The Use of AFLP Markers for Cultivar Identification in Hydrangea macrophylla

  • The principal morphological characters used for identification of hydrangea cultivars are often dependent on agroclimatic conditions. Furthermore, information on the selection or the genetic background of the hydrangea breeding is so rare that a molecular marker system for cultivar identification is needed. Amplified fragment length polymorphism (AFLP) markers were employed for fingerprinting Hydrangea macrophylla cultivars and candidate cultivars of H. macrophylla selected in Korea. One AFLP primer combination was sufficient to distinguish 17 H. macrophylla cultivars and 4 candidate cultivars. The profile of 19 loci that can minimize the error of amplification peak detection was constructed. AFLP markers were efficient for identification, estimation of genetic distances between cultivars, and cultivar discrimination. Based on the observed AFLP markers, genetic relationship was reconstructed by the UPGMA method. Seventeen H. macrophylla cultivars and H. macrophylla for. normalis formed a major cluster, and candidate cultivars selected in Korea formed another cluster.

Exotic Mahogany Leaf Litter Hinders Growth of Philippine Native Tree Seedlings

  • With continuous decline of Philippine forest cover, sustainable forest management and restoration are essential to restore destroyed forest ecosystems. Unfortunately, of ten most planted trees in reforestation projects in the Philippines, eight are exotic species, with large leaf mahogany (Swietenia macrophylla) being the most dominant. In this study, effect of Swietenia macrophylla in reforestation projects on native tree species was evaluated. Effects of S. macrophylla leaf litter, frequency, and canopy closure on the growth of the Philippine native species Pterocarpus indicus were investigated. Results showed that S. macrophylla leaf litter significantly inhibited the growth of P. indicus seedlings based on root collar-to-shoot height. The standardized growth rate of seedlings in plots without S. macrophylla leaf litter was significantly higher than the growth rate of seedlings in plots with leaf litter. Furthermore, there was no significant difference in the standardized growth rate of seedlings between plots without leaf litter and a control plot. On the contrary, S. macrophylla tree frequency and canopy closure showed no significant effect. These results attest to the negative effect of widely planted S. macrophylla to a valuable Philippine native tree P. indicus. With accumulating scientific evidence about negative effects of S. macrophylla on native trees, discontinued use in tree planting and reforestation efforts with active management of restoration sites previously planted with large leaf mahogany are needed.

Chemical Composition and Antimicrobial Efficiency of Swietenia macrophylla Seed Extract on Clinical Wound Pathogens

  • Microbial wound infection prolonged the hospitalization and increase the cost for wound management. Silver is commonly used as antimicrobial wound dressing. However, it causes several adverse side effects. Hence, this study was aimed to evaluate the antimicrobial efficiency of Swietenia macrophylla seed extract on clinical wound pathogens. Besides, the bioactive constituents of the seed extract were also determined. S. macrophylla seeds were extracted with methanol by maceration method. The seed extract inhibited 5 test bacteria and 1 yeast on disc diffusion assay. The antibacterial activity was broad spectrum, as the extract inhibited both Gram positive and Gram negative bacteria. On kill curve analysis, the antibacterial activity of the seed extract was concentration-dependent, the increase of extract concentration resulted in more reduction of bacterial growth. The extract also caused 99.9% growth reduction of Bacillus subtilis relative to control. A total of 21 compounds were detected in gas chromatography- mass spectrometry analysis. The predominant compounds present in the extract were oleic acid (18.56%) and linoleic acid (17.72%). In conclusion, the methanolic extract of S. macrophylla seeds exhibited significant antimicrobial activity on clinical wound pathogens. Further investigations should be conducted to purify other bioactive compounds from the seeds of S. macrophylla.

보리밥나무(Elaeagnus macrophylla)의 종자 산포와 발아율에 미치는 조류의 영향 (Effects of Bird Ingestion on Seed Dispersal and Germination of the Elaeagnus macrophylla)

  • 보리밥나무(Elaeagnus macrophylla)는 해안 인접지의 척박한 토양에서 생육하는 상록성 질소고정 식물로서, 열매의 결실기는 조류의 봄철 이동시기와 일치한다. 보리밥나무의 열매를 이용하는 조류를 파악하고 이들이 보리밥나무의 종자 산포와 발아에 미치는 영향을 밝히기 위하여 2007년 3월부터 4월까지 이동성 조류의 중간 기착지인 전남 선안군 홍도에서 조류 조사 및 종자 발아실험을 실시하였다. 그 결과 찌르레기(Sturnus cineraceus), 직박구리(Hypsipetes amaurotis), 개똥지빠귀(Turdus naumanni) 등 8종의 조류가 보리밥나무 열매를 섭식하였으며, 조류에 의해 소화된 종자는 자연 상태로 파종된 열매에 비해 발아시기가 단축되고 발아율도 높아진 것으로 나타났다. 또 열매를 주로 이용한 찌르레기류의 행동권 분석을 통해 보리밥나무 종자의 잠재적인 산포 범위는 6.9 ha에 이를 수 있는 것으로 예측되었다. 따라서 보리밥나무는 이동시기의 조류에게 중요한 먹이자원을 제공하고, 조류는 보리밥나무 종자의 산포자로서 새로운 지역으로 정착할 수 있는 기회를 주는 것으로 판단된다. 이와 같은 개척자 식물과 종자 산포자간의 상호작용은 척박한 해안 및 도서지역의 훼손된 상록활엽수림과 난대림 생태계의 자연 복원에 활용될 수 있을 것으로 기대된다.

Genetic differentiation and antioxidant activities of Bouea macrophylla Griffith in Nakhon Nayok province

  • Genetic differentiation and antioxidant activities in ethanolic extracts from leaves of Bouea macrophylla Griffith were determined. The result revealed genetic differentiation among sour ma-praang, ma-yong and sweet ma-praang of B. macrophylla Griffith (${\phi}_{PT}=0.772$, p-value=0.000). In addition, high genetic diversities were found in sour ma-praang and sweet ma-praang populations (P: 51.4 and 57.1 %; He: 0.1900.035 and 0.2240.036, respectively), but low genetic diversity was found in ma-yong population (P: 8.6 %; He: 0.0350.021). Total phenolic contents of sour ma-praang, ma-yong and sweet ma-praang were estimated as $680.51{\pm}89.81$, $701.03{\pm}59.89$ and $530.85{\pm}41.23mg$ gallic acid/g extract, respectively. Free radical scavenging activities of sour mapraang, ma-yong and sweet ma-praang were found (1/EC50=4.17, 1.43 and 1.37, respectively) corresponding with metalchelating activities (1/EC50=0.83, 0.65 and 0.17, respectively). Therefore, the obtained data may be applied to cultivation and utilization of their leaves as source of natural phenolics and antioxidants.

Analgesic and antiinflammatory activity of Alstonia macrophylla and Mallotus peltatus leaf extracts: Two popular ethnomedicines of Onge, a Negrito tribe of little Andaman

  • Two popular ethnomedicines of the Onge, a Negrito tribe of Andaman Islands, were evaluated for analgesic and antiinflammatory activity. The methanol extract as well as the different fractions of methanol extract of both Alstonia macrophylla and Mallotus peltatus leaves were studied using Swiss albino mice and Wistar albino rats. Acetic acid induced writhing, Tail flick and Tail immersion; Carrageenin- and Dextran-induced paw oedema tests were used. Dose-dependent analgesic and antiinflammatory activity were demonstrated for both methanol leaf extracts as well as fractions. Results were highly comparable with that of the standard drug pethidine.

Protective Effect of Methanol Extract of Swietenia macrophylla Seeds on Oxidative States Associated with Streptozotocin Induced Diabetic Rats

  • The methanol extract of seeds of Swietenia macrophylla King. (MESM) was studied for its antidiabetic activity in streptozotocin induced diabetic rats. It was principally aimed to correlate the efficacious role of MESM on reduction of oxidative state associated with diabetes. The extract was found to be potent antidiabetic evidenced by significant reduction of blood glucose level in diabetic rats (47.96% reduction of blood glucose level, at 300 mg/kg, on day 10). It was found that, MESM at 300 mg/kg, significantly decreased TBARS (35.03 and 22.22%) whilst increased GSH (86.75 and 31.45%), SOD (93.05 and 45.88%) and CAT (56.99 and 68.46%) levels in liver and kidney respectively in diabetic rats.

산수국 잎의 대식세포 활성화를 통한 면역증진활성 (Immune-Enhancing Activity of Hydrangea macrophylla subsp. serrata Leaves through Macrophage Activation)

  • In this study, we investigated the immune-enhancing activity of water extracts from Hydrangea macrophylla subsp. serrata (WE-HML). WE-HML increased cell viability and production of immunomodulators, which contributed to activating phagocytic activity in RAW264.7 cells. Inhibition of JNK and NF-κB reduced the production of immunomodulators by WE-HML. ROS inhibition suppressed the production of immunomodulators, and the activation of JNK and NF-κB signaling by WE-HML. TLR4 inhibition attenuated the production of immunomodulators, and activation of JNK and NF-κB signaling by WE-HML. In the immunosuppressed mouse model, WE-HML increased the spleen index, the levels of the cytokines, the numbers of white blood cells, lymphocytes, and neutrophils. However, WE-HML inhibited LPS-mediated overproduction of pro-inflammatory mediators in RAW264.7 cells, which indicated that WE-HML may have anti-inflammatory activity under excessive inflammatory conditions. Taken together, WE-HML may be considered to have immune-enhancing activity and expected to be used as a potential immune-enhancing agent.

잡종 기원 녹보리똥나무와 큰보리장나무의 형태학적 및 분자적 다양성 분석 및 평가 (Analysis and evaluation of morphological and molecular polymorphism in the hybridization of Elaeagnus ×maritima and E. ×submacrophylla)

  • 녹보리똥나무와 큰보리장나무는 형태적 특성에 기반하여 잡종 분류군으로 제안된 바 있으나, 이에 대한 분류학적 실체가 불명확하다. 본 연구에서는 녹보리똥나무와 큰보리장나무의 잡종 기원을 밝히기 위하여 현장 조사와 표본관에 소장된 표본을 검토하여 형태적 특징을 관찰하였으며, 핵리보솜 구간(internal transcribed spacer, 5S non-transcribed spacer)과 엽록체 구간(matK)의 염기서열을 비교·분석하였다. 형태적 특징을 관찰한 결과, 녹보리똥나무는 보리장나무와, 큰보리똥나무는 통영볼레나무와 형태적으로 유사성을 보였으나, 분자 분석 결과, 녹보리똥나무는 핵리보솜 구간에서 보리장나무와 보리밥나무의 서열의 혼성화가 관찰되었다. 큰보리장나무는 다양한 양상이 관찰되었는데, 일부 개체는 핵리보솜 구간에서 통영볼레나무와 보리밥나무의 서열의 혼성화가, 엽록체 구간에서는 보리밥나무 서열이 관찰되었다. 다른 개체는 핵리보솜 구간에서는 보리밥나무의 서열을 보였으나, 엽록체 구간에서는 통영볼레나무의 서열이 관찰되어 핵과 엽록체간 불일치를 보였다. 일부 개체는 통영볼레나무와 보리밥나무의 중간형을 보였으나, 핵리보솜과 엽록체 구간 모두 보리밥나무 서열이 관찰되었다. 이러한 결과는 두 종이 잡종기원이며, 부모종 또는 잡종 개체간 교배가 빈번히 일어나는 것을 시사한다.

Effect of Swietenia macrophylla seeds on normal and alloxan-induced diabetic rats

  • The methanol extract of the seed of Swietenia macrophylla (Meliaceae) seeds were investigated for its hypoglycaemic activity in normal and alloxan induced diabetic rats. Seed extract suspended in aqueous Tween 80 solution, the graded doses were administered to normal and experimental diabetic rats for 10 days. Fasting blood glucose levels, serum lipid profiles and changes in body weight were evaluated in normal rats while the hepatic marker enzymes and liver glycogen levels were evaluated additionally in diabetic rats. The diabetic groups treated with the seed extract were compared with standard drug glibenclamide. The findings showed the significant antidiabetic potential of the extract in ameliorating the diabetic conditions in diabetic rats. The extract also has hypoglycaemic action in normal rats. A toxicity study has been performed for the extract, which revealed that the extract is safe to use even at the doses of 3.2 mg/kg of body weight orally. The result from the present investigation indicates good correlation between the folk use and scientific investigation.
