Atypical Clinical Course of after Glyphosate Herbicide Intoxication (Glyphosate 음독의 임상양상)

  • Purpose: Although glyphosate-surfactant herbicide intoxication is relatively mild toxic in humans, we encountered an atypical clinical presentation. We performed this study to understand the atypical clinical course and factors associated with severe intoxication after glyphosate ingestion. Methods: We conducted a retrospective study of 61 patients (male 43, mean age $54{\pm}18.8$ years) who were presented with glyphosate ingestion between March 1997 and March 2011. The severe intoxication group was defined as patients with systolic blood pressure less than 90 mmHg, respiratory distress needing intubation, or altered mental state. Results: Of the 61 patients, 22 patients (36.1%) had a severe clinical course, 1 patient (1.6%) had died and 1 patient (1.6%) had moribund discharge. The most common symptoms were nausea with or without vomiting which occurred in 30 patients (49.2%). Twenty-seven patients had metabolic acidosis that was the second most common medical complication. Advanced age, pH, base excess, $HCO_3$, Sat, creatinine, X-ray abnormalities and ECG abnormalities were significant factors. Hemoglobin, platelet, $pO_2$, $pCO_2$, BUN, sodium, potassium and AST levels were not different comparing the laboratory characteristics between the severe and mild intoxication groups. Conclusion: The results of this study showed that severe intoxication occurred in 22 patients (36.1%) after glyphosate intoxication. Advanced age, pH, base excess, $HCO_3$, Sat, creatinine, X-ray abnormality and ECG abnormalities were significant predictive factors for severe intoxication in patients with glyphosate surfactant herbicide poisoning.

Regulation of Magnesium Release by cAMP during Chemical Hypoxia in the Rat Heart and Isolated Ventricular Myocytes

  • Chemically induced hypoxia has been shown to induce a depletion of ATP. Since intracellular free $Mg^{2+}\;([Mg^{2+}]_i)$ appears to be tightly regulated following cellular energy depletion, we hypothesized that the increase in $[Mg^{2+}]_i$ would result in $Mg^{2+}$ extrusion following hormonal stimulation. To determine the relation between $Mg^{2+}$ efflux and cellular energy state in a hypoxic rat heart and isolated myocytes, $[Mg^{2+}]_i,$ ATP and $Mg^{2+}$ content were measured by using mag-fura-2, luciferin-luciferase and atomic absorbance spectrophotometry. $Mg^{2+}$ effluxes were stimulated by norepinephrine (NE) or cAMP analogues, respectively. $Mg^{2+}$ effluxes induced by NE or cAMP were more stimulated in the presence of metabolic inhibitors (MI). Chemical hypoxia with NaCN (2 mM) caused a rapid decrease of cellular ATP within 1 min. Measurement of $[Mg^{2+}]_i$ confirmed that ATP depletion was accompanied by an increase in $[Mg^{2+}]_i.$ No change in $Mg^{2+}$ efflux was observed when cells were incubated with MI. In the presence of MI, the cAMP-induced $Mg^{2+}$ effluxes were inhibited by quinidine, imipramine, and removal of extracellular $Na^+.$ In addition, after several min of perfusion with $Na^+-free$ buffer, a large increase in $Mg^{2+}$ efflux occurred when $Na^+-free$ buffer was switched to 120 mM $Na^+$ containing buffer. A similar $Mg^{2+}$ efflux was observed in myocytes. These effluxes were inhibited by quinidine and imipramine. These results indicate that the activation of $Mg^{2+}$ effluxes by hormonal stimulation is directly dependent on intracellular $Mg^{2+}$ contents and that these $Mg^{2+}$ effluxes appear to occur through the $Na^+-dependent\;Na^+/Mg^{2+}$ exchange system during chemical hypoxia.

Widdrol Blocks 3T3-L1 Preadipocytes Growth and Differentiation Due to Inhibition of Mitotic Clonal Expansion

  • Adipocyte differentiation is strongly associated with obesity, which causes metabolic disorders. In this study, we investigated the inhibitory effects of widdrol on 3T3-L1 preadipocyte growth and differentiation. Widdrol decreased lipid droplet accumulation and down-regulated adipogenic transcription factors such as C/$EBP{\alpha}$, C/$EBP{\beta}$, and $PPAR{\gamma}$. Widdrol blocked preadipocyte proliferation and differentiation through the inhibition of mitotic clonal expansion, which was accompanied by the failure of degradation of p21, a cyclin-dependent kinase inhibitor. Cell-cycle analysis clearly indicated that widdrol actively induces cell-cycle arrest at the G1-S phage transition, causing cells to remain in the preadipocyte state. Moreover, widdrol increased p21 expression and inhibited Rb phosphorylation in preadipocyte incubated in a hormone medium. Therefore, these findings clearly suggest that widdrol blocks preadipocyte growth and differentiation through the inhibition of mitotic clonal expansion by p21-and Rb-dependent G1 arrest and can be developed as a potent anti-adipogenic agent for reducing obesity.

Studies on the Enzyme Activities and Heavy metals of Forest Soil in Mt. Nam. Seoul (남산 삼림 토양에서의 효소 활성도와 중금속 함량에 관한 연구)

  • This study was carried out to investigate to determine seasonal variation of dehydrogenase activity, phosphatase activity, adenosine tri-phosphate content and some physicochemical properties, such as soil pH, moisture content, organic matter and several heavy metal concentrations from Apr. 1997 to jan. 1998 in Pinus densiflora and Quercus mongolica forest in Mt. Nam, to explain a relationship between enzyme activity and the soil factors. There were ranges of 4.03-4.65 in soil pH, 18.65-51.09% in moisture content and 6.69-95.95% in orgainc matter. The organic matter content decreased with soil horizon, showing the higher values in Q. mongolica forest. In comparison to the results of Kawngneung site as control area, there were slightly differences due to a development level of forest ecosystem and microbial degradation of organic matter. The heavy metal concentrations showed 32.50-75.55 ${\mu}g/g$ in Cu, 69.33-134.84 ${\mu}g/g$ in Zn, 57.02-150.32 ${\mu}g/g$ in Pb, and 0.36-1.00 ${\mu}g/g$ in Mt. Nam. These values are higher than in Kwangneung site because of long-term exposure to air pollutants from central city. On the other hand, ATP contents in Mt. Nam were lower than in Kawngneung site in relation to soil organic matter, moisture content and relatively high heavy metal concentrations. ATP contents per soil weight was largest in F+H layer and in spring time of other seasons. Dehydrogenase activity as an index of soil microbial activity had a ranges of 170.67-1,221.66 ${\mu}g$ TPF/g that showed lower values than in Kawngneung site. However, phophatase activity had a contray tendency due to P fertilization for a continuous management. Those values increased through spring to a maximum in the summer and fall in autumn. This is basically caused by metabolic state of soil on the biological activity and several and several factors, such as aeration, soil temperature, vegetation and microflora.

  • Leucine-responsive regulatory protein (LRP) is a regulatory protein of molecular weight 18.8 kDa and is widely known to regulate many metabolic and functional activities of operons in Escherichia coli. The gene for Lrp from Escherichia coli in pQE system of 6 ${\times}$ His-tagging was expressed and $^3H$-labeled protein, as well as the wild type Lrp, was purified. The crosslink experiments were performed to analyze the quaternary structure of Lrp at high of $5{\mu}M$ and at low concentrations below $0.3{\mu}M$ with cross linkers, such as glutaraldehyde, 1, 2, 3, 4-diepoxy-butane (DEB), and ethylene glycol bis (succinimidyl succinate) (EGS). In the experiments, we found that the Lrp protein can be formed higher conformation states of tetramer, hexamer, octamer, as well as dimeric state when incubated with the above cross linkers.

The Role of Rice Vacuolar Invertase2 in Seed Size Control

  • Sink strength optimizes sucrose import, which is fundamental to support developing seed grains and increase crop yields, including those of rice (Oryza sativa). In this regard, little is known about the function of vacuolar invertase (VIN) in controlling sink strength and thereby seed size. Here, in rice we analyzed mutants of two VINs, OsVIN1 and OsVIN2, to examine their role during seed development. In a phenotypic analysis of the T-DNA insertion mutants, only the OsVIN2 mutant osvin2-1 exhibited reduced seed size and grain weight. Scanning electron microscopy analysis revealed that the small seed grains of osvin2-1 can be attributed to a reduction in spikelet size. A significant decrease in VIN activity and hexose level in the osvin2-1 spikelets interfered with spikelet growth. In addition, significant reduction in starch and increase in sucrose, which are characteristic features of reduced turnover and flux of sucrose due to impaired sink strength, were evident in the pre-storage stage of osvin2-1 developing grains. In situ hybridization analysis found that expression of OsVIN2 was predominant in the endocarp of developing grains. A genetically complemented line with a native genomic clone of OsVIN2 rescued reduced VIN activity and seed size. Two additional mutants, osvin2-2 and osvin2-3 generated by the CRISPR/Cas9 method, exhibited phenotypes similar to those of osvin2-1 in spikelet and seed size, VIN activity, and sugar metabolites. These results clearly demonstrate an important role of OsVIN2 as sink strength modulator that is critical for the maintenance of sucrose flux into developing seed grains.

Wastewater-based epidemiology for the management of community lifestyle and health: An unexplored value of water infrastructure (하수기반역학을 이용한 커뮤니티 생활상 및 건강 관리: 물 인프라의 새로운 가치)

  • Traditional wastewater research mainly focuses on 1) estimating the amount of waste entering sewage treatment facilities, 2) evaluating the treatment efficiency of sewage facilities, 3) investigating the role of sewage treatment effluent as a point source, and 4) designing and managing sewage treatment facilities. However, since wastewater contains a variety of chemical and biological substances due to the discharge of human excreta and material used for daily living into it, the collective constituents of wastewater are likely a reflection of a community's status. Wastewater-based epidemiology (WBE), an emerging and promising field of study that involves the analysis of substances in wastewater, can be applied to monitor the state of a defined community. WBE provides opportunities for exploiting indicators in wastewater to fulfill various objectives. The data analyzed under WBE are those pertaining to selected natural and anthropogenic substances in wastewater that are a result of the discharge of metabolic excreta, illicit or legal drugs, and infectious pathogens into the wastewater. This paper reviews recent progress in WBE and addresses current challenges in the field. It primarily discusses several representative applications including the investigation of drug consumption across different communities and the management of community disease and health. Finally, it summarizes established indicators for WBE.

Divergence of Granular Sludges and Microbial Communities in Two Types of Anaerobic Reactors Treating Different Wastewaters

  • An advanced anaerobic expanded granular sludge bed (AnaEG) and an internal circulation (IC) reactor, which were adopted to treat starch processing wastewater (SPW) and ethanol processing wastewater (EPW), were comprehensively analyzed to determine the key factors that affected the granules and microbial communities in the bioreactors. The granule size of $900{\mu}m$ in the AnaEG reactor was smaller than that in the IC reactor, and the internal and external morphological structures of the granular sludge were also significantly different between the two types of reactors. The biodiversity, which was higher in the AnaEG reactor, was mainly affected by reactor type. However, the specific microbial community structure was determined by the type of wastewater. Furthermore, the dominant methanogens of EPW were mainly Methanosaeta and Methanobacterium, but only Methanosaeta was a major constituent in SPW. Compared with the IC reactor, characteristics common to the AnaEG reactor were smaller granules, higher biodiversity and larger proportion of unknown species. The comparison of characteristics between these two reactors not only aids in understanding the novel AnaEG reactor type, but also elucidates the effects of reactor type and wastewater type on the microbial community and sludge structure. This information would be helpful in the application of the novel AnaEG reactor.

Subacute Oral Toxicity and Bacterial Mutagenicity Study of a Mixture of Korean Red Ginseng (Panax ginseng C.A. Meyer) and Salvia plebeia R. Br. Extracts

  • As various populations are rapidly becoming an aging society worldwide and interest in health issues has increased, demand for functional foods including herbal products has increased markedly to maintain a healthy state which has led to safety issues about their intake as an inevitable result. The objective of this study was to identify the safety profile of a Korean red ginseng and Salvia plebeia R. Br. extract mixture (KGC-03-PS) which is a valuable ingredient that can be used as a functional food. In the present study, the subacute oral toxicity and bacterial reverse mutagenicity of KGC-03-PS were evaluated. Sprague Dawley rats were administered KGC-03-PS orally for 28 days by gavage. Daily KGC-03-PS dose concentrations were 0, 500, 1,000, or 2,000 mg/kg body weight (bw) per day. Bacterial reverse mutation test with KGC-03-PS dose levels ranging from 312.5 to $5,000{\mu}g/plate$ was carried out by OECD test guideline No. 471. Five bacterial strains (Salmonella typhimurium TA98, TA100, TA1535, TA1537, and Escherichia coli WP2) were tested in the presence or absence of metabolic activation by plate incorporation method. There were no toxicological effects related with test substance in the clinical evaluation of subacute oral toxicity test including clinical signs, body weight, and food consumption. Moreover, no toxicological changes related to KGC-03-PS were observed in the hematological and serum biochemical characteristics as well as in the pathological examinations, which included organ weight measurements and in the gross- or histopathological findings. KGC-03-PS did not induce an increase in the number of revertant colonies in all bacterial strains of the bacterial reverse mutation test. The no-observed-adverse-effect level of KGC-03-PS is greater than 2,000 mg/kg bw/day, and KGC-03-PS did not induce genotoxicity related to bacterial reverse mutations under the conditions used in this study.

Definition, Role and Method of Yinyang Pattern Differentiation (음양변증(陰陽辨證)의 정의와 기능 및 판별방법 연구)

  • In order to ensure the fact that eight principle pattern differentiation is used clinically as a basic guideline for Korean medicine practice, the definition, role and method of yin-yang pattern differentiation with its case report were explored at first. Yinyang Pattern Differentiation is a method of discriminating human tendencies or morbidity based on the yin and yang characteristics expressed in living bodies. And yin and yang are the two contrasting characteristics and aspects of the interaction when certain physical conditions that have a lasting effect on the human physiological metabolic function are correlated with the morbidity. Specific methods of yinyang pattern differentiation can be divided into several types of yin and yang indicators. First, time and space factors like day and night, hot and cold seasons, above and below, topographical districts. Second, colors and pulse and their/or relative clearness and muddiness, hardness and softness, moving and resting. Third, diagnose yin and yang patterns through distinguishing the true and false of a fever and cold in an emergency phase such as increase of brain pressure and shock state. Fourth, general characteristics of the propensity and constitution of a subject such as body type, speech, behavior, and physiological metabolism. And for clinical use, these were summarized again as a symptom indicators of physical signs and color, pulse, tongue and questionnaire indicators of propensity, body type, and space-time characteristics. Conclusively, it was confirmed that yinyang pattern differentiation has its own diagnostic significance which is distinct from exterior-interior, cold-heat and deficiency-excess pattern differentiation.
