The composition of the bacterial populations in Gorosoe(Acer mono Max.) sap was characterized during storage with different heat treatments($63^{\circ}C$ for 30 min and $73^{\circ}C$ for 15 sec). The saps were aseptically collected at 0, 15 and 30 days of storage and analyzed by dilution plating and 16S rDNA PCR-DGGE analysis. There were significant differences in the total number of colony forming units(CFUs) of bacteria between heated and nonheated saps. Bacteria of nonheated sap were present at a level of $3.4{\times}10^7CFU/m{\ell}^{-1}$, whereas living bacteria were not detected in the heated sap. According to the 16S rDNA sequence and DGGE analysis, Pseudomonas sp. was the most abundant bacterial strain in the samlpes, and the bacterial community structures become more simplified with time and were composed of the Chryseobacterium sp. with time. These results allowed us to characterize the dominant bacteria involved in Gorosoe sap and to better understand their dynamics throughout storage.
Borosilicate glass was prepared in the composition of 81% $SiO_2$, 4% $Na_2O$, 2% $Al_2O_3$, 13% $B_2O_3$. The albite phase($NaAlSi_3O_8$) increased with the $ZrO_2$(0~10wt.%) addition. For measurement of glass transition temperature($T_g$), crystallization temperature($T_{c,max}$) measured by differential thermal analysis. The $T_g$ and $T_{c,max}$ were $510{\sim}530^{\circ}C$$650{\sim}670^{\circ}C$, respectively. The crystallized glass was heated at various conditions(temperature, time). After nucleation at $550^{\circ}C$ for 2hours prior to crystal growth at $650^{\circ}C$ for 4hours, the resulting Vickers hardness, fracture toughness and bending strength were about $736H_v$, $1.0779MPa{\cdot}m^{1/2}$, and 493MPa, which were 17%, 45% and 149% higher than parent borosilicate glass, respectively. Crystal size and transmittance of crystallized borosilicate glass were analyzed by FE-SEM, EDX and UV-VIS-NIR spectrophotometer. Transmittance of crystallized borosilicate glass was decreased with increasing $ZrO_2$(wt%) at visible-range. The results prove that light-weight bulletproof can be fabricated by the crystallization of borosilicate glass.
연어알(Salmo Salar)에서 carotenoprotein을 CM-세룰로스, 50% 황산암모늄 침전, DEAE-세룰로스 및 세파덱스 G-75젤을 사용하여 충분히 정제하였다. 인산염 완충용액(pH 7.0)에서 자외선 흡수극대는 ${\lambda}_{max}$ 409, 540 그리고 580nm이었다. 세파덱스 G-200젤 여과법으로 측정된 분자량은 50, 200rhk 26,000달톤으로 heterogeneous한 두가지 형태였다. 전자는 적자색으로 SDS-PAG 전기영동 결과 분자량이 12,500달톤인 4개의 동일한 subunit로 구성되었고, 상당량의 지질과 만노오스, 갈락토스와 글루코사민을 함유하고 있음을 밝혔다. carotenoprotein의 색소를 추출하여 tlc로 정제한 다음 분배비 시험, 요오드 시험, 에폭시 그룹 시험, 알릴릭 시험, 환원 시험 그리고 아세틸화 시험등의 유기반응과 uv/vis, ir 및 nmr 데이타를 얻어 구조를 결정한 결과 carotenoid는 astaxanthin ester임을 확인하였으며 그 ester는 스테아르산(47.9%)과 팔미트산 (21.4%)이 대부분이었다.
Objective of present study was to prepare and characterize self-nanoemulsifying drug delivery system (SNEDDS) of lutein and to evaluate its effect on bioavailability of warfarin. The SNEDDS was prepared using an oil, a surfactant, and co-surfactants with optimal composition based on pseudo-ternary phase diagram. Effect of the SNEDDS on the bioavailability of warfarin was performed using Sprague Dawley rats. Lutein was successfully formulated as SNEDDS for immediate self-emulsification and dissolution by using combination of Peceol as oil, Labrasol as surfactant, and Transcutol-HP or Lutrol-E400 as co-surfactant. Almost complete dissolution was achieved after 15 min while lutein was not detectable from the lutein powder or intra-capsule content of a commercial formulation. SNEDDS formulation of lutein affected bioavailability of warfarin, showing about 10% increase in $C_{max}$ and AUC of the drug in rats while lutein as non-SNEDDS did not alter these parameters. Although exact mechanism is not yet elucidated, it appears that surfactant and co-surfactant used for SNEDDS formulation caused disturbance in the anatomy of small intestinal microvilli, leading to permeability change of the mucosal membrane. Based on this finding, it is suggested that drugs with narrow therapeutic range such as warfarin be administered with caution to avoid undesirable drug interaction due to large amount of surfactants contained in SNEDDS.
The anthracite coalfields of Korea are confined to the areas where sedimentary rocks of Permian and Jurassic are preserved. The Chungnam coalfield lies in the sedimentary rocks of Jurassic which belongs to the Daedong Supergroup (the Nampo group). For the property analysis of each coal seam interbeded in Daedong Supergroup, Seongju area is chosen and twelve coalseams are taken. Many standard tests have been established for optical analysis (maceral analysis, coalification degree measurement), chemical analysis (proximate, ultimate analysis) and physical analysis (ignition temperature, ash fusion temperature, hardgrove grindability index and X-ray diffraction). The Jurassic anthracite mainly consist of vitrinite and macrinite and the range of the reflectance is $R_{max}$ 5.0-6.5 which means metaanthracite rank. By the chemical composition analysis, it shows low H/C and high O/C value compare with international average value. By the physical analysis, it has very high ignition temperature ($531-584^{\circ}C$) and ash fusion temperature ($1510-1700^{\circ}C$) and very low combustion velocity (0.2-1.9 mg/min). The very wide range of the hardgrove grindability index (46-132) means that the grindability controlled mainly by the structural conditions of coal bearing strata.

A peroxidase (PD-cP; 0.47 mg/100 g leaves) was purified from autumn leaves of Phytolacca dioica L. and characterized. PD-cP was obtained by acid precipitation followed by gel-filtration and cation exchange chromatography. Amino acid composition and N-terminal sequence of PD-cP up to residue 15 were similar to that of Spinacia oleracea (N-terminal pairwise comparison showing four amino acid differences). PD-cP showed a molecular mass of approx. 36 kDa by SDS-PAGE, pH and temperature optima at 3.0 and $50.0^{\circ}C$, respectively and seasonal variation. The Michaelis-Menten constant ($K_M$) for $H_2O_2$ was 5.27 mM, and the velocity maximum ($V_{max}$) $1.31\;nmol\;min^{-1}$, while the enzyme turnover was $0.148\;s^{-1}$. Finally, the presence of $Ca^{2+}$ and $Mg^{2+}$ enhanced the PD-cP activity, with $Mg^{2+}$ 1.4-fold more effective than $Ca^{2+}$.
본 연구에서는 아이소플라본 함량이 서로 다른 한국산 4 품종(아가 8호, 우람, 다원콩 및 청자 3호)의 콩에 대하여 발아에 따른 아이소플라본 조성 및 에스트로겐 유사활성의 변화를 살펴보았다. 아이소플라본 조성은 발아에 따라 총아이소플라본, glycosides 계열, acetyl glycoside 계열 및 비배당체는 증가하였지만, malonyl glycosides 계열은 감소하였다. 품종에 따라 발아 전에는 아가 8호> 청자3호> 우람> 다원 순으로 총아이소플라본 함량이 높게 나타났고, 발아 후에는 다원 품종이 우람 품종에 비해 높게 나타났다. 에스트로겐 유사활성은 아이소플라본 함량이 높은 아가 8호 및 청자 3호가 $25{\mu}g/mL$의 농도에서 발아 후 135.34 및 121.05%의 높은 세포 증식 효과를 보였다. 또한, 에스트로겐 유사활성이 높게 나타난 아가 8호 및 청자 3호의 발아콩 추출물은 에스트로겐과 병용 처리 시 낮은 세포생존률을 보여 항에스트로겐 유사활성을 보였다. 이러한 결과로부터 아이소플라본 함량이 높은 아가 8호 및 청자 3호에서 에스트로겐 유사활성이 높게 나타나 품종에 따른 영향을 크게 받는 것으로 나타났으며, 아이소플라본 함량과 에스트로겐 유사활성간의 높은 상관관계를 확인하였다.
본 연구의 목적은 유산소 운동이 비만과 제1형 당뇨병 어린이들에게 adipokines과 염증반응에 미치는 영향을 알아보고자 실시되었다. 피험자들은 비만군(OG, n=9), 제1형 당뇨병군(DG, n=9), 그리고 정상군(NG, n=9) 어린이로 연구하였다. 모든 인자는 체중, 체지방률, 체질량지수(BMI), 최대산소섭취량(ml/kg/min), 지질 인자와 adipokines를 분석하였다. 그 결과 체중, 체지방률, BMI, 최대산소섭취량에서 OG에 비해 DG와 NG는 유의한 차이가 나타났다(p<0.05). LDL-C와 HDL-C 역시 OG에 비해 DG와 NG에서 유의한 차이가 나타났다(p<0.05). 게다가 adiponectin과 retinol binding protein (RBP)-4는 12주 운동 후에 DG와 NG에서 유의한 변화가 나타났으며 (p<0.05), OG에 비해 DG와 NG에서 유의한 차이가 나타났다(p<0.05). Monocyte chemoattractant protein (MCP)-1에서 모든 그룹은 12주 운동후 유의하게 증가하였다(p<0.05). 결론적으로 규칙적인 운동은 비만과 제1형 당뇨병 어린이에게 신체조성의 변화는 가져오지 못했으나 제1형 당뇨병 어린이의 혈관 염증을 줄여줄 수 있으며, 비만어린이에서는 대사 증후군을 보호할 수 있을 것으로 사료된다.
The radical centers detected in the reaction of metmyoglobin (MetMb) with hydrogen peroxide ($H_2O_2$) have been studied by using a spin trapping technique. A broad 5-line asymmetric electron spin resonance (ESR) spectrum, with $2A_{max}=4.07\;mT$ and $2A_{min}=2.97\;mT$, obtained after incubation of MetMb with $H_2O_2$ in the presence of a spin trap, 5,5-dimethyl pyrroline-N-oxide (DMPO) was gradually weakened with time and disappeared completely by 6 min after addition of guanidine-HCl (14 M). When a higher concentration (6 M) of the agent was added, the signal disappeared within 40 see and the DMPO/OH signal appeared immediately. Then, a new 8-line signal with similar intensities grew gradually and was fixed by 45 min, coexisting with the DMPO/OH signal. This new signal was found to be composite, consisting of two different radical species. One of the 6-line signals, with $a_N$ 1.49 mT and $a_H$ 0.988 mT, was assigned to the DMPO/phenoxyl radical adduct. The second 6-line signal with $a_N$ 1.55 mT and $a_H$ 2.22 mT was assigned to carbon-centered radical adduct. When 3,3,5,5-tetramethylpyrrolin-N-oxide (TMPO), was employed in the place of DMPO, another broad asymmetric 5-line signal was detected with $2A_{max}=3.99\;mT$ and $2A_{min}=3.04\;mT$, which is virtually identical to that obtained from the DMPO system The shape of the spectrum of the TMPO adduct changed drastically, with lapse of time resulting in a broad singlet after 40 min. The broad singlet was assigned to the porphyrin radical adduct. Incubation of globin with Fenton reagent in the presence of DMPO initially gave a DMPO/OH signal. Then, a new 12-line signal began to grow after one minute and fixed after 15 min. coexisting with the DMPO/OH signal, This 12-line signal was assigned to DMPO/phenoxyl with $a_N$ 1.47 mT, $a_{{\beta}H}$ 0.99 mT and $a_{{\gamma}H}$ 0.13 mT. A minor concentration of carbon-centered radical adduct was also detected. This radical composition is identical to that of guanidine HCl treated MetMb/DMPO/$H_2O_2$ system, indicating that the radical producing conditions are somehow common in both systems. Heme iron can be released by excess $H_2O_2$ in the MetMb/$H_2O_2$ system, providing for Fenton reagent. When MetMb was pretreated with tyrosine blocking agent, $KI_3$ the broad 5.line MetMb-derived signal was not detected in the MetMb/DMPO/$H_2O_2$ system, whereas no such effect was detected on such system of Hb in which the radical center was assigned to cysteine residue not tyrosine, indicating that tyrosine residue is a main radical center produced in the MetMb/$H_2O_2$ system Thus, the present data strongly support the previous indication that the apomyoglobin-derived radical center formed in the reaction of MetMb with $H_2O_2$ is a tyrosine residue.
본 연구는 해양사고 피해규모에 의한 위험수준을 평가하였다. 이러한 위험수준 분석은 정량적분석보다 전문가에 의한 정성적분석이 용이하다. 우리나라 RCC 및 RSC 수색${\cdot}$구조구역에 대한 위험수준 평가를 위해 본 연구에서는 전문가 지식에 기반한 퍼지이론과 계층분석법을 이용하였다. 본 연구의 퍼지이론은 퍼지 확장원리에 의한 최대최소화 합성이고, 비퍼지화는 무게중심법을 이용하였다. 그 결과 목포 수색${\cdot}$구조 구역에 위험수준이 가장 놀은 것으로 평가되어, 향후 위험수준을 경감 하기위해 많은 구조선과 구조장비가 필요 할 것으로 판단된다.
