Changes in Antioxidative Activity and Total Polyphenols of Crude and Defatted Grape Seed Extract by Extraction Condition and Storage (포도종자의 처리별 추출조건 및 저장에 따른 항산화활성 및 총페놀함량의 변화)

  • This study was conducted to determine the yield, free radical scavengering effect and total phenol contents of various solvent fractions on the crude and defatted grape seed extract during storage. The optimal condition for the extraction yield, free radical scavengering effect and total phenol contents was $90\%$ ethanol for 6 hour at $70^{\circ}C$. The extraction yield for crude and defatted grape seed at optimal condition was $8.9\%\;and\;9.16\%$, respectively. Also, the strongest free radical scavengering effect with $41.52\;{\mu}g/mL$ was observed in $95\%$ ethanol of defatted grape seed extracted for 6 hour at $70^{\circ}C$. Similar result was observed in total phenol contents of defatted grape seed. The ethyl acetate fraction obtained from ethanol extract of defatted grape seed showed the strongest RC50($12.35\;{\mu}g/mL$) compared to other organic fractions. Free radical scavengering effect of crude and defatted grape seed extracts treated with alkali condition(pH 10) was reduced compared to that of acidic condition(pH 2) during storage far 1 month at $50^{\circ}C$. Overall, more stronger free radical scavengering effect and higher total phenol contents in defatted grape seed extracts was observed than that of crude grape seed.

Bioactive Compounds and Antioxidant Activity of Jeju Camellia Mistletoe (Korthalsella japonica Engl.) (제주 동백나무 겨우살이의 용매별 기능성 성분 및 항산화 작용)

  • Mistletoes are hemi-parasitic plant growing on different host tree and shrubs. They are traditionally used in folkloric medicine for the treatment of diarrhea, cough, diabetes, hypertension, cancer and skin infection. The purpose of this study was to determine the contents of phenolics and antioxidant activity of 70% ethanol, 100% methanol and hot water extracts of Jeju camellia mistletoe (Korthalsella japonica Engl.). Ethanol was most effective in extracting total phenols (7,427 mg gallic acid equivalent (GAE)/100 g) and flavonoid (1,777 mg rutin equivalent (RE)/100 g). The free radical scavenging activity, 1,1-diphenyl-2-picryl hydrazyl (DPPH) (EC50 = 7.8 mg/ml) and hydrogen peroxide (H2O2) (EC50 = 1.4 mg/ml), and the capacity for chelating metal ions (EC50 = 8.0 mg/ml) and reducing power (EC50 = 14.9 mg/ml) of the samples also higher in ethanolic extracts. The strong correlation (r2 = −0.996~−0.881) between antioxidant capacities and the phenolic contents implied that phenolic compounds are a major contributor to the antioxidant activity of the ethanolic extracts of Jeju camellia mistletoe. As conclusions, Jeju camellia mistletoe contains bioactive substances with a potential for reducing the physiological as well as oxidative stress and this could explain the suggested cancer preventive effect of these plants as well as their protective role on other major diseases.

Antimicrobial Activity of Quercus mongolica Leaf Ethanol Extract and Organic Acids against Food-borne microorganisms (식중독균에 대한 신갈나무 잎 추출물과 유기산의 항균효과)

  • This study was conducted to determine the optimal extraction condition of temperatures and kinds of extraction solvent, and antimicrobial activity of ethanol extract of Quercus mongolica leaf and organic acid against foodborne microorganisms. The antimicrobial activity of ethanol extract at $60^{\circ}C$ for 6 hour against foodborne microorganisms was stronger than those at $30^{\circ}C\;and\;90^{\circ}C$. Also, the ethanol extract showed stronger antimicrobial activity than those of water and methanol extract. The minimal inhibitory concentrations of the ethanol extract of Quercus mongolica leaf against B. cereus and L. monocytogenes, was $62.5{\sim}125\;{\mu}g/mL$ but, the minimal inhibitory concentration was $250\;{\mu}g/mL$ against S. typhimurium and P. aeruginosa and over $500\;{\mu}g/mL$ against E. coli O157:H7, respectively. The minimal inhibitory concentrations of the lactic, citric, acetic acid and the ethanol extract of Quercus mongolica leaf against B. cereus and L. monocytogenes was 2500, 5000, 1250 and $125\;{\mu}g/mL$, respectively. The combined effect of each organic acid and the ethanol extract against B. cereus was not observed but, synergistic effect was observed against L. monocytogenes. In the meantime, when the ethanol extract was combined with each organic acid at sub-lethal concentration, the combination did not increase the inhibitory effect of the most active single compound alone against E. coli O157:H7, respectively.

Triglyceride Composition of Walnut Oil (호도기름의 Triglyceride 조성(組成)에 관한 연구(硏究))

  • The present study was conducted to elucidate the triglyceride composition of walnut oil. The triglycerides were separated by thin layer chromatography (TLC) and fractionated on the basis of partition numbers by reverse phase high performance liquid chromatography (HPLC) on a column packed with $\mu$-bondapak $C_{18}$ using methanol-chloroform mixture as a solvent system. Each of these collected fractions was fractionated again on the basis of acyl carbon number of triglyceride by gas liquid chromatography (GLC). The fatty acid composition of triglycerides for each partition numbered group was also analyzed by GLC. From the results, it was found that walnut oil consists of ten kinds of triglycerides, and the patterns of major ones in walnut oil were as follows: 53.3% of $(C_{18:2},\;C_{18:2},\;C_{18:2})$, 10.1% of $(C_{18:1},\;C_{18:2},\;C_{18:2})$, 5.4% of $(C_{18:1},\;C_{18:1},\;C_{18:2})$, 4.3% of $(C_{18:1},\;C_{18:2},\;C_{18:3})$, 3.9% of $(C_{18:0},\;C_{18:2},\;C_{18:2})$, 2.0% of $(C_{18:0},\;C_{18:1},\;C_{18:2})$, 1.8% of $(C_{18:0},\;C_{18:2},\;C_{18:2})$.

Effect of Growth Inhibition and Quinone Reductase Activity Stimulation of Makgeoly Fractions in Various Cancer Cells (막걸리 분획물에 의한 암세포 성장 억제 및 Quinone Reductase 활성 증가 효과)

  • In this study, we investigated the anticancer activity of Makgeoly (MG). MG was fractionated into four fractions by using solvent partition method, affording hexane (MGH), methanol (MGM), butanol (MGB) and aquous (MGA) soluble fractions. We determined the cytotoxicity of these four fractions in four kinds of cancer cell lines, such as HepG2, MCF-7, B16-F10 and HT29 by MTT assay. Among the various fractions, the MGM showed the strongest cytotoxic effects on all cancer cell lines. The morphological changes such as membrane shrinking and blebbing of cells were also observed by MGM treatment in HepG2 cell. In addition, we observed quinone reductase (QR) activity stimulating effects in all fraction layers of MG on HepG2 cells. QR activity increased approximately 2.6 and 2.1 times in MGM and MGH treated HepG2 cell at $100{\mu}g/mL$, respectively, compared to that in control value. Although further studies are needed, the present work could suggest that the fin of MG has a potential to be used as a chemopreventive agent against cancer.

Polyacetylene Compounds from Panax ginseng C.A. Meyer (인삼의 Polyacetylene 화합물)

  • Several major polyacetylene compounds were isolated from the petroleum-ether fraction of fresh Korean ginseng roots through solvent fractionation. partition and silica gel column chromatography. Further separation of acetylenic compounds was accomplished by bonded normal phase HPLC utilizing a moderately nonpolar microparticulate column. The preparative separation for the various spectral measurements was carried out by low pressure preparative liquid chromatography. The chemical structure of these polyacetylenes separated was determined by UV. IR/FTIR. $^{1}H$ NMR. mass spectral and elemental analysis. These are identified to be heptadeca-1-en-4.6-diyn-3.9.l0.-triol [1] heptadeca-1.9-dien-4.6-diyn-3-ol. heptadeca-1.8-dien-4.6-diyn-3.10-diol and the 4th was denatured polyacetylene. heptadeca-1.4-dien-6.8-diyn-3.10-diol. Two different p-substituted benzoates of panaxynol were synthesized for the determination of exciton chirality. The circular dichroism spectra in the UV region show that panaxynol p-bromobenzoate and p-dimethyl-aminobenzoate constitute negative exciton chirality [2]. Isolated major polyacetylene compounds were irradiated in aerated solution with 300 nm UV light to obtain the oxidized product at the allylic alcohol center to corresponding carbonyl compounds such as heptadeca-1-en-4.6-diyn-9.10-diol-3-one and heptadeca-1.9-dien-4.6-diyn-3-one. These photooxidation compounds have en-on-diyne chromophore and undergo nucleophilic addition reaction with methanol to yield ${\beta}-methoxy$ carbonyl compounds such as heptadeca-9-en-4.6-diyn-1-methoxy-3-one and heptadeca-4.6-diyn-1-methoxy-9.10-diol-3-one.

Antioxidant Properties and Protective Effects of Inula britannica var. chinensis Regel on Oxidative Stress-induced Neuronal Cell Damage (금불초 추출물의 항산화 효과 및 산화 스트레스에 대한 신경세포 보호작용)

  • The antioxidant properties and protective effects of Inula britannica on ${H_2}{O_2}$-induced SH-SY5Y neuroblastoma cell damage were investigated. A series of solvent fractions, including hexane(Fr.H), petroleum ether, chloroform, ethyl acetate(Fr.EA), and water fraction(Fr.W), were prepared from the 70% methanol extracts of Inula britannica. Fr.W had the highest total contents of phenolics and flavonoids, followed by Fr.EA. The antioxidant properties of the fractions were also evaluated by analyzing their scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radicals, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals, and nitric oxide. Fr.W showed the strongest activities in all assays. The concentrations of Fr.W that resulted in 50% reductions of the DPPH and ABTS radicals were 20.7 ${\mu}g$/mL and 39.4 ${\mu}g$/mL, respectively. Fr.W showed the weakest cytotoxic activities on the SH-SY5Y cells, whereas it effectively protected ${H_2}{O_2}$-induced cell death, increasing cell survival by 35.0-77.0% at a concentration range of 62.5-250 ${\mu}g$/mL. In this range, Fr.W also significantly decreased intracellular ROS levels by 34-39%. Overall, the antioxidant properties of Inula britannica can contribute to rescuring neuronal cells from oxidative stress-induced cell injury.

A Study on Hypoglycemic Effects of Crude Extracts of Schizandrae Fructus (오미자 추출물의 혈당 강하 효과에 관한 연구)

  • Hypoglycemic effect of Schizandrae Fructus (SF) extract containing in Okchun-san was determined on 3T3-L1 fibroblasts and adipocytes by investigating insulin-like activity, insulin sensitizing activity and ${\alpha}-glucoamylase$ suppressing activity. SF were extracted by using 70% ethanol followed by XAD-4 column chromatography with a mixture solvent of methanol and water, and the fractional extractions were utilized for assaying hypoglycemic effect. No inhibition of ${\alpha}-glucoamylase$ activity of SF was observed. Insulin-like activity 3T3-L1 adipocytes was not shown by SF. A significant insulin sensitizing activity of SF extractions was observed in 3T3-L1 adipocytes, giving SF extractions with 1 ng/ml insulin to reach glucose uptake level increased by 50 ng/ml of insulin alone. When cells were treated with SF (Fr. 4 or 5) plus 1 ng/ml insulin, glucose uptake was increased more than seven times as compared to 1 ng/ml of insulin alone, suggesting that SF extracts increased GLUT4 content by enhancing insulin signaling. These data suggest that SF extracts (especially Fr. 4 and 5) contains an effective insulin sensitizing compounds for hypoglycemic activity in 3T3-L1 adipocytes.

Preparation of PMSQ/TiO2 Composite Fine Powder by Sol-Gel Process (Sol-Gel Process를 이용한 PMSQ/TiO2 복합 미립자의 합성)

  • Monodisperse, spherical $PMSQ/TiO_2$ composite fine powders were prepared by modified sol-gel process where 300 nm $TiO_2$ fine powders were used as seed particles for hetero-condensation with hydrolyzed MTMS (Methyltrimethoxysilane). The reaction was carried out under $N_2$ atmosphere at ambient temperature using $NH_3$ as a catalyst. Methanol was used as a solvent. Powder was obtained by the filtration of the solution with a glass filter and washing with acetone. The stirring rate, reaction temperature, $[H_2O]/[MTMS]$ and $[MTMS]/[TiO_2]$ ratio were varied to investigate shapes and sizes of particles. Monodisperse particles of $1-2{\mu}m$ diameter were obtained with [MTMS]=0.2 M, $[NH_3]=0.6M$, $[H_2O]/[MTMS]=100$, $[MTMS]/[TiO_2]=10-50$ at ambient temperature with mild stirring condition. These composite particles had a contact angle of almost 180 degree contact angle with water, which proves their excellent hydrophobicity. The study of UV absorption spectra showed that they have UV protecting effect.

  • Light cycle oil (LCO), one of the by-products of the catalytic cracking gasoline manufacturing process, contains a lot of valuable aromatics. In particular, 2,6-dimethylnaphthalene (2,6-DMN) contained in LCO has been becoming important as the basic material of polyethylene naphthalate plastic and liquid crystal polymer, etc. If it were possible to separate and purify the valuable aromatic hydrocarbons (such as 2,6-DMN) from LCO, which have only been used as fuel mixed with heavy oil, it would be very meaningful in terms of the efficient use of resources. We investigated the high-purity purification of 2,6-DMN by the combined method of melt crystallization (MC) and solute crystallization (SC). The enriched DMN isomer mixtures (concentration of 2,6-DMN : 10.43%) recovered from LCO by distillation-extraction combination and the crystal recovered by MC used as raw materials of MC and SC, respectively. The solvent of SC used was a mixture of methanol and acetone (60 : 40 wt%). The crystal of 2,6-DMN with a high-purity of 99.5% was recovered by MC-SC combination. We confirmed that the MC-SC combination was one of the very useful combinations for the high-purity purification of 2,6-DMN contained in the enriched DMN isomer mixtures.
