Bush Growth and Fruit Quality of 'Duke' Blueberry Influenced by Nutritional Composition in Unheated Plastic House (블루베리 '듀크' 품종의 무가온 하우스 재배에서 질소비율 조절에 따른 수체생육 및 과실품질 변화)

  • The aim of the present study was to determine the influence of different fertilizer combinations on the growth, yield, and fruit quality of 'Duke' blueberry cultivar and the water quality of growth medium. The experiment was carried out with three year old 'Duke' blueberry bushes which were cultivated in containers ($60{\times}80{\times}40cm$) filled with 130 L peat moss and 40 L pearlite (v/v). Sawdust was used as the mulch in growth containers. Three different fertilizer combinations (FC) i.e., FC-1 consisted with standard solution, FC-2 consisted with nitrogen reduced by 10% from FC-1, and FC-3 consisted with nitrogen reduced by 20% from FC-1 were tested while, the ground water used as the control. The effects of different fertilizer combinations on shoot diameter, shoot length, number of shoots, leaf length, SPAD value (the relative content of chlorophyll), berry weight, soluble solids content, titratable acidity, and yield per bush in 'Duke' blueberry were examined. Also, the effects of different fertilizer combinations on pH, EC, $NH_4$ and $NO_3$ in 'Duke' blueberry growth medium were monitored. The highest pH and lowest EC, $NH_4$ and $NO_3$ in growth medium was recorded with control treatment during the experiment period. The maximum shoot diameter (3.7 mm) and shoot length (35.7 cm) was recorded for the FC-1. Highest number of shoots (47%) were recorded from 'Duke' blueberry bushes supplemented with FC-1 compared to other treatments. The fertilizer combinations supplemented with nitrogen showed significant influence on leaf length and SPAD value compared to control 'Duke' blueberry bushes. However, the fruit quality attributes, i.e., berry weight, soluble solids content, and titratable acidity were not significant different among fertilizer treatments. The significantly highest yields per bush were recorded for FC-1, FC-2, and FC-3, as 2.2, 2.9, and 2.7 kg, respectively compared to control (0.2 kg). Although, the FC-1 was supplemented with highest nitrogen content it resulted low yield per bush while having high number of shoots and vigorous growth.

Oral Health and Related Factors for the Elderly (Structural Equation Modeling을 통한 노인(老人)의 구강건강(口腔健康) 관련요인(關聯要因) 분석(分析))

  • This study aims to analyze realities of oral health and related factors, and establish Structural Equation Modeling. The subjects of study were 9,340 elderly over age 65 who took the health examination(the first) for the local insured which National Health Insurance Corporation carried out in the survey area mentioned below from January 2002 to December 2002. The areas surveyed were 4 big cities including Seongbuk-ku, Seoul, 5 medium cities including Wonjusi, Gangwon-do, and 5 Counties including Yeong deok County, Kyeongbuk. Considering location and the scale of population, firstly, big unit areas(metropolitan city, province) were selected according to convenience, secondly, low unit areas(city, county, district) were selected randomly. The subjects were the elderly who took all tests including an oral examination and filled in the questionnaire. Major results from analysis are as follows: 1. Review of Composition Conception Validity As a result of analyzing composition conception validity of SEM including posture test, urine test, blood test, habits of eating, drinking and smoking, oral symptoms, and oral health status, using fit index such as GFI, CFI, TLI, and RMSEA, all were within fit range and composition conception validity was recognized. 2. As a result of analyzing SEM to find the relationship between each factor and oral health status, it was confirmed that all factors except urine test affected oral health status and the synthetic SEM to explain it could be established. In result, we could verify that the elderly of rural areas who had lesser experience of visiting a clinic and oral prophylaxis had a higher rate of caries, missing teeth, and denture need, and drinking and smoking negatively affected the rate of caries, periodontal, and missing teeth. Also, periodontal diseases were observed from 43.2% of the total elderly and much from the lower age. Most of oral disease can be prevented by right oral health behavior. Therefore through oral health professionals from each district public health center of the nation, oral health education for the elderly about right eating habits and oral health care should be carried out systematically and policy change to increase access to dental service is required lest that visiting a dental clinic should be impossible or oral health behaviors such as oral prophylaxis and denture wearing should be neglected by economic, geographical barriers. Also, to establish SEM to explain the relationship between oral health status and systemic health, more accurate test methods and effective index development should be preceded. Because items developed by National Health Insurance Corporation applied to this study without alteration, structuring a model had the uppermost limit. Continual study seems to be needed.

The Manufacturing Techniques of the Stone Standing Maitreya Bodhisattva Bronze Wind Chimes of Gwanchoksa Temple, Nonsan (자연과학적 분석을 통한 논산 관촉사 석조미륵보살입상(論山 灌燭寺 石造彌勒菩薩立像) 청동풍탁(靑銅風鐸)의 제작 기법 연구)

  • The wind chime is a longstanding Jangeomgu (majestic article) found in Korea, China, and Japan. However, basic research on wind chimes is currently inadequate as it is difficult to estimate the time of production, and there are few relics. Therefore, this research morphologically classifies the eight bronze wind chimes decorating the baldachin of the Stone Standing Maitreya Bodhisattva of Gwanchoksa Temple, Nonsan. Based on this, the manufacturing techniques and production period are scientifically demonstrated. The synthesis of the research results reveals that the structure and characteristics of the wind chimes of the Stone Standing Maitreya Bodhisattva of Gwanchoksa Temple, Nonsan differ depending on their location on the baldachin. The four large-sized wind chimes on the lower-baldachin were manufactured by casting a Cu-Sn-Pb alloy, and they are estimated to have been made during the early period of Goryeo. The two medium-sized wind chimes of the upper-baldachin's northern direction were manufactured through forging a Cu-Sn or Cu-Sn-Pb alloy, and they appear to have a similar structure to the cylindrical wind chimes appearing during the latter period of Goryeo and the Joseon period. The two small-sized wind chimes of the upper-baldachin's southern direction were manufactured by casting a Cu-Sn-Pb alloy containing Zn, and based on the chemical composition of the alloy and the shape of the clapper, they are estimated to have been manufactured during the latter period of Joseon. Through the observation of microstructures and a chemical composition analysis, it is demonstrated that two wind chimes of the lowerbaldachin were manufactured by casting and slow cooling the alloy with an alloy ratio of Cu:Sn:Pb≒80:15:5. In addition, it is estimated that the wind chimes of the upper-baldachin's northeast direction were manufactured by forging an alloy of Cu-Sn with a similar alloy ratio to that of forged high tin bronze. The results of a comparative analysis of prior research on domestic wind chimes confirm that two wind chimes of the lower-baldachin have a similar composition ratio to the wind chime excavated from Wolnamsaji in Gangjin, containing an amount of tin that corresponds with ancient records. Having a similar alloy ratio to forged high tin bronze, the wind chimes of the upper-baldachin's northeast direction are the only instances among all of the wind chimes that have been examined to date that were manufactured using this forging method. The purpose of this research is to collect baseline data to verify and classify the manufacturing period of wind chimes according to their morphological characteristics based on scientific evidence. It is hoped that this data can be utilized for the restoration and conservation processes of the wind chimes of the Stone Standing Maitreya Bodhisattva of Gwanchoksa Temple, Nonsan.

Effect of Treatment and Samples of Human Follicular Fluid on Development In Vitro of Mouse Embryos (사람 난포액의 처리 방법과 Sample이 생쥐 수정란의 체외 발달율에 미치는 영향)

  • The present studt was performed to investigate the effect of treatment and samples of human follicular fluid (hFF) on the development in vitro of mouse embryos. The two cell stage embryos collected at 40 h post-hCG injection were cultured in the modified human tubal fluid (m-RTF) containing 15% synthetic serum substitute (SSS) or human tubal fluid (hFF) for up to 3 days at $37^{\circ}C$ in 5% $CO_2$ incubator. Also the composition of hormone, total protein and protein pattern of hFF samples were analyzed. The developmental rate of mouse embryos developed to blastocyst were not significant difference in the m-RTF containing 15% hFF filtered with 0.22 or 0.8 ${\mu}m$ syringe filter, however, the embryos cultured in the m-RTF containing inactivated hFF were significantly (p<0.05) developed at the high rate to blastocyst than those containing fresh hFF and SSS. The in vitro developmental rate to blastocyst and hatched blastocyst in the m-RTF containing 15% hFF sample A (90.5 and 85.4%, respectively) and SSS (79.4 and 75.3, respectively) were significantly (p<0.05) increased, compared with hFF sample B (64.2 and 54.1 %, respectively). The hFF sample A tended to be higher concentration of LH, FSR, total protein and the ratio of progesterone/$E_2$ and lower concentration of $E_2$ and progesterone than the hFF sample B, but there were no differences in the protein pattern between the two hFF samples. The results of these study suggest that the addition of hFF to the culture medium enhances the development in vitro to blastocyst and hatched blastocyst, but the in vitro developmental rate of mouse embryos is different between hFF samples.

Optimal conditions and effects of prebiotics for growth and antimicrobial substances production of Lactobacillus brevis BK11 (Lactobacillus brevis BK11의 증식과 항균물질 생산을 위한 최적 배양조건 및 prebiotics의 영향)

  • Lactobacillus brevis BK11 obtained from Baikkimchi was selected to study the effects of culture medium, initial pH, atmosphere composition, incubation temperature and time, and prebiotics on growth and production of antimicrobial substances. Growth and antimicrobial substances production of L. brevis BK11 were significantly higher in MRS broth than in BHI or M17 broth. The production of cell mass, lactic acid, and bacteriocin by BK11 strain was at maximum in MRS broth adjusted to pH 6.0. Aerobic and microaerobic conditions were favored cell growth and antimicrobial substances production than anaerobic condition. Biomass and lactic acid production and antimicrobial substances activity of BK 11 were significantly better at 30 and $37^{\circ}C$ than at $25^{\circ}C$. Growth of the strain BK11 entered the stationary growth stage at 24 h after inoculation, and decreased after 36 h. Antimicrobial activities of cell-free culture supernatant and bacteriocin solution were highest when cultured in MRS broth with an initial pH 6.0 for 24-30 h at $37^{\circ}C$. In addition, the highest cell number and lactic acid and bacteriocin production were recorded in the presence of 1 and 2% (w/v) fructooligosaccharide (FOS), however, inulin and raffinose did not affect biological and physicochemical characteristics and antimicrobial activities of L. brevis BK11 cultures. According to these results, production of antimicrobial substances by L. brevis KB11 was closely associated with cell density. Under optimal conditions for antimicrobial substances production, L. brevis BK11 effectively inhibited the growth of Helicobacter pylori ATCC 43504.

Effect of Buffer Composition, Sephadex Grade and Column Size on Filtration Based Quality Improvement of Semen from Murrah Buffalo Bull

  • Sixty semen ejaculates collected at weekly interval from four Murrah Buffalo bulls over a period of seven months (Nov.1999 to May 2000) were used in the present study. Three buffer medium (sodium citrate, TES and Tris) were used for soaking of sephadex. Three grades of sephadex (G-15,G-100, and G-200) were used for preparation of columns. Columns of three different height (one, two and three cm) were used for separation of semen. Twenty semen ejaculates were used in each project. In the first experiment each semen ejaculates was divided into four parts. One part was kept as control and other three parts were passed thought one cm column of sephadex G-15 prepared in three different buffers. There was significant (p<0.05) increase in percent progressive sperm motility and percent live spermatozoa and decrease in percent abnormal spermatozoa and percent spermatozoa with damaged acrosome as well as sperm numbers after filtration through all the three columns. Sperm quality obtained in the filtrate of column prepared in Tris buffer was better in comparison to other two buffers. So the Tris buffer was used in the second trial. Twenty semen ejaculates were used in this experiment. Each semen ejaculate was divided into four parts. One part was kept as control (non-filtered) and other three parts were passed through columns of different grade of sephadex (G-15, G-100 and G-200). Progressive sperm motility and live sperm percentage improved significantly while decline in percent abnormal spermatozoa and percent spermatozoa with damaged acrosome and sperm concentration was observed after filtration through all the columns as compared to control (non-filtered) semen. Since post filtration quality of semen was better in the sephadex G-100 column, therefore it was selected for the next experiment. In third experiment, Tris buffer and sephadex G-100 were used for preparing columns of different height (one, two and three cm) and twenty semen ejaculates were filtered. The quality characteristics of semen (percent progressive sperm motility, percent live spermatozoa and sperm concentration) after filtration through one cm column were significantly (p<0.05) higher than after filtration through columns of two and three cm height. However non -significant (p>0.05) difference due to height of columns was observed for percent abnormal and percent damaged acrosome but 1 cm column comparatively gave better result than of 2 and 3 cm column height.

Effect of Pregnancy on Lactation Milk Value in Dairy Buffaloes

  • Buffalo are a major source of milk production, contributing 12.1% in the World and 38.0% in Asia. The buffaloes are kept under peri-urban farming systems to produce milk for urban populations. Breeding is delayed in these herds to get more economic benefit because farmers believe that the pregnancy decreases milk production. The lactation milk value has been studied in this paper as an economic indicator. Complete milk yield records of 3,304 buffaloes was collected from a group of state farms. Economic traits including lactation yield, lactation length, calving interval (CI), dry period and milk yield per day of calving interval (MYPDCI) were derived from the data. The animals were grouped according to parity number (1-3), service period (G1 to G4, conceiving during <150, 150-200, 200-300 and >300 days post calving) and yield levels (HMY>2,500; MMY 2,001-2,500; and LMY 1,500-2,000 liters/ lactation). To study the effect of pregnancy on milk composition a research trial was conducted at a medium size private dairy farm, using forty lactating buffaloes of three yield levels and four service period groups, as described already. Milk was sampled on alternate weeks and analyzed for fat and protein contents (%). For quantifying the value of milk produced during a lactation period, the value corrected milk (VCM) was determined and converted to lactation milk value (LMV). Group means were compared for varicous parameters. Highest milk yield ($2,836.50{\pm}15.68$ liters/lactation) was recorded in the HMY animals of G4 group while lowest milk yield of $1,657.04{\pm}8.34$ liters/lactation was found in LMY of G1. Lactation was significantly increased with the extending of service period. The shortest dry period was recorded in HMY, parity 1, G1 animals and the longest in parity 2, MMY, G4.The CI was shortest in HMY, parity 1, and G1 animals and longest in LMY, parity 3, G4 buffaloes. The HMY, parity 2, G1 buffaloes showed the highest MYPDCI and the lowest value was recorded ($6.53{\pm}0.17$ vs. $2.76{\pm}0.04$ liter/day) for LMY, parity 1, G4 buffaloes. The VCM decreased with the delayed conception. This decreasing trend was higher in respect of the total yield but decrease in the VCM was smaller due to the increasing levels of fat and protein in the milk. The gap between the various production classes was reduced based on the VCM as compared with the yield per day of CI. LMV showed a consistent decline with extending service period in all three production groups. The study suggests that CI increased with delayed conception, showing a consistent trend in the low, moderate and high yielding buffaloes. There was a coherent declining pattern of milk yield with delaying conception, associated with prolonged CI. An animal conceiving at a later stage of lactation showed a decline in financial returns of 24 to 27% compared with those conceiving earlier.

Effects of Fragmentation on the Bird Community in Agricultural Landscapes (농촌 경관에서 파편화가 조류 군집에 미치는 영향)

  • This study was conducted to investigate the effects of fragmentation on the bird community in Seongduck-ri and Hanggum-ri agricultural landscapes of Kangha-myon, Yangpyong-gun, Kyonggido. We analyzed the composition of tree species, forest environmental structures, structures of agricultural landscapes, avifaunas and guild structures. Number and density of tree species ranging from 6 to 10cm in D.B.H. were high in Seongduck-ri, but basal area was high in Hanggum-ri. Coverages of foliage layers under two meters and over 12 meters were high in Hanggum-ri. Meanwhile, Seongduck-ri agricultural landscapes consisted of twelve patches, dominated by Larix leptolepsis and Robinia pseudoacacia Hanggum-ri landscapes was composed of eleven patches, and dominated by Pinus rigida, Castanea crenate, Larix leptolepsis and paddy field. Dominant bird species showed the same order in two study areas, thirty-six species of birds were observed in Hanggum-ri landscapes, but twenty species were in Seongduck-ri landscapes. Number of species and density belonging to hole-, bush-, ground-nesting and water-, bush-, ground-foraging guild were high in Hanggum-ri landscapes. Number of species and density belonging to hole-nesting guild showed the high value in Hanggum-ri landscapes that was not fragmented. It could be attributed to the low human disturbance and relative high density of medium and large trees in D.B.H. Also, number of species and density belonging to bush-, ground- and water- foraging guild showed the high value in Hanggum-ri landscapes that had the high foliage amounts in the foliage layer under two meters. It will be related with the paddy field, dry field and brook, which patches covered the 28.4% of Hanggum-ri landscapes. Therefore, road not only dominate the heterogeneous patch in agricultural landscapes, but also easily cause the human disturbance. Thus, road construction would decrease the natural patch diversity and edge effects that are important for sustaining the diverse avifauna in agricultural landscapes.

Present Status of Rooftop Gardening in Sylhet City Corporation of Bangladesh: an Assessment Based on Ecological and Economic Perspectives

  • Present study analyzes the rooftop gardening status, floristic composition and cost and return of the rooftop garden in Sylhet City Corporation of northeastern Bangladesh. Data was collected from 450 rooftop gardeners randomly during July-September 2010. Study reveal that rooftop gardening is generally for mental satisfaction (95.3%) followed by leisure time activity (87.8%) in the study area and almost all the family members of gardeners' were involved; while collection of planting materials, sites preparation and marketing of products were reported to be carried out by males only (male 71.33%). Middle income classes were most interested in rooftop gardening (43.78%). The survey recorded 53 plant species (35 families) of which Cucurbitaceae family represented highest eight species. Shrubs (28%) were highest followed by herbs (26%) among agri-crops (36%) and flower species (30%). About 89% of the rooftop gardeners procured planting materials from nursery, market, fair, neighbor, relative and friends and they mostly prefer to use seedlings (48%) for roof gardening followed by direct seed sowing (21%). Gardeners sell products sporadically in different local markets, directly or through intermediaries, with no uniform pricing for system. Rooftop gardening improves the food security and meet nutritional deficiency to the gardeners. Survey revealed that generally very few people consider rooftop gardening commercially to get profit and from the cost-return analysis this gardening system can be economically viable if proper and scientifically managed. The study conclude that active government and NGOs could play vital role to increasing this activities by providing training and motivate people with technical aspects of rooftop gardening.

Effects of Gonadotropins and Steroid Hormones on Follicular Oocyte Maturation in Vitro and Fertilizing Ability In Vivo of Korean Native Cattle (성선자극호르몬과 스테로이드호르몬의 첨가가 한우난포란의 체외성숙과 수정능력에 미치는 영향)

  • This experiment was carried out to investigate the effects of hormone addition(FSH, HCG, estrogen and progesterone) and composition (BSA and FCS) of mKRB on the in vitro maturation and fertilizability of follicular oocytes of the Korean native cattle. The ovaries were removed at a slaughterhouse, returned to laboratory in a thermostat (30-35$^{\circ}C$) within 4 hr, and collected by aspirating normal follicles which had diameters of 1 to 6 mm. The oocytes with cumulus cells were cultured for 8, 16, 24 and 30 hr in a modified KRB solution containing BSA or FCS and hormones. The in vitro matured oocytes in mKRB containing FCS, FSH and steroids were transferred in the rabbit uterus for examination of their in vivo fertilizability with bovine sperm preincubated 4 to 6 hr in the rabbit uterus. 1. The mean number of oocytes collected per cattle was 6.5 from 1-3mm follicles, 1.3 from 4-6mm follicles, and total was 7.7. 2. The meiotic division at 16hr-cuture in the oocytes from 1-3mm follicles was slightly stimulated by the addition of FSH in mKRB + BSA solution compared with the control. At 30hr-culture, their maturation rates(%Met II) were also increased by FSH of 1 $\mu\textrm{g}$/ml(38.4%) and 5$\mu\textrm{g}$/ml(35.7%) as compared with the control (21.4%). The maturation rate at 30hr-culture in the oocytes from 4-6mm follicles was 53.8% and 57.1% by the FSH addition of 1$\mu\textrm{g}$/ml and 5$\mu\textrm{g}$/ml, respectively. These rates were similar with the control(57.1%), but higher than those of oocytes from 1-3mm follicles. 3. The meiotic division at 16hr-culture in the oocytes from 1-3mm follicles was stimulated by the HCG addition of 1IU/ml and 5IU/ml. However, the maturation rate at 30hr-culture was greatly decreased by the HCG addtion (26.6% and 13.3%) compared with the control(53.3%) and these rates (30.8%) in the oocytes from 4-6mm follicles were also lower than that fo the control(58.3%). 4. Low maturation rate (37.5%) of the oocytes cultured in mKRB containing BSA and 5IU/ml HCG was increased (55.0%) when 15% FCS with HCG was added to mKRB instead of BSA. 5. When 16hr-cultured oocytes in mKRB containing BSA and gonadotropins (5$\mu\textrm{g}$/ml FSH and 5IU/ml HCG) were transferred in the medium without gonadotropins and recultured for 16hr, the maturation rate of HCG-treated oocytes was greatly improved. 6. The maturation rates of oocytes were greatly affected by steroids. The combined addition of FCS+FSH+estrogen or +progesterone to mKRB increased the maturation rate compared with the combination of BSA+FSH or FCS+FSH in mKRB. 7. The fertilization rate, presence of pronuclei, was increased by the combination of FCS+FSH+p in mKRB as compared with that (5.6%) of BSA+FSH and the rates of FCS+FSH+steroids ranged from 12.5 to 17.6%.
