B16 Melanoma 세포에서 Chitosan Oligosaccharide가 Melanin 생성에 미치는 영향 (Effect of Chitosan Oligosaccharide on Melanin Production in B16 Melanoma Cells)

  • To investigate the effect of chitosan oligosaccharide on melanin synthesis, we measured tyrosinase activity and melanin production in B16 melanoma cells. Chitosan oligosacchaide itself did not have any anti-oxidant activity in DPPH radical scavenging, and did not affect the proliferation of B16 melanoma cells. Chitosan oligosaccharide dose-dependently increased melanin production in the absence or presence of MSH. However, chitosan oligosaccharide did not have any influence on the tyrosinase activity and tyrosinase expression in B16 melanoma cells. These results suggest that chitosan oligosaccharide-induced melanin production may be independent on tyrosinase activity in B16 melanoma cells. From the above results. chitosan oligosaccharide dose-dependently appears to increase melanin production in B16 melanoma cells, suggesting that chitosan oligosaccharide may be used as a tanning agent.

Padina boryana, a brown alga from the Maldives: inhibition of α-MSH-stimulated melanogenesis via the activation of ERK in B16F10 cells

  • Background: The present study investigates the potent skin whitening ability of ethanol extract from the brown alga, Padina boryana (PBE) which was collected in the shores of Fulhadhoo Island, the Maldives, and its specific pathways of action. The effect of PBE which contains a rich amount of polyphenols was evaluated using B16F10 murine melanoma cells and provides insight to the underlying mechanisms with reference to the inhibition of melanin formation. Methods: Melanin synthesis and cellular tyrosinase inhibition were assessed in the α-MSH-stimulated melanocytes. Melanogenic pathway-related protein expressions were investigated via Western blotting. ERK 42/44 was particularly examined considering its involvement in the melanogenic pathway. Further, RT-qPCR techniques were involved in gene expression analysis. Results: PBE dose-dependently inhibited the cellular melanin synthesis and tyrosinase levels. Western blotting revealed the potential of PBE to downregulate microphthalmia-associated transcription factor (MITF), tyrosinase, and tyrosinase-related protein-1 and protein-2 (TRP-1 and TRP-2). Moreover, results explained the phosphorylation of ERK was sustained via PBE and hence declined the ultimate melanin synthesis. Gene expression analysis reinforced the results obtained. Conclusions: The study provides substantial evidence to express the potential of PBE to inhibit B16F10 melanoma cell melanin synthesis. Concisely, results suggest the ability of PBE to be involved in medicinal and cosmeceutical applications.

  • 인체피부의 색소침착에 있어서 다양한 차이는 주로 표피의 멜라닌생성 세포에 의한 멜라닌 합성 비율, 합성된 eumelanin과 pheomelanin의 상대적인 양과 melanocyte에서 keratinocyte로 melanosomes의 이동 속도와 그 방법에서 기인된다. 색소침착은 유전적, 환경적으로 조절되어지는 복합적 특성이다. 많은 관심이 집중되었던 하나의 유전자인 melanocortin 1 receptor 유전자가 있다. 인간집단에서 이 유전자의 다앙한 polymorphism은 색소침착의 다양성에 있어서 중요하다. 자외선(UV)에 대한 노출은 다양한 성장 요인, cytokines과 호르몬의 합성이 증가되고, 표피에서 그들 수용체들의 환경적응등이 나타난다. 색소침착 조절에 관한 정보는 과다색소 침착된 피부손상의 임상치료를 위한 전략들에서 이끌어 냈다. 주된 3개의 전략은 다음과 같다. 1) melanin 합성경로를 방해하는 화합물의 사용 2) eumelanin 합성을 조절하는 호르몬의 구조에서 기인된 peptides 또는 peptide-mimetics개발 3) melanocytes에서 keratinocytes로의 melanosome 이동을 감소시키는 물질의 개발. 이 모든 3가지의 전략은 각각 전체 멜라닌 합성, eumelanin 생성 또는 피부의 멜라닌 단위를 억제시킴으로 미백작용을 유도시킬 것으로 기대되어 진다.

천궁 메탄올 추출액의 멜라닌 형성 억제 효과 (Inhibitory Effect of Methanolic Extract of Cnidii Rhizoma on the Melanogenesis)

  • Down-regulation of melanin synthesis is required for recovery of pigmentary disorders and it is known that direct inhibitors of tyrosinase suppress melanin synthesis. We screened several oriental medicinal plants using B16/Fl0 cells and found that the methanolic extract of Cnidii Rhizoma down-regulated melanin synthesis effectively. Although the proliferation of Bl6/Fl0 cells was decresed by the methanolic extract of Cnidii Rhizoma, it did not appear necrosis. Bl6/F10 cells incubated with the methanolic extract of Cnidii Rhizoma showed reduced pigmentation and tyrosinase activity. Western blotting revealed that the amount of tyrosinase was decreased by the methanolic extract of Cnidii Rhizoma. These results suggest that the inhibitary effect of the methanolic extract of Cnidii Rhizoma on melanogenesis is due to the suppression of tyrosinase in Bl6/F10 cells and Cnidii Rhizoma is a candidate for an efficient whitening agent.

측백엽(側柏葉) 메탄올 추출물이 멜라닌 형성에 미치는 영향 (Effect of Methanolic Extract from Biota Orientalis Folium on Melanin Synthesis)

  • Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin bio-synthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) stimulates melanogenesis and enhances the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Biota Orientalis Folium on the basal melanogenic activities of B16 mouse melanoma cells, and on the ${\alpha}$-MSH or tyrosinase-induced melanogenesis. Biota Orientalis Folium alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. The decrease of cell propagation was observed in B16 cells treated with 200${\mu}$g/ml dose of Biota Orientalis Folium, indicating that Biota Orientalis Folium-induced depigmenting effect was caused by inhibition of melanin synthesis, not due to destruction of B16 cells. Pretreatment of the cells with Biota Orientalis Folium also suppressed the increase of ${\alpha}$-MSH (10 nM) induced melanin content and tyrosinase activity. Biota Orientalis Folium inhibited the revelation of ${\alpha}$-MSH induced tyrosinase protein and tyrosinase related protein and mRNA of tyrosinase in B16 melanoma cell. These results suggest that Biota Orientalis Folium inhibits melanogenesis and abrogates ${\alpha}$-MSH and tyrosinase-induced melanogenesis in B16 melanoma cells.

감마선 조사가 베타글루칸의 미백활성에 미치는 영향 (Effect of Gamma-Irradiation on the Whitening Activity of β-Glucan)

  • This study evaluated the change in whitening activity of ${\beta}-glucan$ by gamma-irradiation. Tyrosinase inhibition was significantly increased in the samples with 30, 50, 100 kGy irradiated ${\beta}-glucan$. Melanin synthesis of irradiated ${\beta}-glucan$ was measured from B16BL6 melanoma cell line treated with ${\alpha}-melanin$ stimulating hormone. Melanin synthesis was increased in the ${\alpha}-melanin$ stimulating hormone added group. However, it was decreased in the groups of 30, 50 and 100 kGy gamma-irradiated ${\beta}-glucan$ treated with ${\alpha}-melanin$ stimulating hormone. These results indicate that gamma irradiated ${\beta}-glucan$ may elevate the whitening activity. Therefore, gamma-irradiated ${\beta}-glucan$ could be used for nutraceutical foods in cosmetic industry.

인진쑥 물추출물의 조다당제에 의한 Melanin 생성억제 효과 (Inhibitory Effects of Crude Polysaccharide of Water Extract of Art on Melanin Biosynthesis)

  • Melanogenesis is a physiological process resulting in the synthesis of melanin pigment. We investigated the inhibitory effect of crude polysaccharide of water extract of Artemisia iwayomgi Kitamura (AI) on melanin biosystheies in B16/F10 melanoma cells. Crude polysaccharide of water extract of Artemisia iwayomgi Kitamura significantly inhibited tyrosinase activity and melanin contents with or without $\alpha$-MSH in vivo. Melanin contents and tyrosinase activity were decreased in a dose-dependent manner,These results show that crude polysaccharide of water extract of Artemisia iwayomgi Kitamura could be developed as skin whitening components of cosmetics.

만형자 용매 분획물의 미백 개선 효과 (Whitening Effects of Solvent Fractions Isolated from Vitex rotundifolia)

  • 최근에는 hyper-pigmentation에 대한 제품 개발을 위해 melanogenesis의 경로에 대한 이해를 위해 많은 노력을 하고 있다. 이에 따라 본 연구에서는 만형자(Vitex rotundifolia)의 depigmentation에 대한 효과를 조사하였다. B16F10 mouse melanoma cell을 이용하여 tyrosinase, MITF, TRP-1, TRP-2 그리고 melanin synthesis의 저해를 확인하였고, 만형자 ethyl acetate 분획물(VR-EA)을 처리하였을 때 농도의존적으로 감소한다는 것을 알 수 있었다. 특히, VR-EA는 tyrosinase와 TRP-1의 경우 각각 53.2, 88.4%의 저해율을 보여 미백에 있어서 효과적인 활성을 보여주었다. 따라서 만형자는 melanin synthesis를 효과적으로 막아주어 미백소재로서 활용 가능성이 충분하다고 사료된다.

  • In this study, we demonstrated the whitening effect of Lagenaria leucantha through the melanin biosynthesis of S bikiniensis and inhibition of melanogenesis in cultured Bl6 melanocytes. And we confirmed the whitening effect of Lagenaria leucantha through the depigmentation of gold fish in vivo. The melanogenesis of B$_{16}$ melanocytes was founded to be activated dose and time dependently by the treatment of u- MSH. When the B$_{16}$ melanocytes was treated with 200nM of $\alpha$-MSH, the morphology of melanocytes was remarkably changed. The melanin content and the synthesis of tyrosinase were strikingly increased. Lagenaria ieucantha inhibited the melanin formation stimulated by $\alpha$-MSH without affection of cell viability. However, Lagenaria leucantha didn't inhibit tyrosinase activity and showed weak suppression on the synthesis of tyrosinase. These results suggest Lagenaria leucantha might inhibit melanin formation with tyrosinase independent manner. Lagenaria ieucantha also inhibition melanin biosynthesis with 18mm inhibition zone in S.bikiniensis. To evaluate the inhibitory activity of melanogenesis of Lagenaria leucantha in vivo, we examined its effect on depigmentation of gold fish. Lagenaria ieucantha remarkably reduced the size and density of melanophores in gold fish. These results suggest that Lagenaria ieucantha can be used as a whitening agent in cosmetics.ics.s.

N-Linked Glycosylation 저해에 의한 닭의장풀 추출물의 미백효능 (Whitening Effect of Dayflower (Commelina communis L.) Extract by Inhibition of N-Linked Glycosylation Process and Melanogenesis)

  • 닭의장풀(Commelina communis L. Dayflower) 추출물의 미백효능을 확인하기 위하여 B16 melanoma 세포를 이용하여 melanin 생성에 관련된 다양한 실험을 실시하였다. 닭의장풀 추출물은 버섯 tyrosinase 활성 실험에서 저해 효과를 보이지 않았으나, melanin 생성 저해효과를 나타내었다. B16-F10 멜라노마 세포를 이용한 활성시험 결과에서, 닭의장풀 추출물은 1,000 ${\mu}g/mL$의 농도에서 약 32%의 멜라닌 생성을 억제하였으며, 세포내 티로시나제 활성 억제 능도 우수하여 닭의장풀 추출물 250 ${\mu}/mL$ 이상의 농도에서 50% 이상의 저해 효과를 보였다. 추출물의 생성 기작에 대한 영향을 조사한 결과, melanin 합성의 key protein 인 tyrosinase 발현의 우수한 저해 능력을 보였고, tyrosinase related protein-1 (TRP-1)의 발현 억제와 tyrosinase related protein-2 (TRP-2)의 N-glycosylatin 억제를 통해 melanin 합성이 억제되는 것으로 분석되었다. 따라서 닭의장풀 추출물은 melanin 합성에 필수적인 효소(tyrosinase, TRP-1)의 발현 저해 및 TRP-2의 N-glycosylation 억제를 통해 미백 효과를 나타내는 것으로 확인되었으며, 이에 따라 본 추출물은 melanin 합성의 효소 경로를 저해하는 미백 소재로 활용할 수 있을 것으로 사료된다.
