Optimization of Experimental Conditions for In vitro P-glycoprotein Assay Using LLC-GA5 Cells

  • Identification of compounds that function as P-glycoprotein (P-gp) substrates or inhibitors can facilitate the selection and optimization of new drug candidates. The purpose of this study is to optimize the experimental conditions for in vitro P-gp assay using LLC-GA5 cells, which is a well-known transformant cell line derived by transfecting LLC-PK1 with human MDR1. The amount of rhodamine123 transported by the LLC-GA5 and LLC-PK1 cells was evaluated under the following experimental conditions: 3 different types of transport media, colchicine pretreatment or nontreatment of the cells in the culture media, and with and without poly-L-lysine coating of the culture plates. The assay sensitivity was found to considerably differ depending on the diluents used in the transport media. P-gp-mediated transport in LLC-GA5 cells was most clearly characterized in the Hanks' balanced salt solution based transport media. The sensitivity of P-gp-mediated transport was not changed by colchicine pretreatment or poly-L-lysine coating of the culture plates.

  • Streptococcus pneumoniae (pneumococcus), the most common cause of bacterial pneumonia, has an ample polysaccharide(PS) capsule that is highly antigenic and is the source of PS vaccine. This investigation was undertaken to optimize the culture conditions for the production of capsular PS by type 1 pneumococcus. Among several culture media, brain heart infusion (BHI) and Casitone based media were found to support luxuriant growth of pneumococcus type 1 at the same level. Because BHI medium is rather expensive and more complex than the Casitone based media, the Casitone based media was used to study optimization of the culture condition. The phase of growth which accomodated maximum PS production was logarithmic phase. Concentrations of glucose greater than 0.2% did not enhance growth or PS production. Substitution of nitrogen sources with other resources or supplemention of various concentrations of metal ion (with the exception of calcium ion) had adverse effects on growth and PS production. On the other hand, low level aeration was beneficial for increased PS production. Addition of 3 mg/I concentration of methionine, phenylalanine, and threonine were found to enhance growth and PS production. The synergistic effect of all the favorable conditions observed in pneumococcal growth assays provided a two-fold cumulative increase in capsular PS production.

  • Nowadays the widely used media in architecture include visualizations, animations and three-dimensional models. An optimized 3D digital method using active CAM(Computer Aided Manufacturing) and CNC(Computerized Numerical Control) imaging is developed for accurate shape and 3D measurements in freeform buildings in this paper. In contrast to a conventional building using auto CAD system and others, the proposed active digital optimization is based on a combination of 3D numerical data and parametric 3D model. The objective of this paper is therefore to present digital optimization method for constructability of freeform building. The 3D digital optimization method is appropriate to serious variations in freeform shape. The developed digital optimization method is necessary to be carried out to verify the robustness and accuracy for constructability.

  • The strategies of commercial development have been focused on the economy of scale for a process. The design of media has been recognized as a key in assuring mass production of entomopathogenic nematodes. Media optimization was conducted with insect host, proteins, lipids, and symbiotic bacteria mass. G. mellonella (insect host) produced about 290,000 infective juveniles per one. Complex media produced about 250,000 infective juveniles / ml in liquid culture within 8 days (one generation).

  • The Digital Multimedia Broadcasting(DMB) system is developed to offer high quality audio-visual multimedia contents to the mobile environment. The system adopts MPEG-4 standard for the main video, audio and other media format. It also adopts the MPEG-4 scene description for interactive multimedia contents. The animated and interactive contents can be actualized by BIFS(Binary Format for Scene), the binary format for scene description that refers to the spatio-temporal specifications and behaviors of the individual objects. As more interactive contents are, the scene description is also needed more high bitrate. However, the bandwidth for allocating meta data such as scene description is restrictive in mobile environment. On one hand, the DMB terminal starts demultiplexing content and decodes individual media by its own decoder. After decoding each media, rendering module presents each media stream according to the scene description. Thus the BIFS stream corresponding to the scene description should be decoded and parsed in advance of presenting media data. With these reason, the transmission delay of BIFS stream causes the delay of whole audio-visual scene presentation although the audio or video streams are encoded in very low bitrate. This paper presents the effective optimization technique for adapting BIFS stream into expected MPEG-2 TS bitrate without any bandwidth waste and avoiding the transmission delay of the initial scene description for interactive DMB contents.

  • The growth-strain method was applied to cutout optimization in laminated composite plates. Since the growth-strain method optimizes a shape by generating the bulk strain to make the distributed parameter uniform, the distributed parameter was chosen as Tsai-Hill value. In this study, of particular interest is to see whether the growth-strain method developed for shape optimization in isotropic media would work for laminated composite Plates. In volume control of the growth-strain method, it makes Tsai-Hill value at each element uniform in laminated composite plates under the predetermined volume. The shapes optimized by Tsai-Hill fracture index were compared with those of the initial shapes for the various load conditions and predetermined volumes of laminated composite plates. As a result, it was verified that volume control of the growth-strain method worked very well for cutout optimization in laminated composite plates.

  • In this paper, we propose nonlinear optimization method for feature matching from multiple view image. Typical solution of feature matching is by solving linear equation. However this solution has large error due to nonlinearity of image formation model. If typical nonlinear optimization method is used, complexity grows exponentially over the number of features. To make complexity lower, we use sparse Levenberg-Marquardt nonlinear optimization for matching of features over multiple view image.

Optimization of Media Composition and Culture Conditions for the Mycelial Growth of Coriolus versicolor and Lentinus edodes (Coriolus versicolor와 Lentinus erodes의 영양배지 조성 및 배양조건의 최적화)

  • The media compositions and culture conditions were optimized for mycelial growth of Coriolus versicolor and Lentinus edodes. Media composition for optimal growth of Coriolus versicolor was 2.0% glucose 0.4% peptone and 0.6% yeast extract. Media composition for optimal growth of Lenttnus edodes was 2.0% glucose 2.0% starch 0.4% bacto-soytone and 0.6% yeast extract. The media supplemented with KH2PO4, 0.046% KH2PO4 0.1% and MgSO4, .7H2O 0.05% supported better mycelial growth than the media without mineral salts. The optimum temperature for mycelial growth ranged from $25^{\circ}C$-28$^{\circ}C$. The optimum pH range for mycelial growth of Coriolus versicolor was 5.2~5.6 while that of Lentinus edodes appeared to be 5.75.

  • With the remarkable growth in rich media in recent years, people are increasingly exposed to visual information from the environment. Visual information continues to play a vital role in rich media because people's real interests lie in dynamic information. This paper proposes a novel discrete dynamic swarm optimization (DDSO) algorithm for video object tracking using invariant features. The proposed approach is designed to track objects more robustly than other traditional algorithms in terms of illumination changes, background noise, and occlusions. DDSO is integrated with a matching procedure to eliminate inappropriate feature points geographically. The proposed novel fitness function can aid in excluding the influence of some noisy mismatched feature points. The test results showed that our approach can overcome changes in illumination, background noise, and occlusions more effectively than other traditional methods, including color-tracking and invariant feature-tracking methods.

  • In this paper, we evaluate the correlation properties and the optimization of PN sequence phase for multi-media DS/CDMA system with variable spreading gain (VSG) scheme. In multi-media multi-rate DS/CDMA systems, the optimization of PN sequence phase is not a tractable problem, since the sequences should be optimized against both sequences of the same length and other sequences with different length. Hence, we verify the correlation properties of PN sequence phase in multi-rate system environment and furthermore, we propose the new phase criterion, MIN-AIP (minimum-average interference parameter), to minimize the bit error rate (BER). As the results of performance evaluations, it is shown that the performance of MIN-AIP criteria gives the best results.
