  • A total of 23 springs distributed in the southern and northern sides of Mt. Hala in Jeju Island were seasonally sampled and analyzed for their major ion chemistry and oxygen and hydrogen isotope compositions to investigate their hydrogeochemical and isotopic characteristics. Dissolved ion concentrations of the south-side springs slightly increase with decreasing altitude. This indicates that dissolved ion concentrations of groundwater recharged at higher altitudes increase by water-rock interaction during the downgradient migration of groundwater through highly permeable volcanic aquifer. Dissolved ion concentrations of the north-side springs also slightly increase with decreasing altitude, but dramatically increase at ~300 m.a.s.l. This may indicate a sudden input of contaminants to the north-side groundwater system around ~300 m.a.s.l. Springs located in areas above ~300 m.a.s.l. have very low concentrations of dissolved ions, showing little seasonal variations. Whereas springs located in areas below ~300 m.a.s.l. show a big seasonal variation in the concentration of dissolved ions. Seasonal variation of oxygen isotope compositions of springs is ~3$\textperthousand$ for high-altitude springs (~1700 m.a.s.l.) and is ~2$\textperthousand$ near shore, indicating an attenuation of the variation through mixing with other groundwater bodies during migration.

  • This study was conducted to analyze the stand structure and growth characteristics along different elevations of Pinus koraiensis natural forests located at mean elevation 1,300 m (site1), 900 m (site2) and 600 m (site3) on Mt. Seorak. Mean DBH and height of P. koraiensis were 25.4 cm and 12.3 m in site1, while that of site2 and 3 were over 40 cm and 20 m, respectively. For this reason, there was 5 times difference of single tree volume between site1 and other sites(p<0.05). Mean ringwidth increment of P. koraiensis was 1.1 mm/yr in site1, 1.5 mm/yr in site2 and 2.6 mm/yr in site3. However, recent growth of ringwidth has increased by 8.6% in site1, whereas site2 and 3 have fallen by approximately 50% over the last 10 years. These results signify that the growth and distribution of natural P. koraiensis are effected by different climatic environment along elevations.

  • Soil erosion at Jawoon-Ri in Hongcheon highland is one of serious problems since saprolite piling on farmland has been typically practiced at 2-3 year's intervals. The objective of the case study was to survey management practices such as tillage, application of saprolite, and cultivating crops and to propose best management practices (BMP) to reduce soil loss in Jawoon-Ri, Hongcheon-Gun. Jawoon-Ri is located in the upper stream of Naerinchun. Upland areas of Jawoon 2 and 4Ri were 206.9 and 142.3 hectare, respectively. Estimation of soil loss in this study was based on USLE (Universal soil loss equation). Annual averaged soil losses were 15.6 MT per hectare in Jawoon-2Ri and 9.0 MT per hectare in Jawoon-4Ri, respectively. This case study tried to find methods to reduce soil erosion below tolerant soil loss level which is $11MT\;ha^{-1}\;yr^{-1}$. Estimated soil losses in more than 40% of uplands in Jawoon-2Ri and 4Ri were higher than tolerant soil loss level. Especially, edge of uplands undergone excessive soil erosion by concentrated runoff water. Therefore consolidation of upland edge was included as one of the proposed Best management practices BMP). The proposed BMP in this area were buffer strips, contour and mulching, diversion drain channel, grassed water-way, detour watet-way and cover crops and so on. Amounts for BMP requirements were 7,680 m for buffer strips, 123 ha (35%) for contour and mulching, 201 ha (57%) for diversion drain channel, 13,880 m for grassed water-way, 3,860 m for detour drainage, 8,365 m for sloping side consolidation and 3,492 ha for cover crops, respectively. Application of BMP are urgently needed in uplands which is direct conjunction with stream.

  • To extend our detailed knowledge for the Hwasan caldera, we carried out magnetotelluric (MT) survey, which is pretty sensitive to electrical property variation in both horizontal and vertical direction of subsurface, across the Hwasan caldera with the direction of EW. The 2-D inversion results of observed MT data lead to following conclusions. Firstly, the depth of the basin basement inferred by the MT inversion results matches well with that suggested by previous potential studies, but the basement resistivity seems fairly low when compared to that of general case. This feature might be related with the large-scaled, highly conductive layer beneath the Euisung Sub-basin suggested by the previous MT study. Secondly, the high resistivity zones reaching to 4000 $\Omega{\cdot}m$ are imaged around two external ring fault boundaries. These zones are thought of as the response of the rhyolitic dykes intruding along the ring fault, and in the previous gravity data correspond to relatively high density anomalies. Thirdly, low resistivity zone reaching to 200 $\Omega{\cdot}m$ is detected around a depth of 1km beneath the central part of the caldera, which has not been yet reported in korean geophysical literatures. If we take account of the evolution model of the Hwasan caldera, this zone is regarded as the past sedimentary layer that subsided during the period of forming external ring fault system. In addition, the relatively low density anomaly observed in the central part of the caldera may be attributed to this sedimentary layer.

  • The production and decomposition rate of litters from the three different locations, Quercus acutissium forest at 630 m, Q. mongolica forests at 1, 005m and 1, 490 m of Mt. Dokyoo, were estimated by Olson model. The contents of N, P, K, Ca and Na in soils were measured and the relationships among them were elucidated. The amounts of litter production in Q. mangolica were the lowest, 378.96g/$m^2$ at 1, 490 m and the highest, 876.12g/$m^2$ at 1, 005 m. And the amounts of litter production in Q. acutissima at 630 m was 686.16 g/$m^2$. The decay rate of litters in Q. mongolica was the smallest, 0.123 at 1, 490 m, and the largest, 0.222 at 1, 005 m. And that in Q. acutissima was 0.169 at 630 m which was the medium rate. The production and decay rate of litters decreased with the ascending altitude. The values at 630 m were maller than those at 1, 005 m. This might be due to the fact that the tree species at 630 m was Q. acutissima was 0.169 at 630 m which was the medium rate. The production and decay rate of litters decreased with the ascending altitude. The values at 630 m was Q. acutissima which was different from Q. mongolica at 1, 005 m and 1, 490 m. The half-0life of litter decay in Q. monglica was 5, 634 years at 1, 490 m and 3.134 years at 1, 005 m. And that in Q. acutissima was 4.132 years at 630 m. The decay rates of litters were tend to be inversely proportional to the ascending altitude. The annual standing stocks of mineral and their amounts returned to the soil were proportional to the decay rate of organic matters.

  • The present study was carried out to examine the efficiency of cloning of transgenic embryos by nuclear transfer(NT) using gene-injected rabbit embryos. The rabbit embryos at pronuclear stage were microinjected with methallothionein-human growth hormone(MT-hGH) gene and cultured to 8- and 16-cell in TCM-199 containing 10% FCS with a monolayer of rabbit oviductal epithelial cells in a 5% $CO_2$incubator. The recipient oocytes were collected from the oviducts 14~16 h after hCG injection. The oocytes were enucleated and activated with 5$\mu$M ionomycin and 2mM 6-dimethylaminopurine. Blastomeres form gene-injected embryos were transferred into the enucleated oocytes by micromanipulation. The nuclear transplant oocytes were electrofused and co-cultured with rabbit oviductal cells. Following 120 h of culture, blastocysts were prepared for gene analysis by polymerase chain reaction(PCR). In previous experiment, the rate of gene-positive embryos detected by the nested PCR analysis was significantly decreased while developing to blastocyst(25%)(Kang et al., 1998). The fusion rate of gene-injected blastomeres was significantly(P<0.05) lower than non-injected blastomeres(66% vs 80%). However, the NT embryos that were derived from gene-injected donor embryos did not differ from control embryos in development to the blastocyst stage(39% vs 31%). Of the 43 NT blastocysts developed from the gene-injected donor embryos, twelve(28%) were positive for the injected DNA. The results indicate that NT with gene-injected embryos can be successfully used for cloning and multiplication of transgenic embryos, furthermore applicable to improvement of transgenic animal production.

  • To obtain biological basic data for the conservation strategies establishment of a rare mistletoe species, Loranthus tanakae(Loranthaceae) in Korea, the distribution range, habitat characteristics and an assessment of the conservation status on the natural populations of L. tanakae were investigated. As a result, the natural populations of L. tanakae were distributed in Bakdudaegan from Mt. Chiri to Mt Seorak in Korea penninsula, and 97.8% of surveyed individuals of the mistletoe species were found in Gangwon province. In natural populations, 1,385 individuals of L. tanakae were parasitic on 480 host trees, and distributed in sunshiny ridges of mountains of altitude range of 353 m to 1,250 m. The range of host trees of L. tanakae were composed of 5 families, 6 genera, 9 species, 1 subspecies, and of these, Quercus mongolica was preferred with 81.5% (389 trees among 480 host trees). As a result of assessing the conservation status through IUCN, L. tanakae was evaluated as Vulnerable (VU). It was considered that Mt. Seorak, Mt. Taegi and Mt. Odae population as habitats with the highest density of distribution of natural populations of L. tanakae should be designated as the protected areas. Thus, conservation strategies and related methods of the natural populations of L. tanakae were also discussed for the sustainable conservation.

  • Investigating and evaluating regionally distributed plant resources provide fundamental information (inventory) to conserve plant diversity in target areas. Nevertheless, the examination of inventory to collect information in areas with little interest has been insufficient. In this study, the vicinity of Mt. Baegamsan(1,004m) was investigated for plant resources in the central and southern part of the Korean peninsula, which has received low interest. Nine surveys identified a total of 638 taxa consisting of 108 families, 339 genera, 572 species, 12 subspecies, 49 varieties, and 5 forms. One taxon was designated to be the endangered wild plant by the Ministry of Environment, and a total of 29 taxa were designated to be endemic species in Korea. Floristic target species identified by the surveys included 1 taxon for grade V, 13 taxa for grade IV, 36 taxa for grade III, 29s taxa for grade II, and 34 taxa for grade I. A total of 38 taxa were identified to be naturalized plants, and 3 of them were designated as invasive alien plants by the Ministry of Environment. There was a difference in plant distribution between western and eastern areas of Mt. Baegamsan. The endangered and valuable plants for conservation were mainly identified from Mt. Baegamsan peak area, the ridge, and the western area. Considering the plant distribution, the Mt. Baegamsan region, located on Nakdong-jungmaek, was estimated to be dispersal routes for plant expansion and retreat.
