The detection limit of our GC/FPD system is ca 1 ng of DMS. Details of our analytical system are presented.

Occurrence of aflatoxin $M_1$ in milk determined by HPLC with derivatization method in Korea (1999-2000)

  • In this study, the levels of aflatoxin $M_1$ ($AFM_1$) in milk were determined by HPLC with derivatization method. Milk samples were purified using $C_{18}$ disposable cartridge followed by derivatization with trifluoroacetic acid and analysed using HPLC with fluorescence detection. The recoveries of AFM1 from milk samples added $AFM_1$ at a level of 0.025~0.1 ng/ml were 94.7~98.0% with detection limit of 0.009 ng/ml. The amounts of $AFM_1$ were determined below 0.05 ng/ml for all tested samples of commercial milk collected in 1999 and 2000.

Voltammetric Determination of Bisphenol A Using a Carbon Paste Electrode Based on the Enhancement Effect of Cetyltrimethylammonium Bromide (CTAB)

  • The influence of cetyltrimethylammonium bromide (CTAB) on the electrochemical behavior of bisphenol A at the carbon paste electrode (CPE) was investigated. CTAB, with a hydrophobic C-H chain, can adsorb at the CPE surface via hydrophobic interaction and then change the electrode/solution interface, and finally affects the electrochemical response of bisphenol A, confirming from the remarkable oxidation peak current enhancement. The electrode process of bisphenol A was examined, and then all the experimental parameters which affects the electrochemical response of bisphenol A, such as pH value of the supporting electrolyte, accumulation potential and time, potential scan rate and the concentration of CTAB, were examined. Finally, a sensitive and simple voltammetric method was developed for the determination of bisphenol A. Under the optimum conditions, the oxidation peak current of bisphenol A varied linearly with its concentration over the range from $2.5\;{\times}\;10^{-8}\;to\;1\;{\times}\;10^{-6}$ mol/L, and the detection limit was found to be $7.5\;{\times}\;10^{-9}$ mol/L. This method was successfully employed to determine bisphenol A in some waste plastic samples.

Determination of Pd(II) and Pt(II) Metal Cyano Complexes Using Capillary Electrophoresis

  • Mixtures of cyano complexes of palladium(II) and platinum(II) were separated by capillary electrophoresis using a fused silica capillary as a separation column and 30 mM phosphate buffer (pH 7) containing 15 wt. % acetonitrile as a running buffer. By virtue of the high ionic mobilities of the negatively charged cyano complexes of Pd(II) and Pt(II), they were separated using a cathodic injection and anodic detection scheme. The metal complexes eluted through the capillary were detected by direct UV absorption at 214 nm. A linear relationship between peak area and concentration was obtained for both ions and the detection limit was lower than $10^{-14}$ mole. The proposed method was applied to real sample, e.g., anode slime obtained from an electrolytic copper refinary, as a method for the simultaneous determination of palladium and platinum.
