Reduction of Perchlorate and Nitrate by Citrobacter Amalonaticus Strain JB101 : Kinetics and the Applicability of MBR (Citrobacter Amalonaticus Strain JB101에 의한 과염소산염과 질산염의 환원 : Kinetics 및 MBR을 이용한 처리 가능성)

  • This study was performed to evaluate the characteristics of the competition between two electron acceptors, perchlorate and nitrate, with Citrobacter Amalonaticus strain JB101. In addition, the applicability of membrane bioreactor(MBR) for perchlorate removal was evaluated. The maximum growth rate of strain JB101 on perchlorate and nitrate are 0.27 and 0.58 $hr^{-1}$, and maximum substrate utilization rates were 35.1 mg $ClO_4^-/g$ protein-day and 45.6 mg $NO_3^-/g$ protein-day, respectively. Nitrate was a competitive inhibitor for perchlorate, and strain JB101 prefer nitrate to perchlorate as electron acceptor. Complete removal of perchlorate could be achieved up to the surface leading rate of 4.6 g $ClO_4^-/m^2-day$ with the MBR fed with 20 mg $ClO_4^-/L$(HCMBR). When 5 mg/L of nitrate was added to the same influent, perchlorate removal efficiency decreased to 96.5%, while nitrate was completely removed. For the MBR fed with 0.7 mg/L of perchlorate (LCMBR), the maximum perchlorate removal efficiency was 100% up to the loading rate of 0.23 g $ClO_4^-/m^2-day$. Membrane fouling was found to be a problem at high leading rate for both MBRs. The acetate consumption ratio per perchlorate was $13.7{\sim}51.7\;e^-eq./e^-eq.$ in LCMBR, while the value was $2.5{\sim}3.6\;e^-eq./e^-eq.$ in HCMBR. This difference could be related to the acetate consumption with oxygen as electron acceptor. Therefore, the amount of acetate addition must be determined considering the concentrations of other electron acceptors in the influent.

  • 본 연구는 membrane bioreactor (MBR)을 질산화 위주로 운영하면서 질소폐수 처리 과정에서 나타나는 MBR의 운전특성 및 처리효율을 검토하고, 아울러 경제적인 질소처리 방법인 아질산화를 유도하기 위한 조건을 탐색하는 것이다. 반응기의 유효부피 8 L, 체류시간을 12시간으로 고정하고 유입폐수 중의 암모니아 농도를 증가시키는 방법으로 암모니아 부하를 $2\;kg/m^3{\cdot}day$까지 증가시켜 보았다. 운전 중반부에 이유를 명확히 알 수 없는 슬러지의 활성 저하 기간을 제외하고는 거의 98% 이상의 질산화 효율을 나타냈으며, 2 kg 이상의 암모니아 부하에서는 효율이 감소하였다. pH 조절, 유입수의 암모니아 농도 및 부하 조절에 의해 MBR 내의 free ammonia (FA) 농도를 변화시켜, FA 저해에 의한 유도한 아질산화는 매우 불안정하게 이루어졌다. 반면에, 공기 공급량을 변화시켜 반응기 내의 DO를 낮추는 방법으로 유도한 아질산 축적은 안정적인 경향을 보여 MBR에서 적절한 운전전략으로 판단된다.

  • Chitosan (deacetylated form of chitin) possesses strong antibacterial activities such as antimicrobial effect, antifungal effect and the induction of plant defense response. Chitosan itself, however, has high molecular weight and viscosity as well as water-insolubility, These natures may restrict applications in various fields, especially in in vivo system. While the hydrolysates of chitosan, chitooligosaccharides (COS) are not only lower in the molecula. weight and viscosity, but also water-soluble. Thus, they would be expected more efficient absorption in vivo. Besides several documents have been reported antibacterial activities of COS against microorganisms (Kendra et al., 1989; Uchida et al., 1989). (omitted)

  • Despite a variety of development in fish farming during the last decades, fish diseases by bacteria, virus, and parasites are still major problems in aquaculture. Aquaculture of Hounder fish is widely performed around Korea as well as Jeju island, due to relatively stable seed production, short farming period, and a higher value in market. Fouling Mitigation for Pressurized Membrane of Side-Stream MBR Process at Abnormal Operation Condition (가압식 분리막을 이용한 Side-Stream MBR 공정의 비정상 운전조건에서 막 오염 저감)

  • Pressurized membrane used for side-stream MBR process requires fouling control strategy both for normal and abnormal operation conditions for stable operation of the facilities. In this study, $85m^3/day$ of pilot-scale side-stream MBR process was constructed for the evaluation of fouling mitigation by air bubble injection into the membrane module. In addition, fouling phenomena at abnormal operation conditions of low influent and/or loading rate were also investigated. Injection of air bubble was found to be effective in delaying transmembrane pressure (TMP) increase mainly due to scouring effect on the membrane surface, resulting in expanded filtration cycle at a high flux of $40L/m^2{\cdot}h$ (LMH). At abnormal operation condition, injection of PACl (53 mg/L as Al) into the bioreactor showed 19% reduction of TMP increase. However, inhibition of nitrifying bacteria by continuous PACl injection was observed from batch experiments. In contrast, injection of powdered activated carbon (PAC, 0.6 g/L) was able to maintain the initial TMP of $0.2kg/cm^2$ for 5 days at the abnormal conditions. It may have been caused from the adsorption of extracellular polymeric substances (EPS), which was known to be excessively released during growth inhibition condition and act as the major foulants in MBR operations.

  • Coarse pore filter could be an alternative of membrane for solid-liquid separation in an activated sludge reactor because of inexpensive cost of the filter material and high flux at low filtration pressure. However such filter module has much less specific filtration area compared to the membrane. Therefore a certain effort is required to increase the specific filtration area in the module design of such coarse pore filter for solid-liquid separation in an activated sludge reactor. In this study, tubular type coarse pore filter was designed at various diameter and configuration. The filtration performance was investigated to separate solid in the activated sludge reactor for domestic wastewater treatment. Tubular type coarse pore filter module could be successfully applicable to solid separation in the activated sludge reactor. The design parameters were the tube diameter of 10mm and vertical installation. Smaller diameter of the tube caused faster increase of the filtration pressure because of the hydraulic head loss in the tube channel.

Dynamic Respiratory Measurements of Corynebacterium glutamicum using Membrane Mass Spectormetry

  • The present work presents a novel approach for the dynamic quantification of respiration rates on a small scale by using lysine-producing Corynebacterium glutamicum ATCC 21253. Cells sampeld from batch cultures at different times were incubated ina 12-ml scale bioreactor equipped with a membrane mass spectrometer. Under dynamic conditions, gas exchange across the gas-liquid phase, specific respiration rates, and RQ values were precisely measured. For this purpose, suitable mass balances were formulated. The transport coefficients for $O_2$ and $CO_2$, crucial for calculating the respiration activity, were determined as $k_La_{O2}=9.18h^{-1}$ and $k_La_{CO2}=5.10h^{-1}$ at 400 rpm. The application of the proposed method to batch cultures of C. glutamicum ATCC 21253 revealed the maximum specific respiration rates of $q_{O2}=8.4\;mmol\;g^{-1}h^{-1}\;and\;q_{CO2}=8.7\;mmol\;g^{-1}h^{-1}$ in the middle of the exponential growth phase after 5 h of cultivation. When the cells changed from growth to lysine production due to the depletion of the essential amino acids theonine, methionine, and leucine, $q_{O2}\;and\;q_{CO2}$ decreased significantly and RQ increased. The respiration data exhibited an excellent agreement with previous cultivations of the strain [13]. This confirms the potential of the developed approach to realistically reflect the metabolic activities of cells at their point of sampling. The short-term influence of added threonine, methionine, and leucine was highest during the shift from growth to lysine production, where $q_{O2}\;and\;q_{CO2}$ increased 50% within one minute after the pulse addition of these compounds. Non-growing, yet lysine-producing cells taken from the end of the batch cultivation revealed no metabolic stimulation with the addition of threonine, methionine, and leucine.

  • Ethanol fermentration of glucose by a strain of Zymomonas mobilis KCTC 1535 was studied in membrane recycle bioreactor, which was coupled with closs flow hollow fiber membrane. The maximum values of product yields and productivity are 0.4685g total ethanol/ g glucose, 14.05g total ethanol/ L/h, respectively The pervaporation performance of the PDMS menbrane has been investigated for the separation of binary mixtures of EtOH/water at $50^{\circ}C{\sim}70^{\circ}C$. The optimum conditions of feed concentration, temperature, feed solution flow rates is determined to be 8%, $70^{\circ}C$, 492ml/min, respectively.   • In this study, effluent water was produced through Submerged Membrane Bio-Reactor(SMBR) process, which is a simple system and decomposes organic matter contained in wastewater with biological treatment process and performs solid-liquid separation, Especially, ozone oxidation treatment process is applied to effluent water containing fluorescent whitening agent, which is a trace pollutant which is not removed by biological treatment, and influences the quality of reused water. The concentration of $COD_{Cr}$ in the SMBR was $449.3mg/{\ell}-COD_{Cr}$, and the concentration of permeate water was $100.3mg/{\ell}-COD_{Cr}$. The removal efficiency was about 70.1%. The amount of ozone required for the removal of the fluorescent whitening agent in the permeated water in SMBR was $6.67g-O_3/min$, and the amount of ozone required to remove $COD_{Mn}$ relative to the permeate water was calculated to remove $0.997mg-COD_{Mn}$ for 1mg of $O_3$.
