Objectives : The purpose of this study was to research the whitening effects and developing by cosmetics of the extract fromDioscoreae Rhizoma, which is one of the most popular health-promoting herb in herbal medications. Methods : We performed tyrosinase inhibition assay, reverse transcription-polymerase chain reaction (RT-PCR) and western blot for whitening effects. Also we measured MTT assay for cell viability. Results : The results were obtained as follows : For whitening effect, tyrosinase inhibition rate of extract fromDioscoreae Rhizomashowed more than 42.28% at 1,000 ㎍/㎖ concentration. Cell toxicity effect on melanoma cells (B16F10) of extract fromDioscoreae Rhizomashowed 81.97% with toxicity at 50 ㎍/㎖ concentration. So we were measured at a concentrations of 5, 10 and 50 ㎍/㎖ in all experiments involving cell. In addition, whitening related mRNAs including microphthalmia associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), tyrosinase were reduced byDioscoreae Rhizoma. We also foundDioscoreae Rhizomatransiently decreased protein kinase A (PKA) which is known to be upstream to the down regulation of MITF and tyrosinase. But phosphorylation of extracellular signal related kinase (pERK) were increased byDioscoreae Rhizoma. These results imply thatDioscoreae Rhizomadecrease melanogenesis via ERK activation and subsequent down regulation of MITF and tyrosinase. Conclusions : Therefore, all these findings suggested the potent usage ofDioscoreae Rhizomaas materials of functional cosmetics by confirming whitening activity related with melanin content.
The need for natural cosmetic ingredients has been increasing over the world nowadays. Agarose, a natural polymer from red seaweeds, has high hydrophilic character and a function of scaffolder. As skin moisturizer, agarose is adequate for percutaneous absorption. While, blueberry fruits extract possesses rich procyanidins and anthocyanins which show health benefits, anti-oxidant effect, anti-aging and anti-melanogenesis. Stability, sensory preference, skin trouble of the emulsion formula are important for cosmetic product development. In this study, we manufactured an emulsion formula for skin moisturizers using the two ingredients and tested emulsion stability and skin trouble. Total phenolic contents of the blueberry fruits extract were evaluated as well as tyrosinase inhibitory and collagenase inhibitory activities. $IC_{50}$ values of blueberry fruits extract for anti-tyrosinase and anti-collagenase activities were 168 and $112{\mu}g/mL$, respectively using gallic acid as a control ($64.8{\mu}g/mL$). The stability (pH and viscosity) of the formula containing 2% blueberry fruits extracts and 0.1% agarose was measured at five different temperatures (room temp., $25^{\circ}C$, $55^{\circ}C$, $45^{\circ}C$, $55^{\circ}C$) under the sun light at 2 day intervals for 12 days. There has been little pH change at the different temperatures. According to the sensory evaluation, there was no significant flavor, discoloration and physical changes of the formula at $25-65^{\circ}C$. These results suggest that emulsion formula containing blueberry extract and agarose could be used as a candidate for lotion and essence products.
In this study we investigated antioxidant activity of against several free radicals and skin whitening effect of 70% ethanol extract (leaf extracts and branch/stem mixed) of Lindera obtusiloba BL. Antioxidant activity was assessed by DPPH, superoxide radical and hydroxyl radical assays. The Lindera obtusiloba BL. extract had antioxidant activity dose dependently with an ${IC}_{50}$ value of 243.14 and 181.10 ${\mu}g$/ml for DPPH, 165.77 and >1500 ${\mu}g$/ml for non-enzymatic system of superoxide radical assay, 35.47 and >100 ${\mu}g$/ml for enzymatic system of superoxide radical assay, 1.21 mg/ml for hydroxyl radical assay. In addition we tested tyrosinase inhibition activity and melanin contents on B16 melanoma F10. B16 melanoma cell was treated by such sample as 1, 5, 10 and 50 ${\mu}g$/ml for 72 hr and tyrosinase inhibition was tested. Melanogenesis was inhibited to 22% at the dose of 50 ${\mu}g$/ml and tyrosinase was inhibited to 45.2% at the same dose. In conclusion Lindera obtusiloba BL had potent antioxidant activity and inhibitory activity of tyrosinase and melanin formation. It could be developed as the health functional food and functional cosmetic resources.
Tyrosinase ubiquitously existing from microorganisms to animals and plants is known to be the most critical and rate limiting enzyme during melanin biosynthesis. In order to develop new tyrosinase inhibitor we have synthesized 14 diphenyl ether compounds possessing hydroxyl, bromo, and nitro groups in the structure. Among the compounds prepared, 18 and 19 have shown much stronger inhibition of tyrosinase monophenolase function than arbutin used as a positive control. Both compounds 18 and 19 possess para-hydroxyphenyl moiety in their structure, which might reinforce the importance of p-hydroxyphenyl group in the tyrosinase inhibitory process. In the DPPH radical scavenging activity test, none of the compounds even 18 and 19 showed significant antioxidant activity. The results suggest that elaborate adjustment of diphenyl ether analogues with proper substituents have potential to be developed as new skin whitening agents working on the tyrosinase function.

In this study, we investigated the effect of branch extracts from Vaccinium oldhamii (VOB) on melanin synthesis in B16F10 cells. VOB promoted melanin production in absence or presence of ${\alpha}$-melanocyte-stimulating hormone (${\alpha}-MSH$) in B16F10 cells. However, VOB did not affect the expression of tyrosinase and TRP-1 associated with melanin synthesis at the mRNA and protein level in B16F10. But, VOB decreased TRP-2 protein level and induced tyrosinase activation in B16F10 cells. Inhibition of tyrosinase activity and tyrosinase knockdown attenuated VOB-mediated melanin synthesis. In conclusion, it is thought that VOB may stimulate melanin synthesis through activating tyrosinase activity.

To identify the natural and novel cosmeceutical ingredients which have whitening function, we examined the effects of tyrosinase activity and melanin production on halophyte Limonium tetragonum (Thunb.) A. A. Bullock extracts. As a result, the extracts showed whitening effect with no cytotoxicity. Melanin contents of B16F10 melanoma cells were decreased in a dose-dependent manner at 50, 100, $200{\mu}g/mL$ treatment of the extracts. In tyrosinase activity inhibition test, L. tetragonum (Thunb.) A. A. Bullock extracts showed decreased tyrosinase activity as the concentration of ${\alpha}-MSH$ was increased. Furthermore, it was observed that the tyrosinase and MITF expression was significantly downregulated by adding L. tetragonum (Thunb.) A. A. Bullock extracts in ${\alpha}-MSH$, a melanogenesis inducing material, treated B16F10 melanoma cells. These results indicate that L. tetragonum (Thunb.) A. A. Bullock extract might be an effective whitening agent by inhibit melanin formation.

In this study, we have investigated the hypo-pigmentary mechanism of methanol extract of Forsythiae Fructus in human melanocyte cell line, HM3KO. Treatment of HM3KO cells with Forsythiae Fructus extract markedly inhibited melanin biosynthesis in a dose-dependent manner. Decreased melanin contents occurred through the decrease of tyrosinase protein and activity. The mRNA levels of tyrosinase and tyrosinase-related protein 1 (TRP-1) were also reduced by Forsythiae Fructus extract. Moreover, the level of intracellular cyclic AMP (cAMP) was significantly decreased by treatment of Forsythiae Fructus extract. These results suggest that Forsythiae Fructus reduces melanin synthesis by down regulation of tyrosinase mRNA transcription, and this is closely related to the cAMP-dependent pathway.

Objective : This experimental study was preformed to investigate the effect of ethanol extract of Belamcandae Rhizoma (EBR) on dermal bioactive properties related to cosmeceuticals such as radical scavenging activity and depigmentation. Methods : The free radical scavenging activity of EBR was assayed in cell free systems using 1,1-diphenyl-2-picrylhydrazyl (DPPH). Antibacterial potency was measured by the size of inhibition zone with change of volume. Cell viability was measured by MTT assay and tyrosinase activity was assessed using L-DOPA. Results : EBR showed slightly radical scavenging activity and protected against UVB-induced cell cytotoxicity. Futhermore, EBR showed antibacterial activities on S. epidermidis and S. aureus. Although the proliferation of B16/F10 cells was decreased by EBR, necrosis was not revealed. EBR augmented tyrosinase activity and dendrite outgrowth in B16/F10 cells. Conclusions : These results suggest that EBR has antioxidative & antibacterial activities, and stimulate melanogenesis.
