The Flower Morphological Characteristics of Salix caprea×Salix gracilistyla

  • The interspecific hybrid of Salix caprea and Salix gracilistyla has never been identified or studied in Korea. Accordingly, this study investigated the flower morphological characteristics of the interspecific hybrid between S. caprea and S. gracilistyla and compared the interspecific hybrid with S. caprea and S. gracilistyla, respectively. The female flowers were investigated for 12 characteristics and the male flowers were investigated for nine. For the female flowers, those of the hybrids were larger than those of S. caprea and S. gracilistyla in terms of catkin length (CL), bract length (BL), and bract width (BW). The hybrids are intermediates between S. caprea and S. gracilistyla in terms of ovary length, width, and stipitate length as well as gland length (GL). For the male flowers, those of the hybrids were bigger than those of S. caprea and S. gracilistyla in terms of CL, BL, and BW. The hybrids are intermediates between S. caprea and S. gracilistyla in terms of catkin width and stamen length (SL). A principal component analysis (PCA) of the female data showed that the first principal component (PC) explained 57.5% of the total variation. The first PC highly correlated the ovary stipitate and pistil style lengths. The analysis was divided into three groups of S. caprea, S. gracilistyla, and the hybrid by the first PC. The results of a PCA of the male data showed that the first PC explained 35.7% of the total variation. The first PC highly correlated with the adelphous SL and was divided into three groups of S. caprea, S. gracilistyla, and the hybrid. The results of the discriminant analysis showed that S. caprea, S. gracilistyla, and the hybrid were distinguishable by flower morphological characteristics. Therefore, the hybrid was distinctly separated from S. caprea and S. gracilistyla by flower characteristics.

동백 발효 추출물 단기 투여의 활성에 대한 연구 (Biological Activities of Extracts of Fermented Camellia japonica Leaf and Flower)

  • Objectives : This study was carried out to investigate the biological activities of methanol extracts of fermented Camellia japonica leaf and flower. Methods : Methanol extracts of fermented Camellia japonica leaf and flower were prepared and a dose of 100 and 400mg/kg/day was administered orally into mice. And after appropriate weeks, changes of serum enzyme activities were investigated to confirm its effects on serum glucose, cholesterol and short term administration safety. Results : Fermented flower extract showed significant decrease of serum level of cholesterol. And showed no toxicity on kidney and liver within the dose of 400mg/kg/day. Conclusion : Thus above result showed no toxicity on kidney and liver in male and female mice.

De novo genome assembly and single nucleotide variations for Soybean yellow common mosaic virus using soybean flower bud transcriptome data

  • The soybean (Glycine max L.), also known as the soya bean, is an economically important legume species. Pathogens are always major threats for soybean cultivation. Several pathogens negatively affect soybean production. The soybean is also known as a susceptible host to many viruses. Recently, we carried out systematic analyses to identify viruses infecting soybeans using soybean transcriptome data. Our screening results showed that only few soybean transcriptomes contained virus-associated sequences. In this study, we further carried out bioinformatics analyses using a soybean flower bud transcriptome for virus identification, genome assembly, and single nucleotide variations (SNVs). We assembled the genome of Soybean yellow common mosaic virus (SYCMV) isolate China and revealed two SNVs. Phylogenetic analyses using three viral proteins suggested that SYCMV isolate China is closely related to SYCMV isolates from South Korea. Furthermore, we found that replication and mutation of SYCMV is relatively low, which might be associated with flower bud tissue. The most interesting finding was that SYCMV was not detected in the cytoplasmic male sterility (CMS) line derived from the non-CMS line that was severely infected by SYCMV. In summary, in silico analyses identified SYCMV from the soybean flower bud transcriptome, and a nearly complete genome of SYCMV was successfully assembled. Our results suggest that the low level of virus replication and mutation for SYCMV might be associated with plant tissues. Moreover, we provide the first evidence that male sterility might be used to eliminate viruses in crop plants.

'캠벨얼리'와 '탐나라' 포도의 꽃과 소포자 발달 (Flower and Microspore Development in 'Campbell Early' (Vitis labruscana) and 'Tamnara' (V. spp.) Grapes)

  • 상업적으로 재배되고 있는 대부분의 포도 꽃은 완전화로서 개개의 꽃들이 모여 화서를 이룬다. 포도 꽃은 5개의 수술, 1개의 암술, 5개의 꽃잎이 융합된 구조인 화관, 그리고 융합된 형태의 꽃받침을 가지며 화방 내 각각의 꽃은 수정하여 과립으로 발달한다. 포도에서는 과립 형성이나 과립비대, 당도의 축적에 대한 연구는 많이 수행된 반면 배우체의 형태 형성, 꽃 기관의 구조, 그리고 꽃 발달과 같은 꽃의 형태적 연구는 거의 이루어지지 않고 있다. 본 연구에서는 '캠벨얼리'와 '탐나라'의 두 품종에서 꽃의 구조와 발달에 따른 특징들을 조사하였다. 포도 꽃의 발달 단계 결정은 애기장대에서 소포자의 형성부터 수 배우체의 발달 시기인 꽃발달 9단계부터 13단계로만 한정하였다. 포도 꽃 발달 9단계에서는 4분포자가 나타났고 꽃 발달 10에서 11단계 동안에는 단세포성 소포자에서부터 중기 2세포성 화분으로 발달하였다. 개화 직전의 꽃 발달 12단계 말에는 성숙한 3세포성화분으로 발달하였다. 꽃 발달 13단계에는 화관이 탈락하고 개화가 일어났다. 꽃 발달 9단계부터 12단계까지 꽃받침 길이는 거의 변하지 않았지만 화관의 길이는 계속 증가했다. 두 포도 품종에서 '꽃받침 길이에 대한 화관 길이의 비율'의 평균은 꽃 발달 9단계에서는 2.0, 꽃 발달 10단계에서는 3.0, 꽃 발달 11단계에서는 4.5, 꽃 발달 12단계에서는 6.5로 나타났으며, '꽃받침 길이에 대한 화관 길이의 비율'을 사용하여 꽃 발달 단계를 판별할 수 있었다. 본 연구 결과로 '캠벨얼리'와 '탐나라'에서 꽃 발달 단계를 정확하게 추정하는 형태적 지표가 개발되었다. 이를 이용하면 두 품종에서 소포자 배양 시기의 판정이나 '탐나라'에서 무핵과 유도를 위한 지베렐린 처리 시기의 판정 등에 유용하게 활용될 수 있을 것이다.

The Potential Regressive Role of Syzygium aromaticum on the Reproduction of Male Golden Hamsters

  • The flower buds of Syzygium aromaticum (clove) have been used as traditional medicine for the treatment of male sexual disorders in Asian countries. Recently, there are some reports about the effects of the clove on reproductive activities in mammals. Therefore, its effect on testicular function was examined in male golden hamsters whose reproductive activity is inhibited by photoperiod such as winter climate. The male animals were given by daily oral administrations (56 consecutive days) in three doses (4 mg, 20 mg, and 100 mg/kg BW) of the alcoholic extract of the clove. Generally lower dose (4 mg) of the extract continued to keep the reproductive activities of testes. The both middle and high doses (20 mg and 100 mg) of the extract completely inhibited the testicular activity in some animals. Taken together, these results suggest a possible biphasic action of alcoholic extract of Syzygium aromaticum flower bud on testicular function.

Pattern of Sexual Dimorphism in Garcinia kola (Heckel) Plantation

  • A study was designed to investigate the pattern of sexual dimorphism in a plantation of Garcinia kola. Twenty trees were randomly selected for the study and have been observed to flower regularly. A total of 100 inflorescence were randomly collected from the crown of each tree and 500 flowers randomly assessed within the period of four (4) flowering seasons. Floral sex assessment was done visually and with a hand magnifying lens; floral morphometric measurements (i.e. pedicel and perianth length and breadth), inflorescence length, and breadth) was taken using a veneer caliper; number of flowers per inflorescence and inflorescence per twig was counted; while, data analysis was conducted on excel using analysis of variance and pairwise t-test comparison. Four floral sexes were identified in the G. kola plantation studied which were unisexual male flowers, unisexual female flowers, cosexual unisexual male flowers, and cosexual hermaphrodite flowers. Three tree sexes were identified viz: inconstant male, invariant female, and cosexual trees. The plantation was significantly sexually dimorphic in floral sex and phenotypic traits (i.e. pedicel and perianth size), and as well as sexually dimorphic in tree sex and reproductive phenotypic traits (i.e. inflorescence size, number of inflorescences per twig, and number of flower bud per inflorescence). The sexual system of the plantation was therefore trioecious with features suggestive of evolving dioecy through the gynodioecious pathway.

  • Jatropha curcas is an oil bearing species with multiple uses and considerable economic potential as a biofuel plant. Plant flowering and breeding characteristics are important for us to understand the reproduction of plant populations. The present study describes the floral biology and flowering phenology of J. curcas which is a prerequisite for hybridization program for genetic improvement through conventional breeding. The plant produces flowers in dichasial inflorescences. Normally, the flowers are unisexual, and male and female flowers are produced in the same inflorescence. Only a few male flowers are produced in an inflorescence, and fruits are produced only through pollination between different flowers from the same or different plants. This study includes a description of the inflorescence, flower anatomy of both male and female flowers, female : male ratio, pollen : ovule ratio, flowering phenology, pollen viability, stigma receptivity, comparison of selfing methods and a comparison of geitonogamy and xenogamy. This information may be useful in J. curcas breeding programmes.

Environmental Risk Assessment of Watermelon Grafted onto Transgenic Rootstock Resistant to Cucumber Green Mottle Mosaic Virus (CGMMV) on Non-Target Insects in Conventional Agro-Ecosystem

  • We investigated the impact of watermelon grafted onto Cucumber Green Mottle Mosaic Virus (CGMMV)-resistant transgenic watermelon rootstock on insects as non-target organisms in a greenhouse in 2005. We quantitatively collected insect assemblages living on leaves and flowers, and we used sticky traps to collect alate insects. We compared the patterns of insect assemblages and community composition, cotton aphid (Aphis gossypii Glover) on watermelon leaves and western flower thrip (Frankliniella occidentalis Trybom) on watermelon male flowers, between CGMMV-resistant transgenic watermelon (TR) and non-transgenic watermelon (nTR). Non-parametric multidimensional scaling (NMS) ordination verified that insect assemblages on leaves and sticky traps were different between TR and nTR (P<0.05). The insect assemblages on male flowers were not statistically significant. Multi-response permutation procedures proofed our results from NMS results (P>0.05). Conclusively, TR watermelons appear to have some adverse effects on the population of cotton aphids on leaves and sticky traps, but watermelon male flowers do not show an adverse effect. Further research is required to assess the effect of TR on the aphid and western flower thrip. Life table experiments might support the specific reason for the adverse effects from leaf assemblages. Assessment of non-target impacts is an essential part of the risk assessment of non-target insects for the impact of transgenic organisms.

Confirmation of $F_1$ Hybridity Using RAPD Markers in Soybean

  • Molecular markers are useful to confirm the hybridity of F1 plant derived from cross of two homozygous parents with similar morphological traits. RAPD markers were used to test F1 hybrid plant obtained from cross of two homozygous soybean (Glycine max) parents. Fl plant for cross I was made from the mating of Hobbit87 (female) and L63-1889 (male) and Fl plant for cross II was obtained from the mating of H1053 (female) and L63-1889 (male). Selfing plant per each cross was also obtained. Among 20 Operon primers used, OPA04 and OPA09 show polymorphism between cross I and II parent. Band in size 1Kb of OPA04 and 2.1Kb of OPA09 primer was polymorphic band. This fragment identified Fl hybrid plant and selfing plant in cross I and II. Female parent Hobbit87 in cross I and H1053 in cross II has no this fragment (recessive allele). However, male parent L63-1889 and Fl hybrid plant in cross I and II has this size of polymorphic band (dominant allele). This indicated that Fl hybrid and selfing plants were detected by RAPD marker before phenotypic marker would be used to identify Fl hybridity. Amplification products of selfing plant for cross I and II were completely same to the those of female parent. When mature, flower color of Fl hybrid plant in cross I and II was purple and flower color of selfing plant in cross I and II was white. Purple flower is dominant trait. Fl hybridity was successfully detected at very early growth stage using RAPD marker. Therefore, RAPD marker can be used broadly to confirm Fl hybridity in many crops.

감 수꽃 착생 실생개체 선발 마커의 효율성 검정 (Efficiency of Sex-linked Molecular Markers for the Selection of Seedlings Bearing Male Flowers in Persimmon (Diospyros kaki Thunb.))

  • 감 꽃은 과실을 맺는 암꽃과 꽃가루를 내는 수꽃, 암술과 수술을 모두 가진 양성화가 있다. 관행 교배육종법으로 부본용 수꽃 신품종을 육성한다면 최종 선발까지 15년 정도의 장기간과 넓은 시험포장이 요구된다. 유묘기에 수꽃 착생 개체를 조기에 선발하여 육종효율을 증진하고자 꽃의 성표현을 알고 있는 주요 품종 88개체를 대상으로 Akagi 등(2014)이 보고한 수꽃 선발 마커들의 적용 효과를 검증하였다. OGI locus marker와 DISx-AF4S marker 적용 결과 각각 83품종, 72품종에서 꽃의 성표현과 분석 결과가 일치하였다. 육성품종인 '미감조생'을 모본으로 한 교배조합 등으로부터 나온 실생 2,509개체에 OGI locus marker 수꽃 선발 마커를 적용하여 890개체를 선발하였고, 분석 완료한 교배실생 중 아직 착과가 되지 않았거나 고사한 개체를 제외한 1,186개체를 대상으로 꽃의 성표현과 마커 분석 결과를 비교하였을 때 401개체가 불일치한 것을 확인하였다(33.8%). 다양한 교배조합으로부터 나온 실생 889개체에 DISx-AF4S marker를 적용하여 분석한 결과 636개체를 선발하였다. 분석 완료한 교배실생 중 아직 착과가 되지 않았거나 고사한 개체를 제외한 379개체를 대상으로 꽃의 성 표현과 마커 분석 결과를 비교하였고 247개체가 불일치한 것을 확인하였다(65.2%). 이상의 결과들을 종합하여 볼 때, 검토한 DISx-AF4S, OGI locus marker는 육종현장에서 활용하기에 유용하지 않을 것으로 판단된다.
