Field tests were carried out with five different mesh sizes (86.6, 101.0, 121.2, 137.7, 151.5mm) to study the relative efficiency of monofilament and multifilament gill net for Marbled sole, Pleuronectes yokohamae, in the western sea of Korea, 2007-2009. The relative efficiency was analyzed by length distribution, catch in number, catch in weight and weight per individual for Marbled sole. Statistical T-test was done to verify the efficiency between two gears. In the field tests, the total body length range, catch in number and weight of Marbled sole was 15.8-48.2cm, 728 and 254,939g, respectively. During the field tests, Marbled sole less than 15cm length which is prohibition size of fishing was not caught, but the caught number of Marbled sole less than 19.5cm length which is first maturity length was 13 (1.8%) and the caught number of Marbled sole more than 19.5cm was 715 (98.2%). Catches of monofilament gill net was average 1.4 times more efficient than multifilament gill net. The weight per individual and total length by mesh sizes in the two gears tended to gradually increase according to mesh size increasing. But there was little difference of weight per individual and total length by mesh sizes between two gears.
The physico-chemical properties of fresh milt of marbled sole, Limanda yokohamae, brown sole, Limanda herzensteini, starry flounder, Platichthys stellatus and olive flounder, Paralichthys olivaceus among flatfishes, and the fine structure of their cryopresserved spermatozoa were investigated. The highest concentration of sperm among these four species was $3.60\pm1.35\times10^{10}/mL$ in marbled sole. Osmolality and pH of seminal plasma in four flatfish species were approximately 330 mOsm/kg and 7.6-8.1, respectively. Seminal plasma compositions showed interspecific differences. The sperm heads of marbled sole, brown sole and starry flounder were ellipsoidal and that of olive flounder was round. The numbers of mitochondria of these four species were eight in marbled sole, seven in brown sole and starry flounder, and six in olive flounder. Cross-sectional view of flagellum showed typical 9+ 2 structure in all species. Most of sperms cryopreserved with a proper method had no visible ultrastructural changes after freeze-thawing, compared with the fresh sperm, but in a few cases, swelling of their heads and midpiece regions were observed.

The proper conditions for cold storage of milt in four species of flatfish were investigated. Various diluents were tested for the preservation of marbled sole, brown sole, starry flounder and olive flounder milt at $1\pm0.5^{\circ}C$ for 7 days. The most effective diluent was $1\%$ NaCl for marbled sole and Stein's extender for brown sole, starry flounder and olive flounder. After 7 days of storage in different dilution ratios (milt: diluent), marbled sole milt diluted with 1:3-10 and brown sole, starry flounder and olive flounder milt diluted 1:1-10 maintained good sperm activity index (SAI). SAI was affected by storage volume and presence of antibiotic. SAI was lower after 7 days storage in the volume of 1.5 mL than in 0.5 or 1.0 mL. SAI after 29 days storage was markedly enhanced by addition of antibiotic in marbled sole (400 ppm gentamicin, 400 ppm neomycin, 200-400 ppm streptomycin), brown sole (600 ppm gentamicin), starry flounder (1,000 ppm gentamicin) and olive flounder (200-1,000 ppm gentamicin, 200-400 ppm neomycin, 200-1,000 ppm streptomycin).

Vitellogenin (VTG) was purified from the blood plasma of estradiol-17$\beta$ ($E_2$)-treated male marbled sole (Limanda yokohamae) using gel filtration and anion exchange chromatography. The purity of the marbled sole VTG (msVTG) was confirmed by polyacrylamide gel electrophoresis (SDS-PAGE) and N-terminal amino acid sequencing. The purified msVTG was used to produce monoclonal and polyclonal antibodies in mice and rabbits, respectively, and the specificity of the polyclonal antisera for msVTG was confirmed by Western blot analysis. The antibodies crossreacted with a protein of molecular mass approximately 160 kDa in the plasma samples of mature female marbled sole. No cross-reactivity was observed with the plasma of male fish. A direct non-competitive sandwich enzyme-linked immunosorbent assay (ELISA) was developed using the monoclonal anti-msVTG and polyclonal anti-msVTG antibodies, with purified msVTG as the standard protein. The values of the intra- and inter-assay variations were within the ranges of $8.l-9.8\%$ and $8.5-12.2\%$, respectively. The sensitivity was about 0.3 ng/mL. Serial dilutions of plasma from mature female sole reacted with the msVTG-antibodies in the sandwich ELISA, whereas the plasma from male fish did not. The results indicate that the maturation status of female marbled sole can be identified using a sandwich ELISA for msVTG.
To estimate the mesh selectivity of monofilament and multifilament gill net for Marbled sole, Pleuronectes yokohamae, the field testes were carried out 12 times with five different mesh sizes (86.6, 101.0, 121.2, 137.7, 151.5mm) in the western sea of Korea, 2007-2009. The master curve of selectivity was estimated by the extended Kitahara's method. In the field testes, the total number of species and catch were 26 and 987, respectively. The catch number of marbled sole was 728 and occupied 73.8% in total catch. The optimum values of l/m for 1.0 of retention probability in monofilament and multifilament gill net were estimated 0.288 and 0.307, respectively and l/m was estimated to be 0.189, 0.203, 0.213, 0.222 and 0.230 in case of monofilament gill net and 0.171, 0.191, 0.205, 0.216 and 0.227 in case of multifilament gill net when the retention probability were 0.1, 0.2, 0.3, 0.4 and 0.5, respectively. The 0.5 selection range of multifilament gill net was wider about 1.5 times than that of monofilament gill net according as multifilament gill net was 0.216 and monofilament gill net was 0.148. So the multifilament gill net has a low selectivity than that of the monofilament gill net. To estimate the optimum mesh size on first maturity length 19.5cm of marbled sole, the retention probability value of 0.1 was adopted in consideration of using the gill net for marbled sole at present. The optimum mesh size were estimated to be 103.2mm and 114.0mm in monofilament gill net and multifilament gill net, respectively, on first maturity length 19.5cm of marbled sole.
A series of experiments were conducted to compare the effects of various diluents in cold storage on the marbled sole, Limanda yokohamae sperm. Various diluents of glucose, L. yokohamae serum, marine fish Ringer's solution, sodium citrate and Ca2+ free artificial seawater (ASM) (tris-HCI, pH 7.4) containing 3 mM ethylene glycol-bis (2-aminoethyl ether) tetraacetic acid (EGTA) were used to store the sperm at 4℃. The storage effect was evaluated using motility index and survival rate of sperm. Glucose and sodium citrate were found to be better diluents which maintained high motility and survival rate of sperm for a storage period of 10 days. Some morphological changes of spermatozoa were observed during the cold storage with diluents. In particular, a detachment of the nuclear enveloped and of the plasma membrane from the nucleus in spermatozoa was observed. Morphological normality of the stored spermatozoa diluted with 0.3 M glucose was better than that of the stored spermatozoa undiluted or diluted with Ca2+ free ASW containing 3 mM EGTA.
A series of field tests to estimate the size selectivity of monofilament and multifilament gill net for marbled sole, Pleuronectes yokohamae, were carried out 12 times with five different mesh sizes (86.6, 101.0, 121.2, 137.7 and 151.5mm) in the western sea of Korea from 2007 to 2009. The master selection curve was estimated by the extended Kitahara's method. The total number of catch species was 23 and that of catch was 1,688 in the field tests. Marbled sole of total catch was 1,150 with 68.1 percent. In the monofilament trammel net, the optimum value of total length (TL) per mesh size (m) for 1.0 of retention probability was estimated 0.280 and the values of TL/m were estimated to be 0.187, 0.201, 0.210, 0.218 and 0.226 when the retention probability were 0.1, 0.2, 0.3, 0.4 and 0.5, respectively. In the multifilament trammel net, the optimum value of TL/m for 1.0 of retention probability was estimated 0.307 and the values of TL/m were estimated to be 0.195, 0.211, 0.222, 0.232 and 0.241 when the retention probability were 0.1, 0.2, 0.3, 0.4 and 0.5, respectively.
This study correlated changes in estradiol-l7$\beta$ ($E_2$), testosterone (T), 17$\alpha$,20-dihydroxy-4-pregnen-3-one (DHP), and vitellogenin (VTG) levels with changes in the gonadosomatic index (GSI) and ovarian histology during the annual reproductive cycle of the wild marbled sole, Limanda yokohamae. Synchronous oocyte development occurs in this fish. Ovary maturity was classified into four periods, based on histological observations: the spawning (December to February), post-spawning (February to April), recovery (May to August), and vitellogenic (September to November) periods. Seasonal changes in the GSI were inversely correlated with water temperatures and reflected the degree of ovarian maturity. Plasma VTG levels were correlated with changes in the GSI, which increased from September to a peak in January, and levels remained comparatively high until February. Estradiol-17$\beta$ was at baseline levels (<0.11 ng/mL) during the spring and summer, and peaked rapidly (1.55$\pm$0.445 ng/mL) from October to January. Plasma T and DHP levels had a similar profile; they rose markedly during the spawning period and remained low (or were not detectable) from spring through autumn. These data indicate that changes in plsama steroid hormones and VTG levels are correlated with the annual ovarian activity of the marbled sole. Based on these results and published reports, it appears that in this species DHP is the most important maturation-inducing steroid and that T is also related to final maturation.
The age and growth of the Marbled sole, Pleuronectes yokohamae were investigated using left sagittal otoliths of 832 individuals collected in the mid-southern waters of Korea from January 2006 to April 2007. The seasonal changes in the ratio of an translucent zone to interval between annular rings revealed that the boundaries between translucent and opaque parts of the otolith were annual growth rings formed between May and June once a year. The spawning period was November to March and therefore the duration from fertilization to the complete formation of the annual growth ring became fifteen months. The parameters of von Bertalanffy growth equations estimated using the average standard length at each age are the $L_{\infty}$ of 48.04cm, K of 0.1406/year and $t_0$ of -0.5116 for female and 26.14cm, 0.3056/year and -0.8202 for male, respectively. Finally, it is suggested that the growth of female is faster than that of male from age-2.
We determined the maturity and growth of the Marbled sole, Pseudopleuronectes yokohamae based on monthly sampling in the West Sea of Korea from February 2009 to December 2010. Determination of sex of P. yokohamae was by gonadal inspection, and age and growth were determined by analyzing the otolith. The biological minimum size of P. yokohamae female was 24.5 cm. The seasonal changes in the ratio of a translucent zone to an opaque zone revealed that the end of the translucent zone of the otolith were annuli formed in May once a year. We compared 8 type growth equations with several length-at-age data. By using the length-at-monthly age data of these, the calculated von Bertalanffy growth equations were $L_t=41.7(1-e^{-0.47(t+0.24)})$ for females and $L_t=32.7(1-e^{-0.75(t+0.01)})$ for males. We estimated that 24.5 cm was the total length of 2 year old P. yokohamae spawns in the cold water season from January to April, and grows to 8 years old for females and 6 years old for males in the West Sea of Korea.
