Medicinal food로 활용하기 위한 산사(山査)에 관한 연구(1) -활용 형태에 따른 영양학적 특성 및 식품으로의 활용방안 모색- (Study of Crataegi Fructus for Medicinal Foods Applications -Nutrition Composition and Scheme for Foods-)

  • Crataegi Fructus has often been cited in medical literature for its medicinal effects. The purpose of this study was to investigate the possibility of Crataegi Fructus application as an edible medicinal (nutritional supplement) food resource. In this study, Crataegi Fructus, which has been used in oriental medicine and folks remedy, was investigated to characteristics of nutritional composition(protein, lipid, ash, fiber, free sugar and minerals). The approximate composition of low Crataegi Fructus was crude protein, 0.26%, crude lipid, 0.30% and crude ash, 0.66%. And total dietary fiber 5.60%(insoluble 4.66%, soluble 0.94%), contents of glucose and fructose were 5.02mg% and 6.21 mg%, Nutritional composition of fermented liquid was crude protein 0.24%, crude lipid, 0.03%, crude ash, 0.53% and total dietary fiber, 0.24%. And glucose, fructose contents were 14.77mg% and 7.30mg%. The other hand, nutrition contents in water extract of Crataegi Fructus were significantly lower than low Crataegi fructus and fermented liquid. The above results showed that Crataegi Fructus and fermented liquid have sufficient values to use as a food stuff for medicinal food and nutritional supplement.

지황(地黃)과 발효(醱酵) 지황(地黃)의 생리활성 비교 연구 (Comparison of Biological Activities on Rehmanniae Radix and Fermented Rehmanniae Radix)

  • Herbal medicines are medicinal products containing a single or a mixture of two or more different herbal substances or herbal preparations as active principles. Recently, much attention has been paid to developing various kinds of fermented herbal extracts, a new type of traditional herbal medicine in the field of Korean traditional medicine. The fermentation of medicinal herbs is intended to exert a favorable influence on bioestability, bioavaliablilty and pharmacological activity of herbal extract in the gastrointestinal tract as well as intensifying the nutritional and pharmacological aspects of the medicinal herbs. The purpose of this study was to investigate biological activities of fermented Rehmanniae Radix by lactic acid bacteria at $30^{\circ}C$ for 3 days in comparison with those for Rehmanniae Radix The fermented Rehmanniae Radix exhibited different chemical profile to Rehmanniae Radix generated with HPLC, indicating production of new ingredients during fermentation. Rehmanniae Radix served as good nutritional sources for the growth of lactic acid bacteria showing increased number of bacteria during fermentation. Toxic effect of the fermented Rehmanniae Radix to cells were not seen judged by the MTT assay. The fermented Rehmanniae Radix exhibited better antioxidant effect than non-fermented Rehmanniae Radix analyzed by a SOD-likely assay. Both hypoglycemic and hypotensive effects of the fermented Rehmanniae Radix were also detected and better than those for Rehmanniae Radix in showing dose-dependent inhibitory effects on alpha-glucosidase and ACE, respectively. In conclusion, fermented Rehmanniae Radix appears to have more biological activities than non-feremented Rehmanniae Radix showing not only antioxidant effect but also cardiovascular protection.

건조방법과 한약재 추출물 첨가가 육포의 미세구조에 미치는 영향 (Effects of Drying Method and Medicinal Herb Extract Addition on the Microstructure of Beef Jerky)

  • 본 연구의 목적은 육포의 제조방법이 품질특성에 미치는 영향을 규명하기 위하여 기본 양념(간장, 물엿, 참기름, 정종, 마늘, 양파, 배즙)에 설탕(A), 감초(B), 3종류의 향신료(정향;C, 회향;D, 청양고추;E) 추출물과 여러 추출물을 혼합 첨가(F)한 6군으로 나누어 쇠고기를 절인후, 열풍건조와 송풍건조하는 육포의 제조과정에서 근육의 미세구조 변화를 주사전자현미경(SEM)과 투과전자현미경(TEM)으로 관찰하였다. 투과전자현미경(TEM)으로 관찰한 결과, 쇠고기는 액틴과 미오신의 근육라인과 근원섬유가 평행으로 발달되어 있었고 세포의 미토콘드리아와 내막을 볼 수 있었으나 절임과정에서 근섬유가 절단되고 근육라인의 분해 및 세포 구조물들의 퇴행이 일어났다. 주사전자현미경(SEM)으로 관찰한 결과, 자연건조한 한약재 추출물 첨가 육포는 근원 섬유와 공간구조가 남아있고 육질이 보존되어 있었으나 열풍건조한 육포는 근원섬유의 배열과 공간구조가 전혀 관찰되지 않았다. 이상의 주사전자현미경과 투과전자현미경으로 육포의 미세구조를 관찰한 결과에서 한약재 추출물은 근섬유의 결을 보존하는데 도움이 되며 송풍건조는 열풍 건조에 비하여 육포의 물성을 좋게 함으로써 품질을 향상시킬 수 있는 건조방법으로 생각된다.

당뇨병 예방을 위하여 한약재 추출액으로 제조한 현미밥의 품질특성 (Quality characteristics of brown rice boiled with medicinal herbs extract for diabetes prevention)

  • 한국인의 식생활에서 매일 섭취 할 수 있는 방안을 제시하기 위해 당뇨병에 효능이 있는 한약재를 이용하여 열수추출액을 제조하고 현미밥을 제조하여 그 품질특성을 연구한 결과는 아래와 같다. 수분함량은 현미의 첨가량이 증가할수록 수분함량은 낮아졌으며, $H_2$에서도 같은 수분함량을 보였다. 물성특성은 $H_1$$H_2$ 모두가 유사한 경향을 보였는데 단단한 정도(hardness)는 대조구인 백미 100%의 경우가 가장 낮았고, 현미 첨가량이 높아질수록 증가하였다. 탄력성(springiness), 응집성(cohesiveness), 점착성(gumminess)은 현미 첨가량이 증가할수록 감소하는 경향을 보였으며, 씹힘성(chewiness)은 현미 첨가량이 증가할수록 증가하는 경향을 보였다. 색도는 $H_1$$H_2$ 모두가 유사한 경향을 보였으며 L값은 현미 첨가량이 증가할수록 감소하였으며 a값은 현미 첨가량이 증가할수록 감소하는 경향을 보였고 b값은 반대로 현미 첨가량이 증가할수록 증가하는 경향을 보였다. $H_2$를 이용하여 제조한 현미밥의 관능평가 결과는 외관과 맛, texture, 전반적 기호도는 현미 첨가량이 증가할수록 감소하는 경향을 보였다. 전체적인 기호도는 $H_1$과 유사한 경향을 보여 현미 30% 첨가구에서 가장 높게 나타났다.

한약재 주정 추출물이 종자발아와 유묘생장에 미치는 영향 (Effect of Ethanol Extract of Herbal Medicine on Crop Growth Inhibitor)

  • BACKGROUND: Herbal extracts have been screened for their inhibitory effect of seed germination and root development on weeds, but there is a scarcity of reports for crop growth regulation. The objective of this research was to develop a growth inhibitor on Brassica campestris, and its effective extraction method from herbal medicine extract. METHODS AND RESULTS: Eighty four herbal medicine extracts were tested for their plant growth inhibition activity on B. campestris. The alcohol extracts of Artemisia annua, Cinnamomum cassia, and Mentha arvensis inhibited over 30% of germination and the extract of A. annua, and C. cassia inhibited over 70% of radicle growth at 0.1 % w/w treatment. The partially purified extracts of A. annua, and C. cassia with dichloromethane and hexane showed stronger radicle growth inhibition than the crude extracts on B. campestris. The diethyl ether extract of A. annua showed a similar 50% radicle growth inhibition ($RI_{50}$ = 45 mg/L) to its partially purified extract with dichloromethane or hexane, but the diethyl ether extract of C. cassia showed a worse $RI_{50}$ than the purified extract. CONCLUSION: The alcohol extracts of A. annua, and C. cassia showed potent radicle growth inhibition properties on B. campestris. Diethyl ether proved to be a good solvent for simple extraction from A. annua.

잇꽃(Carthamus tinctorius)으로부터 Flavonol glycoside 화합물들의 분리 및 항당뇨 효과 (Flavonol glycosides from the flowers of Carthamus tinctorius and their anti-diabetic activity)

  • 잇꽃을 80% MeOH 수용액으로 추출하고, 얻어진 추출물을 EtOAc, n-BuOH 및 물로 용매 분획 하였다. 이 중 n-BuOH 분획으로부터 silica gel (SiO2), octadecyl silica gel (ODS) column chromatography, 및 Prep-LC로 정제하여 4종의 화합물을 분리하였다. Nuclear magnetic resornance, mass spectroscopy 및 infrarad spectroscopy 등의 스펙트럼 데이터를 통해 화합물의 화학구조를 astragalin (1), isoquercetin (2), nicotiflorin (3), 그리고 rutin (4) 로 동정하였다. 본 연구를 통해 잇꽃 추출물, 용매 분획물 및 모든 분리 화합물의 alloxan에 의해 손상된 zebrafish 유충 췌도 보호 효과를 평가하기 위한 실험을 수행하였다. EtOAc (CTE), n-BuOH (CTB) 및 H2O (CTW) 분획 모두 통계 유의적으로 우수한 항당뇨 효능을 확인하였으며, 분리된 화합물 1-4로 처리된 췌도 크기는 알록산 유도군 대비 각각 87.0, 88.5, 88.7, 및 89.3% 로 유의적으로 증가했다. 분리된 4종의 화합물 중 nicotiflorin은 6.752 mg/추출물(g)으로 높은 함량을 나타내었다. 이 결과를 통해 잇꽃 유래 화합물은 항당뇨 소재의 기능 지표성분으로서 활용 가능성을 확인하였다.

Herbal Extract Prevents Bone Loss in Ovariectomized Rats

  • This research aims to test a new drug candidate based on a traditional medicinal herb, F1, an herbal extract obtained from Astragalus membranaceus and its main ingredient, 1-monolinolein that may have fewer side effects and less uterine hypertrophy. In vitro experiments, human osteoblast-like cell lines, MG-63 and Saos-2, were analyzed by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) and an alkaline phosphatase (ALP) assays. Mouse osteoclasts were induced through a calcium-deficient diet and inhibition effects were measured. In vivo experiments were done using ovariectomized (OVX) rats for 9 weeks. At necropsy, uterus weights were measured, trabecular bone area (TBA) of tibia and lumbar vertebra were measured bone histomorphology. In results, cell proliferation and ALP activity in Saos-2 by ether F1 or 1-monolinolein did not increased significantly compared to the control. The F1 inhibited osteoclast development ($IC_{25}=3.37{\times}10^{-5}$mg/mL) less than 17$\beta$-estradiol. The OVX rats administered F1 (2 mg/kg/day and 10 mg/kg/day) showed an increase in TBA of the tibia significantly (136.3${\pm}4.2% and 138.5{\pm}$10.3% of control). In conclusions, the herbal extract, F1 inhibited tibia and lumbar bone loss and did not cause uterine hypertrophy. However, 1-monolinolein, the main ingredient of the herbal extract, did not inhibit bone loss.

털부처꽃 채취부위별 추출물이 만성 알코올 투여 흰쥐에 미치는 영향 (Effect of Plant Part Extracts of Lythrum salicaria L. on Chronically Alcohol-Administrated Rat)

  • The study was done to investigate the effects of the extracts from the different parts of Lythrum salicaria (LS) on liver protective activities in chronically alcohol-treated rats. SD male rats except normal animals were administrated with alcohol ($30m{\ell}$ of 30%~40% ethanol/kg/day) and the extracts (300 mg/kg/day) for 10 weeks. Chronic alcohol administration decreased body weight, high density lipoprotein (HDL)-cholesterol and the reduced form-glutathione (GSH), whereas increased the ethanol content, glutamic-oxaloacetic transaminase (GOT), total cholesterol, low density lipoprotein (LDL)- cholesterol, triglyceride in blood/serum and the ratio of the oxidized form of glutathione (GSSG) and total GSH (GSSG/total GSH) in liver tissue. Groups treated with the extracts of leaf, root and stem, showed decrease in GOT, total cholesterol and GSSG/total GSH and increase in hepatic aldehyde dehydrogenase (ALDH), total GSH and serum albumin. Administration with the root extract of LS decreased blood ethanol content compared with the other part extracts. But, serum triglyceride values in rats treated with root and stem extract were higher than that of the negative control animals. Flower extract-fed group showed decrease in body weight and serum triglyceride, but increase in the ratio of GOT and glutamic-pyruvic transaminase (GPT), and GSSG/total GSH. From the results, we conclude that the extracts of root and leaf among the plant parts of LS might be useful for the amelioration of the chronic alcohol-induced liver demage of rat.
