Candida glabrata infection of urinary bladder in a Chinchilla Persian cat

  • A 5-year-old castrated male Chinchilla Persian cat weighing 4.84 kg was referred for hematuria. The cat had a history of urethrostomy and bacterial cystitis. In urine culture, Candida glabrata was cultured on Sabouraud dextrose agar. Based on these results, the cat was diagnosed with Candida cystitis. Subsequently, oral administration of fluconazole was initiated. Urine culture was negative at 31 days after administration. This case describes the diagnosis and treatment of Candida glabrata infection of urinary bladder in a cat with a history of urethrostomy.

Propagation of Human Embryonic Stem Cells on Human Amniotic Fluid Cells as Feeder Cells in Xeno-Free Culture Conditions

  • Human embryonic stem cells (hESCs) have been routinely cultured on mouse embryonic fibroblast feeder layers with a medium containing animal materials. For clinical application of hESCs, animal-derived products from the animal feeder cells, animal substrates such as gelatin or Matrigel and animal serum are strictly to be eliminated in the culture system. In this study, we performed that SNUhES32 and H1 were cultured on human amniotic fluid cells (hAFCs) with KO-SR XenoFree and a humanized substrate. All of hESCs were relatively well propagated on hAFCs feeders with xeno-free conditions and they expressed pluripotent stem cell markers, alkaline phosphatase, SSEA-4, TRA1-60, TRA1-81, Oct-4, and Nanog like hESCs cultured on STO or human foreskin fibroblast feeders. In addition, we observed the expression of nonhuman N-glycolylneuraminic acid (Neu5GC) molecules by flow cytometry, which was xenotransplantation components of contamination in hESCs cultured on animal feeder conditions, was not detected in this xeno-free condition. In conclusion, SNUhES32 and H1 could be maintained on hAFCs for humanized culture conditions, therefore, we suggested that new xeno-free conditions for clinical grade hESCs culture will be useful data in future clinical studies.

보건용 마스크의 요구사항 및 만족도 (Requirements and satisfaction with medical masks)

  • This study analyzed requirements for medical masks and difference in satisfaction based on demographic characteristics and side effects experience caused by wearing medical masks. Three factors related to requirements for medical masks were identified: wearing comfort and usability, blocking function for harmfulness, and design. As a result of the difference analysis for requirements of medical masks based on demographic characteristics, all three factors showed a significant difference in gender and occupation. Design did in age and monthly income. As a result of the presence or absence of breathing difficulties experience, design factor was significant. As a result of the presence or absence of skin trouble experience, wearing comfort and usability showed a significant difference. As a result of the difference in satisfaction with medical masks based on demographic characteristics and side effect experience caused by wearing medical masks, breathing, ear string tightness, blocking harmful substances, price, and over all wearing comfort showed a significant difference by gender, marital status, age, occupation, and monthly income, and the presence of absence of breathing difficulty experience and skin trouble experience, respectively. As a result of the interaction effect analysis between demographic characteristics and side effects experience caused by wearing medical masks, it showed a significant interaction effect between gender and monthly income, between marital status and monthly income, between monthly income and breathing difficulties experience, and between monthly income and skin trouble experience.

한국과 중국의 뜸 문화가 일본 뜸 문화 형성 단계에 미친 영향 분석 (The analysis of Korea and China's moxibustion culture effects to Japan's process of forming moxibustion culture)

  • 본 연구에서는 역사적으로 한국과 중국의 뜸 문화가 일본 뜸 문화 형성에 끼친 영향과 일본이 이를 수용하고 제도화 하여 현재 일본사회에 끼친 영향에 대해 연구하고자 한다. 연구방법은 삼국, 특히 일본의 뜸 관련 문헌을 중심으로 그 특징과 발달과정을 시기별로 구분하여 연구, 문헌 분석하였다. 연구결과 첫째, 일본의 의료문제를 해결하기 위하여 중국, 한국과의 교류를 통하여 각종 의료기술과 서적의 수입으로 자국의 전통의학 체계를 확립 것으로 나타났다. 둘째는 한국은 중국의 선진 의학지식을 수용하고 체계화하는 과정에서 이를 다시 일본에 전파하는 가교 역할을 하여 일본 전통의학 발달에 커다란 역할을 한 것으로 나타났다. 결과적으로 일본은 뜸 문화가 가장 활발한 것을 알 수 있었다. 본 연구의 시사점은 일본 뜸 문화 발전과정의 연구를 통하여 우리의 전통의학인 뜸에 대한 새로운 인식과 발전 방향에 기초 자료가 될 것으로 사료된다.

High efficiency of homemade culture medium supplemented with GDF9-β in human oocytes for rescue in vitro maturation

  • Objective: Optimizing culture media for the incubation of immature oocytes is a vital strategy to increase the oocyte maturation rate during in vitro maturation (IVM) programs. This study evaluated the IVM and fertilization rates of human germinal vesicle (GV) and metaphase I (MI) oocytes using two different maturation media (commercial and homemade) with or without growth differentiation factor 9-β (GDF9-β). supplementation. Methods: Immature oocytes from intracytoplasmic sperm injection (ICSI) cycles were collected and assigned to one of two IVM culture media (commercial or homemade; cleavage-stage base). After maturation, MII oocytes were examined under an inverted microscope for the presence of the polar body, zona pellucida (ZP) birefringence, and meiotic spindle (MS) visualization after maturation in four conditions (commercial or homemade medium, with or without GDF9-β. ICSI was done for matured oocytes, and fertilization was confirmed by the visualization of two distinct pronuclei and two polar bodies. Results: No significant differences were found between the two culture media in terms of the time and rate of oocyte maturation or the rate of fertilization (p>0.05). Growth factor supplementation increased the 24-hour maturation rate for both GV and MI oocytes only in homemade medium. The maturation rate after 24 hours was higher for MI oocytes (p<0.05). Similar results were observed for MS visualization and ZP structure in both types of media (p>0.05). Conclusion: Higher rates of oocyte maturation and fertilization were observed after application of homemade medium supplemented with GDF9-β. Therefore, this combination may be recommended as an alternative for clinical IVM programs.

한국전통주인 막걸리로부터 분리한 Lactobacillus plantarum subsp. plantarum B-134의 gamma-aminobutyric acid (GABA)의 생산 (Production of gamma-Aminobutyric Acid (GABA) by Lactobacillus plantarum subsp. plantarum B-134 Isolated from Makgeolli, Traditional Korean Rice Wine)

  • 본 연구는 막걸리로부터 ${\gamma}$-amino butyric acid (GABA) 생성 유산균을 분리 및 동정하고 최적 GABA 생산조건을 확립하는데 그 목적이 있다. 막걸리로부터 64균주의 유산균은 MRS 배지에서 성장된 집락의 색과 모양의 특성에 따라 분리하였다. 분리균주의 GABA 생산은 1% MSG가 첨가된 MRS 액체배지에서 배양하여 TLC와 HPLC 방법에 의해 평가되었다. B-134 균주는 GABA생성을 위한 우수균주로 선발하였다. 16S rRNA 유전자 및 glutamate decarboxylase B (gadB) 유전자의 염기서열분석을 통하여, B-134 균주는 Lactobacillus plantarum subsp. plantarum B-134 균주로 명명하였다. GABA 생성을 위한 온도, pH, NaCl 및 MSG 농도를 달리하여 최적배양조건을 조사하였다. 그 결과 B-134 균주의 최적배양 조건은 온도 $37^{\circ}C$, pH 5.7, NaCl 농도 0% (w/v), 그리고 MSG 농도 3% (w/v)로 결정되었으며 본 조건에서 48시간 배양시 25 mM의 GABA를 생산하였다. 이러한 결과로부터 B-134 균주는 GABA함유 건강기능식품개발을 위한 유용한 균주로 판단된다.

Adverse events following immunisation with the first dose of sputnik V among Iranian health care providers

  • Purpose: Since late 2019, the novel coronavirus disease has been a global concern, and alongside preventive strategies, including social distancing and personal hygiene, vaccination is now the primary hope for controlling the pandemic. Sputnik V is an adenovirus vector vaccine used against coronavirus disease 2019 (COVID-19) among Iranian health care providers, and there is a lack of information regarding the Adverse Events Following Immunisation (AEFI) by Sputnik V among the Iranian population. The present study aimed to evaluate AEFI by Sputnik V vaccine among Iranian population. Materials and Methods: Every member of the Islamic Republic of Iran Medical Council received their first dose of the Sputnik V vaccine in Mashhad (Iran) and was referred to receive their second dose enrolled in the present study and asked to fill an English language checklist asking about development of any AEFI following immunization with the first dose of Sputnik V vaccine. Results: A total number of 1,347 with a mean±standard deviation age of 56.2±9.6 years filled the checklist. Most of the participants were male (838 [62.2%]). The present study demonstrated that immunization with the first dose of Sputnik V results in at least one AEFI in 32.8% of the Iranian medical council members. Most of the AEFI was related to musculoskeletal symptoms, including myalgia. By considering the age of 55 years as a cut-off point, individuals younger than 55 had a higher rate of AEFI (41.3% vs. 22.5%, p=0.0001). Male gender, use of analgesics, beta-blockers, and previous COVID-19 infection have a lower chance of developing AEFI (p<0.05). Conclusion: The present study demonstrated that most of the AEFI was related to musculoskeletal symptoms, including myalgia, and older individuals, male gender and those receiving analgesics and beta-blockers were less likely to develop AEFI following immunization with the first dose of Sputnik V.

Analysis of Microbial Composition Associated with Freshwater and Seawater

  • Knowledge of the distribution and biodiversity of environmental bacteria and the ecosystem that influences them is crucial for predicting an ecosystem. However, bacterial culture methods can only analyze approximately 0.1% of the existing microorganisms, those that are readily cultured under laboratory conditions. By contrast, next-generation sequencing (NGS) has generally been known to obtain more diverse profiling of bacterial composition. We compared the bacterial communities using both a culture-dependent (MALDI-TOF) and culture-independent (NGS) methods. Environmental specimens were obtained from both freshwater and seawater. Water samples were also analyzed by both pyrosequencing and MiSeq sequencing, in order to select one NGS platform which could analyze comparatively more diverse microbiota. Bacterial distribution analyzed with MALDI-TOF showed no difference between the microbiota of freshwater and seawater, whereas the results analyzed with NGS distinguished between the two. The diversity indexes of MiSeq sequencing were higher than for Pyrosequencing. This indicated that MiSeq sequencing is capable of analyzing a comparatively wider diversity of bacteria. The genus of Flavobacterium and Planktophila were identified as being unique to freshwater, whereas EU801223 and OM43 were found in the seawater. Difference between the bacterial composition of the freshwater and seawater environments was identified by MiSeq sequencing analysis.

Effect of feeding mixed microbial culture fortified with trace minerals on ruminal fermentation, nutrient digestibility, nitrogen and trace mineral balance in Sheep

  • Background: The aim of the present study was to determine the effects of feeding trace mineralsfortified mixed microbial culture (TMC) on ruminal fermentation, nutrient digestibility, blood electrolyte status, nitrogen balance, and trace mineral balance in sheep. Methods: Mixed microbes [0.6 % (v/w) of Enterobacter sp., Bacillus sp., Lactobacillus sp., and Saccharomyces sp.] were cultured with 99 % feedstuffs and 0.4 % trace minerals including zinc and copper for ensiling. Six sheep (a mean body weight of $46.5{\pm}1.2kg$) were fed two diets: a control diet (concentrate mix and rye straw) and an experimental diet (a control diet + 3.1 % TMC). Results: TMC feeding did not induce negative effects on ruminal fermentation, nutrient digestibility, blood electrolytes, and nitrogen balance in sheep. Feeding with TMC increased the intake of trace minerals (p < 0.05) and did not affect absorption of trace minerals in the whole digestive tract. Feeding with TMC increased fecal excretion and absorbable intake, and retention of zinc and copper (p < 0.05) by 71 % and 77 %, respectively. Conclusion: Feeding with TMC resulted in higher zinc and copper bioavailability and retention without any adverse effects on sheep performance.

응급실 간호사의 감염관리 조직문화, 감염관리 피로도와 코로나19 감염관리 수행도와의 상관관계 (Correlation Among Organizational Culture, Fatigue for Infection Control, and Infection Control Compliance of COVID-19 Among Emergency Nurses)

  • Purpose: The purpose of this study was to investigate the levels and the relationship of emergency nurses' organizational culture and fatigue for infection control, and infection control compliance of COVID-19. Methods: Data was collected from 114 nurses caring for patients suspected of COVID-19 at two emergency medical centers in Gyeonggi-do and Seoul June 18-July 23, 2021. Results: Emergency nurses caring for patients suspected of COVID-19 suffered from high fatigue for infection control. The mean scores of the organizational culture and fatigue for infection control, and infection control compliance of COVID-19 were 51.80 ± 8.37 of 70 points, 78.46± 12.28 of 100 points and 71.02± 7.84 of 80 points, respectively. The higher infection control compliance of COVID-19 is significantly related to the higher organizational culture for infection control (r= .42, p< .001). Conclusion: In the COVID-19 pandemic, the formation of a positive organizational culture for infection control may be a priority as a major strategy to improve the infection control compliance of emergency nurses. It is also necessary to manage the high level of fatigue for infection control among emergency nurses.
