Effects of Siegesbckiae Herba on the Brain Ischemia (희렴 이 뇌허혈에 미치는 효과)

  • This experimental study was designed to investigate the effects of SIEGESBECKIAE HERBA extract (SHE) on the change of cerebral hemodynamics 〔regional cerebral blood flow (rCBF) and mean arterial blood pressure(MABP)〕 in normal condition and cerebral ischemic rats, and to determine the mechanism of action of SHE. This study was designed to investigate whether or not SHE inhibit lactate dehydrogenase (LDH) activity in neuronal cells and cytokines production in serum of cerebral ischemic rats. The results were as follows SHE increased rCBF significantly in a dose-dependent manner, but MABP was not changed by SHE in normal rats. The SHE-induced increase in rCBF was significantly inhibited by pretreatment with indomethacin (IDN), an inhibitor of cyclooxygenase but was increased by methylene blue (MTB), an inhibitor of guanylate cyclase. SHE inhibited lactate dehydrogenase (LDH) activity significantly in neuronal cells. rCBF was increased significantly and stably by SHE(10㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group in ischemic rats. In serum by drawing from femoral arterial blood after middle cerebral arterial occlusion(MCAO) for 1hr and reperfusion for 1hr, the sample group was decreased IL-1β production significantly compared to that of the control group. In serum by drawing from femoral arterial blood after MCAO 1hr and reperfusion 1hr, sample group decreased TNF-α production significantly compared to that of the control grolilp. In serum by drawing from femoral arterial blood after reperfusion 1hr, sample group increased TGF-β production significantly compared to that of the control group. In serum by drawing from femoral arterial blood after MCAO for 1hr and reperfusion for 1hr, IL-10 production of the sample group was similar to that of control group. These results suggested that SHE had inhibitive effect on the brain damage by inhibited LDH activity, IL-1β and TNF-α production, but accelerated TGF-β production.

  • Study of the effects of the methanolic extract of Tagetes patula roots on blood pressure led to the isolation of well known citric (1) and malic acid (7) as hypotensive, and pyridine hydrochloride (4) as hypertensive constituents of the plant along with a new constituent, 2-hydroxy, 5- hydroxymethyl furan (9). Citric acid and malic acid caused 71% and 43% fall in Mean Arterial Blood Pressure (MABP) of rats at the doses of 15 mg/kg and 30mg/kg respectively while pyridine hydrochloride produced 34% rise in the MABP of rats at the dose of 30mg/kg. $LD_{50}\;and\;LD_{100}$ of citric acid in mice have been determined as 545 mg/kg and 1000 mg/kg, respectively.

Experimental Effects of Sibjeondaebo-tang and Gamy-Sibjeondaebo-tang on Cerebral Hemodynamics in Cerebral Ischemia Rats (십전대보탕(十全大補湯)과 가미십전대보탕(加味十全大補湯)이 뇌허혈 흰쥐의 뇌혈류역학에 미치는 실험적 영향)

  • This Study was designed to investigate the effects of Sibjeondaebo-tang (SDT) and Gamy-Sibjeondaebo-tang (GST, Sibjeondaebo-tang adding Cervi Pantotrichum Cornu) on the improvement in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. And, It was to investigate the effects of the SDT and GST with the change of histologic examination through the BDNF in the hippocampus CA1. In changes of cerebral hemodynamics, SDT and GST significantly increased rCBF in a dose-dependent manner but decreased MABP in normal rats. In mechanism of cerebral hemodynamics, Increase of GST-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and Decrease of GST-induced MABP was significantly increased by pretreatment with methylene. These results suggested that the action of GST was mediated by guantlate cyclase pathway. In cerebral ischemics, the rCBF was stably improved by SDT (10 mg/kg, i.p.) significantly and stably increased by GST (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrast with the findings of rapid and marked increase in Control group. These results suggested that GST had anti-ischemic action in cerebral ischemic state. In histological examination through TTC stain, Sample A group and Sample B group decreased discoloration in the cortical part at $28^{th}$ day after MCAO induction. In immunohistochemistric response of BDNF, Sample A group and Sample B group increased respondent effect at $28^{th}$ day after MCAO induction. These results suggest that GST can Increase rCBF in normal state, as well as improve the stability of rCBF in cerebral ischemic state. Furthermore, methylene blue inhibitor study suggested the mechanism of blood flow enhancement by GST may be mediated by guanylate cyclase pathway.

  • Cheonghunhwadam-tang have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CHT Gamypang(CTG) was CHT adding Aurantii Fructus(AF). The effects of CTG on the regional cerebral blood f1ow(rCBF) and mean arterial blood pressure(MABP) is not known. The mechanical Study of CTG on the cerebral hemodynamics is not known too. Therefore, purpose of this Study was to investigate effects of CTG on the rCBF and MABP, mechanism of CTG on the cerebral hemodynamics in rats. The changes of rCBF and BP was determinated by Laser-Doppler Flowmetry(LDF). The results were as follows ; CTG extract was significantly decreased rCBF in a dose-dependent, but the change of MABP was not shown. Pretreatment with propranolol(3mg/kg, i.v.) was significantly increased CTG(10 mg/kg, i.v.) induced decrease of rCBF. Pretreatment with indomethacin(3mg/kg, i.v.) and methylene blue(10㎍/kg, i.v.) were increased CTG induced decrease of rCBF too. This results suggest that the mechanism of CTG is mediated by adrenergic β - receptor, guanylate cyclase and cyclooxygenase.

Effects of Tonggok($BL_{66}$) ${\cdot}$ Hyeopgye($GB_{43}$) Supplement and Sangyang($LI_1$) ${\cdot}$ Gyueum($GB_{44}$) Draining in Acupuncture on the Improvement of Cerebral Hemodynamics in Rats (족소양담경(足少陽膽經) 정격(正格) 척침(刺鍼)이 백서(白鼠)의 뇌혈류력학(腦血流力學) 변동(變動)에 미치는 영향)

  • Objectives: The purpose of this report is to examine the effect of $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and LIl ${\cdot}$ $GB_{44}$ draining in acupuncture for cerebral apoplexy treatment. Methods : I designed to investigate whether $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture affects cerebral hemodynamics [regional cerebral blood flow(rCBF), mean arterial blood pressure(MABP)] in normal rats, and to make manifest whether $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture is mediated by cyclooxygenase or guanylate cyclase. The changes of rCBF and MABP were determinated by laser-doppler flowmetry(LDF). Results : 1. $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and LI1 ${\cdot}$ $GB_{44}$ draining in acupuncture increased rCBF significantly, but decreased MABP. These results suggest that $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture increased significantly rCBF by dilating pial arterial diameter(PAD). 2. By pretreatment with indomethacin(1 mg/kg, i.p.), the effect of $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture induced increase of rCBF was significantly inhibited, the decreased MABP by treatment with $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture was increased. 3. By pretreatment with methylene blue($10{\mu}/kg$, i.p.), the increased rCBF by treatment with $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture was decreased conversely, the decreased MABP by treatment with $BL_{66}$ ${\cdot}$ $GB_{43}$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture was decreased. and then increased. Conclusion : I suggest that $BL_{66}$ ${\cdot}$ $GB_43$ supplement and $LI_1$ ${\cdot}$ $GB_{44}$ draining in acupuncture has an anti-ischemic effect through the improvement of cerebral hemodynamics.

Effects of Joongjeo($TE_3$) Supplementation Aekmoon($TE_2$) Draining on Changes in Cerebral Blood Flow and Blood Pressure in Normal Rats (수소양삼초경(手少陽三焦經) 정격(正格)의 자경보사(自經補瀉)(중저 보(中渚 補), 액문 사(液門 瀉)) 자침(刺鍼)이 정상 흰쥐의 뇌혈류량(腦血流量) 및 혈압(血壓)에 미치는 영향(影響))

  • Objectives : Joongjeo($TE_3$) Supplementation Aekmoon($TE_2$) Draining is a method belongs to Ohaeng-acupuncture, using directional supplementation and draining. Methods : This study was designed to investigate the effects of $TE_3$ supplementation $TE_2$ draining on changes in cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in normal rats. For these reasons, the present author investigated rCBF and MABP using laser doppler flowmeter in normal rats. In addtion, the present author also investigated action mechanisms of $TE_3$ supplementation $TE_3$ draining on changes in rCBF and MABP too. Results : In this results, $TE_3$ supplementation $TE_2$ draining elevated rCBF in time-dependent manner, but MABP levels decresed by $TE_3$ supplementation $TE_2$ draining. Pre-treatment with indomethacin (IDM), an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. But pre-treatment with methylene blue(MTB), an inhibitor of guanylate cyclase, decreased rCBF levels. In addition, pre-treatment with IDM also decreased MABP levels, but pre-treatement with MTB increased MABP levels. Conclusions : In conclusion, these results suggest that $TE_3$ supplementation $TE_2$ draining is effective to treat patient with disease related to cerebral ischemia, because $TE_3$ supplementation $TE_2$ draining can increase rCBF. In addition, the mechanisms are thought to be related to guanylate cyclase pathways.

The Effects of Perpendicular Needling Laogong ($PC_8$) on the Improvement of Cerebral Hemodynamics in Rats (노궁(勞宮)($PC_8$) 직자(直刺)가 백서(白鼠)의 뇌혈류력학(腦血流力學)에 미치는 영향)

  • Objectives : This study was designed to investigate the effects of acupuncturing $PC_8$ used perpendicular needling method determine the mechanism of action of acupuncturing $PC_8$ by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. Methods : This study also investigated the effects of acupuncturing $PC_8$ on the change of rCBF in cerebral ischemic rats, and revealed the mechanism of its action. In addition, the effects of acupuncturing $PC_8$ on focal ischemic brain injury was studied in cerebral ischemic rats. Results : 1. Acupuncturing $PC_8$ significantly increase rCBF but decreased MABP in normal rats. 2. Acupuncturing $PC_8$ increased of rCBF was significantly inhibited by pretreatment with indomethacin (1mg/kg, i.p.), an inhibitor of cyclooxygenase in normal rats. 3. Acupuncturing $PC_8$ increased of rCBF was significantly inhibited by pretreatment methylene blue (10 ${\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase in normal rats. 4. Acupuncturing $PC_8$ was significantly improved the rCBF than control group increased unstable in cerebral ischemic rats. 5. Acupuncturing $PC_8$ was not significantly improved the rCBF than control group by pretreatment with indomethacin (1mg/kg, i.p.), an inhibitor of cyclooxygenase in cerebral ischemic rats. 6. Acupuncturing $PC_8$ was significantly increased the rCBF than control group by pretreatment methylene blue ($10{\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase in cerebral ischemic rats. Conclusions : In conclusion, our study suggested that acupuncturing $PC_8$ can increase rCBF in normal state, and improve stability of rCBF in ischemic state. In addition, we suggested that mechanisms related with acupuncturing $PC_8$ was involved in the guanylate cyclase pathway.

Effects of Yangsim-tang on the Changes of Cerebral Hemodynamics in Rats (양심탕이 백서의 뇌혈류역학 변화에 미치는 영향)

  • Objectives : This study was performed to evaluate effects of Yangsim-tang extract (YST) on hemodynamics (regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) ) in normal rats, and effects of cyclooxygenase and guanylate cyclase under YST. Methods : Laser-Doppler flowmetry (LDF) measured changes of rCBF, and a data acquisition system assembled with MacLab and Macintosh measured changes of MABP. Results : YST significantly increased rCBF, but did not change MABP. Pretreatment with indomethacin significantly inhibited rCBF increased by YST, but pretreatment with methylene blue did not significantly inhibit rCBF increased by YST. Conclusions : YST increases rCBF, and the action of this response is mediated by cyclooxygenase.

Experimental Study of Patholobi Caulis on the Transient Cerebral Ischemia in Rats (계혈등(鷄血藤) 추출물이 뇌허혈에 미치는 실험적 연구)

  • The study was designed to investigate the mechanism of Patholoobi Caulis freeze dried powder (PCF) on the regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats and cytokines production ($IL-1{\beta}$, $TNF-{\alpha}$, IL-10, $TGF-{\beta}$) in cerebral ischemic rats. The results in normal rats were as follows ; Increase of PCF-induced rCBF was significantly inhibited by pretreatment with methylene blue (10 ${\mu}g/kg$, I.p.), an inhibitor of guanylate cyclase, and was inhibited by indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. Increase of PCF-induced MABP was decreased by pretreatment with methylene blue, but was increased by indomethacin. These results suggested that the mechanism of action PCF was mediated by cyclic 3',5'-guanosine monophosphate. The results in cerebral ischemic rats were as follows ; In cytokine production in serum from femoral arterial blood 1 hr after middle cerebral arterial occlusion, PCF (10 mg/kg. i.p.) significantly decreased $IL-1{\beta}$ and $TNF-{\alpha}$ production, and increased IL-10 production compared with control group. In cytokine production in serum from femoral arterial blood 1 hr 1 hr after reperfusion, PCF (10 mg/kg, i.p.) significantly decreased $IL-1{\beta}$ production, and incresed IL-10 production compared with control group. These results suggested that PCF was significantly and stably increased regional cerebral blood flow by inhibiting $IL-1{\beta}$ production, and by accelerating IL-10 production.

Experimental Study of Dadun(LR1)·Shaofu(HT8) Reduction in Acupuncture on the Improvement of Cerebral Hemodynamics (대돈(大敦)·소부(少府) 사법자침(瀉法刺鍼)이 뇌혈류역학(腦血流力學) 변동(變動) 개선(改善)에 미치는 영향)

  • Objective: The aim of this study was to investigate anti-ischemic effect of LR1 HT8 Reduction in Acupuncture Methods: I designed to investigate whether LR1 HT8 Reduction in Acupuncture affects cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD), mean arterial blood pressure(MABP)] in normal rats and to make manifest whether LR1 HT8 Reduction in Acupuncture is mediated by cyclooxygenase or guanylate cyclase. The changes of rCBF and MABP were determinated by laser-doppler flowmetry(LDF), and the change of PAD was de terminated by video microscope and width analyzer. Results: The results were as follows; 1. LR1 Reduction in Acupuncture was increased rCBF and PAD, but decreased MABP. 2. HT8 Reduction in Acupuncture was significantly increased rCBF, but decreased MABP, and increased PAD. 3. LR1 HT8 Reduction in Acupuncture was significantly increased rCBF, PAD, but decreased MABP after withdrawing of the needle. This results suggest that LR1 HT8 Reduction in Acupuncture increased significantly rCBF by dilating PAD. 4. Pretreatment with indomethacin(1mg/kg, i,v.) was significantly inhibited LR1 HT8 Reduction in Acupuncture induced increase of rCBF and PAD, but increased LR1 HT8 Reduction in Acupuncture induced decrease of MABP after withdrawing of the needle. 5. Pretreatment with methylene blue($10{\mu}g/kg$, i,v.) was decreased LR1 HT8 Reduction in Acupuncture induced increase of rCBF and PAD, but accelerated LR1 HT8 Reduction in Acupuncture induced decrease of MABP. Conclusions: I suggest that LR1 HT8 Reduction in Acupuncture has an anti-ischemic effect through the improvement of cerebral hemodynamics, and the mechanism is mediated by cyclooxygenase.
