Water Potential and Photosynthesis of Korean Ginseng (Panax ginseng C. A. Meyer) Leaves (인삼(人蔘)잎의 수분(水分) Potential과 광합성(光合成))

  • This study was carried out to get information on the cause of low photosynthesis ofKorean ginseng, a shade plant. Photosynthesis, water content, stomatal conductance and water potential in leaves of ginseng and soybean were investigated. The light intensity for maximum photosynthesis was about $300{\mu}E\;/\;m^2\;/\;s^2$ in ginseng and about $800{\mu}E\;/\;m^2\;/\;s^2$ in soybean, respectively Photosynthesis was remarkably lower in ginseng than in soybean under the same light intensity and $temperature(at\;20^{\circ}C)$. Photosynthesis of detached leaves was stopped in shorter time in ginseng than in soybean particularly at high $temperature(30^{\circ}C)$. The decreasing rate of water content in detached leaves was slower in ginseng than that of soybean, while the remained water level in the leaves was much higher in $ginseng(70{\sim}71%)\;than\;in\;soybean(50{\sim}53%)$ when photosynthesis was stopped. Water content had a positive correlationwith photosynthesis in both plants. However, at the same water level, the ratio of photosynthesis to water content was remarkably lower in ginseng than In soybean. The relationship between the stomatal conductance and photosynthesis was significantly positive correlation in the both plants. The ratio of photosynthesis to stomatal conductance was similar in booth plants below about 40 $mmol\;/\;m^2\;/\;s\;at\;20^{\circ}C$. Water potential was remarkably lower in ginseng than in soybean, and waterpotential had a significantly positive correlation with water content, stomatal conductance and photosynthesis in both plant. These results suggested that the low stomatal conductance and low water potential might cause the low photosynthesis in ginseng compared to soybean.

Optimal HPLC Condition for Simultaneous Determination of Anthocyanins in Black Soybean Seed Coats (검정콩 함유 안토시아닌의 동시분석을 위한 최적 HPLC 분석 조건)

  • Black soybean has been widely utilized as foods and oriental medicinal materials. The pigmentation in the seed coat of black soybean is due to accumulate anthocyanins in the epidermis palisade layer. The anthocyanins of black soybean seed coat are considered as a parameter of quality evaluation of black soybean. Therefore, the purpose of this study was to investigate the most suitable HPLC condition for simultaneous determination of anthocyanins in black soybean seed coats extracts. The efficient HPLC analytical condition of D3G, C3G, and Pt3G contained extracts of black soybean seed coats was developed. The gradient elution employed a $250\;mm\;{\times}\;4.6\;mm$ i.d. YMC-pak ODS-AM 303 column. The gradient system was used two mobile phases. A gradient elution was performed with mobile phase A, consisting of 5% aqueous formic acid, and mobile phase B, comprising 5% formic acid - acetonitrile, and delivered at a flow rate of 0.7 mL/min as follows: $0{\sim}35\;min$, $90%\;A{\sim}60%\;A$; 36 min, 90% A; 46 min, 90% A. The UV-VIS. detection wavelength was set at 520 nm. The limit of detection (LOD) for D3G, C3G, and Pt3G were under 10 ng/mL.

Biochemical Characterization of a Psychrophilic Phytase from an Artificially Cultivable Morel Morchella importuna

  • Psychrophilic phytases suitable for aquaculture are rare. In this study, a phytase of the histidine acid phosphatase (HAP) family was identified in Morchella importuna, a psychrophilic mushroom. The phytase showed 38% identity with Aspergillus niger PhyB, which was the closest hit. The M. importuna phytase was overexpressed in Pichia pastoris, purified, and characterized. The phytase had an optimum temperature at $25^{\circ}C$, which is the lowest among all the known phytases to our best knowledge. The optimum pH (6.5) is higher than most of the known HAP phytases, which is fit for the weak acidic condition in fish gut. At the optimum pH and temperature, MiPhyA showed the maximum activity level ($2,384.6{\pm}90.4{\mu}mol{\cdot}min^{-1}{\cdot}mg^{-1}$, suggesting that the enzyme possesses a higher activity level over many known phytases at low temperatures. The phytate-degrading efficacy was tested on three common feed materials (soybean meal/rapeseed meal/corn meal) and was compared with the well-known phytases of Escherichia coli and A. niger. When using the same amount of activity units, MiPhyA could yield at least $3{\times}$ more inorganic phosphate than the two reference phytases. When using the same weight of protein, MiPhyA could yield at least $5{\times}$ more inorganic phosphate than the other two. Since it could degrade phytate in feed materials efficiently under low temperature and weak acidic conditions, which are common for aquacultural application, MiPhyA might be a promising candidate as a feed additive enzyme.

Heavy Metal Accumulation in Edible Part of Eleven Crops Cultivated in Metal Contaminated Soils and Their Bio-concentration Factor (중금속 오염 토양에서 재배한 주요 작물별 가식부 중금속 축적 농도 및 생물농축계수)

  • BACKGROUND: The current study was conducted to examine the species specific accumulation of Cd and Pb in 11 crop species (Soybean, Sesame, Corn, Polished rice, Carrot, Potato, Garlic, Spring onion, Chinese leek, Red pepper, Eggplant), through cultivating them under the same condition with metal contaminated soils.METHODS AND RESULTS: Eleven crop species were cultivated in three different soils contaminated with Cd and Pb and harvested. Edible parts of each crop was pretreated and analyzed to determine Cd and Pb concentrations, and subsequently bioconcentration factors (BCFs) were calculated. In general, the crops of which seeds are used as food showed high concentrations of both Cd and Pb. For instance, Cd concentrations in crops cultivated in Soil A was in the order of soybean (0.432 mg kg-1) > sesame (0.385) > polished rice (0.176) > carrot (0.116) > corn (0.060) > red pepper > (0.047) > potato (0.044) > egg plant (0.025) > garlic (0.023) > spring onion (0.016) > Chinese leek (0.011). BCFs showed the same order.CONCLUSION: From this study, it can be conclude that seeds plants should not be cultivated in Cd and Pb contaminated soils to secure food safety from metal contaminated soils.

Evaluation of Toxicity of Anthocyanin from Black Soybean by Feeding Test in Mice (검정콩 안토시아닌의 마우스를 이용한 안전성 연구)

  • Over the past few years, anthocyanin has been demonstrated to exert potentially anti-oxidative and bioavailability effects including anti-cardiovascular disease and cancer-preventive effects in humans. This study was conducted to assess the toxicity of anthocyanin extracts from black soybean seed coats via oral administration in a mice feeding test. The final anthocyanin-containing products induced signs of toxicity on mortality in the 4-week feeding test. The incidence of clinical signs and changes in body and organ weight were also not observed in all anthocyanin-treated groups as compared with the control groups. In hematology analysis, RBC indices concluding MCV, MCH, and MCHC and WBC differential counting such a NEU, LYM, MONO, EOS, and BASO evidenced no significant differences between the anthocyanin treatment and control groups. The biochemical parameters in serum analysis - ALT, AST, BUN, Crea etc.- were also not significantly altered in those groups. Absolute and relative organ weights were not increased after 4 weeks of treatment with anthocyanin extracts in mice. In conclusion, anthocyanin extracts from the black soybean has biological activity without any toxic effects, which also suggests that the consumption of soy containing anthocyanin products may be a good source for health and medical supplies.
