Effect of Ipomoea aquatica extract on anti-melanogenesis and skin barrier function (공심채 추출물의 멜라닌 생성 저해 및 피부장벽 개선 효과)

  • We investigated the applications of functional materials through the examination of a variety of physiological activities of Ipomoea aquatica extract. I. aquatica extract showed low cytotoxicity against murine melanoma B16F10 cells. At concentrations that exerted little or no cytotoxicity to the cells, I. aquatica extract showed high DPPH radical scavenging activity ($ID_{50}$, $7.84{\mu}g/mL$), inhibited tyrosinase activity ($ID_{50}$, $106.56{\mu}g/mL$), and decreased melanin content ($ID_{50}$, $41.75{\mu}g/mL$). The treatment of B16F10 cells with I. aquatica extract suppressed the protein expression of tyrosinase in a dose-dependent manner. These findings suggested that I. aquatica extract inhibited melanin synthesis in murine melanoma B16F10 cells through the suppression of intracellular tyrosinase expression, as well as the simultaneous direct inhibition of tyrosinase activity. Additionally, I. aquatica extract promoted the expression of involucrin, which is related to skin barrier protection. These results indicate that I. aquatica extract may be an appropriate material for the improvement of skin barrier function.

Antioxidant and Whitening Effects of Agrimonia pilosa Ledeb Water Extract (짚신나물 물 추출물의 항산화 활성 및 미백효과에 관한 연구)

  • This study was performed to assess the antioxidant activities and whitening effects of Agrimonia pilosa Ledeb on melanin synthesis. The whitening effects of Agrimonia pilosa Ledeb water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effect of Agrimonia pilosa Ledeb water extract on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effect of Agrimonia pilosa Ledeb onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activitie. Our results indicated that Agrimonia pilosa Ledeb water extract effectively inhibited free radical generation. In DPPH and hydroxy radical scavenging activity, Agrimonia pilosa Ledeb water extract had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Agrimonia pilosa Ledeb suppressed the expression of tyrosinase in B16BL6 melanoma cells. These results show that Agrimonia pilosa Ledeb inhibited melanin production on the melanogenesis. The underlying mechanism of Agrimonia pilosa Ledeb on whitening activity may be due to the inhibition of tyrosinase activity. We suggest that Agrimonia pilosa Ledeb may be useful as new natural active ingredients for antioxidant and whitening cosmetics.

Anti-melanogenic effect of Eruca sativa extract (루꼴라(Eruca sativa) 추출물의 멜라닌 생성 저해 효과)

  • Eruca sativa (known as rocket plant) is a member of the Brassicaceae, which is considered an important chemo-preventive plant family. Although Eruca sativa has positive biological effects such as antioxidant and renal protective activities, the effect of the Eruca sativa extract as a therapeutic agent for skin whitening has not been reported. In this study, we investigated the applicability of the extract of Eruca sativa as a functional materials by examining the its physiological activities. The Eruca sativa extract showed low cytotoxicity against murine melanoma B16F10 cells. At concentrations (below 100 mg/L) that showed none or little cytotoxicity, the Eruca sativa extract showed high DPPH radical scavenging activity (ID50, 17.60 mg/L). In addition, the Eruca sativa extract inhibited tyrosinase activity ($ID_{50}$, 132.54 mg/L) and decreased melanin content ($ID_{50}$, 158.90 mg/L). Finally, the treatment with the Eruca sativa extract suppressed the protein expression of tyrosinase in a concentration-dependent manner. These findings suggested that the Eruca sativa extract inhibited melanin synthesis by not only suppressing intracellular tyrosinase expression but also directly inhibiting tyrosinase activity. Therefore, these results indicate that the Eruca sativa extract may be an effective material for functional cosmetics such as skin whitening materials.

  • To develop a functional beverage from Korean cherry (Prunus tomentosa Thunberg), inhibition effect of ethanol extract and juice of the korean cherry on melanin systhesis and tyrosinase activity in vitro was investigated. Inhibition ratio of tyrosinase activity increased as the concentration of solid of korean cherry juice increased, and inhibition affect was high in initiation step of enzyme reaction and then gradually decreased. Inhibition ratio of tyrosinase activity was high in the 70% (v/v) ethanol extract of the cherry and the highest in the ethyl acetate fraction of the 70% (v/v) ethanol extract. Ultimatly, the amounts of functional matter (melanin synthesis inhibitor) in the cherry was highest in ethyl acetate fraction of the ethanol extract.

CoMFA Analysis on Inhibitory Effect of $3{\beta}$-Hydroxy-12-oleanen-28-oic Acid Analogues on PTP-1B Activity and Prediction of Active Compounds ($3{\beta}$-Hydroxy-12-oleanen-28-oic Acid 유도체들의 PTP-1B 저해활성에 대한 CoMFA 분석과 활성 분자들의 예측)

  • The comparative molecular field analyses(CoMFA) models between the substituents with changing groups($R_1{\sim}R_4$) of $3{\beta}$-hydroxy-12-oleanen-28-oic acid derivatives as substrate molecule and their inhibitory activities($pI_{50}$) against protein tyrosine phosphatase(PTP)-1B were derived and discussed quantitatively. The optimized CoMFA F1 model have best predictability and fitness($r^2_{cv.}=0.654$ and $r^2_{ncv.}=0.995$). The order of contribution ratio (%) with CoMFA fields on the inhibitory activities was a steric field(53.0%), electrostatic field(36.2%) and hydrophobic field(10.8%). From the analytical results of CoMFA contour maps, the inhibitory activities were dependent on the R4 group in substrate molecules. Particularly, the new active compounds(P1 & P2) with the inhibitory activity against melanin synthesis were expected.

Inhibitory Effects on Melanin Production of Demethylsuberosin Isolated from Angelica gigas Nakai (참당귀로부터 분리한 Demethylsuberosin의 멜라닌 생성 억제 효과)

  • The anti-melanogenic substance was isolated from the root of Angelica gigas Nakai by silica gel column chromatography, preparative HPLC and TLC. As a result of the structure analysis by mass, $^1H$-NMR, and $^{13}C$-NMR spectrometry, the compound was identified as demethylsuberosin. Demethylsuberosin reduced melanin contents of B16F1 melanoma cells in a dose-dependent manner and decreased to about 74% at a concentration $5{\mu}g/ml$. Demethylsuberosin inhibited the expression in microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein-1 (TRP-1), and tyrosinase related protein-2 (TRP-2) in melanocytes. These results suggest that the whitening activity of demethylsuberosin may be due to the inhibition of the melanin synthesis by down-regulation of MITF, tyrosinase, TRP-1 and TRP-2 expression. Thus, our results provide evidence that demethylsuberosin might be useful as a potential skin-whitening agent.

The Anti-melanogenic Effect of Geranium krameri Extract (선이질풀 추출물의 멜라닌 생성 억제 효과)

  • We investigated the application of functional materials by examining a variety of physiological activities of Geranium krameri extract obtained from the National Institute of Horticultural and Herbal Science. Geranium krameri extract had a low cytotoxicity against murine melanoma B16F10 cells. At concentrations with little or no cytotoxicity, Geranium krameri extract showed high DPPH radical scavenging activity ($IC_{50}$, $8.72{\mu}g/mL$) and anti-microbial activities against Bacillus subtilis, Escherichia coli, and Candida albicans. Additionally, Geranium krameri extract inhibited tyrosinase activity ($IC_{50}$, $456.86{\mu}g/mL$) and decreased melanin content ($IC_{50}$, $50.35{\mu}g/mL$). The treatment of B16F10 cells with Geranium krameri extract suppressed the protein expression of tyrosinase in a dose-dependent manner. These findings suggest that Geranium krameri extract inhibits melanin synthesis in murine melanoma B16F10 cells by suppressing intracellular tyrosinase expression, as well as directly inhibits tyrosinase activity simultaneously.

A. esculentus extract had a low cytotoxicity against murine melanoma B16F10 cells. At concentrations showing little or no cytotoxicity, A. esculentus extract showed high a DPPH radical scavenging activity ($ID_{50}$, $5.24{\mu}g/mL$), inhibited tyrosinase activity ($ID_{50}$, $102.12{\mu}g/mL$), and decreased melanin content ($ID_{50}$, $17.85{\mu}g/mL$). The treatment of B16F10 cells with A. esculentus extract suppressed the protein expression of tyrosinase in a dose-dependent manner. These findings suggest that A. esculentus extract inhibits melanin synthesis in murine melanoma B16F10 cells by suppressing intracellular tyrosinase expression, as well as directly inhibits tyrosinase activity simultaneously. Additionally, A. esculentus extract promotes the expression of involucrin, which is related to skin barrier protection. These results indicate that A. esculentus extract is an appropriate material for improving skin barrier function.

The Analysis of Whitening Effects on Extracts from Ginkgo (Ginkgo biloba L.) Seeds (은행나무 종자 추출물의 미백효능 분석)

  • Ginkgo (Ginkgo biloba L.) seeds, called 'Baekqwa', were extracted from 70% ethanol to investigate the whitening effect and to confirm the application potential as a cosmetic material. Ginkgo seed ethanol extracts (GBE) were treated with B16F10 melanoma cells, and melanin contents and tyrosinase, which is the main enzyme concerning the synthesis process of melanin, inhibitory activity were confirmed. As a result, there were inhibited in a concentration-dependent manner, and GBE also significantly reduced the protein expression and mRNA levels of tyrosinase, tyrosinase-related protein-1, -2 (TRP-1, -2), and their upstream transcription factor, microphthalmia-associated transcription factor (MITF). These results suggest that GBE could be used as an effective whitening agent that has an inhibitory effect on melanin production by regulating the expression and degradation of MITF in melanocytes.

Physiological activities of ethanol extract of Allium tuberosum root (부추뿌리 추출물의 생리활성)

  • This study investigated various physiological activities to examine the applicability of the functional materials of Allium tuberosum root extract. The A. tuberosum root extract showed a low cytotoxicity against murine melanoma B16F10 cells. It also showed high DPPH radical scavenging activity (ID50, 6.2 ㎍/mL), inhibited tyrosinase activity (ID50, 115.4 ㎍/mL), and decreased melanin content (ID50, 31.5 ㎍/mL). Treatment of B16F10 cells with A. tuberosum root extract suppressed the protein expression of tyrosinase in a dose-dependent manner. These findings suggest that A. tuberosum root extract inhibits melanin synthesis by suppressing intracellular tyrosinase expression. Additionally, A. tuberosum root extract inhibited elastase with an ID50 value of 145.1 ㎍/mL and contained isoquercitrin. These results indicate that A. tuberosum root extract is an appropriate natural material.
