향나무 심재 추출물로 염색된 직물의 방미성과 집먼지 진드기 기피효과 (Antifungal activity and house dust mite repellent effect of fabric dyed with Juniperus chinensis heartwood extracts)

  • This study was carried out to investigate the antifungal activity and house dust mite repellent effect of cotton and silk fabrics dyed with Juniperus chinensis heartwood extracts by hot-water and methanol. The chemical composition of Juniperus chinensis heartwood extracts by hot-water and methanol was analyzed by GC-MS. In the case of antifungal activity, dyed fabrics with hot water extract had no effect against chaetomium globosum fungus. But showing no growth in fabrics dyed with methanol extract, dyed fabrics with methanol extract showed very excellent antifungal activity. Silk fabric dyed with methanol extract showed excellent house dust mite repellent effect of 94.3%~96.0% against dermatophagodes farinae. The composition of Juniperus chinensis heartwood extracts from hot-water and methanol was different. There were no terpene and its analogue peaks in hot-water extract. However there were terpene and its analogue peaks in methanol extract. From these GC-MS results, terpene was identified in methanol extract of Juniperus chinensis heartwood. Efficient ingredient of antifungal activity and house dust mite repellent effect was assumed terpene.

대두유의 자동산화 및 가열산화에 미치는 정향추출물의 항산화 효과 (Effects of Clove Extracts on the Autoxidation and Thermal Oxidation of Soybean Oil)

  • This study was conducted to investigate the antioxidant activities of clove extracts in water, methanol and ether. The clove extracts, BHA and ${\alpha}$-tocopherol were added to each oil at a level of 200 ppm. The activities of the substrate oils and controls were tested under autoxidation and thermal oxidation conditions. The degree of the effects of the antioxidant activities under autoxidation condition were in the following order; ether extract 〉 methanol extract 〉BHA 〉 ${\alpha}$-tocopherol 〉 water extract = control group. The induction periods of the control, water, methanol and ether extracts, and BHA and ${\alpha}$-tocopherol were 9.5, 9.6, 10.7 11.8, 10.4 and 9.7 days, respectively. Under thermal oxidation condition, the methanol extract showed stronger antioxidant activity than those of the water and ether extracts. The antioxidant activity of the methanol extract was attributed to ${\alpha}$-tocopherol and BHA.

Phytotoxic Effect, DPPH Radical Scavenging Activity and Chlorogenic Acid Level of Methanol Extracts from Aerial Parts of Several Korean Salad Plants

  • A series of aqueous or methanol extracts from four different Korean salad plants were assayed to determine their allelopathy and antioxidant activity. The extracts applied on filter paper in a Petri-dish bioassay significantly inhibited root growth of against alfalfa (Medicago sativa) seedlings. Leaf extracts from 40 g dry tissue $L^{-1}$ of Aster yomena was most phytotoxic to alfalfa root growth, and followed by that of Cirsium japonicum, Taraxacum officinale, and Ixeris dentate. Methanol extracts of plants dose-dependently increased DPPH free radical scavenging activity in vitro. Antioxidant activity of methanol extracts from the same plant species was investigated, and the result showed high DPPH free radical scavenging activity in Cirsium japonicum, Aster yomena, and Ixeris dentate, however, in Taraxacum officinale was least activity. By means of HPLC analysis, chlorogenic acid, p-coumaric acid, and total phenolics with 7.68, 17.47 and 18.64 mg, $100g^{-1}$, respectively, showed the highest amounts in methanol extracts from Cirsium japonicum leaves. These results suggest that Compositae salad plants contain water-soluble substances with allelopathic potential as well as antioxidant activity.

흰쥐의 지질대사에 미치는 해당화 뿌리 추출물의 효과 (Effects of Rosa rwgosa Radix on Lipid Metabolism in Rats)

  • The feeding effects of Rosa rugosa roots powder extracted with two different extracting solvents on lipid metabolism in rats were evaluated. Rats were fed with purified diets containing methanol or 70% aqueous acetone extracts at a 1% level for 4 weeks. In addition, to examine the effects of age, rats with different ages (4weeks old vs. 3 months old) were fed with purified diets containing methanol extracts at a 1% level for 4weeks. Feeding of methanol extracts decreased the concentration of triacylglycerol in serum and liver, whereas the aqueous acetone extracts lowered the concentration of liver triacylglycerol with concomitant elevation of the concentration of serum triacylglycerol. The methanol extracts ameliorated the degree of increase in the serum triacylglycerol level in relation to age and decreased especially the concentration of free fatty acid in serum of adult rats. The results suggested possibility that the regulatory mechanism responsible for triacylglycerol-lowering effects of Rosa rugosa roots extracts in the liver could be modulated by differences in the components of extracts due to the variation in extracting solvents, and that the methanol extracts might be effective in lowering an age-dependent increase in the concentration of serum and liver triacylglycerol in rats.

In Vitro Assessment on Biological Activities of Methanol Extracts from Several Compositae Edible Plants

  • Phytotoxicity, antioxidant activity, and cytotoxicity of the aqueous or methanol extracts from the young sprouts of the six Compositae medicinal plants were determined. Aqueous leachates at 40g dry tissue $L^{-1}$ (g $L^{-1}$) Cirsium japonicum and Aster yomena showed the highest inhibitory effect on alfalfa (Medicago sativa L.). Total phenolic content showed the highest amount in methanol extracts from Ixeris dentata, and followed by A. yomena, and Cephalonoplos segetum. Methanol extracts of C. segetum and I. dentata at 25${\mu}g$ m$L^{-1}$ exhibited the highest DPPH radical scavenging activity by 87.2, and 52.8%, respectively. By means of HPLC analysis, MeOH extracts of C. segetum had the highest amount of antioxidant chlorogenic acid. Based on MTT assay, the methanol extracts from Y. sonchifolia ($IC_{50}$ = 65.7${\mu}g$ $mL^{-1}$) showed the highest cytotoxicity against Calu-6. These results suggest that plant extracts had a dose-dependent biological potentials including phytotoxicity, antioxidant activity, and anticancer activity, and that their activities exhibited differently depending on plant species.

대두 및 대두발효식품의 항돌연변이성

  • To investigate the inhibitory effect of soybean and Korean traditional fermented soybean products on the chemically induced mutagenesis, we extracted soybean, Kanjang, Doenjang, Kochujang, and Chongkukjang with water, methanol and hexane. Inhibitory effect of the extracts was assayed by the SOS chromotest using Escherichia coli PQ37 as a test strain. 4-nitroquinoline-1-oxide(4NQO), N-methyl-N'-nitro-N-nitrosoquanidine(MNNG), and aflatoxin B$_{1}$(AFB$_{1}$) were used as mutagens. Methanol extracts showed relatively higher inhibitory effect than water and hexane extracts. Methanol extracts of soybean, Doenjang, Kochujang, and Chonhkukjang showed inhibitory effect of 68.4, 96.3, 17.5, and 100.9% against MNNG, and 28.6, 109.1, 41.3, and 101.8% against AFB$_{1}$, respectively. Doenjang methanol extract showed inhibitory effect of 51.0, 96.3, and 109.1% against 4NQO, MNNG, and AFB$_{1}$, respectively. Methanol extract of Doenjang showed dose-dependent inhibitory effect against 4NQO, MNNG, and AFB$_{1}$. Inhibitory effect of heat-treated Doenjang and Chongkukjang methanol extracts on the mutagenecity of MNNG and AFB$_{1}$ was remained over 95% of the inhibitory effect of heat-untreated extracts, demonstrating the heat stability of the potent antimutagenic activity.

곡류 및 두류 추출물로 부터 HMG-CoA reductase 저해활성 검색 (Screening of HMG-CoA Reductase Inhibitory Activity of Ethanol and Methanol Extracts from Cereals and Regumes)

  • 국내산 곡류의 에탄올 및 메탄올 추출물로부터 콜레스테롤 생합성계에서 가장 중요한 율속효소로 알려져 있는 HMG-CoA reductase 저해활성을 검색하였다. HMG-CoA reductase activity 측정용 효소원은 cholestyramin을 급여하여 사육 한 후 밤 12시경에 간율 적출하여 얻어진 microsome을 freeze-thaw방법으로 조제하였고 효소활성은 spectrophotometric assay로 측정하였다. 그 결과 각 시료 추출물을 $100{\;}{\mu}g/assay$ 되게 첨가한 경우 HMG-CoA reductase 억제활성은 에탄올 추출물에서는 수수가 가장 높은 활성을 보였고 다음은 흑임자였으며 나머지는 활성이 미미하거나 전혀 없는 것으로 나타났다. 메탄올 추출물에서는 기장이 가장 높은 활성을 나타내었고 다음은 수수였으며 그 다음은 메밀, 흑미의 순으로 나타났다. 또한 추출물에서 높은 억제활성을 나타내었던 수수와 기장 메탄올 추출물을 각종 용매로 분획하여 활성을 검토한 결과 수수와 기장 모두 헥산 획분에서 가장 높은 활성을 나타내었다.

식중독 유발세균의 증식에 미치는 황련 추출물의 효과 (Antimicrobial Effect of Plagiorhegama dubium Extracts on Food-borne Pathogen)

  • 본 연구에서는 식중독 유발 세균에 대한 항균 활성이 우수한 천연 항균성 물질을 검색하기 위해 예로부터 민간과 한방에서 널리 이용되어온 황련을 각종 유기용매로 추출하여 식중독 유발 세균에 대한 항균 활성을 조사해 보았다. 황련을 petroleum ether, chloroform, ethyl acetate, methanol를 이용하여 실온에서 각각 용매별로 계통 분획하고, 열수추출물을 얻은 후, 9종의 식중독 유발세균(Staphylococcus aureus, Bacillus cereus, Salmonella enteritidis, Shigella flexneri, Escherichia coli, Salmonella typhimurium, Shigella dysenteriae, Pseudomonas aruginosa, Shigella sonnei)에 대하여 항균효과를 조사하였다. 황련 추출물의 농도별 항균 활성 검색에서는 황련의 methanol 추출물이 가장 큰 항균효과를 보였으며 S. aureus와 E. coli가 가장 민감하게 반응하는 균주였다. 황련의 methanol 추출물과 백화사설초의 methanol 추출물을 혼합하여 항균력을 측정해 본 결과 두 추출물을 섞어 첨가했을 경우가 단독으로 사용했을 시 보다 상승효과를 나타내었다. 또한 황련의 methanol 추출물이 식중독 유발세균의 성장에 미치는 효과를 검정하기 위해 S. aureus 및 E. coli의 배양액에 황련의 methanol 추출물을 각각 4,000 ppm 농도로 첨가했을 시, E. coli이 생육이 60시간 이상까지 억제됨을 관찰할 수 있었고, S. aureus의 생육도 36시간까지 지연시킬 수 있었다. 이상의 결과 황련의 methanol 추출물은 S. aureus와 E. coli이 성장을 효과적으로 억제시킬 수 있음을 알 수 있었다.

산사(Crataegus pinnatifida Bunge)의 추출 용매별 항산화 활성 (Antioxidative activities of various solvent extracts from haw (Crataegus pinnatifida Bunge))

  • 산사과육을70% methanol, 70% ethanol, chloroform:methanol(CM, 2:1, v/v), n-butanol, ethyl acetate(EA)로 추출하여 시료농도 0.2 mg/mL, 0.5 mg/mL, 0.8 mg/mL 농도에서 각 추출물의 항산화 활성을 비교하였다. DPPH radical 소거 활성은 70% methanol, 70% ethanol, chloroform:methanol(2:1, v/v), ethyl acetate, n-butanol 추출물 순으로 높게 측정되었고 특히 70% methanol 및 70% ethanol 추출물은 다른 추출물에 비하여 유의적으로 높은 것으로 나타났으며(p<0.05), 0.8 mg/mL 농도에서 각각 92.89%, 91.17%의 높은 활성을 나타내었다. 환원력과 ferric reducing antioxidant power(FRAP)는 동일한 경향을 나타내었는데 활성은 70% methanol, 70% ethanol, n-butanol, ethyl acetate, chloroform: methanol(2:1, v/v) 추출물 순으로 높게 나타났으며, 70% methanol 및 70% ethanol 추출물이 다른 추출물에 비하여 높은 것으로 측정되었다(p<0.05). ABTS radical 소거 활성은 DPPH와 동일한 경향이었으며 0.8 mg/mL 농도에서 70% methanol 및 70% ethanol 추출물은 각각 75.81%, 74.73%의 활성을 나타내었다. NO radical 소거 활성은 70% methanol, 70% ethanol, ethyl acetate, n-butanol, chloroform:methanol(2:1, v/v) 추출물 순으로 나타났고, hydroxyl radical 소거 활성은 n-butanol, 70% methanol, 70% ethanol, chloroform: methanol(2:1, v/v), ethyl acetate 추출물 순으로 측정되었다. Hydroxyl radical 소거 활성을 제외한 다른 실험 결과에서 70% methanol 및 70% ethanol 추출물은 다른 용매 추출물에 비하여 유의적으로 높은 것으로 나타났고 chloroform: methanol(2:1, v/v), ethyl acetate, n-butanol 추출물 간에는 대부분 유의성이 없는 것으로 측정되었다(p<0.05). 본 실험 결과 산사 과육 추출물의 항산화 활성은 우수한 것으로 나타났고 특히, 70% methanol 및 70% ethanol 추출물에서 높은 활성을 보였다. 따라서 산사는 천연 항산화제 및 기능성 식품 소재로서 활용될 수 있을 것으로 생각된다.

녹용의 약효 성분에 관한 연구(VI) 녹용의 지용성 성분 및 Pantocrin이 흰쥐 척수 신경의 Aldolase 활성에 미치는 영향 (Biochemical Studies on Antler (Cervus nippon taiouanus) (VI) Comparative Study on the Effect of Lipid Soluble Fractions of Antler Sponge and Velvet Layers and Pantocrin on the Aldolase Activity in the Rat Spinal Nerves)

  • In the present study, in attempt was made to observe the effect of lipid components in pantocrin and antler velvet and sponge layers extracts on the aldolase activity in the rat spinal nerves. The antler chloroform-methanol (2:1) extracts are known to be composed of neutral lipids, cholesterol, cerebrosides, spingomyelin, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl serine and gangliosides. The antler extracts were studied on the aldolase activity in the rat spinal nerves. The aldolase activity was measured by the method of Lehninger. To investigate the components of tile extracts which affect the enzyme activity, the chloroform-methanol (2:1) extracts of antler were fractionated into petroleum ether soluble and insoluble fractions and their effects on the enzyme activity were compared. It was found that the chloroform-methanol (2:1) extracts of the antler sponge and velvet layers as well as those petroleum ether soluble fractions obtained from the chloroform-methanol (2:1) extracts, decrease the aldolase activity by 18-23%. However, pantocrin showed increasing effect on the aldolase activity by 42.6%. The petroleum ether insoluble fraction of the chloroform-methanol (2:1) -extract showed no significant increasing activity (about 13%) on the spinal nerves aldolase. The components of pantocrin ind the petroleum ether insoluble fractions were attempted to analysize by thin layer (silica gel) and gas liquid chromatography.
