Antibacterial Activity of the Honey Bee Venom against Bacterial Mastitis Pathogens Infecting Dairy Cows

  • The antibacterial activity of Korean honey bee venom (KBV) was examined against seven major bacterial mastitis pathogens, Enterococcus faecium, Escherichia coli, Methicillin resistant Staphylococcus aureus (MRSA), Staphylococcus aureus, Streptococcus intermedius, Streptococcus oralis and Streptococcus uberis isolated from infected mammary quarters of cows. Seven bacterial mastitis pathogens were studied for antibacterial activity of the KBV by disc diffusion assay, minimal inhibitory concentrations (MIC) and bacterial count in milk samples. The KBV showed activity against Ent. faecium, E. coli, MRSA, Staph. aureus, Strep. intermedius. The order of susceptibility of the bacteria against the KBV was Staph. aureus>MRSA>E. coli>Strep. intermedius>Ent. faecium>Strep. oralis>Strep. uberis. The MIC against Staph. aureus, MRSA and E. coli were stronger effect as compared with standard drug. The effect of the KBV ($100{\mu}g\;ml^{-1}$) on the viability of Ent. faecium, E. coli, MRSA, Staph. aureus, Strep. intermedius, Strep. oralis and Strep. uberis in milk differed significantly with each other within 12 h incubation period. The results indicate that KBV has significant antibacterial effects against major bacterial mastitis bacteria, Ent. faecium, E.coli, MRSA, Staph. aureus, Strep. intermedius. Results of the study indicate the potential use of KBV as alternative to antibiotic therapy. Further investigations are needed though to confirm its efficacy and its effects on the animals.

  • 본 연구에서는 원유와 사일리지, 전통발효식품 등으로부터 유방염 원인균에 대한 항균성을 가지는 유산균을 분리하였고, 이들 중 원유에서는 79, 91-3 균주와 전통발효식품에서는 SA131, NK24, NK34, 32, 44-1 균주, 사일리지에서는 253, 261, 262, 263, 265 균주, 닭분변에서 B32, C23 균주에서 유방염 원인균에 대한 비교적 높은 항균력을 확인할 수 있었다. 이를 통해 새로운 항생제 대체물질의 가능성을 확인할 수 있었다.

Successful treatment of recurrent subclinical mastitis in cows caused by enrofloxacin resistant bacteria by means of the sequential intramammary infusion of enrofloxacin HCl-2H2O and ceftiofur HCl: a clinical trial

  • Background: Recurrent subclinical mastitis (RScM) due to resistant bacteria has low clinical and bacteriological cure rates, often requiring the culling of cows. The sequential intramammary administration of enrofloxacin hydrochloride-dihydrate (enro-C) followed by ceftiofur HCl may be useful for treating these cases. Objectives: This study assessed the bacteriological and clinical cure-efficacies of the sequentially intramammary administration of enro-C, followed by ceftiofur HCl to treat RScM in Holstein/Friesian cows. Methods: This trial was conducted in a herd with a high prevalence of RScM, and 20 Holstein/Friesian cows were included: 45% suffering subclinical mastitis and 38.9% of the mammary quarters affected. Twenty-nine bacterial isolates in vitro resistant to enro-C were obtained (coagulase-negative Staphylococcus spp, 55.2%; Staphylococcus aureus, 27.6%; Escherichia coli, 6.9%; Streptococcus uberis, 6.9%; Corynebacterium bovis, 3.4%). Polymerase chain reaction-isolated the following genes linked to enro-C resistance: chromosomal (gyrA) and plasmid (aac(6')-lb-cr). The treatments were as follows: twice-daily intramammary infusions of enro-C (300 mg/10 mL) for 5 days. Cows clinically considered treatment failures were also treated with intramammary ceftiofur (125 mg/10 mL, twice daily for 5 days. The clinical and bacteriological cure rates were carried out when completing each treatment phase and at 14 and 21 days, aided by a California mastitis test, somatic cell count, and failure to identify the initially causative bacteria. Results: Enro-C achieved 65% clinical and bacteriological cure rates, and 100% cure rates were obtained after the rescue treatment with ceftiofur HCl. Conclusions: Outstanding clinical and bacteriological cure rates in cows affected by RScM were achieved with the consecutive intramammary infusions of enro-C, followed by ceftiofur HCl.

유우유방의 유방염에 대한 자연방어기전 (Immunophysiological Defense Mechanism of the Bovine Udder on Mastitis A Review)

  • This paper reviews the mechanisms effecting host defense in the mammary gland and assesses their possible in preventing of bovine mastitis. The streak canal is the first line of defense against invading mastitis pathogens, providing a physical barrier and antibacterial substances. The milk leukocytes are a second defense line by ingesting pathogens breached the streak canal by multiplication, physical passage, and propulsion during milking. Leukocytosis in milk and enhancement of the phagocytic defense machanisms of the udder were accomplished by inserting intramammary devices. Milk antibodies serum derived and synthesized in mamma tissue aggregate and opsonise bacteria, agglutinate and neutralise toxins, and inhibit. binding of bacteria to epitherial surfaces. Vaccination generally has been unsuccessful because protection is not absolute, but immunization is useful in controlling specific pathogens. Immunostimulant to enhance locally the protective nature of antibody-producing plasma cells concentrated in internal teat end tissue may be effective in reducing the occurrence of infection, but ineffective in preventing intramammary infections.

항균제내성의 검사요령과 임상적응용 (Technique of Drug Tolerance Test for Selection of Antibacterial Agents and It′s Clinical Value)

  • The author carried out bovine mastitis test by California Mastitis test(CMT) for the milk from dairy cows in the surburbs of Taejeon. In order to select prefer commercial therapeutic antibacterial agents for mastitis treatment, Staphylococcus aureus were isolated from the CMT positive milk and the strains were tested for the tolerance test to the agents. The results obtained were summarized as follows : 1. The tolerance test appeared graduate tolerance in cases with continuously repeat of therapeutic agents. 2. The antibacterial agents revealed strong tolerance were D-and E-ointments. 3. The antibacterial agents revealed suspect tolerance were A-and F-ointments and ampicillin, penicillin, kanamycin and streptomycin. 4. The antibacterial agents revealed non or rare tolerance were B-and G-ointments and chloramphenicol, erythromycin and oxytetracycline. 5, It is concluded that the use of 3-day-interval in turn with antibacterial agents selected by tolerance test may be beneficial.

Candida krusei에 의한 유우유방염(乳牛乳房炎) (A Case of Bovine Mastitis Caused by Candida krusei)

  • This experiment was undertaken to determine the causative agent of bovine mastitis induced by yeast-like fungi and to know its therapeutic effect. California mastitis test, somatic cell counting and pH titration as well as cultural examination were performed continuously for the quarter milk and the feces obtained from the diseased cattle from November, 1979 to February, 1980. As a result, the causative agent was identified as Candida krusei and intramammary infusion of Nystatin was effective in the udder. Candida krusei was isolated from the feces.

젖소 유방염에서 16S rRNA 파이로시퀀싱을 이용한 우유 내 마이크로바이옴의 동정과 난백의 항균효과 (Identification of microbiome with 16S rRNA gene pyrosequencing and antimicrobial effect of egg white in bovine mastitis)

  • Bovine mastitis is an important microbial disease in the dairy industry. We investigated the frequencies of bacterial pathogens in 62 farms and pathogen antibiotic resistance from mastitis samples (n = 748). We tested the antimicrobial activity of chicken and duck egg white and lysozyme purified from chicken egg white. Moreover, we compared the microbiomes of normal and mastitic raw milk obtained by 16S rRNA gene pyrosequencing and culture methods. The results showed that the frequencies of Gram-positive pathogens (Enterococcus faecalis 37% and Staphylococcus aureus 36%) were higher than that of a Gram-negative pathogen (Escherichia coli 15%). Resistance frequencies to ampicillin and norfloxacin were lowest in Staphylococcus aureus (21%), Enterococcus faecalis (23%), and Escherichia coli (33%), and the antimicrobial activity of chicken egg white was higher than those of lysozyme and duck egg white. Pyrosequencing results revealed clear differences between the microbiomes of mastitic and normal raw milk samples and revealed a slightly similar, but clearly different, composition of pathogens compared to that from the culture method. Thus, pyrosequencing may be useful for elucidating changes in microbiomes during mastitis progression and treatment. A chicken egg white and antibiotic combination may help with mastitis treatment; however, further studies are needed.

임상형 유방염을 가진 홀스테인(Holstein) 소에 대한 은 이온의 적용 연구 (Application of Silver Ion for Clinical Mastitis in Holstein Cows)

  • 유방염은 병원성 미생물들에 의한 유선 내에 염증이 생기는 질환으로, 유방 조직과 우유 내 체세포수 (SCC)와 세균학적 분석이 유방염 진단을 위해서 사용된다. 본 연구에서는 임상형 유방염이 걸린 홀스테인 소의 유선 내로 은 이온(20 ${\mu}g$/ml)을 투입하고, 유방염의 임상 증상의 변화와 우유 내 somatic cell count, 소 혈청 알부민과 lactoferrin 등과 같은 유방염 관련 인자들을 조사하였다. 은 이온의 유선 내 투입은 유방의 부종과 견고함 등의 염증 증상을 완화시켰으며, 우유의 응고와 체세포수를 감소시켰다. 또한, 우유 내 소 혈청 알부민과 lactoferrin의 농도가 은 이온의 투입에 의해 감소되었다, 그러나 임상형 유방염이 걸린 소의 유선 내 은 이온의 투입은 착유된 우유의 양과 혈액 내 백혈구와 적혈구 수를 변화시키지는 않았다. 이러한 결과는 유방염이 걸린 소에 은 이온을 유선 내로 투입 함으로서 염증과 세균의 활성을 감소시킨다는 것을 보여주며, 은 이온을 임상형 유방염의 치료에 적용할 수 있는 가능성을 보여 주는 것이다.

  • Mastitis is an inflammatory condition of the mammary gland, most often caused by bacterial infections, resulting in significant economic losses to the dairy industry. Antimicrobial resistance has been of great concern because of the extensive clinical use of antibiotics. For this reason, the development of new compounds as an alternative treatment to bovine mastitis is needed. Bee venom has been widely used as an oriental treatment for several inflammatory diseases and bacterial infections. The aim of the present study was to evaluate the antimicrobial activity of bee venom on bacteria isolated from bovine mastitis. A total of 107 isolates from bovine mastitic milk samples collected in 2019 and 2020 in Jeonbuk province. All bacterial isolates were tested for susceptibility to bee venom of the honey bee (Apis mellifera). In order to obtain comprehensive antibacterial activities of the bee venom, we measured the minimal inhibitory concentration (MIC) of the bee venom against bacterial strains. Bee venom showed significant inhibition of bacterial growth of Gram-negative bacteria Citrobacter spp., Escherchia coli, Klebsiella spp., Pseudomonas spp., Serratia spp. and Raoultella with MIC values of 96, 81, 72, 230, and 85 ㎍/mL, respectively, and Gram-positive bacterial Enterococcus spp., Staphylococcus spp. and Streptococcus spp. with MIC values of 29, 21 and 16 ㎍/mL, respectively. The results indicated that the MIC values were different depending on the bacterial strains, and those of Gram-positive bacteria were lower than those of Gram-negative bacteria for bee venom. These findings suggested that bee venom could be an effective antimicrobial treatment for bovine mastitis; however, further research is necessary to evaluate the mechanism underlying the antimicrobial action, its effectiveness/safety in vivo and effective application for therapeutic use.

Antimicrobial resistance studies in staphylococci and streptococci isolated from cows with mastitis in Argentina

  • Background: Staphylococcus aureus and Streptococcus agalactiae are the main cause of clinical mastitis in dairy cattle in Argentina, whereas coagulase-negative staphylococci (CNS) and environmental streptococci are the main cause of subclinical mastitis. Bacteria isolated from infected animals show increasing antimicrobial resistance. Objectives: This study aims to determine the antimicrobial resistance of staphylococci and streptococci isolated from milk with mastitis, and to genotypically characterize the methicillin-resistant (MR) staphylococci. Methods: Isolation was performed on blood agar and identification was based on biochemical reactions. Antimicrobial susceptibility was according to the Clinical and Laboratory Standards Institute guidelines. The antimicrobial resistance genes, SCCmec type and spa type were detected by the polymerase chain reaction method. Results: We isolated a total of 185 staphylococci and 28 streptococci from 148 milk samples. Among the staphylococcal isolates, 154 were identified as CNS and 31 as S. aureus. Among the 154 CNS, 24.6% (n = 38) were resistant to penicillin, 14.9% (n = 23) to erythromycin, 17.5% (n = 27) to clindamycin, 6.5% (n = 10) to cefoxitin and oxacillin. Among the S. aureus isolates, 16.1% (n = 5) were resistant to penicillin, 3.2% (n = 1) to cefoxitin and oxacillin (MRSA). Six MR isolates (5 CNS and 1 MRSA) were positive to the mecA gene, and presented the SCCmec IVa. The MRSA strain presented the sequence type 83 and the spa type 002. Among the 28 streptococcal isolates, 14.3% (n = 4) were resistant to penicillin, 10.7% (n = 3) to erythromycin and 14.3% (n = 4) to clindamycin. Conclusions: The present findings of this study indicate a development of antimicrobial resistance in main bacteria isolated from cows with mastitis in Argentina.
