Effect of the Shape and Size of Quorum-Quenching Media on Biofouling Control in Membrane Bioreactors for Wastewater TreatmentS

  • Recently, spherical beads entrapping quorum quenching (QQ) bacteria have been reported as effective moving QQ-media for biofouling control in MBRs for wastewater treatment owing to their combined effects of biological (i.e., quorum quenching) and physical washing. Taking into account both the mass transfer of signal molecules through the QQ-medium and collision efficiencies of the QQ-medium against the filtration membranes in a bioreactor, a cylindrical medium (QQ-cylinder) was developed as a new shape of moving QQ-medium. The QQ-cylinders were compared with previous QQ-beads in terms of the QQ activity and the physical washing effect under identical loading volumes of each medium in batch tests. It was found that the QQ activity of a QQ-medium was highly dependent on its specific surface area, regardless of the shape of the medium. In contrast, the physical washing effect of a QQ-medium was greatly affected by its geometric structure. The enhanced anti-biofouling property of the QQ-cylinders relative to QQ-beads was confirmed in a continuous laboratory-scale MBR with a flat-sheet membrane module.

  • Due to the low biodegradability of dyes, conventional biological wastewater treatment systems are inefficient in treating textile wastewater. In this study, white rot fungus, Trametes versicolor KCTC 16781, were investigated for the decolorization of Reactive black 5 solutions. This fungus was able to degrade the dye solutions by the ligninolytic enzymes (laccase and MnP) produced. The enzyme activity remained constant until the end of reaction. The combined process of biological treatment and ceramic membrane showed better efficiency for decolorization and TOC removal than each single process.

Reduction of energy demand for UF cross-flow membranes in MBR by sponge ball cleaning

  • Sponge ball cleaning can generate an abrasion effect, which leads to an attractive increasing in both permeate flux and membrane rejection. The aim of this study was to investigate the influence of the daily sponge ball cleaning (SBC) on the performance of different UF cross-flow membrane modules integrated with a bioreactor. Two 1"-membrane modules and one 1/2"-membrane module were tested. The parameters measured and controlled are temperature, pH, viscosity, particle size, dissolved organic carbon (DOC), total suspended solids (TSS), and permeate flux. The permeate flux could be improved by 60%, for some modules, after 11 days of daily sponge ball cleaning at a transmembrane pressure of 350 kPa and a flow velocity of 4 m/s. Rejection values of all tested modules were improved by 10%. The highest permeate flux of 195 L/㎡.h was achieved using a 1"-membrane module with the aid of its negatively charged membrane material and the daily sponge ball cleaning. In addition, the enhancement in the permeate flux caused by daily sponge ball cleaning improved the energy specific demand for all tested modules. The negatively charged membrane showed the lowest energy specific demand of 1.31 kWh/㎥ in combination with the highest flux, which is a very competitive result.

Production of Citric Acid in a Flat-type Membrane Bioreactor (평판형 막 생물 반응기를 이용한 구연산의 생산)

  • A flat-type membrane bioreactor(FMBR ) for aerobic whole cell immobilization was developed and its performance for the citric acid production was investigated using Aspergillus niger (KCTC 1232). The reactor consisted of three layers. The top layer contained flowing air for oxygen supply, the middle layer had stationary cells, and the bottom layer had flowing aqueous nutrients. The initial pH of the culture medium played an important role in citric acid production and the lower initial pH of the culture medium resulted in a higher citric acid yield. Under air and pure oxygen aerations the volumetric productivity reached 0.20 and 0.40g/Lh. Furthermore, the productivity improved with the increase of the culture medium feed rate.

Electrochemical and Biochemical Analysis of Ethanol Fermentation of Zymomonas mobilis KCCM11336

  • An electrochemical bioreactor (ECB) composed of a cathode compartment and an air anode was used in this study to characterize the ethanol fermentation of Zymomonas mobilis. The cathode and air anode were constructed of modified graphite felt with neutral red (NR) and a modified porous carbon plate with cellulose acetate and porous ceramic membrane, respectively. The air anode operates as a catalyst to generate protons and electrons from water. The growth and ethanol production of Z. mobilis were 50% higher in the ECB than were observed under anoxic nitrogen conditions. Ethanol production by growing cells and the crude enzyme of Z. mobilis were significantly lower under aerobic conditions than under other conditions. The growing cells and crude enzyme of Z. mobilis did not catalyze ethanol production from pyruvate and acetaldehyde. The membrane fraction of crude enzyme catalyzed ethanol production from glucose, but the soluble fraction did not. NADH was oxidized to $NAD^+$in association with $H_2O_2$reduction, via the catalysis of crude enzyme. Our results suggested that NADH/$NAD^+$balance may be a critical factor for ethanol production from glucose in the metabolism of Z. mobilis, and that the metabolic activity of both growing cells and crude enzyme for ethanol fermentation may be induced in the presence of glucose.

Concentration of 6-Amonopenicillanic Acid from Penicillin Bioconversion Solution and Its Mother Liquor by Nanofiltration Membrane

  • In this study, nanofiltration was applied to the concentration of the 6-aminopenicillinic acid (6-APA) from bioconverted penicillin solution and also to its mother liquor. The 6-APA in the solution was concentrated from 0.211 mol/L to 0.746 mol/L by nanofiltration. The final maximum concentration was 3.6 times higher than the initial concentration an the recovery yield was 97% to 99% of the original 6-APA. The concentrated solution was crystallized with the yields of 88.9-90.2% and the purity of the crystallized product was about 98%. The concentration of 6-APA in the mother liquor after crystallization was 0.014 mol/L and thus was concentrated 20-30 fold by nanofiltration and crystallization. The recovery of 6-APA was over 98%. The salts contained in the mother liquor, such as NH$_4$Cl and KCl, could be removed by allowing them to permeate through the membrane.

Assessment of the Cytotoxic and Apoptotic Effects of Chaetominine in a Human Leukemia Cell Line

  • Chaetominine is a quinazoline alkaloid originating from the endophytic fungus Aspergillus fumigatus CY018. In this study, we showed evidence that chaetominine has cytotoxic and apoptotic effects on human leukemia K562 cells and investigated the pathway involved in chaetominine-induced apoptosis in detail. Chaetominine inhibited K562 cell growth, with an $IC_{50}$ value of 35 nM, but showed little inhibitory effect on the growth of human peripheral blood mononuclear cells. The high apoptosis rates, morphological apoptotic features, and DNA fragmentation caused by chaetominine indicated that the cytotoxicity was partially caused by its pro-apoptotic effect. Under chaetominine treatment, the Bax/Bcl-2 ratio was upregulated (from 0.3 to 8), which was followed by a decrease in mitochondrial membrane potential, release of cytochrome c from mitochondria into the cytosol, and stimulation of Apaf-1. Furthermore, activation of caspase-9 and caspase-3, which are the main executers of the apoptotic process, was observed. These results demonstrated that chaetominine induced cell apoptosis via the mitochondrial pathway. Chaetominine inhibited K562 cell growth and induced apoptotic cell death through the intrinsic pathway, which suggests that chaetominine might be a promising therapeutic for leukemia.

  • Pervaporation can be successfully utilized to recover alcohol from fermentation broths and dilute process streams. Polydimethylsiloxane (thickness: $60\mu$m), such as hydrophobic membrane have been employed in this application to produce an enriched product. The fermented alcohol was continuously extracted, and simultaneously concentrated by pervaporation, from the membrane bioreactor, and the extracted alcohol concentration was 6 to 10 times higher than in the broth. This experimental conditions were 6-10 Torr vacuum line, pH=5, $32^{\circ}C$, 3.5 liters of working volume, 200rpm of agitator speed, and 0.14 wm aeration flow rate.
