Progressive collapse resistance of RC buildings can be analyzed by considering column loss scenarios. Using finite element analysis and a static test, the progressive collapse process of a RC frame under monotonic vertical displacement of a side column was investigated, simulating a column removal scenario. A single-story 1/3 scale RC frame that comprises two spans and two bays was tested and computed, and downward displacement of a side column was placed until failure. Our study offers insight into the failure modes and progressive collapse behavior of a RC frame. It has been noted that the damage of structural members (beams and slabs) occurs only in the bay where the removal side column is located. Greater catenary action and tensile membrane action are mobilized in the frame beams and slabs, respectively, at large deformations, but they mainly happen in the direction where the frame beams and slabs are laterally restrained. Based on the experimental and computational results, the mechanism of progressive collapse resistance of RC frames at different stages was discussed further. With large deformations, a simplified calculation method for catenary action and tensile membrane action is proposed.
Action of anthraquinone on the sodium plus potassium activated ATPase activity in the rabbit red cell membrane has been investigated and the experiments were also designed to determine the mechanism of action of anthraquinone on the ATPase activity. The following results were obtained 1. The activity of the NaK ATPase from red cell membrane is inhibited by anthraquinone and the concentration of anthraquinone for maximal inhibition is about 5mM. 2. The ratio of inhibition of NaK ATPase by anthraquinone, with a giving concentration of sodium in the medium, is increased by raising the potassium concentration. 3. The ratio of inhibition of NaK ATPase by anthraquinone, with a given concentration of potassium in the medium, is increased by raising the sodium concentration. 4. The action of anthraquinone on the NaK ATPase activity is inhibited by calcium ions and the ratio of inhibition is increased by small amounts of calcium but almost constant by larger amounts. 5. The inhibitory action of anthraquinone on the NaK ATPase activity was not related to the amino group of lysine, the hydroxyl group of threonine or the imidazole group of histidine. 6. The inhibitory action of anthraquinone on the ATPase activity is due to sulfhydryl group or the carboxyl group of the enzyme of NaK ATPase.
In this paper, we take advantage of Wavelet Transform to identify damping ratios of membrane structures under wind action. Due to the lightweight and flexibility of membrane structures, they are very sensitive to the wind load, and show a type of fluid-structure interaction phenomenon simultaneously. In this study, we firstly obtain the responses of an air-supported membrane structure by ADINA with the consideration of this characteristic, and then conduct Wavelet Transform on these responses. Based on the Wavelet Transform, damping ratios could be obtained from the slope of Wavelet Transform in a semi-logarithmic scale at a certain dilation coefficient. According to this principle, damping ratios could eventually be obtained. There are two numerical examples in this study. The first one is a simulated signal, which is used to verify the accuracy of the Wavelet Transform method. The second one is an air-supported membrane structure under wind action, damping ratios obtained from this method is about 0.05~0.09. The Wavelet Transform method could be regarded as a very good method for the the damping analysis, especially for the large spatial structures whose natural frequencies are closely spaced.
Periplanetasin-4 is an antimicrobial peptide with 13 amino acids identified in cockroaches. It has been reported to induce fungal cell death by apoptosis and membrane-targeted action. Analogs were designed by substituting arginine residues to modify the electrostatic and hydrophobic interactions accordingly and explore the effect of periplanetasin-4 through the increase of net charge and the decrease of hydrophobicity. The analogs showed lower activity than periplanetasin-4 against gram-positive and gram-negative bacteria. Similar to periplanetasin-4, the analogs exhibited slight hemolytic activity against human erythrocytes. Membrane studies, including determination of changes in membrane potential and permeability, and fluidity assays, revealed that the analogs disrupt less membrane integrity compared to periplanetasin-4. Likewise, when the analogs were treated to the artificial membrane model, the passage of molecules bigger than FD4 was difficult. In conclusion, arginine substitution could not maintain the membrane disruption ability of periplanetasin-4. The results indicated that the attenuation of hydrophobic interactions with the plasma membrane caused a reduction in the accumulation of the analogs on the membrane before the formation of electrostatic interactions. Our findings will assist in the further development of antimicrobial peptides for clinical use.

The activating effect of aconite on the ATPase was not related to the sulfhydryl group of cysteine, the amino group of lysine, the hydroxyl group of threonine or the imidazole group of histidine. 6. The action of aconite on the ATPase activity is due to carboxyl group of the enzyme of NaK ATPase.

Lycopene, an acyclic carotenoid found in tomatoes (Lycopersicon esculentum) and a number off fruits, has shown various biological properties, but its antifungal effects remain poorly understood. The current study investigated the antifungal activity of lycopene and its mode of action. Lycopene showed potent antifungal effects toward pathogenic fungi, tested in an energy-independent manner, with low hemolytic effects against human erythrocytes. To confirm the antifungal effects of lycopene, its effects on the dimorphism of Candida albicans induced by fetal bovine serum (FBS), which plays a key role in the pathogenesis of a host invasion, were investigated. The results showed that lycopene exerted potent antifungal activity on the serum-induced mycelia of C. albicans. To understand the antifungal mode of action of lycopene, the action of lycopene against fungal cell membranes was examined by FACScan analysis and glucose and trehalose-release test. The results indicated that lycopene caused significant membrane damage and inhibited the normal budding process, resulting from the destruction of membrane integrity. The present study indicates that lycopene has considerable antifungal activity, deserving further investigation for clinical applications.

The plasma membrane and microsomes, isolated from the cells treated with hematoporphyrm derivative (HpD) for 1 and 24 h, accumulated the aggregated porphyrin. The quantity of aggregated porphyrin was same in the plasma membrane and microsomes after isolating them from cells treated with HpD for 1 h whereas the microsomes accumulated higher quantity of aggregated porphyrin when cells were treated with HpD for 24 h. Photodynamic action of aggregated porphyrin on plasma membrane and microsomes was investigated using lipid specific fluorescent probes: 1,6-diphenyl-1,3,5-hexatrine (DPH) and 1-(4-trimethylammonium), 6-diphenyl-1,3,5-hexatrine(TMA-DPH). The time dependent anisotropy of these probes in the membranes was measured and the decay of anisotropy was analyzed using wobbling in cone model. Upon irradiation both the plasma membrane and the microsomes showed an increase in the limiting anis~)tropy and order parameter and a decrease in the cone angle of the lipid probes. The increase in the limiting anisotropy was pronounced in membranes isolated from the cells treated with HpD for 24 h. Photoinduced change in the limiting anisotropy was dependent on the duration of incubation of cells with HpD before isolating the membranes. In both the membranes. the membrane core was affected more as compared to the outer leaflet. In addition to the structural changes, a decrease in Na$^+$-K$^+$-ATPase and NADPH cyt c reductase activity was also observed upon irradiation of HpD treated cells. Inhibition in NADPH cyt c reductase was more when cells were treated with HpD for 24 h, however, Na$^+$-K$^+$-ATPase activity did not depend on the duration of the treatment of cells with HpD before irradiation. Our results suggest that the extent of photoinduced perturbations in the membranes varies as a function of duration of the treatment of cells with HpD and the membrane core is more susceptible to the photodynamic action of aggregated porphyrin.
The mechanisms controlling the intensity and duration of synaptic transmission are numerous. Once an action potential reaches a nerve terminal, the stored neurotransmitters are released in a quantum fashion into the synaptic cleft. At that point neurotransmitters can act on post-synaptic receptors to elicit an action on the post-synaptic cell or net at so-called auto-receptors that are located on the presynaptic side and which often regulate the further release of the neutotransmitter. Whereas the action of the neurotransmitter receptors is regulated by desensitization phenomenon, the major mechanism by which the intensity and duration of neurotransmitter action is presumably regulated by either its degradation or its removal from the synaptic cleft. In the central nervous system, specialized proteins located in fe plasma membrane of presynaptic terminals function to rapidly remove neurotransmitters from the synaptic cleft in a sodium chloride-dependent fashion. These proteins have been referred to as uptake sites or neurotransmitter transporters. Once taken up by the plasma membrane transporters, neurotransmitters are repackaged into secretory vesicles by distinct transporters which depend on a proton gradient.
Both polar and gravity-induced lateral transport of auxin was markedly reduced in corn coleoptile segments by octanol treatment. Octanol enhance net auxin uptake without affecting that of benzoic acid, suggesting that the effect did not result from a nonspecific action on general membrane permeability. Since naphthylphthalamic acid (NPA) action on both transport and net uptake of auxin was substantially decreased in the presence of octanol, a specific interaction of octanol with the NPA site (efflux carrier) can be postulated. Studies on in vitro binding of NPA to membrane vesicles indicated that octanol did not interfere with NPA binding. When basipetal transport of auxin was impared by plasmolysis, octanol still inhibited auxin transport in the plasmolyzed tissues. The results ruled out the possibility of octanol acting at the plasmodesmata. Kinetic analysis of growth indicated that IAA-sustained growth was rapidly blocked by octanol implicating a common system by which auxin transport is linked to auxin action. Possible mechanisms for octanol action will be discussed.
