Dracocephalum palmatum Stephan 잎 헥산 분획 추출물의 인간 유래 전립선 암세포 사멸에 대한 작용 (Effects of hexane fraction of Dracocephalum palmatum Stephan leaf on human-derived prostate cancer cell death)

  • Objectives : Dracocephalum palmatum Stephan (Lamiaceae) is a medicinal plant used by the East-Russian nomads but there were few studies on this plant. This study was to evaluate anti-cancer effects of D. palmatum Stephan leaf hexane fraction on human derived prostate cancer cell death. Methods : The dried leaves of D. palmatum were dissolved in methanol, and hexane fraction (DpLH) was again obtained from lyophilized methanol extract (DpLM). DpLH was investigated by measuring by MTT assay and annexin V/PI staining to evaluate its effects on the cell viability and apoptosis of PC-3 cells. The ROS generations were detected by DCF-DA dye. The protein expressions were confirmed by p-AKT, Bcl-2, Bax, procaspase-3 activities. Results : After treatment of DpLH to PC-3 cells, the cell proliferation was significantly inhibited, and in addition, DpLH treatment also accelerated apoptosis of PC-3 cells. When DpLH was treated to the PC-3 cells, its ROS production significantly decreased. The proportion of all proteins (p-AKT/actin, Bcl-2/Bax and procaspase-3/actin ratios) showed decreasing tendency of expression compared with the control group. Conclusions : As shown in the above results, the extract from D. palmatum inhibits ROS production and promotes cell death, which is considered to be a relatively safe induction of cell death when administered to a living body. In conclusion, these results suggested that DpLH may have anti-cancer effect in human prostate cancer cell.

Does Vitex Agnus-Castus L. Have Deleterious Effect on Fertility and Pregnancy Outcome? An Experimental Study on Rats for Prediction of Its Safety

  • Objectives: Herbal medicine is a worldwide health topic. Vitex agnus-castus L. (VAC) is a popular plant used for gynecologic problems due to its hormonal effects. The aim of this study is to reveal VAC extract effect on fetus when this herb is used started from antenatal period or during pregnancy. Methods: Performed from starting day of January 2019 till February 2019, 48 rats were assigned in randomly divided eight-member six groups: control (C1), treated group with 365 mg/kg VAC from initiation of insemination (T1) and 30 days prior to pregnancy (T2), control that underwent caesarean section on 15th day of gestational age (C2) and treated group with 365 mg/kg VAC from initiation of insemination (T3) and 30 days prior to pregnancy (T4) that underwent caesarean section. Weight, sex and number of fetuses, abortion and still birth rate and estradiol level were evaluated using t-test by SPSS software. Results: We showed increased weight among T1 group considering totally and sex-dependent which is significant (all p-value < 0.05). We also detected significantly decreased weight in T2 in total (p-value < 0.0001) and when considering female fetuses (0.043) but not males (0.17). Although the results showed slightly non-significant increased weight among fetuses of T3 (totally or based on the fetus sex) compared to the control group (C2), T4 group had statistically decreased weight compared to control group. Pregnancy rate and pregnancy outcome were affected by VAC usage. The time of VAC initiation also affected live birth and abortion rates. Conclusion: VAC extract may affect pregnancy rate, live birth rate, abortion and stillbirth rates. Its effect on the weight and the sex showed dual pattern depends on the time of initiation and pregnancy trimester of evaluation. Prescribing this medicinal plant for patients being prone to pregnancy should be with caution. Further study is recommended.

녹차 결명자 추출물의 교정용 브라켓과 치면 사이의 경계부에서 분리된 mutans streptococci에 대한 항균작용 (Effect of Leaf-Extract from Camellia sinensis and Seed-Extract from Casia tora on Viability of Mutans Streptococci isolated from the interface between orthodontic brackets and tooth surfaces)

  • 본 연구는 치아우식증에 관련된 Hutans streptococci 표준균주 및 임상분리 균주의 성장억제를 유도할 수 있는 천연생약추출물을 검색하기 위하여 실시되었다. 녹차추출물은 CHMC-2032를 사용하였으며, 결명자 추출물은 $50\%$ 에탄올을 이용하여얻었다. 이들 추출물의 Mutans streptococci표준균주 및 교정환자들에서 브라켓과 치면 사이의 경계부에서 분리된 각각 10 균주씩의 S. mutans 및 S. sobrinus에 대한 최소성장억제농도를 액체배지 희석법으로 구하였다. 그 결과 CHMC-2032의 S. mutans 및 S. sobrinus의 표준 균주, S. sobrinus의 대부분 임상분리 균주에 대한 최소성장 억제농도는 5mg/ml이었다 그러나 결명자 추출물에 의한 S. mutans 및 S. sobrinus에 대한 세균 성장억제 효과는 미미하였다. 본 연구 결과 치아우식증의 예방적 측면에서 결명자차보다는 녹차를 마시는 것이 유리하며, CHMC-2032를 이용하여 구강양치용액을 제조하여 사용할 경우 교정환자를 포함하여 대부분의 사람에게서 치아우식증 예방 효과가 있으리고 추정된다.

  • 식물과 해양미세조류 추출물의 Magnaporthe grisea에 대한 항진균 활성을 조사하였다. 석류껍질, 부추 씨, 옥수수껍질의 아세톤 추출물은 M. grisea에 대하여 살진균제양상의 높은 항 진균 활성을 나타내었다. 석류껍질, 부추 씨, 옥수수껍질 아세톤 추출물들의 M. grisea에 대한 최소 억제 농도는 각각 100 ng/ml, $50\;{\mu}g/ml,\;and\;100\;{\mu}g/ml$이었다. Amphiprora gigantea의 에탄올 추출물은 $100\;{\mu}g/ml$의 최소억제 농도로 M. grisea에 대한 항진균 활성을 나타내었다.

GC/MS and HPLC/PDA characterization of essential oils and phenolic compounds from the aerial parts of common rue (Ruta graveolens)

  • Two different extraction methods were used to evaluate the medical value of common rue, Ruta graveolens L. (RGL). The results of our 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid assays indicated that the antioxidant activity of RGL essential oil extract obtained through steam distillation was very low, whereas ethanol (EtOH) extracts of RGL showed higher antioxidant activity. RGL essential oil was extracted by steam distillation and characterized by GC/MS analysis. Furthermore, EtOH extracts of RGL were obtained under reflux and analyzed by HPLC/PDA. The GC/MS results indicated that the ketone compounds 2-undecanol acetate, nonyl cyclopropanecarboxylate, and 2-nonanone accounted for more than 70% of the composition of RGL essential oil. The HPLC/PDA analyses indicated that the RGL extracts were rich in phenolic compounds such as protocatechuic acid, rutin, psoralen, xanthotoxin, and bergapten, among which rutin was the most abundant. Collectively, our results demonstrated that RGL contains high levels of phenolic compounds and could thus be commercialized as a valuable plant-derived antioxidant.

  • The chromatographic separation of methanolic extract from the twigs of Ilex macropoda (Aquifoliaceae) led to the isolation of three hemiterpenes, and their chemical were elucidated by physico-chemical and spectroscopic analysis. Their structures were characterized as 4,5-dihydroxyprenyl caffeate (1), $4-(6-O-caffeoyl-{\beta}-D-glucucopyranosyloxy)-5-hydoxyprenyl$ caffeate (2), and $4-{\beta}-D-glucucopyranosyloxy-5-hydroxyprenyl$ caffeate (3), respectively. Compound 1 was isolated for the first time from plant.

  • Various bioactive substances are found in Loranthus tanakae, including quercetin 3-rhamnoside (1), kaempferol 3-rhamnoside (2), rhamnetin 3-rhamnoside (3), and rhamnocitrin 3-rhamnoside (4), which inhibit tyrosinase. These compounds are mainly found in the EtOAc fraction of L. tanakae extract and demonstrate higher rates of tyrosinase inhibition than ascorbic acid, which was used as a control. Our results suggest that L. tanakae extracts can be utilized in skin whitening cosmetics.

DNA Damage Protection and Anti-inflammatory Activity of Different Solvent Fractions from Aruncus dioicus var. kamtschaticus

  • This study investigated DNA damage protection and anti-inflammatory activity of different solvent fractions from Aruncus dioicus var. kamtschaticus (A. dioicus) aerial parts water extract. As for DNA damage protection, distilled water ($H_2O$) fraction displayed the most powerful protection for DNA damage at a concentration of 1 mg/ml. As for anti-inflammatory activity, dichloromethane ($CH_2Cl_2$) fraction exhibited the highest NO inhibition activity, ranging from 61% to 19% ($10-40{\mu}g/ml$). Furthermore, the levels of pro-inflammatory cytokines mRNA expressions and intracellular reactive oxygen species (ROS) were employed to verify the anti-inflammatory activity of the $CH_2Cl_2$ fraction on further researches. It could be concluded that A. dioicus had a significantly effect of DNA damage protection and anti-inflammatory activity which also as an essential edible vegetable and medicinal species.

Inhibitory Effect of Scutellaria barbata Don Water-extracts on Growth and DNA Incorporation of Human Cancer Cells

  • The water-extracts of Scutellaria barbata Don (SBDE) were isolated from Chinese medicinal plant sources. The extracts showed strong growth-inhibitory activity and cancer chemopreventive activity on the growth and DNA incorporation of MG63 human osteosarcoma and K562 human leukemia cell lines. The growth of human cancer cells was inhibited in the presence of the extracts (20, 50 and 100 ${\mu}$g/ml), and the effects were concentration-dependent and incubation time-dependent up to 8 days. When 50 ${\mu}$g/ml of the extracts was added to the media of MG63 and K562, cell growth after 8 days or 6 days of incubation was retarded by 93.2 to 97.3% of the control group. Morphological changes of MG63 and K562 cell lines were observed. As the concentration of the extracts increased up to 50 ${\mu}$g/ml, degree of cell aggregation decreased. Moreover, the DNA incorporation of the cells which were labeled with [3H] thymidine was significantly reduced after 3 days of incubation at $37^{\circ}C$ with the extract. Therefore, it is suggested that the extract is highly effective on inhibition of cancer cell growth. The extract also inhibited gene expression of IGF-II in transcriptional level. Since IGF-II works as a mitogenic effector on MG63 and K562 cell lines, these results suggest that the growth inhibition is in part mediated through the inhibition of IGF-II gene expression.

가지 물추출물의 3T3-L1 지방전구세포 분화 억제효능 (Inhibitory Effect of Eggplant Extract on Adipocyte Differentiation in 3T3-L1 Cells)

  • Abnormal growth of adipocyte characterized by increased cell numbers and differentiation is considered as an major pathological characteristic feature in obesity. Thus, inhibition of mitogenesis of preadipocytes and their differentiation to adipocytes would be beneficial for the prevention and inhibition of obesity. In the present study, we attempted to evaluate anti-adipogenic activity of eggplants (the fruits of Solanum melongena L.) employing preadipocytes cell line, 3T3-L1 as an in vitro assay system. Water extract of eggplants significantly inhibited adipocyte differentiation when treated during adipocyte differentiation process, as assessed by measuring fat accumulation using Oil Red O staining. Eggplant extract, however, showed little effects on fully differentiated adipocytes. Eggplant didn't show significant toxicity up to 500 ${\mu}g$/ml to the 3T3-L1 cells. Further studies with interval treatment demonstrated that eggplant exerted inhibitory activity on adipocyte differentiation via acting on early stage of adipogenesis. Conclusively, eggplants are suggested to be beneficial for the prevention of obesity.
