Assessment on the Extraction Efficiency of Explosive Compounds in Soil for Improving the Working Draft of International Standard (국제표준(안) 개선을 위한 토양 중 화약물질 추출 효율성 평가)

  • The ISO document of ISO/TC/190/SC3/WG11/N11 is a working draft of international standard (WD) dealing with analytical method for the determination of explosives and related compounds using high performance liquid chromatography. The scope of this WD covers the storage of samples, preparing test portion, extraction and instrumentation. The main purpose of this study was to improve the extraction conditions which were already adopted in the WD. For this purpose, mechanical shaking method could be corresponded up to 18 hours of ultrasonic bath extraction in the WD was tested. Methanol was also tested with the intention of being added as an extracting solvent other than acetonitrile in the WD. According to the results, 16 hours of mechanical shaking method showed statistically the same effectiveness as that of 18 hours of ultrasonic bath extraction. In case of extracting solvent, methanol also showed statistically the same extraction capability as acetonitrile for DNB, TNT, 2-A-DNT and 2,4-DNT. However, the recovery rate of TNB with methanol extraction was 40% higher than that of acetonitrile extraction. Through adding mechanical shaking method into committee draft (cf. the next stage draft of the WD during the process for making international standard), ISO standard of analyzing explosives and related compounds in soil would become more useful in dealing with huge number of field samples in the laboratory. In other aspect, adopting methanol as an alternative extracting solvent would be very effective in the terms of exchangeability with GC-ECD/MS method which is being developed by German experts.

Changes of Total Polyphenol Content and Antioxidant Activity of Ligularia fischeri Extracts with Different Microwave-Assisted Extraction Conditions (마이크로웨이브 추출조건에 따른 곰취 추출물의 총 폴리페놀 함량 및 항산화작용의 변화)

  • This study was undertaken in order to compare reflux extraction(RE) and microwave-assisted extraction(MAE) in extraction efficiency and establish optimum microwave extraction conditions in obtaining Ligularia fischeri extracts. A considerable reduction in extraction time was accomplished by MAE. When 70% methanol 50% methanol 70% ethanol, or 50% ethanol was used, MAE extract contained equal levels of soluble solid and total polyphenol as obtained by RE. The optimum microwave-assisted extraction conditions for Ligularia fischeri were achieved by 120∼150 watts of microwave energy and 4∼8 minutes of extraction time. No significant changes were found in antioxidant activity with DPPH scavenging method over the variation of microwave energy or extraction time. The use of diluted methanol or ethanol improved soluble solid content(30%), total polyphenol content(2.7%) and antioxidant activity(68%).

  • This study was conducted in order to compare reflux extraction(RE) and microwave-assisted extraction(MAE) in extraction efficiency and establish optimum microwave extraction conditions in obtaining Aster scaber Thunb extracts. Extraction time was reduced considerably in MAE. When 70% methanol, 50% methanol, 70% ethanol, or 50% ethanol was used, no difference was found in the amount of soluble solid and total phenol between MAE and RE. The optimum microwave-assisted extraction conditions for Aster scaber Thunb were achieved by 120-150 watts of microwave energy and 4∼8 minutes of extraction time. No significant changes were found in antioxidant activity using DPPH scavenging method over the variation of microwave power or extraction time. The use of diluted methanol or ethanol improved soluble solid content(30.8%), total polyphenol content(2.9%) and antioxidant activity(69% ).

Microalgal Oil Recovery by Solvent Extraction from Nannochloropsis oceanica (Nannochloropsis oceanica로부터 용매추출법을 이용한 미세조류 오일 회수)

  • In this study, oil as a source of biodiesel from Nannochloropsis oceanica was extracted using organic solvent. The oil extraction yield and efficiency from dry and wet microalgae were investigated. The initial fatty acids content of the N. oceanica was 317.8 mg/g cell showing a high oil content over 30%. The yield from dry microalgae was higher than that from wet microalgae due to the inhibition of water. The yield by chloroform-methanol was the highest and the yield by hexane was the lowest. However, the total fatty acids contents with the chloroform-methanol were 678.7 and 778.2 mg/g oil under dry and wet conditions, respectively. The high oil extraction yield by chloroform-methanol reflected the fact that the extracted oil contained a high level of impurity. The hexane-methanol extraction from dry N. oceanica showed high oil extraction efficiency, 82.6%. The chloroform-methanol extraction under wet condition also showed high efficiency, 88.0%. While the hexane-methanol extraction from dry microalgae is desirable under low drying cost, the chloroform-methanol extraction from wet microalgae is desirable under high drying cost.

Comparative Studies on Methods of Extracting Polyacetylene Compounds from White Ginseng (인삼 Polyacetylene 성분의 추출방법 비교연구)

  • The amounts of panaxynol and panaxydol, which are major polyacetylone compounds In white ginseng were determined by capillary-GC (FID), and the extraction efficiencies when using varictus extraction solvents (petroleum ether, dichloromethane, ether, ethyl acetate, acetone, acetonitrilr and methanol) and various extraction methods (shaking, Soxhlet and reflux) were compared . The GC column was SPB-1 fused silica capillary (0.25 mm id x30 m, Supelco), and the column oven temperature was programmed to rise from $200^{\circ}C$ to $300^{\circ}C$ at the rate of $4^{\circ}C$ per minute. The extraction efficiencies for panaxynol and panaxydol according to extraction souvtints were the highest in methanol and decreased in the order of dichloromethane, acetone, ether, ethyl acetate, acetonitrile and petroleum ether. The extracttion efficiencies for panaxynol and panaxydol according to extraction methods were the highest for reflux and the lowest for shaking, and those with Soxhlet were almost equal to those for reflux. The analytical amounts of panaxynol and panaxydol obtained by reflux with methanol %mere 4.2 and 6.4 mg/g, respectittely in white ginseng.

Modifier Effects on Supercritical $CO_2$ Extraction Efficiency of Cephalotaxine from Cephalotaxus wilsoniana Leaves

  • The effects of modifiers such as methanol, water diethylamine in methanol (10 v/v %), and diethylamine in water (10 v/v %) were investigated at three different concentrations (1, 5, and 10 v/v %) of the modifiers in supercritical $CO_2$ (SC-$CO_2$) in order to enhance the supercritical fluid extraction (SFE) efficiency of cephalotaxine from Cephalotaxus wilsoniana Leaves. Among the modifiers employed, methanol basified with diethylamine was found to greatly enhance the extraction efficiency relative to any other modifiers employed. The results suggest that cephalotaxine in plant matrices may be readily changed from SC-$CO_2$-insoluble salt to SC-$CO_2$-soluble free base by basified modifiers. In addition, SC-$CO_2$modified with basified methanol could enhance the extraction efficiency of cephalotaxine more than 30% when compared to the conventional organic solvent extraction.

  • Natural goods, especially therapeutic plants, are abundant in the World. Because they have the ability to provide all humanity with countless advantages as a source of medicines, medicinal plants are presently receiving more attention than ever. These plants' therapeutic efficacy is based on bioactive phytochemical components that have clear physiological effects on the human body. The drying process is crucial for the preparation of plant materials prior to extraction since freshly harvested plant materials include active enzymes that create active components, intermediates, and metabolic processes. Many of the phytoconstituents may be extracted using the semi-polar solvent methanol. The goal of the current work is to use the GC-MS gas chromatography- mass spectrometry technology to identify the phytochemicals and review their biological activity. In methanol leaf extract, 5 phytocompounds were found in Ricinus communis, 5 phytocompounds in Pongamia pinnata, 12 phytocompounds in Datura metal, 7 phytocompounds in Azadirachta indica, 11 phytocompounds in Acalypha indica.

Development of Microwave Extraction Method for the Active Ingredients and Functional Constituents of Paeonia Root (마이크로웨이브 추출방법을 이용한 작약의 유효성분 추출 및 생리활성 측정)

  • The heat-mediated reflux apparatus extraction of Paeonia lactiflora Pall. has been widely used as a traditional extraction method. In this paper, the microwave apparatus extraction method of Paeonia Radix was performed and the active ingredients and functional constituents were compared with the reference extraction method. The most effective extraction condition of albiflorin using the microwave was 120℃ in 50% methanol, and paeoniflorin was maximally extracted at 60℃. The reduced level of paeoniflorin molecule at high-pressure and high-temperature extraction condition was caused by the molecular instability. Additionally, the microwave extraction of 50% methanol extracts at 150℃ showed the highest functional constituents determined by in vitro DPPH radical scavenging activity, polyphenol concentration, and tyrosinase inhibition assay. The microwave apparatus was adapted as a rapid, low-cost, and environmentally friendly method to extract active ingredients and the practical extraction conditions of Paeonia Radix can be used in industrial applications.

Comparative Analysis of Latex Plants by GC-MS using Methanol Extraction

  • Plants are able to produce a large number of diverse bioactive compounds. Solvent extraction is used for isolation of plant metabolites. The extract yield for plant metabolite extraction strongly depends on the nature of solvent. A review showed the methanol can yield more bioactive compounds. Drying of the sample material is also important for the extraction of plant material. The present study was carried out to analyze the phytocomponents of 5 different latex producing plants. The plants like Calotropis gigantea, Carica papaya, Nerium oleander, Ficus benghalensis and Plumeria alba leaves and latex. The GC-MS analysis of the metabolites revealed phytocomponents. Calotropis gigantea leaves showed 14 compounds and latex produced 5 compounds out of this 4,4,6A,6B,8A,11,11,14B-Octamethyl-1,4,4A,5,6,6A,6B,7,8,8A,9,10,11,12,12A,14,14A,14B-Octadeca-hydro-2 and 2R- Acetoxymethyl-1,3,3-trimethyl-4T-(3-Methyl-2-Buten-1-Yl)-1T-Cyclohexanol compound was present in both latex and leaf extraction. Beta. -carotene compound was present in both latex and leaf of Carica papaya. It was observed that Ficus benghalensis contained 2R-Acetoxymethyl-1,3,3-trimethyl-4T-(3-Methyl-2-Buten-1-Yl)-1T-Cyclohexanol was same in latex and leaf extraction.

Comparison of Conventional Solvent Extraction, Microwave-Assisted Extraction, and Ultrasound-Assisted Extraction Methods for Paclitaxel Recovery from Biomass (바이오매스로부터 파클리탁셀 회수를 위한 전통적 용매 추출, 마이크로웨이브를 이용한 추출, 초음파를 이용한 추출 방법 비교)

  • In this study, conventional solvent extraction (CSE), microwave-assisted extraction (MAE), and ultrasound-assisted extraction (UAE) were compared for the recovery of paclitaxel from biomass. As a result of investigating the effect of the extraction solvent type (acetone, chloroform, ethanol, methanol, methylene chloride), methanol was the most suitable for all extraction methods. In the case of MAE and UAE using methanol, most of the paclitaxel (> 95%) was recovered by only one extraction. The recovery rate of paclitaxel increased with the increase of extraction temperature (25-45 ℃), microwave power (50-150 W), and ultrasonic power (180-380 W) for MAE and UAE. In addition, SEM analysis showed that the biomass surface structure was slightly corrugated in CSE, while in the MAE and UAE, it was very rough and destroyed by strong impact.
