탄소원으로서 입국을 이용한 유산균 발효 (Lactic Acid Fermentation with Rice Koji as a Carbon Source)

  • Recently, several health benefits of rice wine, makgeolli, were known due to the interest on the traditional Korean liquor and the researches on the rice wine are increasing. Organic acids produced during the process of rice wine fermentation play important roles in the taste and flavor. In this study, we have examined the optimal conditions for lactic acid production in rice koji as a carbon source. Skim milk was also used as a supplementary ingredient for the optimization of lactic acid fermentation. Bacterial growth of Lactobacillus sakei was monitored under this condition. The pH, acidity of the culture and the ethanol tolerance of this bacterium were also tested. Through these experiments, we were able to optimize the growth condition of lactic acid bacteria by the addition of skim milk. This was also able to affect the change of pH, acidity, sugar concentration and alcohol tolerance, which might contribute to the improvement of the quality of rice wine. The optimal condition for the growth was 2 days with 10% (w/v) of skim milk concentration. With these results, it was confirmed that rice koji was an effective carbon source for the growth of lactic acid bacteria.

복숭아 막편의 품질 특성 및 항산화성 (Quality Characteristics and Antioxidant Activities of Peach Makphyun)

  • 본 연구는 시판되고 있는 복숭아 통조림을 선택하여 복숭아 페이스트를 만든 후 0, 5, 10 또는 20%를 첨가한 막편을 제조하여 품질 특성 및 항산화성을 분석하였다. 복숭아 첨가량이 증가할수록 pH는 낮아지고 산도는 증가하였다(P<0.05). 복숭아 첨가량이 증가할수록 막편의 환원당이 증가하였다(P<0.05). 색도는 복숭아 첨가량이 증가할수록 명도와 적색도는 감소하고, 황색도는 증가하였다(P<0.05). 총페놀 함량은 복숭아 첨가량이 증가할수록 증가하였다. DPPH 라디칼 소거능 및 hydroxyl 라디칼 소거능은 복숭아 첨가량이 증가할수록 $IC_{50}$ 값이 낮아져 항산화능이 높았다. 기호도는 복숭아를 첨가할수록 외관, 향미, 맛, 조직감, 전체적인 기호도가 증가하는 것으로 나타났다. 특히 전체적 기호도는 복숭아 20% 첨가군이 가장 높은 점수를 받았다(P<0.05). 이상의 결과로 보아 복숭아 첨가가 떡의 항산화능과 기호도를 증가시켜 저장성이 낮은 복숭아의 활용도를 높이고 설기떡의 가치를 높일 수 있을 것으로 사료된다. 복숭아 20%를 첨가하여 복숭아 막편을 제조할 경우 전반적인 기호도와 항산화능에서 가장 적합할 것으로 판단된다. 향후 다양한 복숭아 통조림과 막걸리 제품을 이용한 비교 분석으로 최대 가치를 창출하는 복숭아 막편에 대한 연구도 필요하리라 생각된다.

전통식품 유래 유산균의 해조류 발효 및 Probiotic 특성 (Seaweed Fermentation and Probiotic Properties of Lactic Acid Bacteria Isolated from Korean Traditional Foods)

  • 해조류의 발효가 가능하고 probiotic 특성이 우수한 유산균을 분리 선발한 후 이들의 미역과 다시마 발효능을 검토하였다. 미역 및 다시마 발효가 가능한 균주를 김치 젓갈, 된장으로부터 331 균주를 순수 분리하여 해조류 구성 다당(alginate, cellulose) 분해능, 균의 생육, 항균 활성 등을 비교 검토한 결과 4균주(stain No. 162, 164, 192, 196)가 우수하였다. 선발 균주 모두 인공위액, 인공담즙액, NaCl에 높은 생존율을 나타내었고 이들 4균주 중 No. 192가 가장 우수하였으며 Enterococcus faecium으로 동정되었다. 미역과 다시마를 이용하여 선발 유산균을 배양한 결과 No. 192 균주가 발효특성이 가장 양호하였으며, No. 162, 164, 196 균주도 양호하였다. 선발 유산균을 이용한 미역과 다시마에서 성장이 가능하였으며 발효 후 미역과 다시마 발효물의 항산화 활성이 증진되었다.

막걸리 유래 미생물의 활용을 위한 연구 동향 (Application of Functional Microbial Strains Isolated from Traditional Rice Wine in Korea)

  • 우리나라는 예전부터 전통적인 방법으로 누룩을 제조하고, 이를 발효제로 막걸리를 만들어 왔다. 막걸리는 여과 또는 살균 과정없이 다양한 미생물을 살아 있는 상태로 섭취하기 때문에 영양학적으로나 기능적인 측면에서 가치가 높다. 최근 많은 연구에서 막걸리로부터 미생물을 분리동정하고 다양한 기능성에 대해 스크리닝한 결과, 높은 프로바이오틱스 활성과 다양한 스펙트럼의 항균활성을 가진 균주들이 선별되었다. 특히 일부 유산균들은 GABA와 EPS 등의 기능성 물질을 생성하기도 했다. 또한, 일부 유산균과 효모의 경우 각각 bacteriocin 및 killer toxin을 통해 항균활성을 나타내는 것으로 여겨진다. 이러한 막걸리 유래 기능성 미생물과 그 대사산물은 기능성 식품 소재나 안전한 식품첨가물 및 다양한 산업분야에서 활용될 수 있을 것이다.

막걸리로부터 분리된 Bacillus amyloliquefaciens 균주의 항균 활성 (Antipathogenic Activity of Bacillus amyloliquefaciens Isolated from Korean Traditional Rice Wine)

  • 강원도를 비롯한 전국에서 발효식품 44점으로부터 유해균에 대한 생육억제 활성이 우수한 고초균주를 분리 및 동정하였으며, 생육억제 활성이 가장 우수하였던 Bacillus amyloliquefaciens MBE1283로 명명된 균주의 유해균에 대한 생육억제 활성, 전분분해 효소활성 및 생육특성을 조사하였다. Escherichia coli O157:H7 KCCM40406을 비롯한 8종류의 유해균에 대한 생육억제 활성을 평가한 결과, 막걸리에서 분리된 B. amyloliquefaciens MBE1283 균주는 다른 B. amyloliquefaciens 균주들에 비해 유의적으로 높은 유해균 생육억제 활성을 나타내었다. B. amyloliquefaciens MBE1283 균주의 ${\alpha}$-amylase 효소활성은 대조구 균주들과 비교하였을 때 현저히 낮았으나 glucoamylase 효소활성은 대조구로 사용한 균주와 유사한 수준이었다. B. amyloliquefaciens MBE1283 균주의 최적 생육온도는 $45^{\circ}C$로서 대조구 균주보다 온도에 대한 내성이 있는 것으로 판단되었으며, 균체 성장에 적합한 최적 배지는 pH 6이었다. 본 연구를 통하여 막걸리로부터 분리된 B. amyloliquefaciens MBE1283 균주는 여러 종류의 유해균에 항균활성을 지니며 높은 온도와 낮은 pH에 대하여 내성을 지닌 것을 알 수 있었다.

전통발효유 타락을 이용한 젤리의 품질 특성 (Quality characteristics of Jelly using the Tarak, Traditional Fermented Milk)

  • Tarak is our own traditional fermented milk made through lactic acid fermentation by using makgeolli as inoculum for milk or using already made tarak as inoculum but it is hardly known to people as its related studies are also insufficient. Accordingly, the purpose of this study was to manufacture jelly by using tarak and analyze its quality characteristics for the diversification of consumption promotion and demand of traditional fermented milk tarak. Tarak jelly that was made according to the percentage of adding tarak showed a tendency that the content of crue protein, crude fat and crude ash except for moisture water meaningfully increased as the amount of tarak added increased (P<0.05). In the case of pH, it declines as the amount of tarak added was high. The chromaticity measurement result of tarak jelly was 54.32 in the case of control group for L, and it increased as the amount of tarak added increased. In the case of a, it decreased as tarak was added, whereas b meaningfully increased. There was meaningful difference between samples in the case of chewiness, fracturability and adhesiveness (p<0.05). The sensory test result showed that there was meaningful difference between samples in their appearance and taste but there was no meaningful difference in the case of overall preference, scent and texture.

「임원십육지」의 조리법에 기초하여 재현한 건락의 대사체 분석과 관능평가 (Analysis of Metabolite and Sensory Evaluation on Kunrak, for Reproduced Manufacturing from Old Literature of 「Imwonsibyukji」)

  • Kunrak, a type of Korea traditional cheese, is made using Tarak, a yogurt produced with makgeolli as the fermentation source. Kunrak is produced by removing whey from Tarak, followed by drying process for safe storage and consumption over a longer period. In this study, we produced kunrak based on the method described in "Imwonsibyukji". Prepared Kunrak was ripening for 96 hours at 20, 30, and $40^{\circ}C$. In order to study characteristics of Kunrak, physiochemical properties (pH, acidity, water contents) and contents of metabolites (organic acids, sugars, amino acids, and fatty acids) were analyzed. During ripening, water contents decreased. The main organic acids in Kunrak were citric acid and lactic acid, and the main free sugar was lactose. Main amino acids were glutamate and phenylalanine, and main fatty acid was saturated capric acid. At later ripening, all metabolites increased immediately after preparation. The sensory evaluation score of overall preference was highest for Kunrak, which was ripening at $40^{\circ}C$ for 96 hours. This study was aimed to assay metabolites of Kunrak under various ripening conditions. The results provide basic data to produce conditions for standardized manufacturing of Kunrak.

20대 소비자의 탁주 이용실태 및 만족도 조사 (Consumption and Satisfaction Evaluation of Takju among Consumers Ages 20 to 29)

  • The purpose of this research was to investigate consumption and satisfaction of Takju (Makgeolli) among consumers ages 20 to 29 living in Seoul and Gyeonggi province. It was found that 86.2% of the respondents drank Takju once or twice a month, and the high satisfaction group drank Takju with a frequency of 8 times a month (P<0.01). Respondents selected market (44.4%) and restaurants (34.9%) as the most common place of purchase, whereas pub (42.2%) and Takju bar (32.8%) were the drinking places selected. Regarding the preferred ingredients, respondents liked pear the most followed by Lycium chinense, corn, pine nuts, ginseng, and citrus fruit. The most compatible foods with Takju were identified as Buchimgae (Jeon) (97.4%), Kimchi (45.6%), Bokkeum (36.4%), Namul & Muchim (25.0%), and Jjim (21.5%). Pajeon was mentioned as the most compatible food with Takju, followed by Bulgogi, Gamjatang, Agwijjim, Dubukimchibokkeum, and Ojingeotwigim. Regarding consumers' satisfaction of Takju, taste was the most highly valued, with color, price, alcohol content, function, flavor, diversity of raw materials, and accuracy of label also highly considered. Further, to formulate a plan to encourage consumption of Takju, the high satisfaction group decided that promotion of local festivals and events related to Takju, standardization of quality, creation of brands, expansion of distribution network, and organization of relevant departments are the most important compared to other items (P<0.05).

Analysis of the MAT1-1 and MAT1-2 Gene Ratio in Black Koji Molds Isolated from Meju

  • Aspergillus luchuensis is known as an industrially important fungal species used for making fermented foods such as awamori and shochu in Japan, makgeolli and Meju in Korea, and Pu-erh tea in China. Nonetheless, this species has not yet been widely studied regarding mating-type genes. In this study, we examined the MAT1-1 and MAT1-2 gene ratio in black koji molds (A. luchuensis, Aspergillus niger, and Aspergillus tubingensis) and in Aspergillus welwitschiae isolated from Meju, a fermented soybean starting material for traditional soy sauce and soybean paste in Korea. The number of strains with the MAT1-1 locus was 2 of 23 (A. luchuensis), 6 of 13 (A. tubingensis), 21 of 28 (A. niger), and 5 of 10 (A. welwitschiae). Fungal species A. tubingensis and A. welwitschiae showed a 1 : 1 ratio of MAT1-1 and MAT1-2 mating-type loci. In contrast, A. luchuensis revealed predominance of MAT1-2 (91.3%) and A. niger of MAT1-1 (75%). We isolated and identified 2 A. luchuensis MAT1-1 strains from Meju, although all strains for making shochu in Japan are of the MAT1-2 type. These strains may be a good resource for breeding of A. luchuensis to be used in the Asian fermented-food industry.

한국의 발효식품에 관하여 (Traditional Fermented Food Products in Korea)

  • Fermented foods available in Korea may be classified into four groups, namely, fermented soybean products, fermented cereal products, fermented vegetable products, and fermented fishery products based on raw materials used. The representative fermented foods based on soybean are Kanjang(soysauce), Doenjang(fermented soybean pastes), and Kochujang(red pepper added-fermented soybean paste). Such fermented products are made using Meju(functioning as a starter and prepared by fermentation of steamed soybean mash) as an essential ingredient, and used widely as a soup base and/or in seasoning side-dishes for everyday meals year around. Excepting Sikhae, all fermented products based on rice and other cereal grains are of alcoholic nature. Takju (Makgeolli) used to be made using rice as the major raw material, however, mainly due to the shortage of rice in recent years, other cereals, such as, barely, corn, and wheat flours are also used to replace rice today. Owing to such changes in the raw materials. the popularity of Takju has been somewhat reduced, yet it is still widely consumed in rural areas. Although Chungju is a popular rice wine with superior qualify over Takju, the amount consumed is considerably limited. The highest qualify rice wine, Bupju, in particular, is made by a low temperature fermentation using glutinous rice. Kimchi is an unique fermented vegetable product of long tradition in Korea. Although it was for consumption mainly in winter season serving as a source of vitamins, today it is widely used throughout the year. Except Kkakdugi and Dongchimi, all of the fermented vegetable products contain salted Korean cabbage as an essential item, while they abound in varieties depending on material composition and methods of processing, and also on seasons and localities Next to Kimchi in this category is Kkakdugi made of raddish in popularity and quantity consumed. The four groups of fermented food products described above are reviewed in some detail and evaluated in terms of their nutritional significances, processes and microorganisms involved. and their commercial potentials. Jeotkal (or Jeot) is a name given to all fermented products of fishery origin. A number of Jeot can be prepared by adding salt and allowing fermentation to the raw materials such as shrimp, anchovy octopus, clam, oyster, etc.
