헛개나무내의 Matrix Metalloproteinase-9 활성 억제제에 관한 연구 (A study of matrix metalloproteinase-9 inhibitor in Hovenia dulcis Thunberg)

  • 세포외 기질의 주요 단백질 구성요소들을 가수분해시켜 재편성(turnover)과 재형성(remodeling)에 핵심 역할을 하는 효소가 matrix metalloproteinases (MMPs)이다. MMPs 중에서 MMP-2와 MMP-9는 catalytic domain이 fibronectin-like domain에 의해 hemopexin-like domain 부위와 떨어져 있는 점이 다른 MMP들과 다르다. MMP-9 억제제의 개발로 간암전이를 막을 수 있다고 보고되고 있다. Hovenia dulcis Thunberg에서 MMP-9의 활성을 저해하는 물질을 분리 정제하였고, 분리된 물질에 대한 MMP-9의 활성억제 여부를 확인하였다. Ethyl acetate (EA)에 의해 용출 분리된 두개의 화합물(화합물 A, 화합물 B)과 MeOH로 용출 분리된 한 개의 화합물(화합물 C)에서 MMP-9의 활성에 저해를 보였고, $^1H$$^{13}C$ NMR, GC-MS 그리고 IR로 이들의 구조를 분석하였다. 화합물 A는 hydroxyl기와 methoxyl기로 치환된 벤젠고리를 함유하는 화합물로 catechine 계열로 추정되었으며, 화합물 B와 C는 hydroxyl기와 methoxyl기로 치환된 벤젠 고리와 분자내 carbonyl기를 갖고 있는 nobiletin 계열의 화합물로 추정되었다. 그리고 화합물 A는 MMP-9 활성을 1.0%농도에서 76% 억제하였으며, 화합물 B는 동일한 농도에서 66% 억제하는 것으로 나타났다. 그리고 화합물 C는 1.0%농도에서 71% 억제하는 것으로 나타나 화합물 A가 MMP-9의 활성 저해능이 가장 좋은 것으로 나타났다.

Expression of Matrix Metalloproteinase-2 and -13 and Tissue Inhibitor of Metalloproteinase-4 in Varicose Veins

  • Background: The relationship between the degree of expression of matrix metalloproteinases or tissue inhibitor of metalloproteinases and venous reflux remains to be investigated. Materials and Methods: Primary varicose vein tissues were obtained from 23 patients, 18 females and 5 males, aged from 19 to 73. Cephalic or basilic veins were obtained for the control group from 10 patients who underwent vascular access for maintenance hemodialysis. Two operative techniques (high ligation with stripping or endovenous laser coagulation) were used. The expression of matrix metalloproteinase-2 and 13 and tissue inhibitor of metalloproteinase-4 in the varicose vein group and control group was assessed semi-quantitatively by immunohistochemical slides stained with primary antibodies. Results: Twenty (87%) of the varicose vein group patients had greater or lesser saphenous vein diseases with reflux. The focal weak (+) stain for matrix metalloproteinases-2, and 13, and tissue inhibitor of matrix metalloproteinase-4 was dominant in the varicose vein group; the focal or diffuse strong stain (++ or +++) was prevalent in the control group. The differences were statistically significant (p<0.01). The degree of reflux and the duration of symptoms were not significantly related to the expression of MMP-13 (p=0.317 and p=0.654, respectively). Conclusion: Further study should be performed to investigate the relationship between the clinical characteristics related to venous hypertension or reflux and expression of MMPs and TIMP in varicose veins.

  • In central nervous system. matrix metalloproteinases (MMPs) are produced by neuron as well as glia and implicated in physiological events such as neurite outgrowth and myelination etc. In addition. MMPs also contribute to the pathogenesis of several CNS diseases such as multiple sclerosis, Alzheimer's disease and malignant glioma. In spite of their functional importance, little is known about the signal transduction pathways leading to the induction of MMPs in CNS. Effect of nitric oxide on the expression of matrix metalloproteinases by the UV irradiated human dermal fibroblasts

  • The production of matrix matalloproteinases(MMPs) by the UV irradiated skin fibroblast and the degradation of extracellular matrix(ECM) by these enzymes is known as one of the main reasons of photoaging. Recently, Fisher group showed that the MMP expression is mainly regulated by the mitogen-activated protein(MAP) kinas family, such as extracellular signal-regulated kinase(ERK), c-Jun amino-terminal kinase(JNK) and p38, each of which forms a signaling pathway. In this work we first examined the effect of nitric oxide (NO) on the production of MMP-1 and MMP-2 by the human dermal fibroblasts (HDFs). NO is a multifunctional messenger molecule generated from L-arginine and involved in many kinds of signaling pathway. We found that the treatment of HDF with NO donor, sodium nitroprusside (SNP) enhanced the expression of MMPs with or without UV irradiation and the treatment with nitric oxide synthase (NOS) inhibitors resulted in the significant decrease of MMPs production. From these results, we concluded that the production of MMPs by the UV irradiated HDF is regulated through the signaling pathway involving NO and cyclic GMP.

Fluorescent and bioluminescent nanoprobes for in vitro and in vivo detection of matrix metalloproteinase activity

  • Matrix metalloproteinases (MMPs) are zinc-dependent endopeptidases that degrade the extracellular matrix (ECM) and regulate the extracellular microenvironment. Despite the significant role that MMP activity plays in cell-cell and cell-ECM interactions, migration, and differentiation, analyses of MMPs in vitro and in vivo have relied upon their abundance using conventional immunoassays, rather than their enzymatic activities. To resolve this issue, diverse nanoprobes have emerged and proven useful as effective activity-based detection tools. Here, we review the recent advances in luminescent nanoprobes and their applications in in vitro diagnosis and in vivo imaging of MMP activity. Nanoprobes with the purpose of sensing MMP activity consist of recognition and detection units, which include MMP-specific substrates and luminescent (fluorescent or bioluminescent) nanoparticles, respectively. With further research into improvement of the optical performance, it is anticipated that luminescent nanoprobes will have great potential for the study of the functional roles of proteases in cancer biology and nanomedicine. 자외선으로 유도된 Hs68 섬유아세포의 노화 반응에 대한 영실추출물의 억제 효능 (Inhibitory Effect of Rosa multiflora hip Extract on UVB-induced Skin Photoaging in Hs68 Fibroblasts)

  • 극심하고 지속적인 자외선에의 노출은 정상적인 피부구조를 파괴하는 다양한 피부 광노화 과정을 야기한다. 자외선은 인체 피부에서 세포외기질의 구성성분을 분해시키는 기질 분해효소인 matrix metalloproteinases(MMPs)의 발현을 활성화시키고, 콜라겐 합성은 감소시킴으로써 피부의 탄력과 구조적 치밀도를 약화시켜 궁극적으로 피부주름을 생성한다. 본 연구에서는 이러한 피부 광노화 현상을 완화시키는 소재로서 영실의 효능을 검증하고자 하였다. 먼저 인간 섬유아세포주인 Hs68을 이용하여 영실의 세포증식 촉진효능을 확인하였다. 여기에 더해 영실이 activator protein (AP)-1 전사인자의 억제를 통해 MMP의 발현을 감소시킴을 mRNA 및 단백질 수준에서 검증하였다. 또한, 진피층을 구성하는 타입 I형 콜라겐과 표피-진피 경계부를 단단히 고정시키는 역할을 하는 타입 IV형 콜라겐 역시 영실에 의해 발현이 증가하며, 자외선에 의한 염증반응의 억제에도 영실이 효과적으로 작용하는 것을 확인할 수 있었다. 결론적으로 본 연구를 통해 영실이 자외선에 의한 피부노화와 주름생성을 효과적으로 개선할 수 있는 가능성을 가짐으로써 항노화, 항염증 및 항주름 소재로서 화장품에 응용될 수 있을 것으로 기대된다.

Inhibition of the Expression of Matrix Metalloproteinases in Articular Chondrocytes by Resveratrol through Affecting Nuclear Factor-Kappa B Signaling Pathway

  • In the present study, we tried to examine whether resveratrol regulates the expression of matrix metalloproteinases (MMPs) through affecting nuclear factor-kappa B ($NF-{\kappa}B$) in articular chondrocytes. Rabbit articular chondrocytes were cultured in a monolayer, and reverse transcription-polymerase chain reaction (RT-PCR) was used to measure interleukin-${\beta}$ ($IL-1{\beta}$)-induced gene expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5 and type II collagen. Effect of resveratrol on $IL-1{\beta}$-induced secretion of MMP-3 was investigated in rabbit articular chondrocytes using western blot analysis. To elucidate the action mechanism of resveratrol, effect of resveratrol on $IL-1{\beta}$-induced $NF-{\kappa}B$ signaling pathway was investigated in SW1353, a human chondrosarcoma cell line, by western blot analysis. The results were as follows: (1) resveratrol inhibited the gene expression of MMP-3, MMP-1, MMP-13, ADAMTS-4, and ADAMTS-5, but increased the gene expression of type II collagen; (2) resveratrol reduced the secretion of MMP-3; (3) resveratrol inhibited $IL-1{\beta}$induced activation (phosphorylation) of inhibitory kappa B kinase (IKK), and thus phosphorylation and degradation of inhibitory kappa $B{\alpha}$ ($I{\kappa}B{\alpha}$); (4) resveratrol inhibited $IL-1{\beta}$-induced phosphorylation and nuclear translocation of $NF-{\kappa}B$ p65. This, in turn, led to the down-regulation of gene expression of MMPs in SW1353 cells. These results suggest that resveratrol can regulate the expression of MMPs through affecting $NF-{\kappa}B$ by directly acting on articular chondrocytes.

Avenanthramide C as a novel candidate to alleviate osteoarthritic pathogenesis

  • Osteoarthritis (OA) is a degenerative disorder that can result in the loss of articular cartilage. No effective treatment against OA is currently available. Thus, interest in natural health products to relieve OA symptoms is increasing. However, their qualities such as efficacy, toxicity, and mechanism are poorly understood. In this study, we determined the efficacy of avenanthramide (Avn)-C extracted from oats as a promising candidate to prevent OA progression and its mechanism of action to prevent the expression of matrix-metalloproteinases (MMPs) in OA pathogenesis. Interleukin-1 beta (IL-1β), a proinflammatory cytokine as a main causing factor of cartilage destruction, was used to induce OA-like condition of chondrocytes in vitro. Avn-C restrained IL-1β-mediated expression and activity of MMPs, such as MMP-3, -12, and -13 in mouse articular chondrocytes. Moreover, Avn-C alleviated cartilage destruction in experimental OA mouse model induced by destabilization of the medial meniscus (DMM) surgery. However, Avn-C did not affect the expression of inflammatory mediators (Ptgs2 and Nos) or anabolic factors (Col2a1, Aggrecan, and Sox9), although expression levels of these genes were upregulated or downregulated by IL-1β, respectively. The inhibition of MMP expression by Avn-C in articular chondrocytes was mediated by p38 kinase and c-Jun N-terminal kinase (JNK) signaling, but not by ERK or NF-κB. Interestingly, Avn-C added with SB203580 and SP600125 as specific inhibitors of p38 kinase and JNK, respectively, enhanced its inhibitory effect on the expression of MMPs in IL-1β treated chondrocytes. Taken together, these results suggest that Avn-C is an effective candidate to prevent OA progression and a natural health product to relieve OA pathogenesis.

비소세포폐암에서 Matrix Metalloproteinase(MMPs)-2, 9와 Tissue Inhibitor of Metalloproteinase(TIMPs)-1, 2의 발현과 생존율과의 관계 (The Relationship Between Expression of Matrix Metalloproteinases(MMPs)-2, 9 and Tissue Inhibitors of Metalloproteinase(TIMPs)-1, 2 and Survival Time in Resected Non-Small Cell Lung Cancer)

  • 연구배경 : MMPs는 종양 전이의 주된 장벽인 기저막과 세포외 기질을 분해하여 암세포의 침습 및 전이에 주된 역할을 하며, TIMP는 MMP에 의한 기저막 단백질 분해를 억제하는 인자로 알려져있다. MMP의 발현은 임상 병기가 진행할수록 증가하며 주위 림프절 전이와 관계가 있으며, TIMP의 발현은 주위 림파절로 전이가 일어나면 감소 또는 증가하는 것으로 알려져 있다. 대상 및 방법 : 비소세포폐암으로 진단 받고 근치적 절제술을 시행 받은 74명 환자를 대상으로 paraffin에 보관된 조직에서 면역조직화학염색법을 이용하여 MMP-2, 9, TIMP-l, 2 항체로 발현을 검색하였으며, 발현에 따른 생존율을 Kaplan-Meier, Log-rank로 검색하였다. 결 과 : MMP-2, 9의 발현은 각각 25/74례 (34%), 19/74(26%)였고, TIMP-l, 2의 발현은 각각 27/74(36%), 32/74(43%)였다. 임상 병기에 따른 MMP-2, 9 발현은 병기의 진행에 따라 유의하게 증가하였고, TIMP-2는 유의하게 감소하였다. 임상 림프절의 전이에 따른 발현은 MMP-2, 9에서만 전이의 진행에 따라 유의하게 증가하였다. MMP-2 발현에 따른 중간 생존기간은 양성군이 20개월, 음성군이 34개월로 양성군이 유의하게 낮았으며, TIMP-2은 34개월, 18개월로 양성군이 유의하게 높았다. MMP-2/TIMP-2에 따른 중간 생존기간은 음성/양성이 유의하게 높았다. 결 론 : MMP-2, 9 발현은 종양의 진행과 림프절의 전이에 관여할 것으로 사료되며, MMP-2와 TIMP-2의 발현은 역비레 관계가 있다.
