Contamination and Bioaccumulation of Butyltin Compounds Inside Jeju Harbor of Jeju Island, Korea

  • Butyltin compounds in seawater, sediments and organisms inside Jeju Harbor of Jeju Island, Korea, were quantitatively determined to assess the extent of contamination and to evaluate the bioaccumulation in Thais clavigera (gastropod), Liolophura japonica and Cellana nigrolineata. In addition, imposex in T. clavigera was used to assess the sublethal effect of tributyltin compounds and its usage as a specific biomarker for these compounds was also studied. The main species in seawater was tributyltin (TBT) in March and June, monobutyltin (MBT) in August, but in sediments and organisms it was dibutyltin (DBT) and TBT, respectively, irrespective of survey time. TBT, the most toxic to aquatic organisms among BTs, was found at concentrations which were sufficient to have a serous effect on the sensitive organisms upon chronic exposure. The high correlations (r>0.7) between BTs indicated that DBT and MBT were mainly degraded from TBT based on antifouling paints and their sources were negligible. The sedimentary organic matters have little influence on the distribution of BTs in marine environment and the lipid content in T. clavigera showed a significant correlation with TBT concentrations. Measurement of imposex T. clavigera was expected to be a very helpful tool for preliminary survey of BTs prior to trace analysis of those.

농축수산문 식품원료 및 그 가공식품에 대한 Listeria 균주의 오염실태조사와 Listeriosis 발생억제방법 (Outbreaks and Control of Listeriosis Attributed to Agricultural, Marine and Animal Husbandry Products)

  • The major purpose of the present study is to syrvey vegetables, meats, seafoods, processed foods and imported foods for the presence of Listeria spp. and to prevent listeriosis caused by the contamination of Listeria monocytogenes. Listeria spp. was not found in vegetables and processed foods. The optimum growth condition of isolates indentified as Listeria monocytogenes was pH 7.0 and 37$^{\circ}C$. the antimicrobial effect of grapefruit seed extract(GFSE) was observed in the level of more than 100ppm by disk method. When 1ml(2.5$\times$106 CFU/ml) of Listeria monocytogenes was inoculated and incubated for 3 days at 3$0^{\circ}C$, the total cell number of the organism was 4.5$\times$109 in the control, 7.2$\times$103 in 100 $\mu\textrm{g}$/ml of GFSE medium, and 3.5 $\mu\textrm{g}$/ml of GFSE medium. Direct visualization of microbial cells by using both transmission electron microscope and scanning electron microscope showed microbial cell membrane the function of which was destroyed by treating with the dilute solutions of GFSE. It could be confirmed that GFSE completely inhibited the growth of the test strain of Listeria monocytogenes.

갈조류 감태 (Ecklonia cava Kjellman)의 대량양식을 위한 가이식 및 양성 조건 (Nursery and Main Culture Conditions for Mass Cultivation of the Brown Alga, Ecklonia cava Kjellman)

  • The mass cultivation of Ecklonia cava Kjellman was studied as a potential biomass source for the extract industry in Korea. Experiments were conducted to investigate the optimal conditions for artificial seed production and mass cultivation of this species. Maximum growth and young thalli development in the nursery culture area occurred at 2 m depth, whereas maximum growth of thalli in the main culture area occurred at 1 m depth. Production of E. cava was between 2.6 and 3.6 kg wet wt. $m^{-1}$ after depth control and removal of fouling organism, etc. The relationship between optimal water depth for culture and underwater irradiance during the E. cava cultivation was calculated as: y = -0.718x + 8.042 ($r^2$=0.976). The growth rates achieved in this trial indicate that E. cava cultures could produce and supply sufficient biomass.

Bacillus subtilis Fermentation for Enhancement of Feed Nutritive Value of Soybean Meal

  • In order to increase the nutritional quality of soybean meal (SBM) as an animal feed, Bacillus subtilis TP6, a previously isolated strain from an Indonesian traditional fermented soybean food, Tempeh, was used as a starter organism for solid-state fermentation. In the pre-treated SBM with water content of 60% (v/w), B. subtilis TP6 was grown to a maximum viable cell number of $3.5{\times}10^9CFU/g$. Compared to control, crude protein in Bacillus fermented SBM was increased by 16%, while raffinose, stachyose, and trypsin inhibitors were reduced by 31, 37, and 90%, respectively. The Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis showed that proteins in the fermented SBM were remarkably hydrolyzed into smaller molecular masses, resulting in a decrease in large sized proteins. Our data suggested that B. subtilis fermentation could increase the nutritive value of SBM through reduction of anti-nutritive factors and improvement of protein quality by hydrolysis of soy protein. In addition, B. subtilis TP6 produced a functional ingredient, poly-${\gamma}$-glutamic acid which has various health benefits.

Monitoring Benthic AIgal Communides:A Comparison of Targeted and Coefficient Sampling Methods

  • Choosing an appropriate sample unit is a fundamental decision in the design of ecological studies. While numer-ous methods have been developed to estimate organism abundance, they differ in cost, accuracy and precision.Using both field data and computer simulation modeling, we evaluated the costs and benefits associated with twomethods commonly used to sample benthic organisms in temperatc kelp forests. One of these methods, theTargeted Sampling method, relies on different sample units, each "targeted" for a specific species or group ofspecies while the other method relies on coefficients that represent ranges of bottom cover obtained from visual esti-mates within standardized sample units. Both the field data and the computer simulations suggest that both meth-ods yield remarkably similar estimates of organisnm abundance and among-site variability, although the Coefficientmethod slightly underestimates variability armong sample units when abundances are low. In contrast, the twomethods differ considerably in the effort needed to sample these communities; the Targeted Sampling requiresmore time and twice the persormel to complete. We conclude that the Coeffident Sampling metliod may be bettcrfor environmental monitoring programs where changes in mean abundance are of central conccm and resources arelimiting, but that the Targeted sampling methods may be better for ecological studies where quantitative reIation-ships among species and small-scale variability in abundance are of central concern.

넙치 치어의 사료 내 myo-inositol 요구량 (Myo-inositol Requirement in Diets for Juvenile Olive Flounder (Paralichthys olivaceus))

  • 본 연구는 넙치 치어에 있어서 수용성 비타민인 myo-inositol의 사료 내 적정 요구량을 평가하고, 결핍 시 어류에 미치게 되는 생리적 영향을 조사하고자 수행 되었다. 실험에 사용된 어류는 평균무게가 1.22 g인 넙치 초기치어를 사용하였으며, 총 15개의 35 L 원형수조에 각 수조당 48 마리씩(3반복구) 무작위 배치하였다. 반 정제 사료원을 기초로한 총 5개의 실험사료는 52%의 조단백질과 18.3 MJ/kg diet의 에너지함량을 갖도록 조성되었고 myo-inositol 함량을 각각 0, 0, 400, 800, 1600 mg/kg diet으로 기초사료에 첨가하였다(M0, M0+, M400, M800, M1600). 실험사료 중 M0+ 사료는 항생제인 tetracycline hydrochloride 및 (SIGMA, USA)를 실험사료에 0.4%로 첨가함으로써, myo-inositol이 넙치의 장내 미생물들에 의하여 합성되는지를 알아보기 위해 설계되었다. 26주간의 사료공급 실험결과, 사료 내 myo-inositol 첨가에 의한 어류의 성장률 결과들에서 유의적인 차이가 나타났으며, myo-inositol 결핍사료를 섭취한 실험어류에서 성장지연과 같은 결핍증상을 보였다. Hematocrit과 Hemoglobin을 측정한 혈액분석과 간 지방함량 분석에서는 실험구 사이에서 차이가 나타나지 않았으나, 전어체 지방분석 결과에서는 단지 M0와 M400 사이에서 유의적인 차이를 나타내었다. 초기 넙치 치어의 myo-inositol 요구량은 성장률을 기초로 한 broken-line regression model (Robbins, 1986)에서 반 정제사료를 기초로 하였을 때 800 mg/kg로 결정되었다.

$CO_2$해양처리를 위한 액적 거동 시뮬레이션 기초연구 (Numerical Study of a Droplet Movement for the Ocean $CO_2$ Sequestration)

  • 2004년 11월 러시아의 교토의정서 비준에 의하여 2005년 2월에 교토의정서의 발효를 앞두고 있다. 따라서 1차 감축대상국은 2008년부터 2012년까지 1990년 대비 각 국가당 설정된 GHG(Green House Gas)배출량을 감축해야하는 상황에 처해 있다. 본 논문에서는 대기 중에 뿜어내는 GHG중 가장 많은 비율을 차지하는 CO₂를 심해에 친환경적으로 처리하는 기술로서 해양격리기술에 대해 소개한다. 본 해양격리기술은 4개의 주요단계로 구분할 수 있는데, 해양수송기술, 해양분사기술, 해양저장효율평가기술, 해양처리생태영향 평가기술등으로 나눌 수 있다. 본 논문에서는 CO₂의 해양 분출시에 초기희석율에 영향을 미치는 해양분사기술 중, 액포의 거동과 용해에 관한 기초 연구를 수행하였다. 특히 본 연구에서는 전산유체역학 기법 (Computational Fluid Dynamics)을 통해 해수를 연속상(Continuous phase), 액적을 분산상(Disperse phase)으로 한 이상(Two-phase) 유체를 3차원 이동형 비구조 격자와 유한체적법을 이용하여 상 경계면이 거동하면서 거동에 따른 용해도의 변화를 추정하였다 따라서, 본 연구는 액적의 거동과 용해도의 유체역학적 관계를 밝히는 데 기초적 자료가 될 것으로 판단된다.

직접 전기분해식 선박평형수 처리장치 개발과 시험에 관한 연구 (Development of the Electrolysis Ballast Water Treatment System and Test)

  • 선박의 항해 안전성을 유지하기 위하여 평형수 탱크에 주입 배출되는 선박평형수는 그 안에 포함되어 있는 각종 수중생물로 인하여 지역 해양 환경에 부정적인 영향을 주고 있다. 국제해사기구(IMO)는 선박평형수를 통한 수중생물의 이동을 막기 위해, 2004년 선박평형수와 침전물 통제 및 관리를 위한 국제협약을 채택하고 2016년 9월에 발효하여, 2017년 9월 이후 정기검사가 도래하는 모든 선박은 선박평형수 처리장치를 설치하도록 하였다. 선박평형수 처리 방식에는 활성물질을 이용하여 처리하는 전기분해식, 오존식, 약품식과 물리적인 처리방식인 필터, 자외선식 등으로 나누어 지며, 두 가지 방식을 혼용하여 사용하기도 한다. 일반적으로 비용과 효율 면에서 전기분해방식이 우수한 것으로 알려져 있다. 본 논문에서는 직접식 전기분해 선박평형수 처리장치의 기본 원리, 구성 요소, 육상 시험 내용을 고찰하였다. 육상시험은 정부시험 시설이 설치되어 있는 KIOST 거제분원에서 $300m^3/h$ 처리 용량의 장치로 수행하였다. 이 육상시험을 통해 직접식 전기분해 선박평형수 처리장치가 IMO에서 제시하고 있는 기준을 만족하고 다른 방식에 비해 효율적인 것을 확인하였다.

남해 연안해역의 부영양화 2. 남해 연안해역의 저질 및 수질의 부영양화 실태 (Eutrophication in the Namhae Coastal Sea 2. The Aspects of Eutrophication of Bottom Mud and Surface Seawater in the Namhae Coastal Seas)

  • The aim of this paper is to grasp eutrophication aspects in Namhae coastal seas, statistically analyzing existing data for their surface seawater and bottom mud. A pollution level(ignition loess) of bottom mud, on the whole, trended to increase as moving the coastal sea around Mokpo-Wando toward the east(Gyeongnam Namhae coastal seas). Especially, the pollution level(ignition loss=10.5%) of bottom mud for the coastal sea around Tongyeong-Keoje-Gosung was similar to that(10.3%) for the coastal sea around Masan-Jinhae, whose coastal marine pollution was the severest in Namhae coastal seas. It indicates that large amounts of pollutant from aqualculture facilities have been, thus far, accumulated on the coastal sea around Tongyeong-Keoje-Gosung, considering there was no significant inflow of sewage and industrial wastewater into this coastal sea. A COD, T-N, and T-P level of surface seawater, on the whole trended to increase as moving the coastal sea around Mokpo-Wando toward the east(Gyeongnam Namhae coastal seas). A COD level appeared to be the second grade of coastal water quality over the entire year throughout all Namhae coastal seas A T-N level exceeded the third grade of coastal water quality throughout all Namhae coastal seas except the coastal sea around Mokpo-Wando. Especially, a T-N level exceeded as many as three and six times over the third grade of coastal water quality in the coastal sea around Tongyeong-Keoje-Gosung and Masan-Jinhae, respectively. A T-P level appeared to be the second grade of coastal water quality in the coastal sea around Mokpo-Wando and the third grade of coastal water quality in the coastal sea around Yosu-Narnhae and Tongyeong-Keoje-Gosung, while it exceeded as many as two times over the third grade of coastal water quality. A degree of eutrophication of the surface seawater was 1.5 in the coastal sea around Mokpo-Wando and 11.9 In the coastal sea around Tongyeong-Keoje-Gosung, gradually increasing as moving toward the east(Gyeongnam Narnhae coastal seas). It sharply increased to 146.1 in the coastal sea around Masan-Jinhae. Because the degree of eutrophication throughout all Namhae coastal seas exceeded 1, a red tide organism could pose a possibility of proliferation at any place of Namhae coastal seas if other requirements were satisfied. It indicates that a red tide may move to another place once a red tide breaks out at a place of Namhae coastal seas.

국내 연근해 및 환자로부터 분리된 vibrio vulnificus의 세균학적 특징

  • 국내 연안의 어패류, 해구 및 환자 등으로부터 Vibrio vulnificus를 분리동정하여 생화학적 성상, 생장조건, 혈천형, 약제 감수성 시험 등 세균학적 특성을 밝혔다. V. vulnificus 균의 생화학적 성상은 유당을 발효(95%)하고 서당은 발효 (4%)않은 균주가 대부분이었으며, ornithine 발효능, gelatine 액화능, mannitol 분해능 등이 다른 연구자들과 동일한 결롸를 나타내지 않았다. 따라서 이들 성상이 V. vulnivicus 동정에 그리 유용하지 않다. o 혈청형은 자연계 분리균주의 경우 o2 혈청형이 제일 많았고 (24%) 환자 분리균주의 경우 04 균(42%)이 제일 많았다. V. vulnificus 의 최적 생상조건은 염농도는 306%였고, 온도는 25-35.deg.C, pH는 8로 나타났다. V. vulnificus균은 chloramphenicol, erythromycin, tetracycline, ampicilin 에 높은 감수성을 지니고 있었고 gentamycin 에 대하여는 내성을 가진 것으로 나타났으며, 자연계 분리균주가 환자 분리균주보다 이들 항생제에 더 높은 감수성을 지니고 있었다. chelating agent EDTA가 V. vulnificus에 대하여 생장억제효과가 있음이 밝혀졌으나 치사적으로 작용치는 않았다.
