Bone morphogenetic protein 1 (BMP-1) is part of a complex capable of inducing ectopic bone formation in mammals. Studies on TGF-β1 processing and Drosophila dorsal-ventral patterning have focused attention on BMP-1 as important in mediating the biological activity of this bone inducing complex. Herein, the bacterial expression, refolding, purification, and initial characterization of the BMP-1 proteolytic domain (BPD) are described. A semi-quantitative fluorescence-based thin layer chromatography assay was developed to assist in rapidly screening for optimal renaturation conditions. According to a preliminary screen for optimal conditions for the refolding of BPD , a detectable proteolytic activity against a high turnover substrate for astacin, a homologous protease from crayfish was observed. The conditions identified have allowed the expression of sufficient amounts of BPD for the characterization of the protein. Its proteolytic activity exhibits the same cleavage specificity as astacin against seven substrates that were previously synthesized for studying astacin. Furthermore, this activity is inhibited by the metal chelator 1,10-phenanthroline but not by its analogue 1,7-phenanthroline. The collagenase inhibitor Pro-Leu-Gly hydroxamate was found to inhibit both astacin and BPD activity. The results presented in this paper argue that BMP-1 does in fact possess an intrinsic proteolytic activity.
현대의 주거단지는 공원을 비롯해 호수, 산책로 등 가족과 이웃의 다양한 만남과 소통의 장으로 활용되는데, 더불어 주거 공간에 조형물을 전시하는 것이 하나의 구성요소로 조성되고 있다. 연구자는 이러한 시대요구에 맞춰 주거 공간에 전시하는 조형물의 디자인 전개 방법을 연구하는 목적으로 연구를 진행하였다. 작가의 생각과 메시지를 조형물에 넣음과 함께 관람하는 사람들도 공감할 수 있는 조형물디자인 하기 위해 디자인과정에 비유법을 활용한 은유, 유추방법과 작가의 생각이 공유될 수 있도록 동물형상과 단순화된 디자인을 전개하고, 금속과 돌을 사용하여 조형물의 형태로 완성하였다. 이러한 디자인 조형연구를 통해 거주공간이라는 특수한 장소에 작가의 생각과 관람자 모두가 함께 공감할 수 있는 조형물을 완성할 수 있었으며, 조형물이 하나의 예술분야로서 특수한 장소에 다양한 시각적 요인인 비유적 시각화 방법을 통해 형태화할 수 있는 가능성을 알 수 있었으며, 향후 지속적인 연구로서, 다양한 비유의 시각적 조형화에 대한 연구가 요구된다.
Agar degrading enzyme-agarase-was purified from the culture fluid of Cytophaga so/ ACLJ-18, by acetone precipitation, DEAE-Cellulose, Sephadex G-100 and CM-Sephadex C25 column chromatographies. The molecular weight of purified agarase was estimated to be 24,700 dalton by SDS-polyacrylamide gel electrophoresis. The optimum pH and temperature for agarase activity were 7.0 and 40$^{\circ}C$, respectively. this agarase was stable in the pH range of 6.5 - 8.0 and 40$^{\circ}C$, and required 0.35M NaCl for optimum activity. And this agarase was inhibited by metal ions such as Ba2+, Cu2+, Co2+, Mn2+, Hg2+, Zn2+, and showed specificity on agar.
Our works performed for preparation of oligosaccharides from carrageenan, seaweed polysaccharide, and one active strain for carrageenan was isolated from sea water and identified to Pseudomonas alcaligenes. Carrageenan degrading enzyme was purified from the culture fluid of isolated strain-Pseudomonas alcaligenes JCL-43, by DEAE-Cellulose, Sephadex G-100, Q-Sepharose and CM Sepharose CL-6B column chromatography. Two enzyme-F-I, F-II- was identified this purifying process, and the molecular weight of the purified carrageenase were estimated to be 23.6kDa and 30.2kDa, respectively. The optimum pH and temperature for two carrageenase activity were 7.0 and 4$0^{\circ}C$. These enzymes were stable in the pH range of 6.0~7.5 and lower than 5$0^{\circ}C$, and required 1.5% NaCl for optimum activity. And these carragennase were inhibited by metal ions such as Cu2+, Zn2+, Hg2+, but increased by Ba2+ and Ca2+, and showed specificity on -carrageenan.
The present study describes the gene cloning and high-level expression of an alkaline and thermostable lipase gene from Trichosporon coremiiforme V3. Nucleotide analysis revealed that this lipase gene has an open reading frame of 1,692 bp without any introns, encoding a protein of 563 amino acid residues. The lipase gene without its signal sequence was cloned into plasmid pPICZαA and overexpressed in Pichia pastoris X33. The maximum lipase activity of recombinant lipase was 5,000 U/ml, which was obtained in fed-batch cultivation after 168 h induction with methanol in a 50 L bioreactor. The purified lipase showed high temperature tolerance, and being stable at 60℃ and kept 45% enzyme activity after 1 h incubation at 70℃. The stability, effects of metal ions and other reagents were also determined. The chain length specificity of the recombinant lipase showed high activity toward triolein (C18:1) and tripalmitin (C16:0).
$\alpha$-Galactosidase was partially purified from the culture filtrate of pichia guilliermondii by Mannobiose-Sepharose affinity column chromatography. The galactosidase exhibited maximum activity at pH 4.5 and 4$0^{\circ}C$, and was stable in the pH and temperature ranges of 4 to 5.5 and 30 to 6$0^{\circ}C$, respectively. The enzyme was inhibited by $Hg^{2+}$ and $Ag^{2+}$. The enzyme activity was ot affected considerably by treatment with other metal compounds. The enzyme hydrolyzed melibiose to galactose and glucose, raffinose to galactose and sucrose, and $Gal^{3}Man_{3}(6^{3}-$\alpha$-galactosyl-1,4-mannotriose)$ to galactose and mannotriose. On the contrary, it could not hydrolyze $Gal^{3}Man_{4}(6^{3}-galactosyl-1,4-$\alpha$-mannotetraose)$.
Glyoxalase I (Ee 4.4.1.5, lactoylglutathione lyase) from Chlamydomonas reinhardtii was purified to homogeneity by ammonium sulfate fractionation, anion-exchange chromatography, hydrophobic interaction chromatography, and affinity chromatography on S-hexylglutathione agarose. The purified enzyme was judged to be homogeneous on SDS-PAGE, and consisted of a single polypeptide chain with a relative molecular weight of 24,000. The enzyme was most active at $40^{\circ}C$ and pH 7.5. It was catalytically most active with methylglyoxal as substrate. A number of properties of the Chlamydomonas glyoxalase I enzyme, such as substrate specificity, molecular mass, kinetic parameters, pi, metal ion effect, have been determined and compared with those reported for preparations from other sources. It had somewhat different characteristics from mammalian enzymes.
Two kinds of new lectin fractions (LOA-I, LOA-II) were obtained from loach (Misgurnus spp.) meat by 0.15 M NaCl extraction, salt fractionation, ion exchange and hydroxyapatite column chromatographies. On polyacrylamide gel electrophoresis, LOA-I exhibited one major and a few minor bands, but LOA-II exhibited three minor bands. The partially purified loach lectins agglutinated not only erythrocytes of human B and AB type, rabbit, dog, but also murine splenic lymphocytes. Agglutinability was relatively labile at various pH and stable at increasing temperature, but was not affected by tested several metal ions. By the sugar specificity test, D-glucosamine and metyl-$\beta$-galactopyranose inhibited agglutinating activity at a final concentration of 3 mM. The lectins contained relatively high amounts of aspartic acid, valine and leucine, but sulfur containing amino acids, cystein, methionine and isoleucine were not determined. LOA-I, LOA-II lectins were nonmitogenic toward murine lymphocytes.
Bioleaching is the process in which insoluble metal sulfide is oxidized by specialized iron- and/or sulfur-oxidizing lithotrophic bacteria in acidic, metal-rich environments. Most of these processes are carried out by the genus Thiobacillus. Three novel Thiobacillus strains (Thiobacillus thiooxidans AZ11, Thiobacillus thiooxidans MET, and thiobacillus thiooxidans TAS) associated with bioleaching have been isolated from soil and sludge (Korean patent No. 1999-0073060 for T. thiooxidans AZ11, Korean patent No. 1999-0005798 for T. thiooxidans MET, and Korean patent No. 1999-0073059 for T. thiooxidans TAS). A partial sequence of 16S ribosomal RNA gene (16S rDNA) and the entire sequence of 16S/23S intergenic spacer region (ISR) were determined in the three above novel strains and in Thiobacillus ferrooxidans ATCC19859 as a reference strain. When phylogenetic analysis was performed based on G+C contents and sequence alignments, T. ferroxidans ATCC19859 was found to be closely related to previously registered T. ferrooxidans strains in a monophyletic manner, while the three novel T. thiooxidans strains were classified in a paraphyletic manner. Close examination on the base composition of 16S/23S ISR revealed that the 5\` part (nucleotide residues 21-200) was specific for the genus Thiobacillus. On the other end, the 3\` part (nucleotide residues 201-520) showed specificity in T. ferrooxidans strains, but not in T. thiooxidans strains. These results suggest that the proximal and distal halves of 16S/23S could be used as a genetic marker for the identification of the genus Thiobacillus and the species T. ferrooxidans, respectively.
조선소에서 선체 블록과 용접 피더의 위치를 알면 작업자의 위치 정보를 쉽게 얻을 수 있다. 이 데이터는 작업장 안전 모니터링 시스템 구축에 매우 유용하다. 그러나 선체 구조와 용접 공정의 특수성 때문에 작업장에 고정 통신망을 적용하기 어렵다. 본 연구에서는 전용 통신선을 대신할 수 있는 유도 전력선 통신을 선체블록과 같은 금속매체에 적용하는 기술을 검토하였다. 용접기의 전원 케이블에 설치할 유도 결합기로 페라이트 코어를 사용하였고, 금속 블록의 지지대에 체결할 결합기로 나노 결정질 코어를 적용하였다. 제안된 커플러는 COMSOL AC/DC 모듈로 3차원 모델링 하였고 동작 원리를 시각화하기 위하여 유한 요소 해석을 수행하였다. 알루미늄 프로파일을 사용한 금속블록 통신 성능 테스트에서 용접 전극의 블록 접촉으로 통신 채널이 형성되었을 때 대역폭은 6 Mbps 이상 유지되었다.
