Protein quality, hematological properties and nutritional status of albino rats fed complementary foods with fermented popcorn, African locust bean, and bambara groundnut flour blends

  • The objective of this study was to determine protein quality and hematological properties of infant diets formulated from local food materials. The food materials were obtained locally, fermented, and milled into flour. The flours were mixed as 70% popcorn and 30% African locust bean (FPA), 70% popcorn and 30% bambara groundnut (FPB), and 70% popcorn, 20% bambara groundnut, and 10% African locust bean (FPAB). Proximate analysis, protein quality, hematological properties, and anthropometric measurements of the animals fed with the formulations were investigated. The protein contents of the formulated diets were significantly higher than that of Cerelac (a commercial preparation) ($15.75{\pm}0.01g$/100 g) and ogi (traditional complementary food) ($6.52{\pm}0.31g$/100 g). The energy value of FPAB ($464.94{\pm}1.22\;kcal$) was higher than those of FPA ($441.41{\pm}3.05\;kcal$) and FPB ($441.48{\pm}3.05\;kcal$). The biological value (BV) of FPAB (60.20%) was the highest followed by FPB (44.24%) and FPA (41.15%); however, BV of the diets was higher than that of ogi (10.03%) but lower than that of Cerelac (70.43%). Net protein utilization (NPU) of the formulations was 41.16-60.20%, whereas true protein digestibility was 41.05-60.05%. Metabolizable energy (232.98 kcal) and digestible energy (83.69 kcal) of FPAB were the highest, whereas that of FPA had the lowest values. The protein digestibility values corrected for amino acid score of the diets (0.22-0.44) were lower than that of Cerelac (0.52), but higher than that of ogi (0.21). The growth patterns and hematological properties (packed cell volume, red blood cells, hemoglobin, mean corpuscular hemoglobin concentration, mean corpuscular hemoglobin, and mean corpuscular volume) of the formulated diets were higher than those of ogi, but lower than those of Cerelac. In conclusion, we established that the FPAB food sample was rated best in terms of protein quality over the other formulated diets. Therefore, a FPAB blend may be used as a substitute for ogi.

  • Male Sprague Dawley rats were exposed to 0, 0.04, 0.2, and 0.8% Pb acetate in drinking water for 13 weeks and fed a commercial diet. Dose-related adverse effects observed at the end of the Pb acetate exposure in the drinking water were as follows: decrease in body weight gain, decrease in hemoglobin, hematocrit(HCT), mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH), increase in serum glucose, decrease in serum testosterone, increase in lead accumulation and $\delta$-ALA release in urine, and decrease in $\delta$-ALAD activities DNA content and histopathlogy (intranuclear inclusion body in kidney proximal tubule cell). Taken together, repeated exposure of lead acetate induced toxicities in hematopoietic system, especially testis and kidney.

포르말린 침지에 의한 붕어(Carassius auratus)의 혈액학 및 혈액화학치에 대한 영향 (Effects of formalin on haematology and blood chemistry in crucian carp (Carassius auratus))

  • To determine the effects of exposure to formalin on the secondary stress indices, changes in haematology and blood chemistry were monitored in healthy crucian carps (Carassius auratus). Fishes were separately exposed in a concentration range of 125 to 500 ppm formalin for 60 min. After exposure, red blood cell (RBC) count and packed cell volume (PCV) were elevated in the 500 ppm formalin exposed group. However, mean corpuscular volume (MCV) and mean corpuscular haemoglobin (MCH) were decreased significantly in the 500 ppm formalin exposed group. Total protein, albumin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, total bilirubin, inorganic phosphorus (IP) and magnesium were significantly increased at a concentration of 500 ppm. Alkaline phosphatase (ALP) and glucose were increased at a concentration of 500 ppm, but this was not significant. Lactate dehydrogenase (LDH) and calcium were significantly decreased at concentrations of 250 and 500 ppm. AST, ALT, glucose and magnesium were significantly increased in the 250 ppm formalin exposed group. These results suggests that formalin exposure might cause some damage in the liver and kidney of crucian carp.

  • The Allium tuberosum is a perennial herb. From the ancient times, it has been used for food or medical purpose. Allium tuberosum is widely distributed in Korea, China, Japan and other countries. As herbal medicine, the Allium tuberosum has medical effects on lumbago, sthenia, diuresis, nocturia, hematemesis, anemia, haemorrhoid, hiccups, diarrhea, eczema, sunstroke, pollution and so on. Recently, the Allium tuberosum has been studied on its components, anti-microorganism action, anti-fungal action, anti-arteriosclerosis action, anti-cancer action. However, there has not been any study on hematological effect of allium tuberosum extract on the blood in large dogs whereas the effects of excess garlic and onion on the blood of cattle, horses, dogs, cats and sheep has been reported by many researchers. Thus, this study was performed to observe the hematological effect of excessive Allium tuberosum extract administrated in large dogs. 1. The Red Blood Cell(R3C) count was significantly decreased(p<0.05), Packed Cell Volume (PCV) count were significantly decreased(P<0.05), and Reticulocyte count was significantly increased(p<0.05) 2. The White Blood Cell(WBC) count was significantly increased(p<0.05).3. The Hemoglobin concentration was significantly decreased(p<0.05) and Mean Corpuscular Hemoglobin Concentration (MCHC) was increased. and Mean Corpuscular Volume(MCV) was significantly increased.(p<0.05). 4. The Reduced glutathione(GSH) and Met-Hemoglobin were significantly increased (p<0.05).

Toxic effects of ammonia exposure on growth performance, hematological parameters, and plasma components in rockfish, Sebastes schlegelii, during thermal stress

  • Rockfish, Sebastes schlegelii (mean length $14.53{\pm}1.14cm$ and mean weight $38.36{\pm}3.45g$), were exposed for 4 weeks with the different levels of ammonia in the concentrations of 0, 0.1, 0.5, and 1.0 mg/L at 19 and $24^{\circ}C$. The indicators of growth performance such as daily length gain, daily weight gain, condition factor, and hematosomatic index were significantly reduced by the ammonia exposure and high temperature. The ammonia exposure induced a significant decrease in hematological parameters, such as red blood cell (RBC) count, white blood cell (WBC) count, hemoglobin (Hb), and hematocrit (Ht), whose trend was more remarkable at $24^{\circ}C$. Mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) were also notably decreased by the ammonia exposure. Blood ammonia concentration was considerably increased by the ammonia concentration exposure. In the serum components, the glucose, glutamic oxalate transaminase (GOT), and glutamic pyruvate transaminase (GPT) were substantially increased by the ammonia exposure, whereas total protein was significantly decreased. But, the calcium and magnesium were not considerably changed.

음양곽 추출물이 사염화탄소 투여로 유발된 간손상 및 연관된 장기 손상의 회복에 미치는 영향 (Effects of Epimedium Koreanum Extract on the Carbon Tetrachloride-Induced Liver Damage and Its Related Organ Damages in Rats)

  • To evaluate hepatoprotective effect of Epimedium Koreanum nakai (EKN) on liver-damaged animal model, rats were intoxicated with carbon tetrachloride (1 ml/kg) for 9 weeks orally. Liver-damaged rats were divided into 2 groups: liver-damaged control (LDC) group and EKN group were administered vehicle (saline), EKN extract per os for 4 weeks respectively. Normal control (NC) group was administered saline as the same process of LDC group. The weights of prostate (absolute), testis (relative), epididymis (relative) and packed cell volume (PCV), mean corpuscular volume (MCV) of EKN group significantly (P < 0.05) increased compared with LDC group. But Mean corpuscular hemoglobin (MCH), mean corpusulcar hemoglobin concentration (MCHC) and aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) were significantly (P < 0.01, P < 0.05) decreased. Fibrotic regions in hepatic parenchyma of EKN group significantly (P < 0.01) decreased compared with LDC group and mean diameters of hepatic lobules significantly (P < 0.01) increased. Percentages of degenerative kidney regions and number of degenerative kidney tubules, number of vasodilated atrophic glomerulus of EKN group was significantly (P < 0.01, P < 0.05) decreased compared with LDC group. Number of atrophic seminiferous tubules and epididymal tubules showing oligospermatozoa of EKN group were significantly (P < 0.01) decreased compared with LDC group. In conclusion, EKN extract has a favorable effect on the $CCl_4$-induced liver damage.

Effects of supplementation cysteine-coated Fe3O4 nanoparticles compared to FeSO4, on reproductive performance in male quail

  • Iron has a crucial role in growth as part of metalo-proteins like haemoglobin or myoglobin, enzymes; they are also involved in energetic reactions. Iron plays a vital role in fertility. At high doses, Iron has a harmful consequence on the reproductive system, which can be strongly reflected the final stage of spermatogenesis. Nutritional products are claiming to use nanotechnology and it is important to recognize the potential toxicity of nano-sized nutrients. Recently iron nanoparticles were proposed as a food additive for poultry. The objective of this study was to investigate the effects of L-cystein coated iron oxide nanoparticles on reproductive performance in male quails. The results of Fourier Transform Infrared Spectrometer, Alternating Gradient Force Magnetometer and Scaning Electron Microscopy showed that iron oxide nanoparticles was produced and have been coated with L-cycstein (Fe3O4-Cys NPs). A total of 100 one-week-old quail chicks were randomly placed to five groups of five replicates. Four quails (two male and two females) were raised in an individual cage for each replicate. The five experimental treatment diets consisted; negative control diet, with no Iron supplementation; positive control diet supplemented with 60 mg/kg of Fe3O4; treatment diets supplemented with 0.6, 6 and 60 mg/kg of L-cystein coated iron oxide nanoparticles. The hemoglobin, Red blood cell, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, gonadal somatic index, daily sperm production, total testicular sperm and sperm viability of the male quails that were fed with diet supplemented by 0.6 mg/kg of Fe3O4-Cys NPs were improved as compare with negative control. This study showed that not only the use of the Fe3O4-Cys nanoparticles had no side effects but also it can be used as a feed additive to improve the reproductive performance in male quails.

Effects of testosterone on the orchidectomized rats

  • The aim of this study was to determine the changes of body weight, organ weight, hematological values and biochemical parameters by testosterone (Testos) on the orchidectomized (Orch) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. Orch group received operation and no treatment. Orch+Testos received operation and testosterone. The body weights of each group increased, but that of Orch+Testos group was significantly lower in Orch+Testos group than in all the other groups. There were significant differences (P<0.05, P<0.001) of body weights between Orch+Testos group and all the other groups. Also, organ weights such as heart, liver, spleen and kidney were measured. The heart weights were significantly lower (P<0.001) in the Orch+Testos group than in all the other groups. The liver weights in the Orch+Testos group were significantly differences in comparison with those in the Sham (P<0.001) and Orch group (P<0.05). On the other hand, there were no significantly differences in the organ weights of spleen and kidney between the Orch+Testos group and the any other groups. The hematological values of white blood cell (WBC), red blood cell (RBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) were no significant differences in any other groups. The concentrations of serum total protein and albumin increased significantly (P<0.05) in the Orch+Testos group as compared to that in the Orch group. However, there were no significant differences in Ca, IP and Mg in any other groups. We conclude that testosterone was significantly decreased the body weight in the orchidectomized rats. Our findings suggest that testosterone may influence the process of lipid packaging and absorption in the orchidectomized rats.

만성설사를 보이는 생후 4개월령의 한우 암컷 송아지에서 도체탕의 치료효과 (The effect of Dochetang for the treatment of chronic diarrhea in a 4 months-old-female Korean native calf)

  • This study was carried out to evaluate the effects of Dochetang for the treatment of chronic diarrhea in a 4 months-old-female Korean native calf. The calf was presented to the Wow Animal Clinic, Iksan with the history of chronic diarrhea for several weeks. The feces test did not reveal the presence of the parasites or microbes causing diarrhea in calf. The blood test was also negative to the virus that causes of diarrhea in calf. Adminstration of parenteral antibiotics resulted in improvement of the condition temporarily but diarrhea was recurred again after 2-3 weeks. Then the calf was treated with Dochetang administered orally once a day in an empty stomach for 15 days. Feces was significantly reduced in moisture on 7 days after initial treatment. On 9 days after initial treatment, the calf had normal appetite and defecation in physiological conditions. Blood samples were collected before administration and on 1, 2 and 3 weeks after initial administration of Dochetang for hematology and biochemistry. A significantly differences were observed in the white blood cell (WBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), albumin (ALB), glutamic pyrubic transaminase (GPT), inorganic phosphorus (IP) and magnesium (Mg), while no significant differences were seen in the red blood cell (RBC), hemoglobin (Hb), hematocrit (HCT), platelet (PLT), glucose (Glu), total protein (T-pro), glutamic oxaloacetic transaminase (GOT), blood urea nitrogen (BUN) and creatine (CRE). This study suggests that Dochetang administration can be a successful alternative therapeutic agent in instead of antibiotics for the treatment of chronic diarrhea in calves.

Effects of vitamin E and selenium (Selevit) on the orchidectomized rats

  • The present study was devised to determine the effects of body weight, organ weight, hematological values and biochemical parameters by vitamin E and selenium (Selevit) on the Orch rats. The kidney weights in the Orch+Selevit group were significantly differences (P<0.01, P<0.001) in comparison with those in all the other groups. On the other hand, there were no significantly differences in the organ weights of spleen between the Orch+Selevit groups and the any other groups. The number of white blood cell (WBC) was significantly higher (P<0.05) in the Orch+Selevit group than in all the other groups. The hematological values of red blood cell (RBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) were not significantly different in any of the groups. The concentrations of serum total protein, albumin and alkaline phosphatase increased significantly (P<0.05, P<0.01) in the Orch+Selevit group as compared to that in the Orch group. However, there were no significant differences in AST and ALT in any other groups. We conclude that Selevit was significantly decreased the body weight in the orchidectomized rats. Our findings suggest that Selevit may influence the process of lipid packaging and absorption in the orchidectomized rats.
